A rare case of breast carcinoma co-existing with axillary mantle cell lymphoma.

ABSTRACT : BACKGROUND : Mantle cell lymphoma (MCL) is a rare variety of non-Hodgkin's lymphoma which originates from CD5+ B-cell population in the mantle zones of lymphoid follicles. Coexistence of such tumours in the axillary lymph nodes with invasive breast cancers without prior history of adjuvant chemotherapy or radiotherapy has not been previously reported in literature. CASE REPORT : We report a rare case of breast cancer co-existing with stage I mantle cell lymphoma of the ipsilateral axillary lymph node detected fortuitously by population screening. CONCLUSION : Though some studies hav

[Co-expression of TrkA and p75 neurotrophin receptor in extracranial olfactory neuroblastoma cells].

OBJECTIVE: To determine whether TrkA and p75 are expressed in olfactory neuroblastoma (ON) and whether the evaluation of their expression by immunohistochemistry is useful to predict the outcome of ON. METHODS: Ten cases of ON and 5 nasopharyngeal carcinoma and 5 normal olfactory neuroepithelial of nasal cavity were examined for the expression of TrkA and p75 neurotrophin receptors using immunohistochemistry and double fluorescence labeling. RESULTS: The 10 cases of ON showed positive immunohistochemical staining for both TrkA and p75. Double staining revealed that almost all TrkA immunoreac

Co-localization of matrix metalloproteinase-1 and mast cell tryptase in the human uterus.

The endometrium displays characteristic cyclical changes involving proliferation and differentiation. The differentiation that takes place requires major tissue remodelling involving the matrix metalloproteinase (MMP) family as key enzymes in this process. Mast cells, containing the tryptase and chymase enzymes that are capable of stimulating the MMP cascade, have been identified in the endometrium, but their role is still unclear. In this study, we observed that the majority of mast cells in the uterus reside in the myometrium and that they co-express mast cell tryptase and MMP-1 in the same

Enhancement of cALL immunogenicity by co-culture with a CD154 expressing 293 cell line.

Pre-B cell acute lymphoblastic leukaemia (cALL) commonly occurs in young patients and although successful conventional therapies are available (such as cytotoxic drugs and bone marrow transplantation) for a proportion of patients (approximately 30%) these are ultimately unsuccessful. Recurrence of disease is a result of the failure of the immune system to recognize these abnormal cells and down-regulation of crucial molecules required for cognate CD4(+) T cell recognition has been postulated as a means of immune escape. In this study we show that an embryonic kidney cell line (293 cells) trans

Amylin-immunoreactivity is co-stored in a serotonin cell subpopulation of the vertebrate stomach and duodenum.

Amylin (or islet amyloid polypeptide) is a 37 amino acid peptide originally isolated from amyloid deposits in the pancreas of non-insulin dependent diabetic patients. It has already been immunohistochemically localised within the B and D cells of pancreatic islets and in endocrine cells of the rat and human stomach and duodenum. In this phylogenetic study, a polyclonal antiserum raised against the carboxy-terminal tridecapeptide amide of human amylin was used to demonstrate and examine the distribution of amylin-immunoreactivity in the stomach and duodenum of various vertebrate species. Possible involvement of T cell co-stimulation in pustulosis palmaris et plantaris via the induction of inducible co-stimulator in chronic focal infections.

BACKGROUND: Inducible co-stimulator (ICOS) is a co-stimulatory receptor on activated T cells that provides the signals needed for Th1 and Th2 responses via its interaction with B7h. Chronic focal infections are closely related to pustulosis palmaris et plantaris (PPP), but the involvement of ICOS in PPP has not been clarified. OBJECTIVE: To investigate the effectiveness of treatments for focal infections on PPP skin lesions and the involvement of ICOS-positive T cells at focal infection sites in the tonsils and in PPP lesional skin. METHODS: In patients that had undergone a tonsillectomy or

Gli pathway-targeted Co(iii) Schiff base complexes inhibit migration of basal cell carcinoma cells.

Basal Cell Carcinoma (BCC) is the most frequently diagnosed cancer globally and affects about one in five Americans. Given the frequency of diagnosis, it is surprising that there are very few therapeutic options. Surgical removal is currently the most common treatment option; however, this can lead to noticeable scarring and cosmetic issues. As a result, there is a compelling interest in developing non-invasive therapeutic approaches to this disease. Here, we introduce a new transition metal-DNA derivative called CoGli-GOPEI that inhibits the migration of murine ASZ BCC cells in laboratory exp

Co-culture of multiple myeloma cell lines and bone marrow mesenchymal stem cells in a 3D microgel environment.

This study reproduces the complex relationships between tumour plasma cells and their bone marrow microenvironment in multiple myeloma in vitro. These relationships are established both with other cells and with the extracellular matrix and are key factors in tumour progression, generating resistance to antitumour drugs in the cellular and non-cellular environments. This paper proposes a 3D microenvironment model designed to capture the main components of the multiple myeloma tumour microenvironment. Multiple myeloma cells (MMCs) were dispersed in a microgel medium formed by gel-textured micro

Extracellular vesicles and co-isolated endogenous retroviruses from murine cancer cells differentially affect dendritic cells.

Cells secrete extracellular vesicles (EVs) and non-vesicular extracellular (nano)particles (NVEPs or ENPs) that may play a role in intercellular communication. Tumor-derived EVs have been proposed to induce immune priming of antigen presenting cells or to be immuno-suppressive agents. We suspect that such disparate functions are due to variable compositions in EV subtypes and ENPs. We aimed to characterize the array of secreted EVs and ENPs of murine tumor cell lines. Unexpectedly, we identified virus-like particles (VLPs) from endogenous murine leukemia virus in preparations of EVs produced b

Co-culture of Liver Parenchymal Cells with Non-parenchymal Cells Under 2D and 3D Culture Systems; A Review.

Hepatocytes are the major parenchymal cells (PC) in the liver and present an important role in liver metabolism. Hepatocytes are considered a gold standard tool for drug toxicity/screening or liver disease modeling. However, the maturation and functions of hepatocytes are lost under routine 2- dimensional (2D) culture conditions. Recent studies revealed that the interactions between hepatocytes and non-parenchyma cells (NPC) under 3D culture conditions can be an alternative option for optimizing hepatocyte maturation. Co-culture of hepatocytes with NPC simplifies the in-vitro liver disease mod

The sodium/myo-inositol co-transporter SLC5A3 promotes non-small cell lung cancer cell growth.

Identification of novel molecular signaling targets for non-small cell lung cancer (NSCLC) is important. The present study examined expression, functions and possible underlying mechanisms of the sodium/myo-inositol co-transporter SLC5A3 in NSCLC. The Cancer Genome Atlas (TCGA) database and local NSCLC tissue results demonstrated that SLC5A3 expression in NSCLC tissues (including patient-derived primary NSCLC cells) was significantly higher than that in normal lung tissues and lung epithelial cells. In primary NSCLC cells and immortalized lines, SLC5A3 depletion, using small hairpin RNA (shRNA

Evaluation of co-cultured spermatogonial stem cells encapsulated in alginate hydrogel with Sertoli cells and their transplantation into azoospermic mice.

An in vitro spermatogonial stem cell (SSC) culture can serve as an effective technique to study spermatogenesis and treatment for male infertility. In this research, we compared the effect of a three-dimensional alginate hydrogel with Sertoli cells in a 3D culture and co-cultured Sertoli cells. After harvest of SSCs from neonatal mice testes, the SSCs were divided into two groups: SSCs on a 3D alginate hydrogel with Sertoli cells and a co-culture of SSCs with Sertoli cells for 1 month. The samples were evaluated by quantitative reverse transcription polymerase chain reaction (qRT-PCR) assays a

Co-Delivery of Docetaxel and Salinomycin to Target Both Breast Cancer Cells and Stem Cells by PLGA/TPGS Nanoparticles.

PURPOSE: Conventional chemotherapy is hampered by the presence of breast cancer stem cells (BCSCs). It is crucial to eradicating both the bulky breast cancer cells and BCSCs, using a combination of conventional chemotherapy and anti-CSCs drugs. However, the synergistic ratio of drug combinations cannot be easily maintained in vivo. In our previous studies, we demonstrated that the simultaneous delivery of two drugs via nanoliposomes could maintain the synergistic drug ratio for 12 h in vivo. However, nanoliposomes have the disadvantage of quick drug release, which makes it difficult to maintai

Intraurethral co-transplantation of bone marrow mesenchymal stem cells and muscle-derived cells improves the urethral closure.

BACKGROUND: Cell therapy constitutes an attractive alternative to treat stress urinary incontinence. Although promising results have been demonstrated in this field, the procedure requires further optimization. The most commonly proposed cell types for intraurethral injections are muscle derived cells (MDCs) and mesenchymal stem/stromal cell (MSCs). The aim of this study was to evaluate the effects of MDC-MSC co-transplantation into the urethra. METHODS: Autologous transplantation of labeled MDCs, bone marrow MSCs or co-transplantation of MDC-MSC were performed in aged multiparous female goat

Transdifferentiation of mouse adipose-derived stromal cells into acinar cells of the submandibular gland using a co-culture system.

A loss of salivary gland function often occurs after radiation therapy in head and neck tumors, though secretion of saliva by the salivary glands is essential for the health and maintenance of the oral environment. Transplantation of salivary acinar cells (ACs), in part, may overcome the side effects of therapy. Here we directly differentiated mouse adipose-derived stromal cells (ADSCs) into ACs using a co-culture system. Multipotent ADSCs can be easily collected from stromal vascular fractions of adipose tissues. The isolated ADSCs showed positive expression of markers such as integrin beta-1

Differentiation induction of mouse cardiac stem cells into sinus node-like cells by co-culturing with sinus node.

Sinus nodal cells can generate a diastolic or "pacemaker" depolarization at the end of an action potential driving the membrane potential slowly up to the threshold for firing the next action potential. It has been proved that adult cardiac stem cells (CSCs) can differentiate into sinus nodal cells by demethylating agent. However, there is no report about adult CSCs-derived sinus nodal cells with pacemaker current (the funny current, I f). In this study, we isolated the mouse adult CSCs from mouse hearts by the method of tissue explants adherence. The expression of c-kit protein indicated the

Importance of B cell co-stimulation in CD4(+) T cell differentiation: X-linked agammaglobulinaemia, a human model.

We were interested in the question of whether the congenital lack of B cells actually had any influence on the development of the T cell compartment in patients with agammaglobulinaemia. Sixteen patients with X-linked agammaglobulinaemia (XLA) due to mutations in Btk, nine patients affected by common variable immune deficiency (CVID) with <2% of peripheral B cells and 20 healthy volunteers were enrolled. The T cell phenotype was determined with FACSCalibur and CellQuest Pro software. Mann-Whitney two-tailed analysis was used for statistical analysis. The CD4 T cell memory compartment was reduc

Induction of drug-metabolizing enzymes by phenobarbital in layered co-culture of a human liver cell line and endothelial cells.

Primary human hepatocytes are extensively used to study the potential of drugs to induce cytochrome P450 (CYP). However, the activities of these enzymes decrease rapidly during culture. Previously we reported that in a layered co-culture system with HepG2 and bovine endothelial cells, the expression levels of various CYP genes were significantly increased compared with the monolayer cultured HepG2 cells. Here, we examined the induction of CYP gene expression by an inducer by examining the effect of phenobarbital treatment on CYP gene expression in the co-culture system. In the layered co-cultu

Co-expression of vimentin and 19S-thyroglobulin in follicular cells located in the C-cell complex of dog thyroid gland.

The localization of vimentin, an intermediate filament protein, in the dog thyroid cells was examined by immunohistochemistry with a monoclonal antibody against vimentin and polyclonal anti-19S-thyroglobulin and anti-calcitonin antibodies. In the dog thyroid gland, C-cells are usually distributed as large cell groups in an interfollicular location. In addition, C-cell complexes that are special C-cell clusters associated with other epithelial elements and cysts are frequently located around the parathyroid gland IV. The C-cell complexes are remnants of embryonic ultimobranchial bodies and reta
