The levels of the aromatase gene and its expression in MCF-7 human breast cancer cells and seven additional cultured cells were investigated. Using normal human foreskin fibroblasts as the control, the aromatase gene appeared to be amplified in MCF-7 cells as shown by Southern and DNA slot blot analyses utilizing human placental aromatase cDNA as the probe. However, the promoter I.1 and the first exon of the aromatase gene were not amplified in MCF-7 cells based on results obtained from DNA slot blot analysis using oligonucleotide probes having sequences derived from those regions of human aro

Acute myeloid leukaemia (AML) is a genetically heterogeneous malignancy associated with poor prognosis and limited treatment options. To identify molecular programs conserved across AML subtypes and perturbations, we analysed three RNA sequencing datasets that captured venetoclax treatment under metabolic stress and the knockdown of chromatin regulators (PSPC1, JMJD1C, and RUNX1). Differential expression analysis was performed using DESeq2, followed by functional enrichment and network analyses. An independent AML cell line dataset was used to validate results. We identified a conserved 73-gen

Microarray analysis is a promising new approach for creating specific expression profiles of multiple genes simultaneously. We quantitatively analyzed differential gene expression patterns in mycosis fungoides-derived clonal T cells and autologous, identically cultured CD4+ lymphocytes using microarrays containing 588 cDNA segments from genes relevant to cell signaling, carcinogenesis, and apoptosis. Among other dissimilarities, neoplastic T cells showed coexpression of CD40 (Bp50) and CD40 ligand (gp39, CD154). These results could be corroborated by reverse transcription-PCR, immunohistochemi

Colorectal cancer (CRC) remains a leading cause of cancer-related mortality worldwide. Effective CRC screening in the United States has reduced incidence of CRC in populations over 50 years of age. Early-onset CRC is emerging as a significant issue. There is need for better understanding of CRC biology to develop screening techniques, diagnostic markers and novel therapies to improve outcomes. MicroRNAs (miRNAs) are involved in several cellular processes and function as tumor suppressors and oncogenes. The miRNA/cytokine networks play a critical role in CRC pathogenesis by contributing to an e

Transcriptional heterogeneity in pancreatic ductal adenocarcinoma (PDAC) arises not only from changes in gene expression but also from dynamic rewiring of gene-gene coordination. Using a divergent-edge framework applied to 77 155 malignant cells from 42 tumors, we identified four reproducible adaptive modules-integrated growth-energy (IGE), stress-adaptive transcription (SAT), IL-2-linked immune evasion (IL2), and multi-pathway collective invasion (MPC)-that cut across canonical PDAC states and reflect distinct regulatory programs. Integrating these modules with CRISPR-Cas9 dependency profiles

MicroRNAs/miRNAs are known epigenetic regulators that function as rheostats to regulate gene expression in cells. miRNA can simultaneously target multiple genes, thus profoundly impacting cellular physiology. Cancer is one of the most complex diseases. It often involves the interplay of various genes, making anti-cancer therapies targeting a single gene less effective. It leads to the development of resistance to treatment over time, thus limiting long-term beneficial outcomes. In contrast, the ability of miRNAs to target multiple genes simultaneously might make them suitable as effective anti

BACKGROUND: Hepatocellular carcinoma (HCC) is a highly aggressive cancer with significant intratumoral heterogeneity, likely contributed by the differentiation of cancer stem-like cells (CSCs) into multiple cancer cell lineages, resulting in therapeutic resistance and increased patient mortality. However, there is limited evidence from patient samples regarding the critical role of CSCs in treatment efficacy and patient outcomes.
METHODS: In this study, through the analysis of single-cell transcriptomic data of HCC patient samples, we identify a rare population of cells with cancer stem-like

The study of gene anomalies linked to thyroid cancer is gaining more and more attention, and these molecular indicators can offer scholarly support for thyroid cancer diagnosis, therapy selection, and prognosis. Genotype-tissue expression pan-cancer data and The Cancer Genome Atlas (TCGA) were used to investigate the expression of GZMK, TREM2, and OR4D10. In order to assess the relationship between GZMK, TREM2, and OR4D10 expression and patient outcome, TCGA clinical survival data were used. We used the clusterProfiler R software tool to conduct enrichment analysis of GZMK, TREM2, and OR4D10.

OBJECTIVE: A dietary compound curcumin hardwires to multiple cellular processes, with suppression of cell proliferation, induction of apoptosis, and inhibition of metastasis considered as the major mechanisms underlying its anticancer properties. Based on our recent evidence that curcumin triggers cell demise in follicular lymphoma (FL) cells, we aimed to identify curcumin-regulated genes of utmost importance for the treatment of follicular lymphoma.
MATERIALS AND METHODS: Large-scale gene-expression profiling was performed during curcumin-triggered apoptosis (8-36 hours) in follicular lympho

We have analyzed gene expression data from three different kinds of samples: normal human tissues, human cancer cell lines, and leukemic cells from lymphoid and myeloid leukemia pediatric patients. We have searched for genes that are overexpressed in human cancer and also show specific patterns of tissue-dependent expression in normal tissues. Using the expression data of the normal tissues, we identified 4,346 genes with a high variability of expression and clustered these genes according to their relative expression level. Of 91 stable clusters obtained, 24 clusters included genes preferenti

This review consists of two parts. In the first part normal mechanisms regulating the progression of cells through the cell cycle are briefly reviewed. Besides mitogenic stimulation, cyclin kinase inhibition, the G1 restriction point and the prb pathway, accuracy of DNA replication and DNA repair, the G2 to M transition, apoptosis and the p 53 pathway, proteolytic, in particular ubiquitin-dependent mechanisms involved in the initiation of DNA synthesis in the separation of sister chromatids and in the telophase to GO/G1 transition, are discussed. In the second part oncogene and tumor suppresso

Ribosomal protein L32 (RPL32) is a core component of the 60S ribosomal subunit and a stable reference gene. Mounting evidence shows that RPL32 is upregulated in hepatocellular carcinoma, lung cancer, breast cancer, prostate cancer, and other malignancies, correlating with poor prognosis. RPL32 promotes cancer cell proliferation, invasion, and progression partly through the MDM2-p53-autophagy axis, and its expression is regulated by promoter methylation, copy number variations, and transcription factors. RPL32 represents a promising prognostic biomarker and candidate intervention target for can

Pancreatic cancer (PC) is one of the most lethal malignancies, with an approximately 5% 5-year survival rate. Understanding the intricate network of interactions among the proteins of key pathways is critical for assessing the PC prognosis. Herein, we have carried out a systematic approach by collecting mutational data of genes involved in 10 PC studies with 6,325 PC samples from cBioPortal. A total of 15,088 genes were obtained, to which a mutational frequency cut-off of > 10 was applied. The 1,617 genes acquired after this filtration. These genes were stratified into five PC tiers (PC-TierII

High-molecular-weight splice variants of the CD44 transmembrane protein family have been implicated in tumorigenesis and metastasis formation. By contrast, in certain tumors--for example, Burkitt's lymphoma, neuroblastomas, and prostate cancer--loss of CD44 expression seems to accompany transformation. Here we describe two modes of action of CD44 proteins. They can bind growth factors and present them to their authentic high-affinity receptors, and thus promote proliferation and invasiveness of cells. Under these conditions the CD44 proteins recruit ERM proteins--for example, ezrin or moesin--

Approximately 40% of chronic myeloid leukemia (CML) patients who discontinue imatinib (IM) therapy maintain undetectable minimal residual disease (UMRD) for more than one year (stopping IM (STOP-IM)). To determine a possible biomarker for STOP-IM CML, we examined plasma miRNA expression in CML patients who were able to discontinue IM. We first screened candidate miRNAs in unselected STOP-IM patients, who had sustained UMRD after discontinuing IM for more than six months, in comparison with healthy volunteers, by using a TaqMan low-density array for plasma or exosomes. Exosomal miR-215 and plas

Human giant cell tumour of bone (GCTB) is a rare low grade of malignancy tumour with tendency to recur. During tumourigenesis the bone remodeling balance is subverted by the tumour cellular components that interacting with bone matrix induce release of growth factors and cytokines, promoting cell proliferation and bone resorption. The master regulators of this positive feed-back are acid and neutral proteases that destroying extracellular matrix increase osteolysis. In contrast, in cats, very few data are reported on GCTB biological activity. In this study, histological features and metallopro

BACKGROUND: Most advanced melanomas initially respond to targeted therapy but eventually relapse. Increasing evidence suggests that drug-tolerant persister cells can adopt a reversible drug-refractory state and represent a key driver of therapeutic resistance. METHODS: We developed MeRLin, a lineage tracing platform that integrates cellular barcoding, single-cell transcriptomic profiling, RNA fluorescence in situ hybridization, and computational analyses to track clonal and transcriptional dynamics in a patient-derived melanoma model during prolonged targeted therapy. Longitudinal analyses ena

Episomal integration is a critical event in human papillomavirus (HPV)-related oncogenesis, although little information is currently available concerning the effect of integration on the host transcriptome. We have used expression microarrays to investigate the effect of integration of HPV16 on gene expression in cervical keratinocytes, using the unique cell line model W12. W12 was generated from a cervical low-grade squamous intraepithelial lesion "naturally" infected with HPV16 and at low passage contains approximately 100 HPV16 episomes/cell. With passage in vitro, integration of viral epis

An animal model of carcinogenesis has been exploited to analyze the various events involved in carcinogen-induced T cell lymphomagenesis. Two carcinogenic agents, the alkylating agent N-methylnitrosourea (NMU) and ionizing gamma-radiation, induce tumors in C57BL/6J mice that are phenotypically and histologically identical. Are the genetic events similar or different in the T cell tumors produced by these two carcinogenic agents? NMU treatment produced a different spectrum of activated oncogenes from gamma-irradiation. The K-ras oncogene was preferentially activated in all of the NMU-induced tu

Epstein-Barr virus (EBV) persists as a life-long latent infection within memory B cells, but how EBV may circumvent the innate immune response within this virus reservoir is unclear. Recent studies suggest that the latency-associated non-coding RNAs of EBV may actually induce type I (antiviral) interferon production, raising the question of how EBV counters the negative consequences this is likely to have on viral persistence. We addressed this by examining the type I interferon response in Burkitt lymphoma (BL) cell lines, the only in vitro model of the restricted program of EBV latency-gene
