Acute phase response in mice experimentally infected with Trypanosoma congolense: a molecular gauge of parasite-host interaction.

Mice infected with Trypanosoma congolense developed a severe anaemia 1 week after infection, which persisted till treatment with diminazine aceturate when the packed cell volume (PCV) recovered to pre-infection levels. This was accompanied by a marked increase in the plasma levels of the acute phase proteins (APP), serum amyloid P-component (SAP) and haptoglobin (Hp). The initial peak levels of Hp and SAP were attained 7 and 12 days post-infection (DPI), respectively. Thereafter SAP levels decreased significantly to near pre-infection levels, but later increased even after treatment to give a

Subcellular distribution of the Entamoeba histolytica 140 kDa FN-binding molecule during host-parasite interaction.

Entamoeba histolytica trophozoites recovered from the host-parasite interface during abscess development obtain different stimuli compared with long-term cultured cells. In order to have a better understanding about the mechanisms in which the 140 kDa fibronectin (FN)-binding molecule (EhFNR) is involved during the invasive process, we decided to compare the regulation process of this molecule among long-term cultured trophozoites, FN-stimulated trophozoites, and trophozoites recently recovered from a liver abscess. A cDNA clone (5A) containing a fragment of the EhFNR that shows identity to th

Early host-parasite interaction models reveal a key role for fibrinolysis in Fasciola hepatica intestinal migration.

BACKGROUND: Fasciola hepatica is the most common etiologic agent of fasciolosis, a parasitic disease that affects millions of ruminants worldwide and a zoonotic human infection of public health concern. Upon ingestion of infective metacercariae, F. hepatica newly excysted juveniles (FhNEJ) emerge in the duodenum and cross the intestinal wall to initiate a migration route that culminates with their establishment within the hepatic bile ducts. The ability of FhNEJ to exploit the broad-spectrum activities of host plasmin, the central protease of the fibrinolytic system, has been proposed as a str

Targeting peptide based therapeutics: Integrated computational and experimental studies of autophagic regulation in host-parasite interaction.

Cutaneous leishmaniasis caused by the intracellular parasite Leishmania major, exhibits significant public health challenge worldwide. With limited treatment options available, the identification of novel therapeutic targets is of paramount importance. Present study manifested the crucial role of ATG8 protein as a potential target in combating L. major infection. Using machine learning algorithms, we identified non-conserved motifs within the ATG8 in L. major. Subsequently, a peptide library was generated based on these motifs, and three peptides were selected for further investigation through

Fungus-insect interactions beyond bilateral regimes: the importance and strategy to outcompete host ectomicrobiomes by fungal parasites.

Fungus-insect interactions have long been investigated at the bilateral level to unveil the factors involved in mediating fungal entomopathogenicity and insect antifungal immunity. Emerging evidence has shown that insect cuticles are inhabited by different bacteria that can delay and deter fungal parasite infections. Entomopathogenic fungi (EPF), however, have evolved strategies to combat the colonization resistance mediated by insect ectomicrobiomes by producing antimicrobial peptides or antibiotic compounds. Deprivation of micronutrients may also be employed by EPF to counteract the ectomicr

Knowledge gaps in host-parasite interaction preclude accurate assessment of meat-borne exposure to Toxoplasma gondii.

Toxoplasma gondii is recognized as a widely prevalent zoonotic parasite worldwide. Although several studies clearly identified meat products as an important source of T. gondii infections in humans, quantitative understanding of the risk posed to humans through the food chain is surprisingly scant. While probabilistic risk assessments for pathogens such as Campylobacter jejuni, Listeria monocytogenes or Escherichia coli have been well established, attempts to quantify the probability of human exposure to T. gondii through consumption of food products of animal origin are at early stages. The b

Leishmania major infection in the fat sand rat Psammomys obesus in Tunisia: interaction of host and parasite populations.

The causative agent of cutaneous leishmaniasis, Leishmania major, was studied in a Tunisian population of the fat sand rat, Psammomys obesus. Seasonal changes in the abundances of parasite and host were monitored in a longitudinal field survey lasting 21 months. Overall, 566 P. obesus, collected during 10 trapping sessions between May 1995 and January 1997, were examined. Analysis of prevalence, using logistic regression, revealed that extrinsic factors, such as season and climatic conditions, and intrinsic factors, such as host age, have a combined effect. Leishmania major showed a seasonal p

Host-parasite interaction and development of infraforms in chicken embryos infected with Coxiella burnetii via the yolk sac.

Two phase I strains of Coxiella burnetii of different virulence were injected into the yolk sacs of chicken embryos, and the yolk sacs and livers were examined at intervals by light, fluorescent, and electron microscopy. The high absorptive and digestive capacities of the yolk endoderm contributed to he entrance of the organisms into endodermal epithelial cells where C. burnetii multiplied. Organisms multiplied not only inside specific vacuoles originating from phagolysosomes but also in the cytoplasm itself. Set up of an in vitro model to study early host-parasite interactions between newly excysted juveniles of Fasciola hepatica and host intestinal cells using a quantitative proteomics approach.

Fasciola hepatica is the causative agent of fasciolosis, a parasitic zoonosis of global distribution causing significant economic losses in animal production and a human public health problem in low-income countries. Hosts are infected by ingestion of aquatic plants carrying metacercariae. Once ingested, the juvenile parasites excyst in the small intestine and, after crossing it, they follow a complex migratory route that lead the parasites to their definitive location in the bile ducts. Despite being a critical event in the progression of the infection, the available data on the cross-talk re

Use of in vitro derived human neuronal models to study host-parasite interactions of Toxoplasma gondii in neurons and neuropathogenesis of chronic toxoplasmosis.

Toxoplasma gondii infects approximately one-third of the world's population resulting in a chronic infection with the parasite located in cysts in neurons in the brain. In most immunocompetent hosts the chronic infection is asymptomatic, but several studies have found correlations between Toxoplasma seropositivity and neuropsychiatric disorders, including Schizophrenia, and some other neurological disorders. Host-parasite interactions of bradyzoites in cysts in neurons is not well understood due in part to the lack of suitable in vitro human neuronal models. The advent of stem cell technologie…

Human peripheral blood mononuclear cells as an ex vivo model to study the host parasite interaction in Toxoplasma gondii.

Toxoplasma gondii is a parasite that can invade any cell in the human body. Here, we implemented and described an ex vivo model with human peripheral blood mononuclear cells (PBMCs) without using culture supplements/antibiotics and without cryopreserved cells (EXMOWS) to study the interactions between T. gondii and human cells. To establish the EXMOWS, three independent tests were carried out. Firstly, blood samples from 5 individuals were included to assess the viability and adherence of PBMCs in plate culture. In a second trial, blood samples from three seropositive and two seronegative indi

Epidemiology and hypothetical transmission cycles of Trichinella infections in the Greater Kruger National Park of South Africa: an example of host-parasite interactions in an environment with minimal human interactions.

Knowledge on the epidemiology, host range and transmission of Trichinella spp. infections in different ecological zones in southern Africa including areas of wildlife-human interface is limited. The majority of reports on Trichinella infections in sub-Saharan Africa were from wildlife resident in protected areas. Elucidation of the epidemiology of the infections and the prediction of hosts involved in the sylvatic cycles within specific ecological niches is critical. Of recent, there have been reports of Trichinella infections in several wildlife species within the Greater Kruger National Park

Environmental and host- and parasite-associated drivers of the flea-mammal interaction network structure differ between biogeographic realms.

We evaluated the effects of climatic factors, host and parasite species richness, and their phylogenetic and functional diversity on the nestedness and modularity of mammal-flea interaction networks from four biogeographic realms. We also tested for the associations between pure structural network dissimilarity (i.e., dissimilarity between host-sharing-by-fleas networks, Dh, or dissimilarity between flea-sharing-by-hosts networks, Df) and environmental dissimilarity, and host and flea compositional, phylogenetic, and functional dissimilarities. We asked whether (a) the relative effects of thes
