JNK/c-Jun pathway activation is essential for HBx-induced IL-35 elevation to promote persistent HBV infection.

BACKGROUND: Immunoregulation plays pivotal roles during chronic hepatitis B virus (HBV) infection. Studies have shown that Interleukin (IL)-35 is an important molecule associated with inadequate immune response against HBV. However, the mechanisms involved in the up-regulation of IL-35 expression during persistent HBV infection remain unknown. METHODS: In this study, we constructed a plasmid expressing the HBV X protein (pCMV-HBx) to evaluate the relationship between HBx and IL-35. Activation of the JNK/c-Jun pathway was analyzed and chromatin immunoprecipitation followed by sequencing and lu

Human T-cell lymphotropic virus oncoprotein Tax represses TGF-beta 1 signaling in human T cells via c-Jun activation: a potential mechanism of HTLV-I leukemogenesis.

Human T-cell leukemia virus I is the etiologic agent of adult T-cell leukemia (ATL), an aggressive T-cell malignancy. The viral oncoprotein Tax, through the activation of nuclear factorkappaB (NF-kappaB), CCAAT-enhancer binding protein (CREB), and activated protein-1 (AP-1) pathways, is a transcriptional regulator of critical genes for T-cell homeostasis. In ATL cells, activated AP-1 complexes induce the production of transforming growth factor beta1 (TGF-beta1). TGF-beta1 is an inhibitor of T-cell proliferation and cytotoxicity. Here we show that, in contrast to normal peripheral T cells, ATL

Network toxicology study and key target validation of chlorpyrifos-induced nonalcoholic fatty liver disease.

Chlorpyrifos (CPF) is a widely used insecticide known for its extended persistence in soil and water. It can induce non-alcoholic fatty liver disease (NAFLD) in mice. However, the specific molecular mechanisms responsible for CPF-induced NAFLD remain partially understood. Utilizing network toxicology analysis, we have identified TP53, HSP90AA1, AKT1, and JUN as pivotal driver genes orchestrating NAFLD progression triggered by CPF. A predictive nomogram constructed on these core genes demonstrates promising translational prospects for NAFLD. Gene-set enrichment analysis (GSEA) underscores the s

Differential expression of c-fos and c-jun in two regions of the rat spinal cord following noxious colorectal distention.

Thoracolumbar and lumbosacral spinal segments that receive afferent input from the descending colon and rectum were stained immunocytochemically for c-Fos- and c-Jun-like proteins following repetitive, noxious colorectal distention (CRD). Noxious CRD (80 mmHg) resulted in significantly more c-Fos- and c-Jun-like immunoreactivity in the sacral dorsal horn than in the thoracic dorsal horn. In both regions of the spinal cord the increase in c-Fos-like immunoreactivity was at least twice that of c-Jun-like immunoreactivity. Basal levels of c-Jun but not c-Fos were observed in the thoracic intermed

N-n-Butyl haloperidol iodide ameliorates liver fibrosis and hepatic stellate cell activation in mice.

N-n-Butyl haloperidol iodide (F2) is a novel compound that has antiproliferative and antifibrogenic activities. In this study we investigated the therapeutic potential of F2 against liver fibrosis in mice and the underlying mechanisms. Two widely used mouse models of fibrosis was established in mice by injection of either carbon tetrachloride (CCl4) or thioacetamide (TAA). The mice received F2 (0.75, 1.5 or 3 mg·kg-1·d-1, ip) for 4 weeks of fibrosis induction. We showed that F2 administration dose-dependently ameliorated CCl4- or TAA-induced liver fibrosis, evidenced by significant decreases i

Superantigen-induced transcriptional activation of the human TNF gene promoter in T cells.

Superantigens, such as staphylococcal enterotoxins (SE), cause food poisoning and shock, which is mediated at least in part by TNF. We have examined the mechanism of superantigen-induced activation of the TNF gene promoter in primary human peripheral blood T lymphocytes and in the leukemic T cell line Jurkat. Like stimulation of the T cell receptor complex through CD3 ligation, SEB induces binding of nuclear proteins to Egr- and Jun/ATF-related consensus sequences present between nucleotides -170 and -100 of the TNF promoter 5' flanking region. By cotransfection of Jurkat T cells with construc

Potential of RAD51C as a hub gene and therapeutic target in bladder cancer: insights from bioinformatics and experimental analysis.

BACKGROUND: The global incidence of tumors is rising sharply. Immune checkpoint inhibitors (ICIs) represent the mainstream of immunotherapeutic approaches. Despite the remarkable clinical benefits of immunotherapy in various tumors, ICIs resistance remains an urgent challenge to be addressed. The transcription factors cAMP response element-binding protein 1(CREB1) and c-Jun converge to regulate oncogenesis and immune modulation. METHODS: Based on the 'CREBP1CJUN_01' gene set from the Molecular Signatures Database and single-cell RNA sequencing data, we identified 116 CREB-c-Jun transcription

Effects of dl-praeruptorin A on cultured neonatal rat ventricular cardiomyocytes with hypertrophy induced by endothelin-1.

The present study investigated whether dl-praeruptorin (Pd-Ia) prevents endothelin-1 (ET-1)-induced cardiomyocyte hypertrophy and the potential pathways that underlie such an effect. We assessed cardiomyocyte surface area, protein synthesis, the expression of Bax/Bcl2 and Jun genes, the expression of atrial natriuretic factor (ANF) and Ca2+/calmodulin-dependent kinase II (CaMK-II) activity in cultured neonatal rat ventricular cardiomyocytes with ET-1-induced hypertrophy. It was found that Pd-Ia decreased the surface area and protein synthesis rate in cardiomyocytes exposed to ET-1. Additionall

Immediate-early gene expression in the brain of the thiamine-deficient rat.

Pyrithiamine-induced thiamine deficiency (PTD) in the rat is associated with neuronal loss in the thalamus and inferior colliculus. Recently, we were able to demonstrate the occurrence of apoptosis in the thalamus of these animals. Given that immediate-early genes (IEGs) participate in signal transduction pathways that mediate programmed cell death, the present study utilized in situ hybridization and immunohistochemistry to examine the expression of four IEGs (c-fos, c-jun, fos-B, and NGFI-A) during the progression of PTD. Elevated c-fos mRNA levels were initially observed in the posterior me

Aldo-keto reductase family 1 member C3 suppresses Glioma progression via inhibiting prostaglandin D2-dependent RhoA/ROCK signaling.

Aldo-keto reductase family 1 member C3 (AKR1C3) is a pivotal metabolic enzyme involved in arachidonic acid (AA) metabolism and cancer metabolic reprogramming, which plays a critical role in the initiation and progression of glioblastoma (GBM). We integrated single-cell and transcriptome data to characterize AA metabolism features in GBM and identify the key gene AKR1C3. Using CCK8 assays, wound-healing assays, transwell assays, and mouse tumor formation experiments, we elucidated the effects of AKR1C3. Dual-luciferase reporter assays and ChIP-PCR experiments were performed to investigate the r

Effect of trichloroethylene and its metabolites, dichloroacetic acid and trichloroacetic acid, on the methylation and expression of c-Jun and c-Myc protooncogenes in mouse liver: prevention by methionine.

Trichloroethylene (TCE), dichloroacetic acid (DCA), and trichloroacetic acid (TCA) are environmental contaminants that are carcinogenic in mouse liver. 5-Methylcytosine (5-MeC) in DNA is a mechanism that controls the transcription of mRNA, including the protooncogenes, c-jun and c-myc. We have previously reported that TCE decreased methylation of the c-jun and c-myc genes and increased the level of their mRNAs. Decreased methylation of the protooncogenes could be a result of a deficiency in S-adenosylmethionine (SAM), so that methionine, by increasing the level of SAM, would prevent hypomethyl
