Absorption, metabolism and health effects of dietary flavonoids in man.

Flavonoids are polyphenolic compounds that occur ubiquitously in foods of plant origin. Over 4,000 different flavonoids have been described, and they are categorized into flavonols, flavones, catechins, flavanones, anthocyanidins and isoflavonoids. Flavonoids have a variety of biological effects in numerous mammalian cell systems, in vitro as well in vivo. Recently, much attention has been paid to their antioxidant properties and to their inhibitory role in various stages of tumour development in animal studies. Quercetin, the major representative of the flavonol subclass, is a strong antioxid

The Role of Flavonoids in Alleviating Mammary Gland Inflammation: A Review.

Mammary gland inflammation, clinically expressed as mastitis in dairy ruminants and lactating women, arises from a self-amplifying loop of pathogen sensing, oxidative stress and innate immune activation, and remains a leading driver of antibiotic use, milk-quality loss and morbidity. Mounting pressure to reduce antimicrobial dependence has refocused attention on flavonoids-structurally diverse plant polyphenols with multi-target bioactivity, derived mainly from in vitro and rodent mastitis models. This review integrates contemporary evidence on the six principal flavonoid subclasses within a u

Classification, Structure-Activity Relationship, and Anti-Osteoarthritis Mechanism of Natural Flavonoids.

Osteoarthritis is a chronic degenerative joint disease. Naturally derived flavonoids have become a hot spot in the field of osteoarthritis drug research and development. This paper summarizes the classification and chemical structure characteristics of flavonoids and the mechanism of anti-osteoarthritis. The key structure-activity relationship (SAR) found that the hydroxyl group of o-diphenol/o-triphenol on the B-ring can scavenge free radicals and chelate pro-oxidant metal ions, which directly determines the antioxidant strength; the C2═C3 double bond on the C-ring is conjugated with the 4-ca

Flavonoids for MASLD: Hepatic Lipid Targets, Biopharmaceutic Barriers, and Formulation Strategies.

Metabolic dysfunction-associated steatotic liver disease (MASLD) develops when hepatic lipid acquisition and synthesis exceed the capacity for oxidation and very-low-density lipoprotein export. Flavonoids act on several components of this network, yet their therapeutic development is constrained by poor aqueous solubility, extensive intestinal and first-pass metabolism, variable activity of circulating metabolites, and limited information on hepatic exposure. Experimental studies link representative flavonoids to AMPK-SREBP-1c and PPARα signaling, mitochondrial quality control, Nrf2-dependent

Elastase inhibition by natural flavonoids: mechanistic insights and potential therapeutic applications.

Overproduction of elastase plays an important role in the progression of inflammatory diseases. In this study, we compared the inhibitory effects of structurally similar bioactive flavonoids (quercetin, hyperoside, luteolin, and luteoloside) on elastase activity and elucidated their mechanisms of action. Enzyme inhibition assays and fluorescence, ultraviolet-visible (UV-vis), Fourier transform infrared (FT-IR), and circular dichroism (CD) spectroscopy examinations assessed the interactions among flavonoids, elastase, and elastase conformational changes. Molecular docking analyzed binding inter

Current insights and future perspectives of flavonoids: A promising antihypertensive approach.

Hypertension, or high blood pressure (BP), is a complex disease influenced by various risk factors. It is characterized by persistent elevation of BP levels, typically exceeding 140/90 mmHg. Endothelial dysfunction and reduced nitric oxide (NO) bioavailability play crucial roles in hypertension development. L-NG-nitro arginine methyl ester (L-NAME), an analog of L-arginine, inhibits endothelial NO synthase (eNOS) enzymes, leading to decreased NO production and increased BP. Animal models exposed to L-NAME manifest hypertension, making it a useful design for studying the hypertension condition.
