Potential of preimplantation genomic selection using the blastomere separation technique in bovine in vitro fertilized embryos.

Preimplantation genomic selection combined with an in vitro embryo production system is expected as a means of accelerating genetic improvement in cattle. While micromanipulation-based biopsy approaches are often used to collect embryonic cells for genetic testing, they require expensive equipment and sophisticated skills, hindering the adoption of this system. In the present study, to develop a simple method for preimplantation genomic selection using the blastomere separation (BS) technique in bovine in vitro fertilized embryos, we examined the accuracy of single nucleotide polymorphism (SNP

In vitro evaluation of sun protection factors of sunscreen agents using a novel UV spectrophotometric technique.

A method for the in vitro determination of low- and high-value sun protection factors (SPF) of sunscreens using artificial substrates and a novel pseudo double beam (PDB) mode of operation of a standard double beam UV spectrophotometer is described. The method allows transmittance to be calculated from detector responses of reference and sample beams measured at different gain levels and facilitates the accurate quantification of low levels of electromagnetic radiation transmitted through highly absorbing samples. The spectrophotometer was modified to hold quartz diffusing plates on which a su

An in vitro biomechanical study comparing the spiral linking technique against the pulvertaft weave for tendon repair.

A new spiral linking technique for tendon repair in which one end of the tendon is spiralled around the other end has been developed. Using pig trotter extensor tendons, the Pulvertaft weave technique was compared with this new technique. Twenty-five repairs using each technique were tested by tensile loading with an Instron testing machine. The spiral linking technique matched the strength of Pulvertaft method: the mean peak loads were 102 and 105 N, respectively. The Pulvertaft weave was stiffer than the spiral linking technique: mean stiffness of 11.1 and 6.7 N/mm, respectively. The spiral

Biomechanical Impact of Cementation Technique Variations on Femoral Stem Stability: An In Vitro Polyurethane Model Study.

Background/Objectives: Achieving optimal primary stability in cemented total hip arthroplasty remains a critical factor influencing long-term implant success. Variability in cementation techniques can significantly affect biomechanical performance, yet consensus on best practices is lacking. This study investigates the influence of cementation parameters on femoral stem fixation. Methods: This in vitro comparative study evaluated four cementation techniques-Classic (line-to-line), Press-Fit (undersized reaming), Overreaming (oversized reaming), and Valgus Malpositioning (15° deviation). An exp

A biomechanical in-vitro study on an alternative fixation technique of the pubic symphysis for open book injuries of the pelvis.

PURPOSE: Implant failure rates remain high after plate fixation in pelvic ring injuries. The aim of this study was to compare an alternative fixation technique with suture-button devices and anterior plate fixation in partially stable open-book injuries. MATERIAL AND METHODS: We acquired 16 human fresh frozen anatomic pelvic specimens. The sacrospinous, sacrotuberous, and anterior sacroiliac ligaments were bilaterally released, and the pubic symphysis transected to simulate a partially stable open-book (AO/OTA 61-B3.1) injury. The specimens were randomly assigned to the two fixation groups. I

Feasibility of Meckel's scan after RBC gastrointestinal bleeding study using in-vitro labeling technique.

The authors report a successful Meckel's scan performed 26 hours after a labeled RBC study using a commercial in-vitro labeling kit, UltraTag (Mallinckrodt Medical, Inc., St. Louis, MO). No abnormal alteration of Tc-99m distribution was observed as would be seen if an in-vivo RBC labeling technique was used. This case demonstrates that it is feasible to perform an in-vitro labeled RBC study for acute gastrointestinal bleeding in pediatric patients and to follow-up with a Meckel's scan if it is necessary for diagnostic confirmation.

Evaluation of the in vitro transformation technique to distinguish Trypanosoma evansi from cyclically transmitted Trypanozoon stocks.

In order to initiate transformation into procyclic forms, bloodstream trypanosomes, were transferred to semi-defined medium at 27 degrees C. All stocks previously classified as Trypanosoma brucei rhodesiense, T.b. gambiense and T.b. brucei transformed into procyclic forms. None of the 31 characterized T. evansi stocks transformed into procyclic forms, but died between day 4 and day 6 in culture. On the other hand, 3 out of 64 stocks of monomorphic brucei subgroup field isolates from camels transformed into procyclic forms, confirming the existence of T.b. brucei infection in camels kept close

Normothermic rapid volume replacement for hypovolemic shock: an in vivo and in vitro study utilizing a new technique.

Hypovolemic shock secondary to intraoperative or traumatic hemorrhage requires urgent, aggressive resuscitation to achieve a successful outcome. Common difficulties encountered include the need for venous access, restoration of blood volume, and most important, maintenance of normothermia. The rapid solution administration set utilized in this study addresses the above limitations. Venous access is quickly accomplished by the percutaneous insertion of a large-bore catheter into the central venous system. The set requires only one central venous entry site for adequate fluid resuscitation. Expe

In vitro and in vivo technique for assessing vagus nerve regeneration after parietal cell vagotomy in the rat.

This study determined if the vagus nerve can regenerate and/or reinnervate the gastric parietal cell mass after parietal cell vagotomy (PCV) and examines tests for assessing vagus nerve regeneration in rats. Microscopic dissection of the neurovascular bundle allowed the vagus nerve to be divided at the gastric body with preservation of the antropyloric nerve and gastric vasculature. Gastric secretory tests were performed under basal and stimulated conditions using secretagogues and insulin hypoglycemia. The candidate hormone, pancreatic polypeptide, was measured in plasma following a mixed mea
