A pharmacokinetic and pharmacodynamic drug-drug interaction study of dorzagliatin and empagliflozin in patients with type 2 diabetes and obesity: an open-label phase I trial.

Combination therapy of glucokinase activators and sodium-glucose cotransporter-2 inhibitors may exhibit potent glucose-lowering effects and improvement of β-cell function and insulin sensitivity in patients with type 2 diabetes mellitus, given their complementary mechanisms of action. However, the safety, pharmacokinetic interactions and pharmacodynamic effects of such combinations remain unexplored. To address this gap, we conduct an open-label, sequential phase I trial (ClinicalTrials.gov, NCT03790787 ) evaluating the dorzagliatin and the sodium-glucose cotransporter-2 inhibitor empagliflozi

Clinical outcome following second-generation drug-eluting stent use for off-label versus on-label indications: insights from the two-year outcome of the TWENTE trial.

Drug-eluting stents (DES) were first used on-label - in simple patients with low clinical risk and easily accessible lesions. Currently, DES are increasingly used off-label - in complex patients undergoing percutaneous coronary interventions (PCI) with historically higher event risk. Therefore, our aim was to investigate whether patients with off-label indications for DES use had similar outcomes compared to patients who were treated for on-label indications only. We analysed two-year follow-up data of 1,387 TWENTE trial patients, treated with second-generation everolimus-eluting XIENCE V or z

Motility-mediated drug interactions with GLP-1 receptor agonists: labeled evidence and a framework for quantitative assessment.

Glucagon-like peptide-1 receptor agonists (GLP-1 RAs) are widely prescribed, and two additional oral products received US approval in 2025 and 2026. All eleven US labels examined describe delayed gastric emptying as potentially affecting the absorption of co-administered oral medications. Since the exenatide program, no study has evaluated a narrow therapeutic index victim across more than one administration schedule; the original warfarin and digoxin studies each used a single schedule. The omission may reflect an interaction paradigm built around enzymes and transporters. Current frameworks

Label-Free SERS Analysis of Glioma Cells to Detect Drug Resistance Using Metabolic Phenylalanine Level.

Glioma is one of the most common malignant brain tumors, and its mainstream clinical treatment regimens mainly include postoperative combined with Temozolomide chemotherapy. Unfortunately, glioma cells tend to mutate when subjected to prolonged Temozolomide treatment, leading to drug resistance and significantly weakening the therapeutic effect. Delaying the resistance time of Temozolomide has become a pressing issue for clinicians to address, and the primary regimen for delaying drug resistance is the combined application of Temozolomide with other therapies. To explore new molecular diagnost

Label-free classification of nanoscale drug delivery systems using hyperspectral imaging and convolutional neural networks.

Label-free characterization of nanoscale drug delivery systems remains a critical challenge in pharmaceutical research. Traditional analytical methods, such as cryo-electron microscopy, are labor-intensive, low-throughput, and often require labeling, which can interfere with nanoparticle functionality. This study introduces a non-invasive hyperspectral imaging (HSI) framework combined with deep learning to classify therapeutic liposomes. A 3D convolutional neural network (3D CNN) was employed to extract spatial-spectral features, while the synthetic minority oversampling technique (SMOTE) addr

Label-Free Analysis Reveals In Vivo Premature Drug Release Governed by Diffusion from Polymeric Micelles.

Premature drug release in the bloodstream significantly limits the clinical use of polymeric micelles (PMs). However, whether this release results primarily from drug diffusion or micelle dissociation is still unclear because current techniques cannot track both the drug and carrier simultaneously and without labels. To address this, we developed an integrated label-free platform combining stable isotope-assisted size-exclusion chromatography, ultrafiltration, and mass spectrometry. This platform enables the absolute quantification of five critical circulatory species: micelle-encapsulated dru

Label-Free Longitudinal Imaging of Single Cell Drug Response with a 3D-Printed Cell Culture Platform.

Image-based phenotypic screening has emerged as a powerful tool for revealing single-cell heterogeneity and dynamic phenotypic responses in preclinical drug discovery. Compared to traditional static end-point assays, live-cell longitudinal imaging captures the temporal trajectories of individual cells, including transient morphological adaptations, motility shifts, and divergent subpopulation behaviors, enabling high content features and more robust early prediction of treatment outcomes. Fluorescence-based screening, while highly specific, is constrained in live-cell contexts by broad spectra

Construction and Preclinical Evaluation of 211At Labeled Anti-mesothelin Antibodies as Potential Targeted Alpha Therapy Drugs.

Targeted alpha therapy (TAT) is a promising tumor therapy that can specifically transport α particle to the vicinity of tumor cells while the normal cells are only slightly irradiated. Mesothelin is a highly promising molecular signature for many types of solid tumors including malignant mesothelioma, pancreatic cancer, ovarian cancer and lung adenocarcinoma etc., while the expression in normal human tissues are limited, thus making mesothelin a promising antigen for TAT. Previously we developed a theoretical model that could predict and optimize in vitro screening of potential TAT drugs. The
