ATF3/SPI1/SLC31A1 Signaling Promotes Cuproptosis Induced by Advanced Glycosylation End Products in Diabetic Myocardial Injury.

Cuproptosis resulting from copper (Cu) overload has not yet been investigated in diabetic cardiomyopathy (DCM). Advanced glycosylation end products (AGEs) induced by persistent hyperglycemia play an essential role in cardiotoxicity. To clarify whether cuproptosis was involved in AGEs-induced cardiotoxicity, we analyzed the toxicity of AGEs and copper in AC16 cardiomyocytes and in STZ-induced or db/db-diabetic mouse models. The results showed that copper ionophore elesclomol induced cuproptosis in cardiomyocytes. It was only rescued by copper chelator tetrathiomolybdate rather than by other cel

Copper nanoregulator with organelle-level precision reprograms COMMD1-Mediated copper homeostasis for myocardial infarction repair.

Copper (Cu) homeostasis dysregulation is one of the key factor exacerbating mitochondrial dysfunction and impairing cardiac repair in myocardial infarction (MI). Herein, by integrating single-cell transcriptomics, clinical specimens, and animal models, we first identified COMMD1 as a key negative regulator of Cu homeostasis in MI. We then constructed a Cu nanoregulator, Qu@Cu-SS31, that selectively accumulate in the mitochondria of ischemic cardiomyocytes and release bioactive Cu2+ in a pH-responsive manner, enhancing mitochondrial function and promoting myocardial recovery. Clinical, biochemical and cell biological characterization of KIDAR syndrome associated with a novel AP1B1 variant.

Adaptor protein (AP) complexes play key roles in escorting transmembrane proteins to various intracellular destinations, including the trans-Golgi compartment, secretory vesicles, and the plasma membrane. The AP-1 complex is heterotetrametric, comprised of four individual subunits: β1, γ1, σ1, and μ1, and encoded by separate genes that interact selectively with distinct cargo proteins. When AP-1 complex assembly is impaired due to loss-of-function variants in any of its component genes, clinical consequences related to altered transmembrane protein trafficking may result. Novel Role of Copper Transporter CTR1 and Therapeutic Potential of Copper Chelators in Retinal Ischemia-Reperfusion Injury.

PURPOSE: Retinal ischemia contributes to vision loss in ischemic and diabetic retinopathies through oxidative stress, neurovascular injury, and inflammation. Copper (Cu), whereas an essential micronutrient, can be toxic in excess and is regulated by Cu transporters such as CTR1. However, the role of CTR1 in ischemic retinopathy remains unclear. METHODS AND RESULTS: Retinal ischemia-reperfusion (IR) injury was induced by elevating intraocular pressure (IOP) to 110 millimeters of mercury (mm Hg) for 40 minutes in the right eyes of Ctr1 heterozygous (Ctr1+/-) and wild-type (WT) mice. In WT mice,

Nitroxyl relieves acute kidney injury by suppressing SLC31A1-mediated cuproptosis in renal tubular epithelial cells.

CONTEXT: Cuproptosis, a copper-dependent form of programmed cell death, has been implicated in the pathogenesis of acute kidney injury (AKI). Nitroxyl (HNO), the one-electron reduced and protonated form of nitric oxide (NO), is an emerging regulator of cellular function. However, the role of HNO in modulating cuproptosis during AKI remains largely unexplored. OBJECTIVE: This study aims to investigate the effect of HNO on cuproptosis in a murine model of renal ischemia-reperfusion (IR) injury. MATERIALS AND METHODS: An in vitro hypoxia/reoxygenation (HR) model using human kidney-2 (HK-2) cell

Femtosecond Single-Particle Spectroscopy of Exciton Diffusion in Individual Copper Phthalocyanine Nanofibers.

Exciton diffusion in individual copper phthalocyanine (CuPc) nanofibers of β- and η-crystalline phases was investigated using femtosecond single-particle spectroscopy. The exciton diffusion coefficient (D) ranged from 1.6 × 10-5 to 0.079 cm2 s-1 for β-CuPc and from 0.040 to 0.48 cm2 s-1 for η-CuPc, with the η phase showing an average D (0.16 cm2 s-1) nearly ten times larger than that of the β phase (0.020 cm2 s-1). The enhanced transport in η-CuPc is attributed to a stronger excitonic coupling and greater intermolecular overlap. The broad distributions of D within each phase strongly correlate

Identification and analysis of mutations in the Wilson disease gene (ATP7B): population frequencies, genotype-phenotype correlation, and functional analyses.

Wilson disease (WD) is an autosomal recessive disorder characterized by toxic accumulation of copper in the liver and subsequently in the brain and other organs. On the basis of sequence homology to known genes, the WD gene (ATP7B) appears to be a copper-transporting P-type ATPase. A search for ATP7B mutations in WD patients from five population samples, including 109 North American patients, revealed 27 distinct mutations, 18 of which are novel. A composite of published findings shows missense mutations in all exons-except in exons 1-5, which encode the six copper-binding motifs, and in exon

Differential Effects of Histidine and Histidinamide versus Cysteine and Cysteinamide on Copper Ion-Induced Oxidative Stress and Cytotoxicity in HaCaT Keratinocytes.

Metal chelators are used for various industrial and medical purposes based on their physicochemical properties and biological activities. In biological systems, copper ions bind to certain enzymes as cofactors to confer catalytic activity or bind to specific proteins for safe storage and transport. However, unbound free copper ions can catalyze the production of reactive oxygen species (ROS), causing oxidative stress and cell death. The present study aims to identify amino acids with copper chelation activities that might mitigate oxidative stress and toxicity in skin cells exposed to copper i

Late neurological presentations of Wilson disease patients in French population and identification of 8 novel mutations in the ATP7B gene.

Wilson disease (WD) is an autosomal recessive disorder of copper biliary excretion caused by an impaired function of ATP7B, a metal-transporting P-type ATPase encoded by WD gene. It results in copper accumulation, mostly in liver and brain tissues. Mutation analysis was carried out on 11 WD French unrelated patients presenting a predominant neurological form of this illness. SSCP and dHPLC analysis followed by sequencing of the 21 exons and their flanking introns were performed. Thirteen different mutations in a total of 17, and, among them, 10 novel variants were evidenced. Two deletions (c.6

Cross-Linking and Functional Analyses for Dimerization of a Cysteine Mutant of Glycine Transporter 1.

Glycine transporter 1 (GlyT1) is responsible for the reuptake of glycine, which regulates glutamate signaling as a co-agonist with N-methyl-D-aspartic acid (NMDA) receptors in the excitatory synapse and has been proposed to be a potential target in the development of therapies for a broad range of disorders of the central nervous system. Despite significant progress in characterizing structure and transport mechanism of the transporter, the regulation of transport function through oligomerization remains to be understood. In the present work, association of two forms of GlyT1 into dimers and h

Cuproptosis and prostate cancer: from molecular mechanisms and microenvironment remodeling to precision therapy.

Prostate cancer (PCa) is a leading malignancy, and progression to castration-resistant prostate cancer (CRPC) remains a central therapeutic challenge. Cuproptosis, a copper-dependent cell death mechanism first characterized in 2022, is triggered by copper binding to lipoylated tricarboxylic acid (TCA)-cycle proteins, inducing their aggregation, Fe-S cluster protein instability, and mitochondrial proteotoxic stress. This review critically evaluates the emerging but still heterogeneous evidence linking cuproptosis to PCa, explicitly distinguishing prostate cancer-specific data from pan-cancer,no

SLC31A1 knockdown mitigates post-MI heart failure via regulation of copper metabolism.

INTRODUCTION: Cuproptosis due to copper overload is a contributor to the progression of cardiovascular diseases, especially heart failure (HF) after acute myocardial infarction (AMI). Solute carrier family 31 member 1 (SLC31A1) is a major Cu2+ transporter responsible for intracellular Cu2+ uptake. In this study, we investigated the role and detailed mechanism of SLC31A1 in post-AMI HF. METHODS: Mouse left anterior descending coronary artery was ligated to produce an in vivo post-AMI HF model. These mice were subjected to treatments with short hairpin RNA targeting SLC31A1, the copper chelator

Flexible electrochemical sensor for highly sensitive and selective non-enzymatic detection of creatinine via electrodeposited copper over polymelamine formaldehyde.

A non-enzymatic electrochemical biosensor was developed for highly sensitive detection of creatinine using copper nanoparticles supported over polymelamine formaldehyde. The synergy between the electrodeposited copper nanoparticles over the highly porous polymer (eCu-PMF) provided a greener platform to boost up the electron transport at the electrode electrolyte interface by eliminating the role of redox species as well as interference of major interferents like glucose, dopamine, and ascorbic acid in physiological media 0.1 M PBS (pH 7.4). The proposed sensor exhibited a wide detection range

A Self-Reinforcing Cuproptosis-Radiotherapy Loop by Copper Carbonate Nanoparticles for Enhanced Immunotherapy.

Radiotherapy leverages radiation-induced immunogenic cell death (ICD) to boost antitumor immunity, showing great potential for localized cancer therapy. However, its immunostimulatory efficacy is limited by suboptimal immunogenicity and the immunosuppressive tumor microenvironment (TME). Herein, we developed a multifunctional therapeutic platform based on copper carbonate nanoparticles (CuCO3@PEG) for enhanced antitumor immunity via remodeling TME and establishing a self-reinforcing cuproptosis-radiotherapy loop. CuCO3@PEG remodels the immunosuppressive TME by neutralizing H+, depleting lactic

Evaluation of divalent metal transporter 1 (DMT1) (rs224589) polymorphism on blood lead levels of occupationally exposed individuals.

Lead (Pb) is an environmental and public health toxicant. It affects various organ systems of the body, thereby disrupting their normal functions. To date, several genes that are known to influence the mechanism of action of lead and toxicity have been studied. Among them, the iron transporter gene, SLC11A2 (Solute Carrier 11 group A member 2) which codes for the transmembrane protein, DMT1 (Divalent Metal Transporter 1) has shown to transport other metals including zinc, copper, and lead. We investigated the influence of DMT1 polymorphism (rs224589) on blood lead (Pb-B) levels. In the present

The substrates of the gamma-aminobutyric acid transporter GAT-1 induce structural rearrangements around the interface of transmembrane domains 1 and 6.

The sodium- and chloride-coupled gamma-aminobutyric acid (GABA) transporter GAT-1 is essential for efficient synaptic transmission by this neurotransmitter. GAT-1 is the first cloned member of the neurotransmitter-sodium-symporter family. Here we address the idea that during transport the extracellular halves of transmembrane domains (TM) 1 and 6, TM 1b/TM 6a, move relative to the binding pocket. Therefore, we have probed the aqueous accessibility of TM 6a and its proximity to TM 1b in the presence and absence of its substrates. Cysteines were introduced, one by one, at all TM 6a positions. Advanced Glycation End Products Induce Cuproptosis and Then Promote the Activation and Migration of Cardiac Fibroblasts.

Excessive advanced glycation end products (AGEs) can lead to cardiovascular diseases such as myocardial fibrosis (MF). Although studies have found that AGEs induce cuproptosis, and cuproptosis promotes fibrosis, it has not been confirmed whether AGEs promote MF through cuproptosis. AGEs increased intracellular copper levels, promoted the expression of solute carrier family 31 member 1 (SLC31A1), and downregulated the expression of ferredoxin 1 (FDX1), lipoic acid synthetase (LIAS), lipoylated (Lip)-dihydrolipoamide S-succinyltransferase (DLST), and Lip-dihydrolipoamide S-acetyltransferase (DLA

Cisplatin nephrotoxicity: a review of the literature.

Cisplatin is a platinum containing drug first approved as an antineoplastic agent in 1978. It remains an important and effective therapy in many forms of cancer today. Cisplatin mediates its tumorcidal effects via a number of different cytotoxic mechanisms. Although it is best known for DNA damage, cisplatin also causes cytoplasmic organelle dysfunction particularly with the endoplasmic reticulum and mitochondria. It also activates apoptotic pathways and inflicts cellular damage via oxidative stress and inflammation. One of its dose limiting toxicities is its effects on the kidney. This includ

A mitochondrion-targeted natural polyphenolic copper carrier overcomes tumor resistance to cisplatin by potentiating cuproptosis.

BACKGROUND: Platinum–based drug resistance remains a major obstacle in cancer therapy. Cuproptosis, a novel form of copper–dependent cell death regulated through mitochondrial pathways, represents a promising strategy to counteract drug resistance in tumors. However, its efficacy is constrained by several physiological barriers, including elevated intracellular glutathione (GSH) levels, inadequate copper accumulation both cytoplasmically and within mitochondria, and the overexpression of copper efflux transporters such as ATP7A/B. METHODS: A series of material characterization techniques (FT-I
