A novel brainstem nucleus orchestrating reward and aversion.

Reward processing is a critical brain function. Zichó and colleagues recently identified a previously unrecognized brainstem nucleus, the subventricular tegmental nucleus (SVTg), as a novel reward center that modulates dopamine release and regulates reward processing by balancing the lateral habenula (LHb)-ventral tegmental area (VTA) axis.

Habenular and interpeduncularis nuclei: shared components in multiple-function networks.

STUDY AIMS: Advance Directives (AD) are central to Advance Care Planning (ACP), allowing patients to document medical preferences in legally binding documents. Despite broad public awareness and support, only one-fifth of the Swiss population has completed an AD. Consequently, the roadmap for ACP implementation by the ACP National Task Force calls for the development of digital applications to promote ACP in Switzerland. This study details the development process for the mobile application "iDecide" for AD creation based on the Swiss Medical Association (FMH) AD template. The primary aim is to

Asymmetry in the epithalamus of vertebrates.

The epithalamus is a major subdivision of the diencephalon constituted by the habenular nuclei and pineal complex. Structural asymmetries in this region are widespread amongst vertebrates and involve differences in size. neuronal organisation, neurochemistry and connectivity. In species that possess a photoreceptive parapineal organ, this structure projects asymmetrically to the left habenula, and in teleosts it is also situated on the left side of the brain. Asymmetries in size between the left and right sides of the habenula are often associated with asymmetries in neuronal organisation, alt…

Distribution of nicotinic receptors in cynomolgus monkey brain and ganglia: localization of alpha 3 subunit mRNA, alpha-bungarotoxin and nicotine binding sites.

The distribution of nicotinic receptors in the brain and ganglia of the Cynomolgus monkey was studied by in situ hybridization and receptor autoradiography. A 35S-labeled antisense riboprobe for the mRNA of the alpha 3 subunit of the human nicotinic receptor, [3H]L-nicotine and [125]alpha-bungarotoxin were used as markers. The highest levels of alpha 3-mRNA were observed in the hippocampus, the medial habenula, the lateral geniculate, the granular layer of the cerebellum, as well as in the pineal gland; moderate levels were found in other nuclei of the thalamus and in the deeper layers of the …

Alterations in Cyclic AMP Concentration and Adenylate Cyclase Activity in Specific Brain Areas of Rats with Inherited Hypothalamic Diabetes Insipidus (Brattleboro Rats).

Abstract The concentration of cyclic AMP (cAMP) and the activity of sodium-fluoride-stimulated adenylate cyclase was measured in 29 microdissected brain areas of homozygous Brattleboro rats and their Long-Evans control rats. In ten of the investigated brain areas a decreased cAMP level was measured in Brattleboro rats. It was particularly decreased in the supraoptic nucleus, cingulate and parietal cortex, hippocampus, habenula and organum vasculosum laminae terminalis. Significantly lower cAMP levels were also found in the periventricular nucleus, bed nucleus of the stria terminalis, area post…

Valproic acid exposure affects social visual lateralization and asymmetric gene expression in zebrafish larvae.

Cerebral asymmetry is critical for typical brain function and development; at the same time, altered brain lateralization seems to be associated with neuropsychiatric disorders. Zebrafish are increasingly emerging as model species to study brain lateralization, using asymmetric development of the habenula, a phylogenetically old brain structure associated with social and emotional processing, to investigate the relationship between brain asymmetry and social behavior. We exposed 5-h post-fertilization zebrafish embryos to valproic acid (VPA), a compound used to model the core signs of ASD in m…

Foxg1 deletion impairs the development of the epithalamus.

The epithalamus, which is dorsal to the thalamus, consists of the habenula, pineal gland and third ventricle choroid plexus and plays important roles in the stress response and sleep-wake cycle in vertebrates. During development, the epithalamus arises from the most dorsal part of prosomere 2. However, the mechanism underlying epithalamic development remains largely unknown. Foxg1 is critical for the development of the telencephalon, but its role in diencephalic development has been under-investigated. Patients suffering from FOXG1-related disorders exhibit severe anxiety, sleep disturbance an…

Pitx2c ensures habenular asymmetry by restricting parapineal cell number.

Left-right (L/R) asymmetries in the brain are thought to underlie lateralised cognitive functions. Understanding how neuroanatomical asymmetries are established has been achieved through the study of the zebrafish epithalamus. Morphological symmetry in the epithalamus is broken by leftward migration of the parapineal, which is required for the subsequent elaboration of left habenular identity; the habenular nuclei flank the midline and show L/R asymmetries in marker expression and connectivity. The Nodal target pitx2c is expressed in the left epithalamus, but nothing is known about its role du…

Awakening Neuropsychiatric Research Into the Stria Medullaris: Development of a Diffusion-Weighted Imaging Tractography Protocol of This Key Limbic Structure.

The Stria medullaris (SM) Thalami is a discrete white matter tract that directly connects frontolimbic areas to the habenula, allowing the forebrain to influence midbrain monoaminergic output. Habenular dysfunction has been shown in various neuropsychiatric conditions. However, there exists a paucity of research into the habenula's principal afferent tract, the SM. Diffusion-weighted tractography may provide insights into the properties of the SM in vivo, opening up investigation of this tract in conditions of monoamine dysregulation such as depression, schizophrenia, addiction and pain. We pr…

Atypical Course of the Habenulo-Interpeduncular Tract in Chick Embryos.

Classical studies of the avian diencephalon hardly mention the habenulo-interpeduncular tract (a.k.a. retroflex tract), although both the habenula (HB) (its origin) and the interpeduncular nuclear complex (its target) are present. Retroflex tract fibers were described at early embryonic stages but seem absent in the adult in routine stains. However, this tract is a salient diencephalic landmark in all other vertebrate lineages. It typically emerges out of the caudal HB, courses dorsoventrally across thalamic alar and basal plates just in front of the thalamo-pretectal boundary, and then sharpl…

Efferent Axonal Projections of the Habenular Complex in the Fire-Bellied Toad Bombina orientalis.

The habenular complex and its associated axonal pathways are often thought of as phylogenetically conserved features of the brain among vertebrates despite the fact that detailed studies of this brain region are limited to a few species. Here, the gross morphology and axonal projection pattern of the habenular complex of an anuran amphibian, the fire-bellied toad Bombina orientalis, was studied to allow comparison with the situation in other vertebrates. Axonal pathways were traced using biocytin applications in dissected brain preparations. The results show that the rostral part of the left d…

Spatial Coordinates of the Rostral Junction Points of the Genu of the Corpus Callosum and the Habenular Commissure in Tree Shrew, Mouse and Rat Brains: A Preliminary Study.

Spatial coordinate systems are essential for precise brain localization and atlas-based comparison, yet current stereotaxic frameworks mainly rely on cranial landmarks such as bregma-lambda (BL) or anterior commissure-posterior commissure (AP). Here, we defined the rostral junction point of the genu of the corpus callosum (GCCrj) that refers to the central convergence point of bilateral genu of the corpus callosum structures at the rostral part on coronal histological slices. The GCCrj and rostral junction point of the habenular commissure (HBCrj) constituted the new GCCrj-HBCrj (GH) coordinat…

Neural mechanism underlying depressive-like state associated with social status loss.

Downward social mobility is a well-known mental risk factor for depression, but its neural mechanism remains elusive. Here, by forcing mice … gh the lateral hypothalamus, the RPE strongly activates the brain's anti-reward center, the lateral habenula (LHb). LHb activation inhibits the medial prefrontal cortex (mPFC) that controls social competitiveness and reinforces retreats in contests. These results reveal the core neural mechanisms mutually promoting social status loss and depressive behavio …

An ultrastructural study of the deep pineal gland of the Sprague Dawley rat using transmission and serial block face scanning electron microscopy: cell types, barriers, and innervation.

The morphology of the deep pineal gland of the Sprague Dawley rat was investigated by serial block face scanning electron microscopy. Cells were three-dimensionally (3-D) reconstructed using the software Fiji TrackEM. The deep pineal gland consisted of 2-5 layers of electron-lucent pinealocytes, with a euchromatic nucleus, endowed with one or two processes. Laterally, the deep pineal merged with the habenula and the stria medullaris thalami, via an intermediate area containing cells with more electron-dense cytoplasm and an indented nucleus with heterochromatin. Neither nerve terminals nor cap…

Specific connections of the interpeduncular subnuclei reveal distinct components of the habenulopeduncular pathway.

The habenulopeduncular pathway consists of the medial habenula (MHb), its output tract, the fasciculus retroflexus, and its principal target, the interpeduncular nucleus (IP). Several IP subnuclei have been described, but their specific projections and relationship to habenula inputs are not well understood. Here we have used viral, transgenic, and conventional anterograde and retrograde tract-tracing methods to better define the relationship between the dorsal and ventral MHb, the IP, and the secondary efferent targets of this system. Although prior studies have reported that the IP has ascen…
