Enhanced transmission at a spinal synapse triggered in vivo by an injury signal independent of altered synaptic activity.

Peripheral nerve crush initiates a robust increase in transmission strength at spinal synapses made by axotomized group IA primary sensory neurons. To study the injury signal that initiates synaptic enhancement in vivo, we designed experiments to manipulate the enlargement of EPSPs produced in spinal motoneurons (MNs) by IA afferents 3 d after nerve crush in anesthetized adult rats. If nerve crush initiates IA EPSP enlargement as proposed by reducing impulse-evoked transmission in crushed IA afferents, then restoring synaptic activity should eliminate enlargement. Optical mapping of spatiotemporal emergence of functional synaptic connections in the embryonic chick olfactory pathway.

In order to understand the functional maturation of the CNS, it is essential to first describe the functional maturation of sensory processing. We have approached this topic by following the ontogenetic patterning of neural circuit formation related to cranial and spinal sensory input using voltage-sensitive dye imaging. In previous studies, we have described the functional maturation of synapses in brainstem/midbrain neural circuits. Here, we elucidate the functional maturation of forebrain circuits by investigating neural networks related to the olfactory nerve (N. I) of chicken embryo. In t

Communication networks in the brain: neurons, receptors, neurotransmitters, and alcohol.

Nerve cells (i.e., neurons) communicate via a combination of electrical and chemical signals. Within the neuron, electrical signals driven by charged particles allow rapid conduction from one end of the cell to the other. Communication between neurons occurs at tiny gaps called synapses, where specialized parts of the two cells (i.e., the presynaptic and postsynaptic neurons) come within nanometers of one another to allow for chemical transmission. The presynaptic neuron releases a chemical (i.e., a neurotransmitter) that is received by the postsynaptic neuron's specialized proteins called neu

Upregulation of brain-derived neurotrophic factor expression in nodose ganglia and the lower brainstem of hypertensive rats.

Hypertension leads to structural and functional changes at baroreceptor synapses in the medial nucleus tractus solitarius (NTS), but the underlying molecular mechanisms remain unknown. Our previous studies show that brain-derived neurotrophic factor (BDNF) is abundantly expressed by rat nodose ganglion (NG) neurons, including baroreceptor afferents and their central terminals in the medial NTS. We hypothesized that hypertension leads to upregulation of BDNF expression in NG neurons. To test this hypothesis, we used two mechanistically distinct models of hypertension, the spontaneously hyperten

Plasticity in an electrosensory system. I. General features of a dynamic sensory filter.

1. In this study we describe changes in neuronal responses within the primary electrosensory processing nucleus of a weakly electric fish that occur when the fish are exposed to repetitive patterns of electrosensory stimuli. Extracellular single-unit recordings show that pyramidal cells within the electrosensory lateral line lobe develop, over a time course of several minutes, an insensitivity to repetitive stimuli applied to a cell's receptive field (local stimulus). The pyramidal cell response cancellation only develops if the local stimulus is applied simultaneously with a diffuse pattern o

Intrinsic excitability mechanisms of neuronal ensemble formation.

Neuronal ensembles are coactive groups of cortical neurons, found in spontaneous and evoked activity, that can mediate perception and behavior. To understand the mechanisms that lead to the formation of ensembles, we co-activated layer 2/3 pyramidal neurons in brain slices from mouse visual cortex, in animals of both sexes, replicating in vitro an optogenetic protocol to generate ensembles in vivo. Using whole-cell and perforated patch-clamp pair recordings we found that, after optogenetic or electrical stimulation, coactivated neurons increased their correlated activity, a hallmark of ensembl

Epilepsy with myoclonus and post-natal development of the motor system in humans: a hypothesis.

Epilepsy with myoclonus is thought to be linked to the motor system. At birth, development of the central nervous system in humans is far from being achieved. Post-natal changes take place at different levels in this neuronal system. These modifications suggest that the motor cortex is a highly dynamic structure during post-natal development. They may account for the age-dependence of various epileptic syndromes. (1) The number of synapses increases during the early post-natal years and then decreases to reach the adult level around puberty. (2) Neurons differentiate and synthesize various neu

The mechanical properties of nerves, the size of the action potential, and consequences for the brain.

The action potential is widely regarded as a purely electrical phenomenon. However, one also finds mechanical and thermal changes that can be observed experimentally. In particular, nerve membranes become thicker and axons contract. The spatial length of the action potential can be quite large, ranging from millimeters to many centimeters. This suggests the use of macroscopic thermodynamics methods to understand its properties. The pulse length is several orders of magnitude larger than the synaptic gap, larger than the distance of the nodes of Ranvier and even larger than the size of many neu

Changes in the sensory processing of olfactory signals induced by birth in sheep.

After giving birth, sheep and many other species form a selective bond with their offspring based on the sense of smell. Processing of olfactory signals is altered to allow the animals to perform this selective recognition. Lamb odors have little effect on either neurotransmitter release or electrical activity of neurons in the olfactory bulb before birth. However, after birth there is an increase in the number of mitral cells, the principal cells of the olfactory bulb, that respond to lamb odors, which is associated with increased cholinergic and noradrenergic neurotransmitter release. Co-existence of blue-sky catastrophe and bistability in a coupled Leech-heart inter-neuron model with both chemical and electrical coupling.

Neuronal activity is characterized by diverse firing patterns that underlie both normal physiological processes and pathological states of the brain. In this study, we explore diverse firing activities, including tonic, bursting and spiking in a coupled Leech Heart inter-neuron model incorporated with both chemical and electrical synapses. These rich dynamics arise due to the emergence of multiple bifurcations, including codimension-one bifurcations (Hopf, saddle-node, and branch point) and codimension-two bifurcations (Bogdanov-Takens and cusp) in the model. Further, due to the changes of che…

Heterojunction nanofluidic memristors based on peptide chain valves for neuromorphic applications.

Memristors exhibit significant potential for neuromorphic computing due to their unique properties. This study introduces a heterojunction nanofluidic memristor (HJNFM) and explores its applications in simulating synapses and constructing neural networks. The HJNFM consists of a SnS2 and MoS2 heterojunction nanochannel with a peptide chain valve. The opening and closing dynamics of peptide chain valve alter ionic conductance of the nanochannel and realize the memristor characteristics. The sequence of the peptide chain also affects the electrical properties of HJNFM. Additionally, by setting u

Does the engram of kindling model the engram of normal long term memory?

The kindling effect is a relatively permanent alteration in brain function which results from repeated electrical or chemical stimulation and culminates in the appearance of electrographic and behavioral convulsions whenever the original stimulus is reapplied. The effect results from tetanic activation in the anterior cortex, limbic system or associated areas of the adult mammalian brain, and the lasting alterations are transynaptic and quite widespread. They are based in part on synaptic facilitation, and they are accompanied by specific alterations in normal behavior. In these and other resp

Methodology for Biomimetic Chemical Neuromodulation of Rat Retinas with the Neurotransmitter Glutamate In Vitro.

Photoreceptor degenerative diseases cause irreparable blindness through the progressive loss of photoreceptor cells in the retina. Retinal prostheses are an emerging treatment for photoreceptor degenerative diseases that seek to restore vision by artificially stimulating the surviving retinal neurons in the hope of eliciting comprehensible visual perception in patients. Current retinal prostheses have demonstrated success in restoring limited vision to patients using an array of electrodes to electrically stimulate the retina but face substantial physical barriers in restoring high acuity, nat

[Auditory neuropathy (auditory neuropathy spectrum disorders): the approaches to diagnostics and rehabilitation].

Auditory neuropathies (auditory neuropathy spectrum disorders, ANSD) may be a consequence of dysfunction of inner hair cells and/or of synapses between these cells and auditory nerve fibers. Another cause of these disorders is supposed to be pathological changes in the auditory nerve itself. The outcome of the rehabilitative treatment of the patients presenting with this disorder depends on the quality of diagnosis and precise location of the pathological process. The present study involved 82 patients with auditory neuropathies. The audiological data obtained in the course of this work were c

Effects of bilirubin on the development and electrical activity of neural circuits.

In the past several decades, bilirubin has attracted great attention for central nervous system (CNS) toxicity in some pathological conditions with severely elevated bilirubin levels. CNS function relies on the structural and functional integrity of neural circuits, which are large and complex electrochemical networks. Neural circuits develop from the proliferation and differentiation of neural stem cells, followed by dendritic and axonal arborization, myelination, and synapse formation. The circuits are immature, but robustly developing, during the neonatal period. It is at the same time that

Temporally Correlated Stochastic Spiking Neural Networks Enabled by 2D Ferroelectric Synapse-Neuron Integration.

Stochastic neural computation is essential for probabilistic inference and brain-inspired information processing. However, most hardware stochastic neurons generate statistically independent spikes and fail to reproduce the temporal correlations observed in biological neurons. We demonstrate an integrated hardware synapse-neuron consisting of a FeFET synapse and a CulnP2S6 neuron, where the neuron exhibits temporally correlated stochastic spiking. The spike probability is continuously tuned by synaptic weight through voltage modulation, enabling hardware stochastic neural computation. Electric

Room-Temperature Skyrmionic Synapse in 2D Ferromagnet Fe3GaTe2 Operating via Collective Spin Texture Transformation.

Magnetic skyrmions, as topologically protected spin textures, hold great potential for energy-efficient neuromorphic systems. While artificial synapses have been demonstrated in magnetic multilayers by electrically controlling skyrmion populations, their probabilistic nucleation severely limits reliability. The recent emergence of 2D van der Waals magnets, with their inherent tunability and novel spintronic phenomena, offers a promising platform to overcome these challenges. Here, we demonstrate an artificial synaptic device in 2D ferromagnet Fe3GaTe2, operating on the fundamentally different

Molecular Cocrystal-Based Neuromorphic Vision System With Near-Infrared Responsivity and High Electronic Performance for Facial Recognition in Scattering Media.

Neuromorphic visual systems are core technologies enabling round-the-clock perception for the Internet of Things (IoT). While reliable sensing in complex lighting conditions requires robust near-infrared (NIR) capabilities, existing optoelectronic devices struggle to simultaneously achieve a broad NIR spectral response and high electronic performance. This limitation severely hinders their practical applications in anti-interference imaging and intelligent recognition. Here, high-performance organic photonic synaptic transistors (OPSTs) are reported, which employ a C8-BTBT channel layer and a

Implementation of Highly Stable Memristive Characteristics in an Organic-Inorganic Hybrid Resistive Switching Layer of Chitosan-Titanium Oxide with Microwave-Assisted Oxidation.

This study proposes a high-performance organic-inorganic hybrid memristor for the development of neuromorphic devices in the memristor-based artificial synapse. The memristor consists of a solid polymer electrolyte (SPE) chitosan layer and a titanium oxide (TiOx) layer grown with a low-thermal-budget, microwave-assisted oxidation. The fabricated Ti/SPE-chitosan/TiOx/Pt-structured memristor exhibited steady bipolar resistive switching (BRS) characteristics and demonstrated excellent endurance in 100-cycle repetition tests. Compared to SPE-chitosan memristors without a TiOx layer, the proposed o
