Assessment of CD3+ T cell density in the intestinal mucosa of dogs with inflammatory bowel disease.

BACKGROUND: T lymphocytes play a central role in the intestinal immune response in dogs with immunomodulator-responsive enteropathy (IRE), but the clinical relevance of mucosal T-cell infiltration remains unclear. HYPOTHESIS/OBJECTIVES: Compare mucosal CD3+ T-cell density between dogs with IRE and healthy comparison dogs across intestinal segments, and assess associations with clinical, biochemical, and histologic severity markers. ANIMALS: Forty-two client-owned dogs with IRE and 12 healthy research dogs. METHODS: Retrospective multicenter study. Archived endoscopic biopsy samples from the

The impact of CD3 affinity-attenuation on T cell engaging bispecific antibodies: is it really that simple?

INTRODUCTION: The first generation of approved T cell engagers (TCEs) showing promising efficacy in hematological and solid tumors relies on high binding affinity to CD3. Treatment of tumors with TCEs has been clinically associated with toxicities related to cytokine release syndrome (CRS). In addition to clinical strategies to mitigate CRS, antibody engineering efforts have been undertaken to generate TCEs with optimized therapeutic index. Strategies pursued in this context include affinity attenuation of CD3 binding arm, to achieve potent tumor cell killing with minimal cytokine secretion.

Bacterial infection triggers CD3ε+ T cell expansion and interaction with macrophages in grass carp (Ctenopharyngodon idella).

While T cells are well established as key players in mammalian adaptive immunity, their functional roles in teleosts remain poorly understood. Here, a monoclonal antibody against grass carp (Ctenopharyngodon idella) CD3ε was generated, and characterized by Western blotting, flow cytometry, and immunofluorescence. In healthy fish, CD3ε+ T cells accounted for 5.1 %, 4.0 %, and 2.6 % of splenic, head kidney, and peripheral blood lymphocytes, respectively. Upon Aeromonas hydrophila infection, CD3ε+ T cell populations elevated significantly in immune tissues such as spleen, head kidney, and posteri

CD3+/CD56+ NKT-like Cells Show Imbalanced Control Immediately after Exercise in Delayed-Onset Muscle Soreness.

The purpose of the study was to carry out an immunophenotypical characterization with a special focus on natural killer cells of junior swimmers from the Hungarian National Swim Team before and after an intensive acute exercise. Nineteen swimmers, ten females and nine males, completed the exercise protocol. Sixteen swimmers experienced delayed-onset muscle soreness. Most of our findings substantiated earlier results, such as the increase in the percentage of the CD3-/CD56+ natural killer cells and the CD3-/CD56dim+ NK cells, and the decrease in the percentage of CD3+ T cells among lymphocytes

Evaluating the distribution of T-lymphocytes and S-phase proliferating cells across the nasal mucosa of dromedary camel (Camelus dromedarius).

The lining mucosa of the nasal cavity performs important roles for the host adaptation to the external environment. Camels are unique in their adaptation to the lifestyle of nomadic deserts. The present study aimed to evaluate the distribution pattern of T lymphocytes and S-phase proliferating cells within the nasal mucosa of camel using antibodies against CD3 and PCNA, respectively. The mucosa of the rostral, middle, and caudal parts of the nasal cavity was collected and processed for immunohistochemical staining. CD3-immunoreactive (-IR) cells were observed within the epithelium and lamina p

Oral treatment with foralumab, a fully human anti-CD3 monoclonal antibody, prevents skin xenograft rejection in humanized mice.

Oral administration of biologics may be a feasible approach for immune therapy that improves drug safety and potentiates mechanisms of tolerance at mucosal barriers. We tested the ability of a fully human non-FcR binding anti-CD3 mAb, foralumab, to prevent skin xenograft rejection in mice with human immune systems. At an intragastric dose of 15μg, the drug could transit through the small bowel. Serum absorption and binding of lymphoid cells was seen and proliferative responses of splenic CD8+ T cells to mitogen were reduced. Five consecutive daily doses, then weekly dosing led to indefinite gr

Colonic immune cells in irritable bowel syndrome: A systematic review and meta-analysis.

BACKGROUND & AIMS: Increases in mucosal immune cells have frequently been observed in irritable bowel syndrome (IBS) patients. However, this finding is not completely consistent between studies, possibly due to a combination of methodological variability, population differences and small sample sizes. We performed a meta-analysis of case-control studies that compared immune cell counts in colonic biopsies of IBS patients and controls. METHODS: PubMed and Embase were searched in February 2017. Results were pooled using standardized mean difference (SMD) and were considered significant when zer

Severe bilaterally symmetrical alopecia in a horse.

A 9-year-old Tennessee Walking Horse gelding was presented for diagnosis of the cause of extensive alopecia. Complete hair loss was noted over the head, neck, shoulder, thigh, and proximal limbs, but the trunk, distal limbs, pelvic area, mane, and tail were unaffected. The alopecic areas were visually noninflammatory with no exudate or crust except on the shoulder and along the back, where multifocal patchy areas of alopecia with scales and crust were evident. The horse was slightly pruritic. Microscopically, the hair bulbs, inner and outer root sheaths of inferior segments, and perifollicular

Three-color flow cytometric study on lymphocytes derived from thymic diseases.

BACKGROUND: Anterior mediastinal masses derive from a variety of diseases. Thymomas have been shown to commonly hold CD4(+)CD8(+) double-positive (DP) lymphocytes, and identification of this subset by two-color flow cytometric study was suggested to help diagnosis of thymoma. Several other thymic diseases, however, possibly hold CD4(+)CD8(+) DP lymphocytes. In this study, we utilized the three-color flow cytometric method for further examination of the phenotypes of lymphocytes in the thymic diseases. MATERIALS AND METHODS: One hundred eight specimens (77 primary and 10 metastatic thymomas, 1

Increased frequency of RAG-expressing, CD4(+)CD3(low) peripheral T lymphocytes in patients with defective responses to DNA damage.

Accumulating evidence indicates that peripheral lymphocyte variants with altered antigen receptor expression may be capable of expressing recombination-activating genes (RAG). We and others recently observed functional RAG gene products in mature T cells with defective TCR expression (MacMahan and Fink, Immunity 1998. 9: 637 - 647; Lantelme et al., J. Immunol., 2000. 164: 3455 - 3459). Here, the association between TCR expression and RAG activity was assessed further in lymphocytes from patients with defective responses to DNA damage. We show that T cells with altered TCR surface expression ar

Scleroderma profunda. Clinicopathological studies.

Scleroderma profunda represents an unusual clinical entity. There are no unified criteria for its diagnosis. Seven patients with scleroderma profunda were studied. The clinical features can be summarised in three groups: 1. hyperpigmented plaques with subcutaneous induration; 2. erythematous plaques with induration of the skin and subcutis; 3. pale indurated plaques with atrophy and sometimes ulcerobullous lesions. Histology showed sclerodermiform changes in the deep dermis and subcutis and vasculitis in one case. Dermal lymphocytic infiltration was well pronounced in 5 cases and consisted of

Spontaneous loss and alteration of antigen receptor expression in mature CD4+ T cells.

The TCR/CD3 complex plays a central role in antigen recognition and activation of mature T cells, and, therefore, abnormalities in the expression of the complex should induce unresponsiveness of T cells to antigen stimulus. Using flow cytometry, we detected and enumerated variant cells with loss or alteration of the surface TCR/CD3 expression among human mature CD4+ T cells. The presence of variant CD4+ T cells was demonstrated by isolating and cloning them from peripheral blood, and their abnormalities can be accounted for by alterations in TCR expression such as defects of protein expression…

Expression and function of the natural cytotoxicity receptor NKp46 on circulating malignant CD4+ T lymphocytes of Sézary syndrome patients.

The natural cytotoxicity receptors NKp30, NKp44, and NKp46 were identified as activating receptors mainly expressed by natural killer (NK) l … correlated with a decreased phosphorylation of the CD3ζ chain associated with NKp46 and/or the TCR/CD3 complexes. Our results reveal that in addition to KIR3DL2/CD158k expression, NKp46 could represent an additional marker on the circulating malignant T lymphocytes of Sézary patients, where it displays an as yet unreported function of inhibitory co-receptor …

Pilot Data Suggest That Obesity and Presence of Malignancy Are Associated with Altered Immune Cell Infiltration in Endometrial Biopsies.

(1) Background: The worldwide endometrial cancer (EC) incidence is rising, amongst others linked to obesity, type 2 diabetes mellitus (T2DM), and metabolic syndrome, possibly due to low-grade adipose tissue inflammation. We studied immune cell infiltration in the endometrium in relation to diagnosis and obesity. (2) Methods: A cohort was created (n = 44) from postmenopausal women, lean (n = 15) and obese (n = 29), with bleeding complaints due to EC (n = 18) or benign pathology (n = 26). Endometrial biopsies were used to study the immune microenvironment and stained for macrophages (CD68 and CD

Enrichment of novel CD3+F4/80+ cells in brown adipose tissue following adrenergic stimulation.

Macrophages play a multifaceted role in maintaining tissue homeostasis, fighting infections, and regulating cold-induced thermogenesis. The brown adipose tissue (BAT) is crucial for maintaining body temperature during cold exposure. Cold stress triggers the sympathetic nervous system to release norepinephrine (NE), which activates BAT via β3-adrenergic receptors, initiating lipolysis and glycolysis. BAT-infiltrating macrophages can either hinder or enhance thermogenesis by controlling the interplay between BAT cells and sympathetic nerves. In this study we report on a unique population of CD3+

Programmed cell death of T lymphocytes during acute viral infection: a mechanism for virus-induced immune deficiency.

Acute viral infections induce immune deficiencies, as shown by unresponsiveness to mitogens and unrelated antigens. T lymphocytes isolated from mice acutely infected with lymphocytic choriomeningitis virus (LCMV) were found in this study to undergo activation-induced apoptosis upon signalling through the T-cell receptor (TcR)-CD3 complex. Kinetic studies demonstrated that this sensitivity to apoptosis directly correlated with the induction of immune deficiency, as measured by impaired proliferation in response to anti-CD3 antibody or to concanavalin A. Cell cycling in interleukin-2 (IL-2) alon…

G-proteins are not directly involved in the CD3-antigen-mediated production of inositol phosphates in HPB-ALL T-leukaemia cells expressing phospholipase C isoforms gamma 1 and beta 3.

The possible involvement of G-proteins in T cell antigen-receptor complex (TCR)-mediated inositol phosphate production was investigated in HPB-ALL T-cells, which were found to express the phospholipase C gamma 1 and beta 3 isoforms. Cross-linking the CD3 antigen on streptolysin-O-permeabilized cells stimulated a dose-dependent increase in inositol phosphate production, as did addition of guanosine 5'-[gamma-thio]triphosphate (GTP[S]) or vanadate, a phosphotyrosine phosphatase inhibitor. It was possible, therefore, that the CD3-antigen-mediated production of inositol phosphates was either via a
