BACKGROUND: Hypopigmentation is an infrequently reported adverse effect of imatinib mesylate (IM) in chronic myeloid leukemia (CML), but there are no reports from Arab or Saudi patients. Thus, we assessed the frequency and impact of hypopigmentation in patients with chronic myeloid leukemia (CML) taking IM in our institution in Riyadh.
PATIENTS AND METHODS: We studied 24 adult CML patients taking IM and followed from March to June 2008. Telephonic interviews with all the CML patients taking IM were conducted and case notes were reviewed. Findings were confirmed on a subsequent clinic visit by…

We report a 3-year-old girl with autosomal dominant inherited Waardenburg syndrome type I showing circumscribed hypopigmentation of the skin, heterochromia iridis, sensorineural deafness, and dental aberrations. Clinical diagnosis was confirmed by the identification of an underlying missense mutation (C811T) in the PAX3 gene. Early diagnosis of Waardenburg syndrome among children with pigment anomalies enables a successful interdisciplinary medical care.

The development of linear hypopigmentation after intralesional or intraarticular injection of triamcinolone acetonide has been reported in the literature as a very rare side effect. This case report describes a patient with linear hypopigmentation and discusses the possible pathophysiology. Clinicians involved in the care of hypertrophic scars and keloids need to be aware of this rare side effect so that they can guide their patients appropriately. They need to understand the pathogenesis of this complication better so that it may become avoidable.

Hypopigmentation of hair and freckles occurred in a patient receiving chloroquine sulfate therapy. This patient had a severe congenital renal failure that presumably increased plasma and tissue levels of the drug substantially, accounting for the hypopigmentation.

INTRODUCTION: We report a case series of pediatric patients with incidentally discovered atypical maculopathy, namely, benign foveal depigmentation of the retinal pigment epithelium (BFD). Longitudinal data, both functional and morphological, were analyzed.
MATERIALS AND METHODS: Functional tests (perimetry, full-field, multifocal and pattern ERG) were performed using the Mon PackOne® unit (Métrovision, Pérenchies, France), and multimodal retinal imaging was carried out using the Heidelberg Retinal Tomograph 2 and Heidelberg Spectralis SD-OCT (Heidelberg Inc., Germany).
RESULTS: Four patient

Accumulating evidence has described the involvement of mTORC1 signaling in pigmentation regulation; however, the precise mechanism is not fully understood. Here, we generated mice with conditional deletion of the mTORC1 suppressor Tsc2 in melanocytes. It resulted in constitutive hyperactivation of mTORC1 and reduced skin pigmentation. Mechanistically, neither the number of melanocytes nor the expression of melanogenesis-related enzymes was decreased; however, endoplasmic reticulum and mitochondrial oxidative stress and lower melanization in melanosomes were observed. By contrast, abrogation of

Cyclin-dependent kinase (CDK) 4/6 inhibitors are targeted therapies widely used in the treatment of hormone receptor-positive, human epidermal growth factor receptor 2 (HER2)-negative advanced breast cancer. Although these agents are generally well tolerated, several cutaneous adverse effects have been reported, including rare cases of vitiligo-like depigmentation. Such pigmentary changes may affect treatment adherence and quality of life. Interestingly, emerging evidence suggests a possible association between these immune-mediated skin manifestations and improved survival outcomes. Further s

Pathogenic variants in the OCA2 gene result in oculocutaneous albinism. In humans and several other mammalian species, OCA2 is transcribed i … redicting that it could exert a dominant-negative effect, which is consistent with a dose-dependent hypopigmentation response in both pathological and physiological contexts. Overall, the modulation of OCA2 exon 10 skipping by both benign and pathogenic variants provides a basis for improving the genetic diagnosis of albinism, and paves the way for research into t …

Neonatal lupus erythematosus (NLE) is an autoimmune disease characterized by a periorbital erythematous rash. Although post-inflammatory hypopigmentation and telangiectasias are known possible sequelae, these features may be particularly noticeable in skin of color. Herein, we describe two infants with skin of color in whom periorbital hypopigmentation and telangiectasias were clues to the diagnosis of NLE.

BACKGROUND: Waardenburg syndrome type I (WS-I) is a rare autosomal-dominant auditory-pigmentary disorder with limited reports in the Ophthalmic literature.
MATERIALS AND METHODS: We describe the history, clinical findings and detailed retinal imaging (ultra-widefield fundus images, fundus autofluorescence and optical coherence tomography) from a patient with WS-I.
CASE DESCRIPTION: Our patient had a history of white forelock and congenital hearing loss. Ophthalmic examination demonstrated iris heterochromia and highly asymmetric choroidal hypopigmentation, with generalised fundus hyperautofl…

Inherited diseases of pigmentation were among the first traits studied in humans because of their easy recognition. The discovery of genes that regulate melanocytic development and function and the identification of disease-causative mutations have greatly improved our understanding of the molecular basis of pigmentary genodermatoses and their underlying pathogenetic mechanisms. Pigmentation mutants can account for hypo-/amelanosis, with or without altered melanocyte number, resulting in different phenotypes, such as Waardenburg syndrome, piebaldism, Hermansky-Pudlak syndrome, Chediak-Higashi

Hypopigmentation is a characteristic of several diseases associated with vesicle traffic defects, like the Hermansky-Pudlak, Chediak-Higashi, and Griscelli syndromes. Hypopigmentation is also a characteristic of the zebrafish mutant vps18(hi2499A), which is affected in the gene vps18, a component of the homotypic fusion and protein sorting complex that is involved in tethering during vesicular traffic. Vps18, as part of this complex, participates in the formation of early endosomes, late endosomes, and lysosomes. Here, we show that Vps18 is also involved in the formation of melanosomes. In the…

Patients with oculocutaneous or ocular albinism have misrouting of optic fibers, with fibers from 20 degrees or more of the temporal retina crossing at the chiasm instead of projecting to the ipsilateral hemisphere. Misrouting can result in strabismus and nystagmus. Because patients with the Prader-Willi syndrome may also have hypopigmentation and strabismus, we wondered whether they too might have misrouting of optic fibers. We therefore studied six patients with Prader-Willi syndrome selected for a history of strabismus, using pattern-onset visually evoked potentials with binocular and monoc…

Q-switched 1064 nm neodymium-doped yttrium aluminium garnet (Qs 1064 nm Nd: YAG) laser plays an important role in the treatment of pigmentary skin disorders, including tattoos. Although it has high efficacy and safety, adverse effect like hypopigmentation may occur causing anxiety to patients. We present a case report of Qs 1064 nm Nd: YAG laser induced hypopigmentation which was successfully treated with ultraviolet B targeted phototherapy, with rapid and satisfactory re-pigmentation.

BACKGROUND: Cutaneous hypopigmentation has been reported after steroid injection. Sub-Tenon triamcinolone injection is often administered to control or prevent ocular inflammation.
METHODS: Observational case report.
RESULTS: Lower eyelid cutaneous hypopigmentation developed 6 weeks after sub-Tenon triamcinolone injection administered at the conclusion of scleral buckle and vitrectomy for retinal detachment repair. The hypopigmentation eventually resolved at 7 months postoperatively.
CONCLUSION: Sub-Tenon triamcinolone injection at the conclusion of retinal surgery can cause temporary cutan…
