Osteopontin and bone sialoprotein expression in regenerating rat periodontal ligament and alveolar bone.

BACKGROUND: Osteopontin (OPN) and bone sialoprotein (BSP) are differentially expressed over time in bone formation and remodelling. We have examined the expression of these proteins as phenotypic markers in studies of periodontal ligament (PL) and alveolar bone (AB) regeneration. METHODS: Window wounds (0.6 mm in diameter through alveolar bone) with either preservation or extirpation of PL were prepared under general anaesthesia in 30 Wistar male rats. Animals were killed on days 1, 3, 7, 14, and 28 after surgery, and OPN and BSP were detected with mouse monoclonal antirat antibodies. RESULT

Bone regeneration adjacent to titanium dental implants using guided tissue regeneration: a report of two cases.

The biologic principle of guided tissue regeneration was applied to regenerate alveolar bone in conjunction with the placement of titanium dental implants. In one case, complete osseointegration of an implant was achieved by the placement of a Teflon membrane over an implant that had been inserted into an alveolus immediately following tooth extraction. In a second case, the same biologic principle was used to increase the volume (height and width) of a resorbed, edentulous alveolar ridge to provide adequate bone dimensions for implant installation. In both cases, the membranes appear to have

Effects of Aging on New Bone Regeneration in a Mandibular Bone Defect in a Rat Model.

The effects of aging on the healing capacity of maxillofacial bone defects have not been studied. The aim of this study was to evaluate the effects of aging on the regeneration of round bony defects in the mandible. We created a round-shaped bony defect in the mandibular angle area in rats of different ages (2-[2 M], 10-[10 M], and 20-month-old [20 M]) and evaluated new bone regeneration in these groups. Changes in bone turnover markers such as alkaline phosphatase, procollagen type I N-terminal propeptide (PINP), cross-linked C-telopeptide of type I collagen, and tartrate-resistant acid phosp

Bone-targeted calcification-photothermal nanoplatform for synergistic tumor ablation and bone regeneration in breast cancer bone metastasis.

Breast cancer bone metastasis is a common and devastating complication among patients with advanced breast cancer, marked by aggressive tumor growth and extensive osteolytic destruction. Effective treatment requires simultaneous elimination of tumor cells and restoration of bone tissue. To address this challenge, we developed a bone-targeting nanomaterial, CPPA NPs, composed of a polydopamine-coated calcium peroxide core embedded with palladium nanoparticles, and functionalized with phytic acid for bone affinity. This design integrates calcification and photothermal activity to achieve synergi

Natural Bone Regeneration-Inspired Poly(Lipoic Acid)-Based Organic-Inorganic Bone Adhesive for Osteoporotic Bone Defect Repair.

Osteoporosis, featured by imbalanced bone remodeling, faces clinical challenges including microenvironment dysregulation, insufficient mineralization, and poor material fixation. Inspired by natural bone regeneration-where ossification centers and surrounding soft tissues orchestrate mineralization and tissue integration-we herein develop a biomimetic adhesive in which strontium titanate (SrTiO3) serves as the mineralization core to mimic ossification centers, boosting biomineralization and osteoblast differentiation. Meanwhile, poly(lipoic acid) (PolyLA), synthesized via one-step polymerizati

Targeting the osteoporotic bone microenvironment: Mechanistic insight and therapeutic biomaterials for accelerating bone regeneration.

Osteoporotic bone defects (OBD) present a major clinical challenge, largely due to a pathological bone microenvironment that severely compromises regenerative capacity. Moving beyond conventional classification schemes that often fail to capture this pathological complexity, our review establishes a conceptual framework that categorizes emerging biomaterials by mirroring the intrinsic functional components of the bone microenvironment itself-namely, ion-releasing, metabolite-based, extracellular matrix (ECM)-mimetic, and cytokine-loaded systems. The component-centric taxonomy not only systemat

Enhanced Bone Regeneration Using Demineralized Dentin Matrix: A Comparative Study in Alveolar Bone Repair.

OBJECTIVES: Alveolar bone resorption following tooth extraction presents significant challenges for implant-supported rehabilitations. Demineralised dentin matrix (DDM) has emerged as a promising scaffold for bone tissue regeneration. This study evaluates the bone-regenerating potential of varying degrees of dentin demineralisation. MATERIALS AND METHODS: Thirty-two male white New Zealand rabbits underwent extraction of the left mandibular anterior tooth and were assigned to 4 groups: undemineralised dentin matrix (UDDM), partially demineralised dentin matrix (PDDM), completely demineralised

Hyperlipidemia impairs bone regeneration of closed bone defects and tooth extraction wounds in mice.

OBJECTIVES: To evaluate the effect of hyperlipidemia on the healing of bone defects. MATERIALS AND METHODS: Apolipoprotein E (ApoE)-deficient mice and wild-type (WT) C57BL/6J mice were fed with an atherogenic high-fat diet (HFD) or a standard chow diet (as control) for 6 weeks. Blood samples were collected to evaluate serum lipid levels. Closed bone defects and open tooth extraction wounds were then created in the mandibles of these animals. One or two weeks after surgery, animals were euthanized. Micro-CT analysis and histomorphometric analysis were conducted to evaluate the healing of bone

Mesenchymal Stem Cells Coated with Synthetic Bone-Targeting Polymers Enhance Osteoporotic Bone Fracture Regeneration.

Osteoporosis is a progressive skeletal disease characterized by reduced bone density leading to bone fragility and an elevated risk of bone fractures. In osteoporotic conditions, decrease in bone density happens due to the augmented osteoclastic activity and the reduced number of osteoblast progenitor cells (mesenchymal stem cells, MSCs). We investigated a new method of cell therapy with membrane-engineered MSCs to restore the osteoblast progenitor pool and to inhibit osteoclastic activity in the fractured osteoporotic bones. The primary active sites of the polymer are the N-hydroxysuccinimide

Periostin-modified bone marrow mesenchymal stem cells from osteoporotic rats promote alveolar bone regeneration.

Bone regeneration is impaired in patients with osteoporosis. Previous studies have shown that periostin (Postn) shows great potential in bone regeneration treatments. However, the role of Postn in bone marrow mesenchymal stem cells (BMMSCs) remains to be elucidated. In this study, we isolated BMMSCs from ovariectomized rats (OVX-BMMSCs) and normal rats. Then, the expression levels of Postn and osteogenesis in OVX-BMMSCs were detected by alizarin red and alkaline phosphatase substrate staining, qPCR, and western blotting. We found that the levels of Postn in OVX-BMMSCs were significantly reduce

Two-step stem cell therapy improves bone regeneration compared to concentrated bone marrow therapy.

Adult stem cells are a promising tool to positively influence bone regeneration. Concentrated bone marrow therapy entails isolating osteoprogenitor cells during surgery with, however, only low cells yield. Two step stem cell therapy requires an additional harvesting procedure but generates high numbers of progenitor cells that facilitate osteogenic pre-differentiation. To further improve bone regeneration, stem cell therapy can be combined with growth factors from platelet rich plasma (PRP) or its lysate (PL) to potentially fostering vascularization. The aim of this study was to investigate th

Aspirin promotes bone marrow mesenchymal stem cell-based calvarial bone regeneration in mini swine.

INTRODUCTION: Stem cells have great therapeutic potential due to their capacity for self-renewal and their potential for differentiating into multiple cell lineages. It has been recently shown that the host immune system has fundamental effects on the fate of transplanted mesenchymal stem cells during bone repair, where the topical administration of aspirin is capable of improving calvarial bone repair in rodents by inhibiting tumor necrosis factor-α (TNF-α) and interferon-γ (IFN-γ) production. This study investigates whether aspirin is capable of accelerating the regenerative potential of bon

[Bone regeneration after filling of extensive cysts by means of bone collagen type 1].

It could be demonstrated by experimental studies on dogs that demineralized bone and bone products have a good osteogenetic potential which gives them a superior rating over artificial materials as replacement of autogenous bone transplants. The results of a prospective clinical study on 52 patients with 56 large mandibular or maxillary cysts are presented showing that bone collagen matrix type I is a good product for the filling of bone defects. Full reossification of a cyst was reaches as early as 8 months postoperatively in 20 cases. 32 defects had become substantially smaller. These result

Microfluidic GelMA/Bone-Derived Extracellular Matrix Microgels for Enhanced Stem Cell-Based Bone Regeneration.

Critical-sized bone defects caused by trauma or surgery exceed the innate regenerative capacity of bone, posing significant clinical challenges. Tissue engineering strategies based on injectable hydrogels and mesenchymal stem cells (MSCs) offer promise due to their adaptability to irregular defects. However, most current systems lack modular regenerative architecture and hierarchical biochemical cues needed to mimic native bone microenvironments, thereby limiting regenerative efficacy. In this study, we engineered modular, cell-laden microgels composed of bone-derived decellularized extracellu

Strontium enhances BMP-2 mediated bone regeneration in a femoral murine bone defect model.

The application of strontium is one option for the clinical treatment of osteoporosis-a disease characterized by reduced bone density and quality-in order to reduce the risk of vertebral and nonvertebral fractures. Unlike other drugs used in osteoporosis therapy, strontium shows a dual effect on bone metabolism by attenuating cellular resorption and simultaneously enhancing new bone tissue formation. Current concerns regarding the systemic application of highly dosed strontium ranelate led to the development of strontium-modified scaffolds based on mineralized collagen (MCM) capable to release

Guided bone regeneration of bone defects associated with implants: an evidence-based outcome assessment.

Guided bone regeneration is a new technique that evolved following the guided tissue regeneration procedure for regeneration of lost periodontium. The objective of guided bone regeneration is to promote bone formation in osseous deformities either before or in conjunction with endosseous implant placement. Osseous defects consist mainly of extraction sites, dehiscences or fenestrations, and localized ridge deformities. In addition, bone defects may either provide natural spacemaking or be nonspacemaking. A plethora of publications indicate that the guided bone regeneration technique can be use

Advances in 3D bioprinting of immunoregulatory biomaterials for bone regeneration.

Bone defects caused by trauma, infection, tumour resection, and degenerative diseases remain major clinical challenges because successful regeneration requires coordinated regulation of inflammation, vascularisation, osteogenesis, and bone remodeling. Advances in osteoimmunology have demonstrated that immune responses are integral to each stage of bone healing, shifting biomaterial design from passive biocompatibility towards active and temporally coordinated immunomodulation. Three dimensional bioprinting provides a versatile platform for integrating material, structural, cellular, and bioche

Advances in Marine Biomineralized Materials in Bone Repair and Regeneration.

Marine biomineralized materials (MBMs), derived from organisms such as corals, mollusks, cuttlefish, sponges, and fish, represent a promising class of natural resources for bone tissue engineering due to their structural similarity to human bone, excellent biocompatibility, and bioactive properties. The composition, hierarchical architecture, biomineralization mechanisms, and species-specific characteristics of MBMs collectively determine their physicochemical properties and regenerative potential. The porous framework of coral bone, the "brick-and-mortar" structure of nacre, the multi-chamber
