Heart failure: advanced development in genetics and epigenetics.

Heart failure (HF) is a complex pathophysiological syndrome that arises from a primary defect in the ability of the heart to take in and/or eject sufficient blood. Genetic mutations associated with familial dilated cardiomyopathy, hypertrophic cardiomyopathy, and arrhythmogenic right ventricular cardiomyopathy can contribute to the various pathologies of HF. Therefore, genetic screening could be an approach for guiding individualized therapies and surveillance. In addition, epigenetic regulation occurs via key mechanisms, including ATP-dependent chromatin remodeling, DNA methylation, histone m

Cracking the fertility code: a comprehensive review of biomarkers and functional measures.

Human fertility is regulated by a complex, integrated network of biological systems that collectively determine reproductive competence in males and females. Conventional fertility assessment relies on established clinical biomarkers, including reproductive hormone profiles, gamete-based functional assessments and imaging indicators of gonadal structure and function. While clinically informative, these measures capture only partial views of molecular and cellular processes underlying gametogenesis and reproductive capacity. Recent advances in reproductive medicine have expanded the biomarker l

Epigenetic processes in Parkinson disease.

Numerous investigations performed over the last 20 years revealed various epigenetic modifications in both postmortem brain samples and in peripheral tissues of individuals with Parkinson disease (PD), as well as in neuronal and animal models of the disease. These include changes in DNA methylation and hydroxymethylation, histone-tail modifications, and a dysregulated expression of both short- and long-noncoding RNA molecules. Overall, there is no doubt that epigenetic modifications occur within the neurodegenerative process linked to PD, resulting in altered expression of PD-linked genes. How

When to Trust Epigenetic Clocks: Avoiding False Positives in Aging Interventions.

Epigenetic clocks have emerged as potential biomarkers of aging due to their association with morbidity and mortality. They have recently gained traction as outcome measurements for anti-aging interventions. However, as the number of new epigenetic clocks continues to grow, different clocks may not agree on the effect of an intervention. This raises the question of when researchers can trust epigenetic clock results. In this study, we find that when extending previously published single-clock intervention studies to include additional clock models, no additional clocks reach statistical signif

Oncogenic role of ERV with therapeutic potential.

Endogenous Retroviruses (ERVs) are originated from ancient exogenous viruses, which integrate into the host genome after infection and persist across all vertebrate lineages. Complete ERV consists of viral genes (gag, pro, pol, and env) in the center and two Long Terminal Repeats (LTRs) at both ends, which encode viral structural proteins, critical enzyme, and glycoprotein. ERVs constitute approximately 8% of human genome and function in a wide range of physiological and pathological processes, including embryonic development, inflammation and infection, neurodegenerative diseases, cancers, et

Epigenetic regulation of non-apoptotic regulated cell death.

For decades, apoptosis has reigned supreme in cell death therapeutics, yet its clinical limitations in resistant cancers and degenerative diseases have unveiled the critical role of non-apoptotic regulated cell death (RCD) pathways, including ferroptosis, necroptosis, pyroptosis, and parthanatos. This review examines chromatin as a key regulatory layer influencing these pathways through dynamic histone modifications, DNA methylation, non-coding RNAs, and 3D genome architecture. We dissect how chromatin landscapes integrate metabolic, oxidative, and inflammatory signals in a cell-type- and line

Epigenetic modulation by essential oils: Potential applications and challenges in palliative care.

Essential oils have long been used in complementary medicine, and recent research has demonstrated their potential to modulate epigenetic mechanisms such as DNA methylation, histone modification and microRNA regulation. These epigenetic effects suggest that essential oils may offer novel therapeutic benefits in palliative care, which primarily focus on symptom management, improvement of quality of life, and support for emotional well-being in patients with life-limiting conditions. This review examined the epigenetic mechanisms by which key compounds in essential oils, such as curcumin, eugeno

Revolutionizing Diabetic Foot Ulcer Management: Exploring the Therapeutic Potential of Natural Products.

Diabetic foot ulcer (DFU) is one of the most common serious complications of diabetes mellitus and often results in amputation. This complication affects 15% of individuals with diabetes over their lifetime. This review investigates the role of epigenetic modifications in DFUs, as many epigenetic mechanisms contribute to the underlying pathogenesis of diabetes and its complications, including DFUs. Therefore, initiatives aimed at understanding these epigenetic mechanisms may represent a paradigm shift in diagnosing and treating DFUs. Molecular-level studies have been conducted to identify the

From Genes to Lives: Integrating the Complexities of Primary Ovarian Insufficiency.

Primary ovarian insufficiency (POI) affects up to 3% of reproductive-aged women and is a critical yet underrecognized contributor to infertility and systemic accelerated aging. While most cases remain idiopathic, advances in genomics increasingly reveal a genetic basis, implicating pathways that govern DNA repair, meiosis, chromosomal stability, and folliculogenesis. This review synthesizes the multifactorial etiology of POI, integrating genetic contributions with emerging evidence on epigenetic dysregulation, mitochondrial dysfunction, and environmental influences such as toxins and lifestyle

Neuroendocrine tumours through an epigenetic lens: Emerging insights for diagnosis and treatment.

Neuroendocrine tumours (NETs) are well-differentiated epithelial neuroendocrine neoplasms that frequently develop in the small intestine, pancreas, and lungs. NETs originate from neuroendocrine cells specialized in hormone secretion implicated in a number of physiological processes. Their malignant transformation is characterized by low mutational burden, suggesting that epigenetic mechanisms may be at play. Recent understanding of epigenetic events driving cancer cell plasticity and tumour initiation has led to advances in the diagnosis and prognosis of NETs. Here, we provide a brief overview

Molecular profile of Cushing's syndrome.

Cushing's syndrome comprises a heterogeneous group of disorders characterized by chronic excess of glucocorticoid release from adrenal glands, caused by altered adrenal function or by ACTH hypersecretion from pituitary or ectopic sources. Advances in high-throughput molecular profiling have substantially refined our understanding of the mechanisms underlying endogenous hypercortisolism. This chapter provides an integrated overview of the genetic, transcriptomic, and epigenetic alterations driving both ACTH-dependent and ACTH-independent forms of the disease. Contrasting epigenetics of Ixodes scapularis populations.

Hard ticks are a source of public health concern, in part due to their ability to inhabit different environments. In the United States (US), blacklegged ticks (Ixodes scapularis Say), the primary vector of Lyme disease, exhibit various phenotypes depending on their geographic origin (i.e. northern and southern US ticks). Although genetics may partially explain how blacklegged tick populations acclimate to different environmental conditions across the US, epigenetics may also contribute to their success. Epigenetic mechanisms, such as DNA methylation, might modulate gene expression allowing for

Genetics and Molecular Mechanisms in Oral Squamous Cell Carcinoma: A Narrative Review.

Oral squamous-cell carcinoma (OSCC) is the most common form of oral cancer, accounting for over 90% of malignancies in the oral cavity. Its pathogenesis is driven by a complex interplay of genetic alterations, transcriptomic dysregulation, epigenetic modifications, environmental exposures, and tumor microenvironment dynamics. Despite advances in therapy, OSCC remains associated with poor survival due to late diagnosis, therapeutic resistance, and tumor heterogeneity. This narrative review explores genetic determinants and molecular mechanisms underlying OSCC, focusing on recurrent mutations, d

Epigenetic regulation of iron metabolism and ferroptosis in Parkinson's disease: Identifying novel epigenetic targets.

Parkinson's disease (PD) is a neurodegenerative disease, and emerging evidence has shown that iron deposition, ferroptosis and epigenetic modifications are implicated in the pathogenesis of PD. However, the interplay among these factors in PD has not been fully understood. In this review, we provide an overview of the current research progress on iron metabolism, ferroptosis and epigenetic alterations associated with PD. Furthermore, we present new frontiers concerning various epigenetic modifications related to iron metabolism and ferroptosis that might contribute to the pathology of PD. Nota

Epigenetic factors associated with peri-implantitis: a review.

Peri-implant diseases are characterized by the resorption of hard tissue and the inflammation of soft tissue. Epigenetics refers to alterations in the expression of genes that are not encoded in the DNA sequence, influencing diverse physiological activities, including immune response, inflammation, and bone metabolism. Epigenetic modifications can lead to tissue-specific gene expression variations among individuals and may initiate or exacerbate inflammation and disease predisposition. However, the impact of these factors on peri-implantitis remains inconclusive. To address this gap, we conduc
