Nephroprotection of plantamajoside in rats treated with cadmium.

Cadmium (Cd), an environmental and industrial pollutant, generates free radicals responsible for oxidative stress. Cd can also lead to various renal toxic damage such as the proximal tubules and glomerulus dysfunction. Plantamajoside (PMS), a major compound of Plantago asiatica (PA), was reported to have the antioxidant effects. In this study, we investigated the protective effects of PMS on Cd-induced renal damage in the NRK-52E cell and rat kidney tissue. Cd exposure increased the ROS generation, lipid peroxidation, serum biochemical values of renal damage, and mRNA and protein expressions o

Immunochromatographic assay of cadmium levels in oysters.

Oysters are one of foodstuffs containing a relatively high amount of cadmium. Here we report on establishment of an immunochromatographic assay (ICA) method of cadmium levels in oysters. Cadmium was extracted with 0.l mol L(-1) HCl from oysters and cleaned up from other metals by the use of an anion-exchange column. The behavior of five metals Mn, Fe, Cu, Zn, and Cd was monitored at each step of extraction and clean-up procedure for the ICA method in an inductively coupled plasma-mass spectrometry (ICP-MS) analysis. The results revealed that a simple extraction method with the HCl solution was

[Biochemical mechanisms of neurotoxicity caused by cadmium].

Cadmium (Cd), which belongs to the heavy metals, is one of the major polluting component of human and animal environment. Exposure to cadmium can lead to absorption of the compounds to the organism and consequently, the toxic effects in the nervous system. The paper presents various views on the biochemical mechanisms of neurotoxicity caused by cadmium. This paper describes the disturbances in the cellular antioxidant system, generation of reactive oxygen and nitrogen species, changes in energy production in the metabolic pathways, changes in the metabolism of biogenic amines, neurotransmitter

Significance of phenols in cadmium and nickel uptake.

The effects of 2-aminoindane-2-phosphonic acid (AIP), a potent phenylalanine ammonia-lyase (PAL) inhibitor, on the accumulation of cadmium and nickel in chamomile (Matricaria chamomilla) were examined in this study. In vitro assay of AIP effect showed a 90% reduction in PAL activity. In plants cultured for 7 days in Cd or Ni solutions with AIP, PAL activity was higher in both shoots and roots (in comparison with metals without AIP), and was correlated with changes in free phenylalanine content. Individual amino acids were both positively and negatively affected by AIP, with the accumulation of

Assessment of biomarkers of cadmium stress in lettuce.

Laboratory and field studies have provided encouraging insights into the capacity of plants to act as biomonitors of environmental quality through the use of biomarkers. However, a better understanding of the overall process of Cd-induced senescence, describing the cascade of Cd effects in plants is needed for a selection of relevant biomarkers of Cd stress. In order to approach this, 5-week old Lactuca sativa L. were exposed for 14 days to 100muM Cd(NO(3))(2) and harvested at days 0, 1, 3, 7 and 14. The parameters measured included classical endpoints (shoot and root growth) and biochemical e

Importance of cadmium speciation in nitrification inhibition.

In this study, the influence of Cd speciation on nitrification inhibition was investigated in batch suspended growth activated sludge systems which contain biomass enriched in terms of nitrifiers. For this purpose, parallel measurements of specific oxygen uptake rates (SOUR), ammonium utilization rates [Formula: see text] and Cd uptake were carried out. Cd speciation was adjusted with a strong complexing agent, ethylenediaminetetraacetic acid (EDTA). Free and biosorbed Cd concentrations were theoretically determined by using the MINEQL+ program and the Cd adsorption constant, whereas labile Cd

Cadmium and children: exposure and health effects.

Cadmium exposure and accumulation in the body start at young age. Exposure routes in children are mainly via food, environmental tobacco smoke and house dust. Excretion from the body is limited. Cadmium accumulation in the kidney is responsible for effects such as nephrotoxicity and osteoporosis which are observed at adult age. Cadmium exposure through inhalation is also associated with lung cancer in adulthood. Although transfer to the neonate through the placenta and through breast milk is limited, teratogenic and developmental effects were observed in experimental animals. The database on h

Molecular and cellular mechanisms of cadmium carcinogenesis.

Cadmium is a heavy metal, which is widely used in industry, affecting human health through occupational and environmental exposure. In mammals, it exerts multiple toxic effects and has been classified as a human carcinogen by the International Agency for Research on Cancer. Cadmium affects cell proliferation, differentiation, apoptosis and other cellular activities. Cd2+ does not catalyze Fenton-type reactions because it does not accept or donate electrons under physiological conditions, and it is only weakly genotoxic. Hence, indirect mechanisms are implicated in the carcinogenicity of cadmiu

Effects of cadmium on trophoblast calcium transport.

Maternal exposure to cadmium (Cd) during pregnancy has been linked to low fetal birthweight, which may be attributed to placental damage and/or dysfunction in nutrient transport. Previous studies have suggested that Cd is accumulated in the placenta, and that placental transport of calcium (Ca) and zinc (Zn) is perturbed by Cd. To investigate the mechanism of Cd perturbation of Ca transport, we used JEG-3, a human choriocarcinoma cell line which exhibits trophoblastic properties, to analyse Cd effects in vitro. Treatment with Cd at low, physiologically relevant concentrations (e.g. 0.04 microM

Effect of cadmium in the zinc deficient rat.

Three-w-old male Sprague-Dawley rats were given a zinc (Zn)-deficient (0 ppm Zn) or a Zn-adequate diet (30 ppm Zn) supplemented with 0, 0.5 or 100 ppm cadmium (Cd) for up to 5 mo. The proximal convoluted tubules of the kidneys had degenerative changes in the rats fed the Zn-deficient diet containing 100 ppm Cd [Zn 0, Cd 100], but there were no lesions in other groups. Electron microscopy showed cytoplasmic vacuolation of the proximal tubules, mitochondrial swelling and coagulative necrosis in Zn 0:Cd 100 rats. The present study revealed diminished bone growth and cortical thinning of the femur

Problems in the analysis of cadmium in autopsied tissues.

A comparison was made between the concentrations of Cd, Zn, Cu, and Fe in the organs at autopsy and after fixation in formalin for anatomicopathological examination. The concentrations of all the metals were lower in the formalin-fixed organs. In particular, the residual rates of Cd in the renal cortex and medulla were 2.3 and 6.1% of those for the fresh organs, respectively. The Cd concentration in the blood collected at autopsy was several hundred times as high as the value measured before death and failed to be an indicator of exposure to Cd. This phenomenon is specific to Cd and is not fou

[Blocking of the latrotoxin channel by cadmium ions].

Influence of cadmium ions on current-voltage characteristics of latrotoxin treated BLM was studied. It was established that the affinity of cadmium ions to cis-mouth of a channel is substantially more than to trans-mouth for all the studied voltages. It was shown than the blocking of latrotoxin channels can occur by screening of the electric potential at the entrance of the pore.

Mobilization of hepatic cadmium in pregnant rats.

Mobilization of hepatic cadmium (Cd) in pregnant rats was investigated. Female Sprague-Dawley rats (200-250 g) were injected subcutaneously, daily with 1.0 mg Cd/kg body weight as CdCl2 for 8 days. A group of these rats was made pregnant. Copper (Cu), zinc (Zn), Cd, and metallothionein (MT) concentrations in the liver, kidney, and plasma of the control and Cd-injected, pregnant and nonpregnant rats, were compared. The hepatic Cd concentration of the Cd-injected rats decreased by 40% during pregnancy and became significantly lower than that of the nonpregnant Cd-injected rats. On the other hand

Thyrotoxicity of the chlorides of cadmium and mercury in rabbit.

Exposure to heavy metals such as cadmium and mercury is of immediate environmental concern. The present study was aimed at establishing a direct relationship between heavy metal poisoning and thyroid dysfunction. Cadmium and mercury treatment at LD50 levels resulted in severe thyrotoxicosis in the rabbit. Within 24 h of intramuscular administration of cadmium chloride 15 mg.kg-1 body weight (bw) and mercury chloride 20 mg.kg-1 bw, thyroid peroxidase activity increased significantly over the control with a concomitant rise in the triiodothyronine (T3) titre. On the other hand, there was a remar

Chromosomal effects of cadmium exposure in humans.

Cytogenetic studies on subjects occupationally or environmentally exposed to cadmium are reviewed and discussed. After a first report of increased chromosome aberrations in lymphocytes of itai-itai patients from Japan, several studies on workers occupationally exposed to cadmium gave conflicting results. These differences may perhaps be explained by the action of co-factors (e.g., nutritional deficiencies in itai-itai patients; concomitant exposure to other metals in some occupational groups), or by different levels of exposure. In a study on cadmium workers with a wide range of cumulative exp

Scientific basis for an occupational standard for cadmium.

The U.S. Occupational Safety and Health Administration (OSHA) has proposed a revised 8-hour permissible exposure limit (PEL) for cadmium in air of either 1 or 5 micrograms/m3, based upon the prevention of lung cancer and kidney dysfunction. To evaluate the scientific basis for these alternative standards, we compare the OSHA estimates of risk, derived from mathematical modelling of selected studies, to empirical data on lung cancer and kidney dysfunction in the published literature. At least seven epidemiologic studies examine renal tubular proteinuria by cumulative cadmium exposure. Three sug
