Combining plasma biomarkers and cognitive challenge tests enhances prediction of functional trajectories of decline among older adults with cognitive impairment.

BackgroundPlasma biomarkers have emerged as promising, less invasive indicators of Alzheimer's disease (AD) pathology. However, biomarkers alone cannot indicate whether actual clinical symptoms are present or progressing. Plasma biomarkers when paired with specific cognitive deficits on brief novel Cognitive Challenge Tests (CCTs), provide excellent sensitivities to underlying AD pathology in preclinical and prodromal stage. There is a lack of data on the extent to which plasma biomarkers and CCTs independently and jointly, predict longitudinal functional decline in the earliest stages of cogn

The Effects of a Single-Session Virtual Rumination Intervention to Enhance Cognitive Functioning in Veterans With Subjective Cognitive Symptoms: Multimethod Pilot Study.

BACKGROUND: Subjective cognitive concerns (SCCs) entail perceived difficulties in thinking or memory, often reported without substantial objective evidence of cognitive impairment. These concerns are prevalent among individuals with a history of brain injuries, neurological conditions, or chronic illnesses, contributing to both psychological distress and functional limitations. They are increasingly considered to be a risk factor for future objective decline. A considerable number of individuals reporting SCCs also exhibit mental health symptoms, such as a history of trauma, depression, or anx

Non-linear demographic correction enhances the diagnostic utility of the Montreal Cognitive Assessment.

BackgroundThe Montreal Cognitive Assessment (MoCA) is widely used for cognitive screening. Despite numerous studies showing that MoCA scores are affected by demographic variables including age, education, and race, the instrument is typically evaluated using a raw score or simple one-point education correction in clinical practice.ObjectiveIn this study, we comprehensively evaluate the diagnostic accuracy of the MoCA in a large cohort of individuals being evaluated for mild cognitive impairment or dementia.MethodsWe used data from the National Alzheimer's Coordinating Center (NACC) database to

Synergistic Actions of Natural Compounds for Enhancing Cognitive and Physical Performance: A Narrative Review.

Energy drinks are widely consumed for cognitive and physical enhancement, yet most commercial formulations rely on a narrow mechanism: rapid stimulation from synthetic caffeine combined with artificial sweeteners. This stimulant-centric strategy overlooks the multidimensional physiology underlying sustained human energy, which emerges from interactions among neurocognitive activation, nitric oxide-mediated vascular function, mitochondrial ATP production, and metabolic homeostasis. This narrative review synthesizes evidence on naturally derived compounds that influence these pathways, including

An Augmented Reality Study for Enhancing Spatial-Temporal Cognition in Indoor Emergency Response.

Fast, accurate spatial-temporal cognition is vital for situational awareness and mission success in indoor emergencies. Complex layouts and occlusions hinder first responders' ability to judge distances, scales, and task completion times. This paper explores the enhancement of these cognitive abilities through the application of Augmented Reality (AR) devices. Utilizing mixed reality and reality capturing technologies, a digital twin model of a three-level indoor structure was built based on LiDAR scans of Turlington Hall at the University of Florida. A mixed reality environment was then const

Combined Ketamine and Cognitive-Behavioral Therapy: Leveraging Metaplasticity to Enhance Treatment Outcomes.

Over the past two decades, ketamine has emerged as a rapid-acting antidepressant, especially among patients with treatment-resistant depression. As an N-methyl-d-aspartate glutamate receptor antagonist, ketamine has reshaped the understanding of the underlying neurobiological mechanisms of depression treatment. Ketamine induces a transient state of enhanced neuroplasticity and likely potentiates metaplasticity-the brain's increased capacity to undergo future plastic changes. These effects suggest a posttreatment window during which patients are more cognitively flexible and responsive to other

Compensatory retinal blood flow enhancement in cognitively normal ApoE ε4 carriers.

This study aims to compare retinal vascular characteristics between ApoE ε4 carriers and non-carriers in healthy individuals. A total of 84 cognitively healthy subjects-64 non-carriers and 20 carriers of the ApoE ε4 allele-were analyzed using swept-source optical coherence tomography angiography (SS-OCTA). In the retinal vascular layer of OCTA images, parameters such as foveal avascular zone (FAZ) area, vessel perimeter, acircularity index (AI), and vascular density (FD) were quantified. Additionally, blood flow areas in the outer retina and choroidal capillaries, as well as the area without p

Mitophagy activation by rapamycin enhances mitochondrial function and cognition in 5×FAD mice.

Alzheimer's disease (AD) is the most prevalent form of dementia, characterized by severe mitochondrial dysfunction, which is an intracellular process that is significantly compromised in the early stages of AD. Mitophagy, the selective removal of damaged mitochondria, is a potential therapeutic strategy for AD. Rapamycin, a mammalian target of rapamycin (mTOR) inhibitor, augmented autophagy and mitigated cognitive impairment. Our study revealed that rapamycin enhances cognitive function by activating mitophagy, alleviating neuronal loss, and improving mitochondrial dysfunction in 5 ×FAD mice.

Cognitive Enhancer Donepezil Attenuates Heroin-Seeking Behavior Induced by Cues in Rats.

PURPOSE: Opioid use disorder is a significant global problem. Chronic heroin use is associated with impairment of cognitive function and conscious control ability. The cholinergic system can be disrupted following heroin administration, indicating that activation of the cholinergic system may prevent chronic heroin misuse. Donepezil as an inhibitor of cholinesterase has been reported to clinically improve cognition and attention. In this study, the inhibition of heroin self-administration and heroin-seeking behaviours by donepezil were evaluated in rats. METHODS: Rats were trained to self-adm

Peripherally administered amylin inhibits stress-like behaviors and enhances cognitive performance.

Amylin, a 37 amino acid peptide pancreatic hormone co-secreted with insulin, normalizes the altered eating patterns induced by chronic stress in the rat. Because these stress-induced changes are driven, in part, by brain corticotropin-releasing factor and corticosterone, and because alterations in the activity of these molecules and the stress system are commonly associated with neuropsychiatric diseases like anxiety, depression, and schizophrenia, we hypothesized that amylin might mitigate behavioral states associated with stress. Therefore, we tested the effects of rat amylin in rodent-based

Using an E-learning activity to enhance student nurses' understanding of cognitive impairment.

BACKGROUND: Nurses often lack the knowledge and expertise to work with older people with cognitive impairment, which includes dementia, delirium and depression. This is due in part to deficits in their basic nursing education related to older people and managing the care needs of people with cognitive impairment. We developed an e-learning activity to facilitate student nurses' knowledge of cognitive impairment. OBJECTIVES: To test if the e-learning activity could improve student nurses' knowledge of cognitive impairment and whether they would find the style of learning beneficial. DESIGN: A

Neuroprotective and Cognition-Enhancing Effects of Compound K Isolated from Red Ginseng.

The present study was aimed at elucidating the effect of compound K derived from red ginseng on memory function in mouse model and glutamate-induced cytotoxicity in mouse hippocampal HT22 cells. Compound K induced antioxidant enzymes in nuclear factor (erythroid-derived 2)-like 2 (Nrf2)-mediated manner, and effectively attenuated cytotoxicity and mitochondrial damage induced by glutamate in HT22 cells. However, the cytoprotective effect by compound K was abolished by heme oxygenase-1 inhibitor, tin protophorphyrin IX, suggesting that neuroprotective effect of compound K was caused by its Nrf2-
