Effects of inoculating autochthonous starter cultures on N-nitrosodimethylamine and its precursors formation during fermentation of Chinese traditional fermented fish.

This study investigated the effects of Lactobacillus plantarum 120, Saccharomyces cerevisiae 2018 and Staphylococcus xylosus 135 inoculation on N-nitrosodimethylamine (NDMA) and its precursors formation, and on microbiological characteristics of Chinese traditional fermented fish products (CTFPs). The results indicated that three strains could directly degrade NDMA in culture broth, and the highest degradation rate was observed in L. plantarum 120. The lactic acid bacteria counts in samples inoculated with L. plantarum 120 and mixed starter cultures were significantly (P < 0.05) higher than th

Crystal defects cause nitrosamine formation in ranitidine under accelerated storage conditions.

The formation of nitrosamines, a class of potent carcinogen impurities, in pharmaceutical products such as ranitidine hydrochloride (RAN) has raised significant public health concerns and led to widespread product recalls. Previous studies have shown that RAN can degrade to N-nitrosodimethylamine (NDMA) after storage at 60 °C for several days. These studies also suggested a link between the degradation rate and the crystal morphology of the RAN. This study shows that NDMA formation in RAN is primarily driven by solid-state reactive species (SSRS) introduced during pharmaceutical manufacturing

Research progress on the generation of NDMA by typical PPCPs in disinfection treatment of water environment in China: A review.

The drugs and personal care products in water sources are potential threats to the ecological environment and drinking water quality. In recent years, the presence of PPCPs has been detected in multiple drinking water sources in China. PPCPs are usually stable and resistant to degradation in aquatic environments. During chlorination, chloramination, and ozonation disinfection processes, PPCPs can act as precursor substances to generate N-nitrosodimethylamine (NDMA) which is the most widely detected nitrosamine byproduct in drinking water. This review provides a comprehensive overview of the im

On the Risk of Nitrosamine Contamination During Drug Product Blister Packaging.

Most N-Nitrosamine compounds are found to be genotoxic in several animal species. Some are classified as probable or possible human carcinogens and very low acceptable daily intake has been established such as 96 ng/day for N-nitrosodimethylamine (NDMA) and 26.5 ng/N-nitrosodiethylamine (NDEA). The pharmaceutical industry has considered all processing areas for potential formation or contamination of N-nitrosamine. One risk is the potential contamination of nitrosamine during drug product blister packaging using lidding foils containing nitrocellulose, and different approaches have been used b

NDMA analytics in metformin products: Comparison of methods and pitfalls.

BACKGROUND: For nearly three years, the concerns regarding trace levels of N-nitrosamines in pharmaceuticals and the associated cancer risk have significantly expanded and are a major issue facing the global pharmaceutical industry. N-nitrosodimethylamine (NDMA) found in formulations of the popular anti-diabetic drug metformin is a prominent example. This has resulted in product recalls raising the profile within the media. Issues of method robustness, sample preparation and several unexpected sources of nitrosamine contamination have been highlighted as false positive risks. It has become app

Alteration of hepatic DNA synthesis in rats treated with N-nitrosodimethylamine during early stages of copper deficiency.

The main aim of this research was to clarify that nutritional dietary copper may participate in the protective action against hepatocarcinogenesis in rats by N-nitrosodimethylamine (NDMA). The copper concentrations in serum and liver from 2 to 8d after rats were first fed a copper deficient diet (copper, 0.6 ppm) decreased significantly compared to those of pair-fed rats (copper in a control diet, 7 ppm). The subcellular distribution of copper in the liver at 5d after feeding of a copper deficient diet began was measured and the copper concentrations in soluble and nuclear fractions decreased

[Granulomatous inflammation of the liver in opisthorchiasis].

Granulomatous liver inflammation was studied in Syrian hamsters in opisthorchiasis alone and in combination with different factors, such as carcinogens, anthelmintics, immunosuppressants and pregnancy. Parasitic ova and metabolites, as well as hepatic cell necrosis are shown to be the initiating factors of granulomatous inflammation. Granulomas originate in ductal walls, hepatic stroma and parenchyma. Four granuloma types are distinguished, i.e. macrophagal, giant-cell, necrotizing and cicatricial one. The biggest granuloma area was observed in superinvasive form of helminthiasis, when infecti

Hepatocyte growth factor suppresses the onset of liver cirrhosis and abrogates lethal hepatic dysfunction in rats.

Heptic fibrosis/cirrhosis is a common hepatic disease characterized by the hyper-accumulation of connective tissue components, and hepatic necrosis. Chronic alcohol ingestion, viral infection, and metabolic disorders are contributing factors and there has been no effective treatment. Hepatocyte growth factor (HGF), originally identified as a potent mitogen for mature hepatocytes, is a long-sought hepatotrophic factor for liver regeneration. Administration of human recombinant HGF into rats with hepatic fibrosis/cirrhosis caused by dimethylnitrosamine (DMN) elicited mitogenic action for hepatoc

Promoting role of Clonorchis sinensis infection on induction of cholangiocarcinoma during two-step carcinogenesis.

Chronic Clonorchis sinensis (CS) infection is etiologically related to cholangiocarcinoma (CHCA) in human and animals. This study was carried out to clarify the role of CS infection on dimethylnitrosamine (DMN)-induced cholangiocarcinogenesis. Fifteen hamsters were administered with 15 ppm DMN for 4 weeks and one week later, the hamsters were infected with 15 metacercariae of CS (DMN-->CS group). The other 15 hamsters were infected with CS and after 5 weeks they were treated with the drug, praziquantel. Again one week later, the hamsters were administered with DMN (CS-->DMN group). The other 1

Paternal exposure to low-dose N-nitrosodimethylamine (NDMA) induces intergenerational alterations in the prostatic microenvironment of rats.

NDMA is a nitrosamine recognized as an environmental contaminant of water and food, unintentionally formed during industrial processes and water treatment. Due to its mobility in aquatic environments, persistence, and carcinogenic potential, NDMA has emerged as a toxicological and public health concern. This study evaluated the effects of low-dose exposure (7.2 ng/kg/day) on the prostate of male Wistar rats across two generations within the Paternal Origins of Health and Disease (POHaD) framework. F0 males were exposed via gavage from postnatal day (PND) 60 to 104. In F1, offspring were evalua

Biological treatment of N-nitrosodimethylamine (NDMA) and N-nitrodimethylamine (NTDMA) in a field-scale fluidized bed bioreactor.

The ex situ treatment of N-nitrosodimethylamine (NDMA) and N-nitrodimethylamine (NTDMA) in groundwater was evaluated in a field-scale fluidized bed bioreactor (FBR). Both of these compounds, which originally entered groundwater at the test site from the use of liquid rocket propellant, are suspected human carcinogens. The objective of this research was to examine the application of a novel field-scale propane-fed fluidized bed bioreactor as an alternative to ultraviolet irradiation (UV) for treating NDMA and NTDMA to low part-per-trillion (ng/L) concentrations. Aerobic treatment of N-nitrosodimethylamine in a propane-fed membrane bioreactor.

N-Nitrosodimethylamine (NDMA) is a suspected human carcinogen that has recently been detected in wastewater, groundwater and drinking water. Treatment of this compound to low part-per-trillion (ng/L) concentrations is required to mitigate cancer risk. Current treatment generally entails UV irradiation, which while effective, is also expensive. The objective of this research was to explore potential bioremediation strategies as alternatives for treating NDMA to ng/L concentrations. Batch studies revealed that the propanotroph Rhodococcus ruber ENV425 was capable of metabolizing NDMA from 8 μg/L

Influence of dithiocarbamates on the metabolism and toxicity of N-nitrosodimethylamine in rats.

Five secondary amines and secondary amino acids were reacted with carbon disulfide to yield dithiocarbamates. These compounds were tested for their influence on biochemical parameters and on some biological effects of N-nitrosodimethylamine (NDMA). The following results were found. (i) All dithiocarbamates tested reduced the activity of N-nitrosodiethylamine-deethylase and completely inhibited the N-nitrosodimethylamine-demethylase in the rat liver. A striking influence on the glutathione content and on the activity of glutathione-S-transferase and glutathione-reductase was not observed. (ii)

New Perspectives on the Interactions between Adsorption and Degradation of Organic Micropollutants in Granular Activated Carbon Filters.

The removal of organic micropollutants in granular activated carbon (GAC) filters can be attributed to adsorption and biological degradation. These two processes can interact with each other or proceed independently. To illustrate the differences in their interaction, three 14C-labeled organic micropollutants with varying potentials for adsorption and biodegradation were selected to study their adsorption and biodegradation in columns with adsorbing (GAC) and non-adsorbing (sand) filter media. Using 14CO2 formation as a marker for biodegradation, we demonstrated that the biodegradation of poor

Histopathology of methylbenzylnitrosamine-induced esophageal carcinoma in the rat: comparison with cytomorphology.

The incidence of human carcinoma of the esophagus is increased in association with environmental nitrosamines and dietary zinc deficiency. For examination of the role of zinc deficiency in esophageal carcinogenesis, methylbenzylnitrosamine [(MBN) CAS: 937-40-6] was used to induce esophageal carcinoma in rats fed either a zinc-deficient or control diet. Histologic examination of the MBN-treated Sprague-Dawley rat esophagi revealed a significantly higher incidence of esophageal carcinoma in the zinc-deficient rats vs. controls (62 vs. 33%). In addition, three distinct cytologic and histologic pa

Effects of intermediate metabolites of 37 xenobiotics on the catalytic activities of reconstituted cytochrome P-450IIB1 and P-450IA1 enzyme systems.

Direct effects of intermediate metabolites of 37 different xenobiotics on the catalytic activities of both reconstituted cytochrome P-450IIB1 and P-450IA1 enzyme systems were studied by determination of NADPH oxidation at various intervals after initiation of the reaction. The results showed that cytochrome P-450IIB1 isozyme was much more likely than cytochrome P-450IA1 isozyme to be attacked by the reactive intermediates formed by some xenobiotics with smaller molecular weight and lose its catalytic activities. These xenobiotics were carbon tetrachloride, chloroform, carbon disulfide, benzene
