Pathogenic antibody recognition of cartilage.

Antibodies against cartilage proteins are highly prevalent in the sera and synovial fluids of rheumatoid arthritis (RA) patients and also precede disease induction in various spontaneous and induced animal models of arthritis. These antibodies play an important role in the induction and perpetuation of the clinical disease. Antibodies binding to cartilage protein(s), especially the major articular cartilage protein, collagen type II (CII) can induce, in naive mice, an acute form of arthritis that can substantially destroy the cartilage and bone architecture. More importantly, these anti-CII an…

[New developments in cartilage imaging].

Development of drugs able to modify the natural course of osteoarthritis is a major research objective. Imaging methods are needed to evaluate the effect of new medications in the experimental and clinical settings. Conventional radiographs are the gold-standard technique for routine imaging. However, magnetic resonance imaging (MRI) has transformed hyaline cartilage imaging by providing direct visualization without exposure to radiation. In addition, MRI can provide direct cartilage volumetry and tissue characterization for research purposes, using specific sequences. MRI can also give inform…

Auricular cartilage in revision rhinoplasty.

Revision rhinoplasty is a challenge in reconstruction to the rhinoplasty surgeon, both in the techniques of repair and the choice of implant material for augmentation grafting. Often, patients seeking revision or reconstructive rhinoplasty have previously undergone septoplasty with sacrifice of major amounts of septal cartilage. These situations confront the surgeon with the need for a decision about the material that will be used for structural grafting. The senior author follows the time-tested approach of generations of surgeons who have used exclusively autogenous material for nasal recons…

Cartilage palisade tympanoplasty and diving.

OBJECTIVE: To determine whether a patient with a serious defect of the tympanic membrane (TM) will be able to dive after surgery. PATIENTS AND METHODS: The authors describe three patients who were divers with serious defects of the TM (more than 75% of TM). In all three cases, cartilage palisade tympanoplasty was used to reconstruct the TM. RESULTS: Six months after surgery, the patients passed clinical examinations, audiograms (hearing restored to normal), tympanometry (increased stiffness of the TM), and pressure tolerance tests in hyperbaric chamber (30-m immersion was simulated for 4 min…

HIV infection of human cartilage.

Infection of human cartilage with HIV in vivo has not previously been reported. Specimens of articular cartilage taken at postmortem from ten patients who were HIV-positive were examined. Two had AIDS and eight were believed to have stage-2 disease. The standard polymerase chain reaction (PCR) protocol was modified to allow semiquantitative analysis of the samples. Oligonucleotide primers labelled with 32P gamma-ATP were used to detect a segment of HIV DNA and a control DNA gene segment (HLA genome) to estimate the ratio of infected cells. The 32P-labelled PCR products were separated on acryla…

Chemical stabilization of cartilage matrix.

Severe destruction of articular cartilage in osteoarthritis manifests clinically when repair processes cannot keep up with the catabolic processes. Loss of proteoglycans, which give the tissue its ability to undergo reversible deformation, precedes and probably contributes significantly to breakdown of the matrix in the most superficial layers of articular cartilage. In this study, we have examined the ability of dithiobis[succinimidyl propionate], a bifunctional reagent with a 1.2-nm span that cross-links proteins at lysine amino acid, and poly-L-lysine of high molecular weight (average MW 36…

Ossiculoplasty with presculpted banked cartilage.

The use of self-stabilizing, presculpted cartilage homografts for middle ear reconstruction has an established track record with long-term results equivalent to reconstruction methods using alloplastic material. This method affords several advantages over other techniques, which will be discussed in detail.

Crushed cartilage in nasal reconstruction.

Experience when using crushed cartilage for restoration of the dorsal nasal contour is presented. The septal cartilage is used as fresh autogenous grafts and as preserved septal cartilage. Implantation of autogenous grafts is preferable. Bank cartilage, however, has the advantage of being easily available and of causing very little reaction in the host organism. Normally, it is absorbed very slowly and replaced by connective tissue, which retains most of the bulk and shape of the implant. The crushed material is pushed in over the undermined nasal dorsum by means of a syringe connected to a pl…

Histochemical properties of cartilage proteoglycans.

Proteoglycan interaction with alcian blue at different concentrations of magnesium chloride was studied both in vitro and in histological sections of paraffin-embedded tissues. Our experiments indicate that a) proteoglycans with different contents of chondroitin sulfate and keratan sulfate, prepared under nondegradative conditions, are not distinguishable on the basis of the critical electrolyte concentrations at which staining is abolished; b) the state of aggregation of proteoglycans only very slightly affects the alcian blue affinity of the macromolecules at different concentrations of magn

A Clinical Study of Decellularized Tissue Engineering Hyaline Cartilage Graft (dLhCG) for Articular Cartilage Repair.

Current clinical approaches for knee cartilage repair are often limited by hypertrophy and fibrosis in the regenerated tissues. A decellularized tissue-engineered hyaline cartilage graft (dLhCG), composed of a type II collagen-based extracellular matrix, has been developed to overcome this limitation. Extensive preclinical studies have demonstrated the ability of dLhCG to achieve hyaline cartilage repair and maintain a stable hyaline phenotype. In this prospective, first-in-human, exploratory clinical study (ChiCTR2400094194, ChiCTR2500100526, and ChiCTR2500105847), dLhCG was implanted to trea…

Injectable microspheres adhering to the cartilage matrix promote rapid reconstruction of partial-thickness cartilage defects.

An effective treatment for the irregular partial-thickness cartilage defect in the early stages of osteoarthritis (OA) is lacking. Cartilage tissue engineering is effective for treating full-thickness cartilage defects with limited area. In this study, we designed an injectable multifunctional poly(lactic-co-glycolic acid) (PLGA) microsphere to repair partial-thickness cartilage defects. The microsphere was grafted with an E7 peptide after loading the microsphere with kartogenin (KGN) and modifying the outer layer through dopamine self-polymerization. The microsphere could adhere to the cartil…

Prediction of Hip Development Using Femoral Head Cartilage and Acetabular Cartilage Diameters on MRI Images.

BACKGROUND: Acetabular dysplasia may persist in developmental dysplasia of the hip (DDH), leading to an increased risk of early hip degeneration. This study aimed to identify predictive factors for residual acetabular dysplasia (RAD) in children aged 2 to 5 years using magnetic resonance imaging (MRI). METHODS: A retrospective study was conducted on 67 hips from 34 patients. The diameters of the femoral head and acetabular cartilages were measured on T2-weighted coronal images, and the difference and ratio of the cartilage diameters were calculated. The outcome measure was the Severin classif…

Cartilage-targeting peptide-modified cerium oxide nanoparticles alleviate oxidative stress and cartilage damage in osteoarthritis.

BACKGROUND: Osteoarthritis (OA) is a degenerative joint disease that leads to a substantial decline in the well-being of older individuals. Chondrocyte senescence and the resultant damage to cartilage tissue, induced by elevated levels of reactive oxygen species within the joint cavity, are significant causative factors in OA development. Cerium oxide nanoparticles (CeONPs) present a promising avenue for therapeutic investigation due to their exceptional antioxidant properties. However, the limited effectiveness of drugs in the joint cavity is often attributed to their rapid clearance by synov…

Evaluation of Articular Cartilage Regeneration Properties of Decellularized Cartilage Powder/Modified Hyaluronic Acid Hydrogel Scaffolds.

The articular cartilage has poor intrinsic healing potential, hence, imposing a great challenge for articular cartilage regeneration in osteoarthritis. Tissue regeneration by scaffolds and bioactive materials has provided a healing potential for degenerated cartilage. In this study, decellularized cartilage powder (DCP) and hyaluronic acid hydrogel modified by aldehyde groups and methacrylate (AHAMA) were fabricated and evaluated in vitro for efficacy in articular cartilage regeneration. In vitro tests such as cell proliferation, cell viability, and cell migration showed that DCP/AHAMA has neg…
