Terfa Safeguards GABAergic Inhibition and Neuronal Excitability in Zebrafish Through a Foxj3-Linked Transcriptional Mechanism.

Neuronal hyperexcitability often results from impaired inhibition; however, the upstream regulators of GABAergic gene programs remain unclear. We identified a role for terfa, the zebrafish ortholog of human TERF2, in maintaining inhibitory tone and network stability. Homozygous terfa mutant larvae exhibit microcephaly, behavioral hyperactivity, and abnormal neuronal calcium dynamics, indicating a hyperexcitable state. Transcriptomic and RT-qPCR analyses showed coordinated downregulation of multiple GABAA receptor subunit genes, accompanied by reduced gad1b reporter activity in the optic tectum

Neuron-specific WDR5 epigenetically upregulates ARID5B to impair GABAergic synaptic transmission and promotes epileptogenesis.

Rationale: Temporal lobe epilepsy (TLE) is a prevalent, drug-resistant neurological disorder that causes severe disability, highlighting the need to identify novel therapeutic targets. Emerging evidence reveals widespread transcriptional dysregulation during epileptogenesis, in which multiple dysregulated genes functionally contribute to disease progression. However, the epigenetic basis of these transcriptomic changes remains poorly characterized. Methods: We established experimental epilepsy models and systematically investigated the remodeling of major histone methylation modifications duri

Distinct effects of volatile and intravenous anaesthetics on presynaptic calcium dynamics in mouse hippocampal GABAergic neurones.

BACKGROUND: General anaesthetics have marked effects on synaptic transmission, but their neuronal and circuit-level effects remain unclear. The volatile anaesthetic isoflurane differentially inhibits synaptic vesicle exocytosis in specific neuronal subtypes, but whether other common anaesthetics also have neurone-subtype-specific actions is unknown. METHODS: We used the genetically encoded fluorescent Ca2+ sensor GCaMP6f to compare the pharmacological effects of isoflurane, sevoflurane, propofol, and ketamine on presynaptic excitability in hippocampal glutamatergic neurones and in hippocampal

Hypoxic preconditioning differentially affects GABAergic and glutamatergic neuronal cells in the injured cerebellum of the neonatal rat.

In this study we examined cerebellar alterations in a neonatal rat model of hypoxic-ischemic brain injury with or without hypoxic preconditioning (Pc). Between postnatal days 7 and 15, the cerebellum is still undergoing intense cellular proliferation, differentiation and migration, dendritogenesis and synaptogenesis. The expression of glutamate decarboxylase 1 (GAD67) and the differentiation factor NeuroD1 were examined as markers of Purkinje and granule cells, respectively. We applied quantitative immunohistochemistry to sagittal cerebellar slices, and Western blot analysis of whole cerebella

GABAergic Inhibition from the Nucleus Tractus Solitarius to Ventrolateral Medulla Phox2b Neurons Modulates Central Respiratory Chemoreflex and Ventilatory Homeostasis.

Central respiratory chemoreception is a vital homeostatic mechanism maintaining arterial blood gas levels. Central respiratory chemoreceptors in the retrotrapezoid nucleus (RTN) and nucleus tractus solitarius (NTS) form interconnected circuits to regulate respiratory homeostasis. We hypothesized that NTS GABAergic neurons (NTSGABA) modulate hypercapnic ventilatory responses by targeting ventrolateral medulla Phox2b neurons (VLMPhox2b), particularly Phox2b-expressing RTN neurons. Stimulation of NTSGABA neurons significantly attenuated CO2-evoked ventilatory responses, accompanied by a reduction…

GABAergic Gbx1 neurons of the superficial dorsal horn are critical elements of a spinal circuit for stress-induced analgesia.

Acute stress is a powerful inducer of endogenous analgesia. Several brainstem and midbrain areas have been identified that are activated dur … rachial nucleus, a key area in supraspinal pain relay. Our results thus identify a subpopulation of GABAergic neurons in the superficial dorsal horn as key elements of a disinhibitory circuit for stress-induced analgesia.

Ephrin-B2 deletion in GABAergic neurons induces cognitive deficits associated with single-nucleus transcriptomic differences in the prefrontal cortex.

BACKGROUND: Ephrin-B2 (EB2) signaling plays a crucial role in regulating memory and synaptic plasticity. Comprehensive identification of cell-type-specific transcriptomic changes in EB2 knockout mice is expected to shed light on potential mechanisms associated with EB2 signaling in cognitive functions. RESULTS: Our study captures changes in cell populations in response to EB2 manipulation and reveals previously uncharacterized cell types (CPA6 + inhibitory neurons) in the mPFC. We validated the differential transcriptomic activity of Pbx1 and Meis1 in CPA6 + neurons using fluorescence in situ

Prepubertal Development of GABAergic Transmission to Gonadotropin-Releasing Hormone (GnRH) Neurons and Postsynaptic Response Are Altered by Prenatal Androgenization.

Gonadotropin-releasing hormone (GnRH) neurons regulate reproduction through pulsatile GnRH release. Women with polycystic ovary syndrome (PCOS) have persistently elevated luteinizing hormone release frequency, reflecting GnRH release; this exacerbates hyperandrogenemia and disrupted reproductive cycles that are characteristic of this disorder. Clinical evidence suggests that neuroendocrine features of PCOS may manifest peripubertally. Adult mice prenatally exposed to androgens (PNA) mimic several reproductive features of PCOS. GnRH neurons from these mice have increased firing activity and rec

Direct conversion of mouse fibroblasts to GABAergic neurons with combined medium without the introduction of transcription factors or miRNAs.

Degeneration or loss of GABAergic neurons frequently may lead to many neuropsychiatric disorders such as epilepsy and autism spectrum disorders. So far no clinically effective therapies can slow and halt the progression of these diseases. Cell-replacement therapy is a promising strategy for treatment of these neuropsychiatric diseases. Although increasing evidence showed that mammalian somatic cells can be directly converted into functional neurons using specific transcription factors or miRNAs via virus delivery, the application of these induced neurons is potentially problematic, due to inte…

T-type channels control the opioidergic descending analgesia at the low threshold-spiking GABAergic neurons in the periaqueductal gray.

Endogenous opioids generate analgesic signals in the periaqueductal gray (PAG). However, because cell types in the PAG are difficult to identify, its neuronal mechanism has remained poorly understood. To address this issue, we characterized PAG neurons by their electrical properties using differentially labeled GABAergic and output neurons in the PAG. We found that GABAergic neurons were mostly fast-spiking cells and could be further divided into two distinct classes: with or without low-threshold spikes (LTS) driven by T-type channels. In contrast, the PAG output neurons lacked LTS and showed…

Presynaptic muscarinic-nicotinic modulation of GABAergic inputs to neurons in the REM sleep executive area of the rostral pons.

A large body of evidence indicates that cholinergic neurons in the brainstem tegmental nuclei play a pivotal role in controlling neurons of the reticular nucleus pontis oralis (PnO), an area considered executive for rapid eye movement (REM) sleep onset and maintenance. More recent data indicate that the PnO is also under GABAergic control-which gates REM sleep-thereby highlighting the importance of interactions between cholinergic and GABAergic processes as a key mechanism in REM sleep regulation. Here, we used a rat mesopontine slice preparation to investigate the modulation of GABAergic inpu

Prenatal stress history modifies adolescent stress effects on adult social behavior and basolateral amygdala GABAergic neurons with perineuronal nets.

Developmental stress is a well-established risk factor for mental health disorders, yet the neural mechanisms underlying these outcomes remain incompletely understood. Inhibitory brain networks, particularly within the amygdala, are disrupted by stress and implicated in stress-related psychopathologies. Using a rodent model, the current study investigated the isolated and combined effects of prenatal and adolescent stress on adult social interactions and GABAergic neurons surrounded by perineuronal nets (PNNs) in the basolateral amygdala (BLA). Male and female rats were exposed to chronic vari…

Integrative genome-scale metabolic model of GABAergic neurons reveals metabolic signatures across the mild cognitive impairment - Alzheimer disease continuum.

INTRODUCTION: Mild cognitive impairment (MCI) represents a prodromal stage of Alzheimer's disease (AD), but the metabolic mechanisms underlying early neuronal dysfunction remain incompletely understood. GABAergic neurons, which maintain excitatory-inhibitory balance and network stability, exhibit early vulnerability during neurodegeneration, although the metabolic alterations associated with their dysfunction remain poorly characterized. METHODS: We developed a context-specific genome-scale metabolic model (GEM) of human GABAergic neurons across the MCI-AD continuum using deconvolved hippocam…
