Biosynthesis of rhizoxin in Burkholderia rhizoxinica affords an unusual polyketide synthase module with ketosynthase and branching domains that install the δ-lactone, conferring antimitotic activity. To investigate their functions in chain branching, we designed chimeric modules with structurally similar domains from a glutarimide-forming module and a dehydratase. Biochemical, kinetic and mutational analyses reveal a structural role of the accessory domains and multifarious catalytic actions of the ketosynthase.

PURPOSE: The small eye mouse mutant (Sey) is caused by a mutation of the Pax6 gene. Previous studies, in which aggregation chimeras were used, have demonstrated that Sey/Sey cells contribute poorly to the neural retina forming small clumps of cells restricted to the inner retina at embryonic day 16.5. In addition, Sey/+ cells are absent from the lens epithelium during this embryonic period and postnatally. By means of a combination of immunosurgery and a modified method of microsurgery, blastocysts were reconstructed to produce viable chimeric fetal-placental units. Reciprocal reconstituted blastocysts were produced using euploid and trisomy 16 blastocysts.
