Delayed effects of low level acute irradiation and chronic environmental radioactive contamination on DNA lymphocytes of people living in Dolon, a settlement located in the vicinity of the Semipalatinsk nuclear test site (Kazakhstan).

During 42 years several hundred nuclear tests were performed by the former USSR at the Semipalatinsk Test Site (STS, Kazakhstan), of which more than 100 were done in the atmosphere. We report here the late genetic damage of external exposure to radiation and environmental radioactive contamination in people living in Dolon, a small settlement situated in the vicinity of the STS. The comet assay was applied on DNA lymphocytes of 20 exposed women and 32 non-exposed women living at 500 km from the STS. We observed a statistically significant difference between the exposed and control groups for m

Biomarkers of genotoxicity of air pollution (the AULIS project): bulky DNA adducts in subjects with moderate to low exposures to airborne polycyclic aromatic hydrocarbons and their relationship to environmental tobacco smoke and other parameters.

The levels of bulky DNA adducts were measured by (32)P-post-labelling in lymphocytes of 194 non-smoking students living in the city of Athens and the region of Halkida, Greece, once in the winter and again in the following summer. Personal exposures to particulate-bound polycyclic aromatic hydrocarbons (PAH) were significantly higher in Athens subjects during both seasons. There was hardly any diagonal radioactive zone in the pattern of DNA adducts observed. Highest adduct levels were observed in a sub-group of subjects living in or near the Halkida Institute campus, which was located in rural

Towards understanding the liver fluke transmission dynamics on farms: Detection of liver fluke transmitting snail and liver fluke-specific environmental DNA in water samples from an irrigated dairy farm in Southeast Australia.

Livestock production around the world is impacted by liver fluke (Fasciola spp.) infection resulting in serious economic losses to the beef, dairy and sheep industries with significant losses of about $90 million per annum in Australia. Triclabendazole (TCBZ) is the most effective anthelmintic treatment available to control liver fluke infections; however, the widespread emergence of TCBZ resistance in livestock threatens liver fluke control. Alternative control measures to lower exposure of livestock to liver fluke infection would help to preserve the usefulness of current anthelmintic treatm

Evaluation of human brain damage in fatalities due to extreme environmental temperature by quantification of basic fibroblast growth factor (bFGF), glial fibrillary acidic protein (GFAP), S100β and single-stranded DNA (ssDNA) immunoreactivities.

Fatalities due to extreme environmental temperatures involving hypothermia (cold exposure) and hyperthermia (heat stroke) might present with poor or nonspecific morphological pathologies, which are insufficient to establish the cause of death in forensic practice. The present study immunohistochemically investigated basic fibroblast growth factor (bFGF), glial fibrillary acidic protein (GFAP), S100β and single-stranded DNA (ssDNA) in the parietal lobe and hippocampus of the brain in fatalities from hypothermia (n=15) and hyperthermia (n=18), and compared them to those of controls (n=39), inclu

Assessment of environmental contamination with Echinococcus spp. through DNA detection in free-roaming canid feces and soil in human echinococcosis hotspots from the Three-River-Source Region of the Qinghai-Tibet Plateau, China.

BACKGROUND: The Three-River-Source Region of the Qinghai-Tibet Plateau is a hyperendemic focus for echinococcosis, with Echinococcus granulosus, E. multilocularis, and E. shiquicus circulating between definitive canid hosts (dogs and foxes) and intermediate hosts (livestock and rodents). However, the extent of environmental contamination by Echinococcus eggs remains understudied and poses significant risks to human and animal health. METHODS: From 2019 to 2021, we collected 631 canid fecal samples (296 from dogs and 335 from foxes) and 398 adjacent soil samples across endemic counties in the

Environmental Epigenetics and Genome Flexibility: Focus on 5-Hydroxymethylcytosine.

Convincing evidence accumulated over the last decades demonstrates the crucial role of epigenetic modifications for mammalian genome regulation and its flexibility. DNA methylation and demethylation is a key mechanism of genome programming and reprogramming. During ontogenesis, the DNA methylome undergoes both programmed changes and those induced by environmental and endogenous factors. The former enable accurate activation of developmental programs; the latter drive epigenetic responses to factors that directly or indirectly affect epigenetic biochemistry leading to alterations in genome regu

Continuous eDNA Monitoring Reveals Rare Fish Diversity and Enables Filtering of Exogenous DNA Signals in a Megariver.

Megarivers support high biodiversity but challenge monitoring efforts, as conventional low-frequency environmental DNA (eDNA) surveys often miss rare taxa and may misclassify transient exogenous DNA. Here, we conducted daily eDNA monitoring for 1 year at a fixed site on the Yangtze River to evaluate the effects of sampling frequency and distinguish native from exogenous signals. Among 210 detected fish taxa, 124 were verified as native. Fish diversity exhibited strong temporal variation, peaking in warm seasons. Downsampling analyses revealed that quarterly sampling recovered only ∼30% of annu

DNA typing of isolates associated with the 1988 sporotrichosis epidemic.

DNA typing techniques were used to examine selected clinical and environmental isolates of Sporothorix spp. recovered from the 1988 sporotrichosis epidemic in multiple states of the United States. Previous studies indicated that isolates in one of the six morphologically or physiologically distinct groups (group I) obtained from environmental sources were Sporothrix schenckii and were the possible etiologic agents responsible for the epidemic. To assess this hypothesis at the genetic level, whole-cell DNA was extracted from selected clinical isolates and representative members of each of the s

Enhanced Extraction of Low-Molecular Weight DNA from Wastewater for Comprehensive Assessment of Antimicrobial Resistance.

Environmental surveillance is recognized as an important tool for assessing public health in the post-pandemic era. Water, in particular wastewater, has emerged as the source of choice to sample pathogen burdens in the environment. Wastewater from open drains and community water treatment plants is a reservoir of both pathogens and antimicrobial resistance (AMR) genes, and frequently comes in contact with humans. While there are many methods of tracking AMR from water, isolating good-quality DNA at high yields from heterogeneous samples remains a challenge. To compensate, sample volumes often

Exploring technical improvements for environmental nucleic acids-based biodiversity assessment and management in coastal ecosystems.

Assessing and conserving marine biodiversity remain critical global challenges, particularly in highly disturbed coastal regions. The use of environmental DNA (eDNA)-metabarcoding has revolutionized biodiversity assessment and management; however, the prevalence of both false positives and negatives continues to be a significant concern. To address these technical errors, we tested two potential methodological improvements in the highly disturbed Guangdong-Hong Kong-Macao Greater Bay Area: (1) the use of random whole-genome amplification (WGA) to reduce false negatives derived from low eDNA co

Effects of sampling strategies and DNA extraction methods on eDNA metabarcoding: A case study of estuarine fish diversity monitoring.

Environmental DNA (eDNA) integrated with metabarcoding is a promising and powerful tool for species composition and biodiversity assessment in aquatic ecosystems and is increasingly applied to evaluate fish diversity. To date, however, no standardized eDNA-based protocol has been established to monitor fish diversity. In this study, we investigated and compared two filtration methods and three DNA extraction methods using three filtration water volumes to determine a suitable approach for eDNA-based fish diversity monitoring in the Pearl River Estuary (PRE), a highly anthropogenically disturbe

Extreme weather events and environmental contamination are associated with case-clusters of melioidosis in the Northern Territory of Australia.

BACKGROUND: Melioidosis, the infection due to the environmental organism Burkholderia pseudomallei, is endemic to northern Australia and South East Asia. It is associated with exposure to mud and pooled surface water, but environmental determinants of this disease are poorly understood. We defined case-clusters in northern Australia, determined their contribution to the observed rate of melioidosis, and explored clinical features and associated environmental factors. METHODS: Using geographical information systems data, we examined clustering of melioidosis cases in time and geographical spac

Nosocomial outbreak of epidemic keratoconjunctivitis accompanying environmental contamination with adenoviruses.

An outbreak of acute keratoconjunctivitis involving 27 patients occurred in the Department of Ophthalmology, Kurume University Hospital. Adenoviral DNA was detected in four inpatients, one outpatient and one healthcare worker. Sequence-based typing of adenoviral DNA indicated serotype 3 from one inpatient, the rest being serotype 37. At a later stage of the outbreak adenoviral DNA types 37 and/or 3 were also detected from almost all environmental instruments and commonly used eye drops, despite thorough disinfection of the environment and enforcement of various infection control measures. An Optimized Method for Quantification of Pathogenic Leptospira in Environmental Water Samples.

Leptospirosis is a zoonotic disease usually acquired by contact with water contaminated with urine of infected animals. However, few molecular methods have been used to monitor or quantify pathogenic Leptospira in environmental water samples. Here we optimized a DNA extraction method for the quantification of leptospires using a previously described Taqman-based qPCR method targeting lipL32, a gene unique to and highly conserved in pathogenic Leptospira. QIAamp DNA mini, MO BIO PowerWater DNA and PowerSoil DNA Isolation kits were evaluated to extract DNA from sewage, pond, river and ultrapure

Salivary 8-hydroxy-2'-deoxyguanosine as a biomarker of oxidative DNA damage in urban outdoor workers exposed to traffic-related air pollution.

Urban air pollution, particularly fine particulate matter (PM2.5), is a major environmental health concern associated with oxidative DNA damage. However, evidence in occupationally exposed and socially vulnerable populations remains limited, and applicability of noninvasive biomarkers requires further investigation. The aim of this study was to determine feasibility of using salivary 8-hydroxy-2'-deoxyguanosine (8-OHdG) as a biomarker of oxidative DNA damage in urban workers exposed to traffic-related air pollution and examine this associations with environmental, biological, and socioeconomic

Optimizing target-to-total DNA ratio in eDNA studies: effects of sampling, preservation, and extraction methods on single-species detection.

There are many decisions to be made when sampling for environmental DNA (eDNA) analysis, whether using a targeted, single-species assay or community-based metabarcoding. Of the entire workflow from sampling water to bioinformatic analyses, the first steps in the process of collecting water, filtering it, and preserving the filter membranes represent major decision points upon which the success of downstream processes depend. Though many previous studies have compared water volume filtered, filter pore size, and preservation and extraction methods, the conclusions are often that they produce di

Current status and topical issues on the use of eDNA-based targeted detection of rare animal species.

Animal detection through DNA present in environmental samples (eDNA) is a valuable tool for detecting rare species, that are difficult to observe and monitor. eDNA-based tools are underpinned by molecular evolutionary principles, key to devising tools to efficiently single out a targeted species from an environmental sample. Here, we present a comprehensive review of the use of eDNA-based methods for the detection of targeted animal species, such as rare, endangered, or invasive species, through the analysis of 549 publications (2008-2022). Aquatic ecosystems have been the most surveyed, in pa

Genotoxic effects of pesticides.

Epidemiologic data showed an increase in the number of cancer cases in persons involved in agricultural production using pesticides. According to IARC, more than 25% of pesticides are classified as oncogens. In recent years, the concept of malignant tumors developing after environmental contamination with chemicals has been accepted. Changes in genetic material are at the basis of this process because many environmental pollutants are chemical carcinogens and mutagens with the capacity of causing DNA damage. DNA damage was proposed as a useful parameter for assessing the genotoxic properties o

Linking the epidemiology of coccidioidomycosis and environmental exposure through targeted genomic enrichment of Coccidioides posadasii.

Valley fever, a disease caused by Coccidioides spp., is a fungal respiratory disease with an expanding range. Methods to culture the pathogen from soil, especially Coccidioides posadasii, are very challenging, limiting the genomics knowledge of environmental strains. In this study, we designed and tested a targeted DNA capture and enrichment system for the characterization of Coccidioides genomes without the need to culture. In this system, RNA probes are hybridized to Coccidioides DNA in a complex sample, followed by DNA amplification, sequencing, and analysis. Our enrichment system was targe
