Oviduct cells express the cyclic AMP-adenosine pathway.

The extracellular cAMP-adenosine pathway refers to the local production of adenosine mediated by cAMP egress into the extracellular space, conversion of cAMP to AMP by ectophosphodiesterase (PDE), and the metabolism of AMP to adenosine by ecto-5'-nucleotidase. The goal of this study was to assess whether the cAMP-adenosine pathway is expressed in oviduct cells. Studies were conducted in cultured bovine oviduct cells (mixed cultures of fibroblasts and epithelial cells, 1:1 ratio). Confluent monolayers of oviduct cells were exposed to cAMP (0.01-100 micromol/L) in the presence and absence of 3-i…

Typhoid toxin sorting and exocytic transport from Salmonella Typhi-infected cells.

Typhoid toxin is an essential virulence factor for Salmonella Typhi, the cause of typhoid fever in humans. This toxin has an unusual biology in that it is produced by Salmonella Typhi only when located within host cells. Once synthesized, the toxin is secreted to the lumen of the Salmonella-containing vacuole from where it is transported to the extracellular space by vesicle carrier intermediates. Here, we report the identification of the typhoid toxin sorting receptor and components of the cellular machinery that packages the toxin into vesicle carriers, and exports it to the extracellular sp…

Comparison between aerobic and anoxic metabolism of denitrifying-EBPR sludge: effect of biomass poly-hydroxyalkanoates content.

Biomass with denitrifying phosphate uptake ability was tested under sequencing anaerobic-aerobic and anaerobic-anoxic conditions. The initial dose of acetate, under anaerobic conditions varied to achieve different PHA (poly-hydroxyalkanoates) saturation of PAO (polyphosphate accumulating organisms) cells. Increased acetate dosage under anaerobic conditions led to higher phosphate release and increased PHA storage by PAOs and, also, to greater phosphate uptake rates under the following aerobic and/or anoxic conditions. The experimental results also indicated that when organic carbon is limited

The ultrastructural basis of periventricular edema: preliminary studies.

The pathophysiology of periventricular lucency (PVL) on CT in primates was evaluated by transmission (TEM) and scanning electron microscopy (SEM). At 100 days after Silastic-induced hydrocephalus, six primate brains showed loss of cilia and ependymal cells over the ventricular surface and abnormal small supraependymal cells. TEM demonstrated communicating pathways between the ventricular lumen and brain parenchyma and the extracellular space was markedly increased. At 1000 days, ependymal changes in two brains were noted at greater distances from the ventricular angle. This study suggests that…

Oral hairy leukoplakia: ultrastructural features.

Ten instances of a white plaque of the lateral tongue unique to homosexual males and referred to as oral hairy leukoplakia were analysed ultrastructurally. The surface epithelial layer exhibited extracellular, intracellular and intranuclear penetration by hyphae of Candida albicans, sometimes accompanied by coccobacilli in the extracellular space. The subcorneal epithelial layer included koilocytoid ballooned cells which had a paucity of cytoplasmic organelles and displayed condensation and emargination of the chromatin. Cells that exhibited these nuclear changes were found to be infected by a…

Extracellular Fluorescence Recording of BacFlash Dynamics Reveals Two Distinct Modes of Proton Extrusion.

BacFlash is a recently described transient bacterial process characterized by synchronized intracellular alkalization, membrane depolarization, and reactive oxygen species (ROS) generation, driven by regulated proton efflux. This phenomenon reveals a previously unrecognized mode of rapid physiological regulation in bacteria. Current techniques for studying BacFlash dynamics rely on intracellular fluorescent labeling with organic dyes or fluorescent proteins sensitive to pH, membrane potential, or ROS. Here, we introduce an extracellular, label-free fluorescence imaging strategy for real-time m

Quantifying the relationship between spreading depolarization and perivascular cerebrospinal fluid flow.

Recent studies have linked spreading depolarization (SD, an electro-chemical wave in the brain following stroke, migraine, traumatic brain i … g is modelled using an empirical relationship between the excess potassium ion concentration in the extracellular space following SD and the vessel radius. We find that the CSF volumetric flow rate depends intricately on the length and width of the PVS, as well as the vessel radius and the angle of incidence of the SD wave. We derive analytical expressions for pressu …

Electrochemical Study on the Extracellular Electron Transfer Pathway from Shewanella Strain Hac319 to Electrodes.

Shewanella can transfer electrons to various extracellular electron acceptors. We electrochemically investigated the pathway of extracellular electron transfer from Shewanella strain Hac319 to electrodes. A resting cell suspension of Shewanella strain Hac319 containing lactate produced a steady-state sigmoidal wave in the presence of flavin mononucleotide (FMN) in cyclic voltammetry, but not in the absence of FMN. A harvested cell suspension without cell-washing also produced a similar catalytic wave without any external addition of free FMN. The midpoint potentials of the two sigmoidal waves

Raffinose Induces Biofilm Formation by Streptococcus mutans in Low Concentrations of Sucrose by Increasing Production of Extracellular DNA and Fructan.

Streptococcus mutans is the primary etiological agent of dental caries and causes tooth decay by forming a firmly attached biofilm on tooth surfaces. Biofilm formation is induced by the presence of sucrose, which is a substrate for the synthesis of extracellular polysaccharides but not in the presence of oligosaccharides. Nonetheless, in this study, we found that raffinose, which is an oligosaccharide with an intestinal regulatory function and antiallergic effect, induced biofilm formation by S. mutans in a mixed culture with sucrose, which was at concentrations less than those required to ind

Articular cartilage vesicles and calcium crystal deposition diseases.

PURPOSE OF REVIEW: Articular cartilage vesicles (ACVs) are small extracellular vesicles that serve as foci of pathologic calcium crystal deposition in articular cartilage matrix. In this review, I have summarized the role of ACVs in calcium crystal formation and discuss recent findings that impact our understanding of the content, behavior, and origin of ACVs in healthy and diseased joints. The burgeoning interest in extracellular vesicles in other fields renders this a timely and relevant topic. RECENT FINDINGS: I have highlighted recent studies demonstrating that some ACVs originate in the

Elevation of amino acids in the interstitial space of the rat brain following infusion of large neutral amino and keto acids by microdialysis: leucine infusion.

A microenvironment similar to that found in maple syrup urine disease was created in the brain of free-moving, awake rats by the infusion of leucine into the brain using microdialysis. To determine the effects on amino acid homeostasis the eluate of the probe was analyzed. Perfusion with leucine elevated the interstitial levels of large neutral amino acids suggesting hetero exchange of large neutral amino acids from neuronal cells into the interstitial space. The data also demonstrated the inter relationship of leucine and glutamine, both of which may be nitrogen sinks in the brain. Elevation

In-vivo microdialysis study of extracellular glutamate response to temperature variance in subarachnoid hemorrhage.

Neurochemical changes may precede the development of clinical signs in neurological disease. Early identification of such changes may offer an opportunity to avoid or treat complications. Under experimental conditions, extracellular levels of glutamate and other amino acids can be monitored by in-vivo microdialysis in cerebral ischemia, head trauma and epilepsy. Data on the release of glutamate under ischemic conditions in humans are limited. There is no published data on the effects of temperature variation or other manipulations on the extracellular glutamate levels in humans. We report for

Pharmacokinetics of antibiotics in natural and experimental superficial compartments in animals and humans.

Transcapillary exchange of antibiotics and other small molecules is diffusion driven and occurs in the capillary beds of the tissues. Small polar molecules are ionized at physiological pH and diffusion is pore-mediated. More lipophilic substances can also leave capillaries by the transcellular route. Splanchnic tissues have fenestrated capillary walls while somatic tissues have mainly micropores in their capillary walls. Under normal physiological conditions the ratio of capillary surface area to volume of fluid present (SA/V) is very large (> 100) and the rate of exchange of substances betwee

Allopurinol does not enhance antioxidant properties of extracellular fluid.

Allopurinol has been shown to provide significant protection against ischemia/reperfusion-induced microvascular and parenchymal cell injury. It has been hypothesized that the protection seen with allopurinol after ischemia/reperfusion (I/R) is caused by inhibition of xanthine oxidase. However, recent reports suggest that the beneficial effects of allopurinol in I/R may be caused by direct free radical scavenging. The objective of this study was to determine whether the regimen of allopurinol administration used in most I/R studies leads to a significant modification of the free radical scaveng

The blood-uterine lumen barrier and exchange between extracellular fluids.

The concept of a blood-tissue barrier defines the rates at which matter exchanges among the vascular and extravascular fluids of the tissue. The remarkably slow rates at which substances such as mannitol (Mr 182) enter uterine fluid from plasma demonstrate the existence of a blood-uterine lumen barrier. Available evidence indicates that the uterine microvascular endothelium and the uterine epithelium behave as lipoid layers interrupted by water-filled channels. Furthermore, both cell layers appear to select actively certain substances over others for exchange with opposing extracellular fluids

Lung liquid and protein exchange: the four inhomogeneities.

William of Ockham, 14th-century scholastic philosopher at Oxford and Munich, emphasized the principle of economy, "pleurality is not to be supposed without necessity" (Ockham's razor). Necessity is the key word. In the modeling of steady-state lung liquid and protein exchange, the desire for simplicity has sometimes outweighed good judgment. In fact, we and others have shown that simple models do not work. It is necessary to include several forms of inhomogeneity. The air-filled lung shows regional (top to bottom) variations of mass, microvascular pressure, and perimicrovascular protein concen

Evaluation of extravascular lung water by single thermal indicator.

Much attention has been directed to developing early therapy that might prevent or ameliorate incipient adult respiratory distress syndrome (ARDS) before respiratory failure ensues. The relationship between increased extravascular lung water (EVLW) and pulmonary microvascular pressure has been used by numerous investigators to confirm alterations in the permeability of the alveolar capillary membrane. In the present study, EVLW was measured in 40 critically ill patients by an indicator dilution technique utilizing thermal indicator alone. Injection of cold saline distinguished intravascular an

Pathophysiology of shock-induced disturbances in tissue homeostasis.

A survey is given on disturbances in tissue homeostasis induced by hypovolemic shock conditions. Fluid shifts taking place between the extra- and intravascular fluid compartments are important early compensatory responses following hypovolemia. Usually the supply-to-demand ratio of oxygen in most tissues can however, not be kept up if the hypovolemic insult is severe due to deterioration of the microcirculation. Cellular hypoxia will ensue and may with time affect the integrity of the cells. Cellular functional disturbances occur earlier and are more pronounced in peripheral non-vital tissues
