The role of oxygen free radicals in postasphyxia cerebral hypoperfusion in newborn lambs.

Previous work in a neonatal lamb model has demonstrated abnormalities in cerebral blood flow (CBF) and oxygen consumption (CMRO2) after asphyxia. Immediately after resuscitation, there was a marked increase in CBF and a significant decrease in CMRO2 compared to control. During the late period after asphyxia (30 min to 4 h), both CBF and CMRO2 were significantly depressed. The same postasphyxia model (n = 16) was used to examine the hypothesis that generation of oxygen free radicals during cerebral reperfusion may be involved in the genesis of late postasphyxia hypoperfusion and depressed CMRO2…

Role of oxygen-derived free radicals in diet-induced hemorrhagic pancreatitis in mice.

The role of oxygen-derived free radicals in the pathogenesis of acute pancreatitis was studied by evaluating the effects of catalase, allopurinol, and dimethylsulfoxide on diet-induced acute hemorrhagic pancreatic necrosis in mice. The mortality rate and degree of hyperamylasemia associated with this model of pancreatitis were reduced by catalase but a similar result followed the administration of heat-denatured catalase, suggesting that the apparent protective effect of catalase was not the result of reductions in free radical levels. Neither allopurinol nor dimethylsulfoxide reduced mortalit…

Effects of melanin-induced free radicals on the isolated rat peritoneal mast cells.

Pheomelanin from human red hair (RHM) produces considerably more cellular damage in Ehrlich ascites carcinoma cells when subjected to radiations of wavelength 320-700 nm than eumelanin from black hair (BHM). Irradiation of RHM generated large amounts of superoxide while BHM did not produce detectable amounts of superoxide. The present investigations describe the effects of irradiation of mast cells in the presence of various natural and synthetic melanins. Irradiation of mast cells in the presence of RHM and red hair melanoprotein released large amounts of histamine while BHM and synthetic mel

Free radical trapping as a therapeutic approach to neuroprotection in stroke: experimental and clinical studies with NXY-059 and free radical scavengers.

There is substantial experimental evidence that free radicals are produced in the brain during ischemia, during reperfusion and during intracranial hemorrhage. Removal of pathologically produced free radicals is therefore a viable approach to neuroprotection. Four compounds with free radical scavenging activity (tirilazad, ebselen, edaravone) or free radical trapping properties (NXY-059) have been examined in experimental models of stroke and evaluated clinically as neuroprotective agents. Both experimental and clinical results are reviewed in this article. Ebselen was a modestly effective neu…

Role of hydroxyl radical formation in neurotoxicity as revealed by in vivo free radical trapping.

Reactive oxygen species have been implicated in dopaminergic toxicity caused by 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) and iron. Although MPTP produces a parkinsonian syndrome after its conversion to 1-methyl-4-phenylpyridine (MPP(+)) by type B monoamine oxidase (MAO-B) in the brain, the etiology of this disease remains obscure. MPP(+) is a highly potent dopaminbergic-releasing agents and dopamine (DA) autoxidation catalyzed by iron and oxidative stress may be involved in the pathogenesis of Parkinson's disease. Neuromelanine synthesis from DA produce highly reactive free radicals…

Move beyond free radical theory of aging.

Aging is a gradual pathophysiological process with the loss of functional integrity and gain of disease vulnerability. Mitochondrial DNA is one essential regulatory factor of aging. The canonical free radical theory attributes aging to oxidative damage of mitochondrial DNA caused by free radicals. However, the general validity of this theory has been challenged by incremental evidences, including some seemingly paradoxical reports, which motivates us to substantiate this theory with our current understandings on free radicals and aging. After reviewing this canonical theory and its adapted mod…

Effects of "On-Water" on the Free Radical Polymerization of Methacrylates.

Water-organic interfaces, referred to as "on-water", have recently attracted considerable attention because of their ability to accelerate various organic reactions. In this study, the effects of "on-water" interface on the conventional free radical polymerization of methacrylates were investigated. Polymerizations were carried out under various conditions, including variations in monomer, solvent, reaction time, and stirring speed, to evaluate the effects of heterogeneity and interfacial area at the water-monomer interface. The results clearly indicate that the "on-water" interface plays a si

Free radical-mediated systemic immunity in plants.

Systemic acquired resistance (SAR) is a form of defense that protects plants against a broad-spectrum of secondary infections by related or unrelated pathogens. SAR related research has witnessed considerable progress in recent years and a number of chemical signals and proteins contributing to SAR have been identified. All of these diverse constituents share their requirement for the phytohormone salicylic acid, an essential downstream component of the SAR pathway. However, recent work demonstrating the essential parallel functioning of nitric oxide (NO)-derived and reactive oxygen species (R

Free radical scavengers in anaesthesiology and critical care.

Free radicals are highly reactive and unstable compounds. These highly reactive molecules cause oxidative damage to cellular components such as DNA, proteins and lipids. They play central role in the mechanism of cell injury and cell death. Free radical scavengers either prevent these reactive species from being formed, or remove them before they can damage vital components of the cell. Oxidative stress defines an imbalance in production of oxidizing chemical species and their effective removal by protective antioxidants and scavenger enzymes. Evidence of massive oxidative stress is well estab…

Tin-free radical addition of acyloxymethyl to imines.

Acyloxymethyl radicals were generated from the corresponding iodomethyl esters and successfully underwent addition to the CN bond of N-Ts, N-PMP, and N-Dpp imines by the action of dimethylzinc or triethylborane. Ethyl acetate, toluene, and benzene as well as dichloromethane were suitable solvents. The utility of acyloxymethyl radicals as a hydroxymethyl anion equivalent was highlighted by the facile hydrolysis of the acyloxy moiety of the adducts to give the corresponding amino alcohol derivatives in good to high yield.

Free radical damage in acute nerve compression.

Nerve compression causes injury by local ischemia and direct mechanical distortion. Peripheral nerves in diabetes mellitus are more prone to injury than those of nondiabetics. We sought to determine whether reperfusion-induced, oxygen-derived free radical injury occurs in peripheral nerves subjected to acute compression in normal and chronically diabetic rats. Female Sprague-Dawley rats weighing 250 to 275 g (N = 347) were divided into two groups: normal and streptozocin-induced diabetics. A total of 187 normal and 160 diabetic nerves were analyzed. After 8 weeks of untreated hyperglycemia, th

The syringe nitroxide free radical--Part II.

Some plastic syringes introduce a nitroxide free radical into the fluids they contain. The EPR spectrum shows a triplet with aN = 17.0 +/- 0.1 G in plasma. We report here that this nitroxide/hydroxylamine couple serves as an antioxidant system, whose redox potential appears to be between that of vitamin C and vitamin E. This adventitious antioxidant can alter the course of free radical processes.

Does ibuprofen treatment in patent ductus arteriosus alter oxygen free radicals in premature infants?

UNLABELLED: Introduction Ibuprofen is used widely to close patent ductus arteriosus in preterm infants. The anti-inflammatory activity of ibuprofen may also be partly due to its ability to scavenge reactive oxygen species and reactive nitrogen species. We evaluated the interaction between oxidative status and the medical treatment of patent ductus arteriosus with two forms of ibuprofen. Materials and methods This study enrolled newborns of gestational age ⩽32 weeks, birth weight ⩽1500 g, and postnatal age 48-96 hours, who received either intravenous or oral ibuprofen to treat patent ductus art

Free radicals properties of gamma-irradiated penicillin-derived antibiotics: piperacillin, ampicillin, and crystalline penicillin.

The aim of this work was to determine the concentrations and properties of free radicals in piperacillin, ampicillin, and crystalline penicillin after gamma irradiation. The radicals were studied by electron paramagnetic resonance (EPR) spectroscopy using an X-band spectrometer (9.3 GHz). Gamma irradiation was performed at a dose of 25 kGy. One- and two-exponential functions were fitted to the experimental data, in order to assess the influence of the antibiotics' storage time on the measured EPR lines. After gamma irradiation, complex EPR lines were recorded confirming the presence of a large…

Potential for misidentification of environmentally persistent free radicals as molecular pollutants in particulate matter.

Environmentally persistent free radicals (EPFRs) have been shown to form on the surfaces of various types of transition metal-containing particulate matter (PM), and it has been demonstrated they are capable of initiating adverse health impacts. Following sonification and solvent extraction for chemical analysis, they are partially converted to molecular species. Alcoholic solvents extracted the EPFRs with near 100% efficiency, while nonpolar hydrocarbon solvents exhibited <20% efficiency and dichloromethane exhibited 20-55% efficiency. The extracted radicals reacted in solution to form multip…
