Developing Novel Tests to Screen for Fetal Growth Restriction.

Fetal growth restriction (FGR) is a major determinant of global morbidity and mortality. There is an unmet need for methods to stratify the pregnant population on the basis of FGR risk. Despite evolutionary divergence in mammalian reproduction, studies of genetically modified mice have identified biomarkers that have been validated in women, and a systematic screen for genes that control fetal growth in animals could help identify novel clinical biomarkers. Current approaches to biomarker identification using human samples include both targeted and discovery approaches (omics). Application of

Embryonic and fetal development of the collared peccary (Pecari tajacu).

The relative timing of the main fetal development events in species determine the extent of fetal development at birth, which range along a gradient of having altricial and precocial traits. The results from this study allow for description of important fetal developments in collared peccary (Pecari tajacu) using data from 118 embryo/fetuses from 68 pregnant peccaries obtained over a period of 15 years through collaborative methods with local hunters in the Amazon. The chronological order of emergence of external characteristics in relation to the total dorsal length (TDL) was: differentiated …

Effect of metronidazole on placental and fetal development in albino rats.

Metronidazole is an antiprotozoal and antibacterial used in gynecology and obstetrics for the treatment of parasitic infections. However, despite having clinical use for more than three decades, questions about the safety of its use during pregnancy is not well understood. Thus, the present study evaluated the effect of metronidazole on placental and fetal development in pregnant rats. Metronidazole was orally administered by gavage at a dosage of 130 mg/kg for 7 and 14 days. Morphological analysis, morphometry and immunohistochemistry were performed at the implantation sites and placentas wit…

Calciotropic and phosphotropic hormones in fetal and neonatal bone development.

There are remarkable differences in bone and mineral metabolism between the fetus and adult. The fetal mineral supply is from active transport across the placenta. Calcium, phosphorus, and magnesium circulate at higher levels in the fetus compared to the mother. These high concentrations enable the skeleton to accrete required minerals before birth. Known key regulators in the adult include parathyroid hormone (PTH), calcitriol, fibroblast growth factor-23, calcitonin, and the sex steroids. But during fetal life, PTH plays a lesser role while the others appear to be unimportant. Instead, PTH-r

The role of the ureteric bud in the development of the ovine fetal kidney.

BACKGROUND: The kidney develops from an intimate interaction between the ureteric bud and the metanephric mass. We attempted to differentially stain the derivatives of the ureteric bud and the metanephric mass in ovine fetuses. METHODS: After appropriate approval, 47 fetal lambs' kidneys at 50 (4), 60 (6), 70 (5), 80 (4), 100 (10), 110 (8), 145 (10) days' gestation (term is 140-145 days) were obtained. After confirming the pregnancy, the sheep were anesthetized, and the fetuses sacrificed. The fetal kidneys were prepared for histological examination, using immunostaining for β-catenin, Lamini

Ultramorphological Characteristics of Podocyte Development in the Human Fetal Metanephros.

The aim of this study was to determine the developmental characteristics of podocytes in the human fetal metanephros using scanning electron microscopy, light microscopy, and transmission electron microscopy. Kidney samples of 15 human fetuses of both sexes (gestational age 10-22 weeks) were analyzed. At the S-shaped body stage, primitive podocytes were arranged in a layer of cuboidal cells beneath the vascular cleft. When observed from Bowman's space, the demarcation between adjacent podocytes was not clear, but mild depressions indicated cell boundaries. At the more advanced S-shaped body st

Effects of maternal anxiety and depression on fetal neuro-development.

BACKGROUND: Fetal development is affected by maternal mental health with research indicating that maternal anxiety and depression are co-morbid; nevertheless differential effects on the fetus have been found. This study examines, prenatally, effects of maternal stress, anxiety and depression on fetal eye-blink reactions to experimental sound and light stimulation. METHODS: Two groups of singleton fetuses (mean 32-weeks gestation) were examined using 4D ultrasound: a control group (N = 14, 7 female) with no stimulation and an experimental group (N = 21, 13 female) exposed to experimental sound…

Impacts of arginine nutrition on embryonic and fetal development in mammals.

Embryonic loss and intrauterine growth restriction (IUGR) are significant problems in humans and other animals. Results from studies involving pigs and sheep have indicated that limited uterine capacity and placental insufficiency are major factors contributing to suboptimal reproduction in mammals. Our discovery of the unusual abundance of the arginine family of amino acids in porcine and ovine allantoic fluids during early gestation led to the novel hypothesis that arginine plays an important role in conceptus (embryo and extra-embryonic membranes) development. Arginine is metabolized to orn

Significance of IGFBP-4 in the development of fetal growth restriction.

BACKGROUND: Fetal growth restriction (FGR) is a leading cause of perinatal mortality and morbidity. Animal studies suggest dysregulation of IGF-binding protein (IGFBP)-4 is significant in the development of FGR, although human data are lacking. We postulated that IGFBP-4 is expressed at the maternal fetal interface and plays a role in regulating IGF bioavailability. Thus, maternal serum levels of IGFBP-4 may be associated with complications of abnormal placental growth and development including FGR. METHODS: Circulating levels of IGFBP-4 and its protease, pregnancy-associated plasma protein-A

Effects of intrauterine infection or inflammation on fetal lung development.

1. Intrauterine infection or inflammation is common in cases of preterm birth. Preterm infants are at risk of acute respiratory distress as a result of lung immaturity; evidence of exposure to infection and/or inflammation before birth is associated with a reduced risk of neonatal respiratory distress syndrome (RDS). Experimentally induced intrauterine inflammation or infection in sheep causes a precocious increase in pulmonary surfactant in the preterm lungs that improves preterm lung function, consistent with the reduced risk of RDS in human infants exposed to infection and/or inflammation b

Development of external genitalia in fetal and neonatal domestic cats.

Development of the external genitalia of fetal and neonatal cat were studied macroscopically, paying attention to the formation of the labia and the sexual differentiation. The female urogenital folds budded from each side of the genital tubercle and, gradually extended to the tip of the genital tubercle by the 6.8 cm stage in crown-rump length. Then, the well-developed urogenital folds ensheathed completely the genital tubercle to form the prepuce of clitoris and the labia, flanking the external opening of vagina as the folds of skin which were equivalent to the labia minora in humans. The ge

Effect of Ascaris trypsin inhibitor on fetal development of mice.

In this study, maternal toxicity and developmental effects of exposure to Ascaris trypsin inhibitor were evaluated in mice. Pregnant BALB/c females were injected intraperitoneally by Ascaris inhibitor /AIT/ at 200, 300, and 400 mg/kg body weight/day, on days 12 to 15 of gestation (stage of fetal development). At day 19 of pregnancy, uterine contents were inspected for implantation sites, early resorptions (moles), living fetuses and dead fetuses. The living fetuses were weighed and examined for external, internal and skeletal abnormalities. The results showed that AIT induced maternal toxicity…

Effect of Ascaris chymotrypsin inhibitor on fetal development of mice.

BALB/c mice were given daily doses of 40-80 mg of Ascaris chymotrypsin inhibitor /AChI/ per kg body weight from 12th until 15th day of gestation (stage of fetal development). It has been found that injection of AChI disturbed the course of pregnancy (bleeding from uterus, abortions, decreased body weight gain as compared to control /p < 0.05/). AChI exhibited embryotoxic effects (a high rate of intrauterine deaths, decreased number of living fetuses and mean fetal weight, delayed skeletal ossification, induced pathological changes of fetal organs and tissues). Congenital malformation (hydronep…

In utero development of fetal thirst and appetite: potential for programming.

Thirst and appetite-mediated ingestive behavior develop and are likely programmed in utero, thus preparing for newborn and adult ingestive behavior. Fetal swallowing activity is markedly different from that of the adult, as spontaneous fetal swallowing occurs at a markedly (six-fold) higher rate compared with spontaneous adult drinking activity. This high rate of fetal swallowing is critical for the regulation of amniotic fluid volume and the development of the fetal gastrointestinal tract. Disordered fetal swallowing has been associated with both a decrease (oligohydramnios) and increase (pol

[Fetal development and postnatal maturation of the longus colli muscle].

In adults, the predominant expression of a slow phenotype in the m. longus colli corresponds to its important postural function. Morphologically, there is a dispersion in fiber size predominating on the fast type 2 fibers which are significantly smaller than the slow type 1 fibers. We deemed it of interest, therefore, to analyze the metabolic differentiation of the muscle longus colli during its development. This study has been carried out on six anatomical samples, in foetuses aged between 16 and 40 weeks of pregnancy and in an 18 month-old child. The histological study combined H&E staining

Development of the cavernous sinus in the fetal period: a morphological study.

The development and morphological structure of the lateral sellar compartment (LSC), an interdural space containing the cavernous sinus (CS), cranial nerves, and internal carotid artery (ICA), was investigated by histological examination of sections of the LSC and cerebral venograms from human fetal specimens. Twenty-eight LSC coronal sections were obtained from 14 fetuses of 13-32 weeks' gestation. Venograms of 11 other fetuses of 13-32 weeks' gestation were studied to observe changes in venous drainage. The CS appeared as a collection of small venous canals with an endothelial layer. These v

Development of the human fetal visceral pleura. An ultrastructural study.

The visceral pleura of human fetuses aged from 9 to 36 weeks of gestation was studied by means of transmission electron microscopy. The main components of the visceral pleura (mesothelium, basal lamina and submesothelial connective tissue layer) are formed in the fetal period. They develop asynchronously in different pleural areas, and do not reach maturity. Fetal differentiation of the lung pleura can be divided in two stages--early (until 17 gestation week) and late stage--up to birth. The high mesothelial cells appear later than the flat cells, but the first type predominates in the final c

The early development of neurosonology: II. Fetal and neonatal echoencephalography.

The obscuring effect of the diploe in the skull for attempts to use ultrasonic energy to image the brain is not present in the fetus, neonate or infant prior to ossification of the skull. Initially, however, the fetal examination was largely confined to a single measurement of the size of the skull, usually in the bi-parietal plane, just prior to labour. In this way it became possible to identify immature fetuses where delivery should be delayed as long as possible, as well as cases of cephalo-pelvic disproportion where the large size of the fetal head would make vaginal delivery difficult.
