Indoxyl glucuronide, a protein-bound uremic toxin, inhibits hypoxia-inducible factor‒dependent erythropoietin expression through activation of aryl hydrocarbon receptor.

Renal anemia is common among chronic kidney disease (CKD) patients, and is mainly caused by inadequate erythropoietin (EPO) production from kidneys due to dysfunction of intracellular hypoxia-inducible factor (HIF) signaling in renal EPO-producing cells. We have previously shown that indoxyl sulfate (IS), a representative protein-bound uremic toxin accumulated in the blood of CKD patients, inhibits hypoxia-induced HIF activation and subsequent EPO production through activation of aryl hydrocarbon receptor (AHR). In this study, we further investigated the effects of other protein-bound uremic t

Platelet MAO-B activity as a marker of behavioural characteristics in dementia disorders.

Both low and high platelet MAO-B (pMAO-B) activity is considered an indicator of increased vulnerability in psychopathology. How the activity of this peripheral enzyme can be linked with the sophisticated functions of the central nervous system (CNS) is not clear; in man, evidence exists that the genetic mechanisms determining the size or capacity of the central serotonin system are common to platelet and brain MAO. In the present study pMAO-B activity was evaluated in demented patients suffering from early-onset Alzheimer's disease (AD), late-onset Alzheimer's disease (SDAT), vascular dementi

Urinary metabolomic profiling to identify the impact of the increased intake of whole grains, vegetables, and fruits in healthy adolescents.

BACKGROUND: A plant-based diet is reportedly effective in preventing chronic and metabolic diseases, primarily because of the antioxidant properties of phytochemicals present in vegetables and fruits. However, systematic metabolomics studies investigating the changes in metabolic phenotypes resulting from plant-based diets in adolescents are lacking. Therefore, this study aimed to examine alterations in urinary metabolomic profiles associated with a plant-based diet to explore its beneficial metabolic effects. METHODS: Eighteen healthy Korean adolescents participated in an 8-week intervention

N,N-dimethyltryptamine (DMT) is neither formed nor retained in serotonin terminals in the rat brain.

Mammalian brain may contain an endogenous pool of the psychedelic substance N,N-dimethyltryptamine (DMT), which may act as a co-transmitter with serotonin (5-HT). We tested the joint hypotheses that endogenous DMT would accumulate in rat brain after inhibiting monoamine oxidase with pargyline, whereas its acidic metabolite 3-indoleacetic acid (3-IAA) would accumulate after pretreatment with the inhibitor of acidic metabolic transport, probenecid. We also tested the hypothesis that pretreatment with inhibitors of plasma membrane 5-HT uptake (escitalopram, ESC) or the vesicular monoamine transpo

A novel indole compound MA-35 attenuates renal fibrosis by inhibiting both TNF-α and TGF-β1 pathways.

Renal fibrosis is closely related to chronic inflammation and is under the control of epigenetic regulations. Because the signaling of transforming growth factor-β1 (TGF-β1) and tumor necrosis factor-α (TNF-α) play key roles in progression of renal fibrosis, dual blockade of TGF-β1 and TNF-α is desired as its therapeutic approach. Here we screened small molecules showing anti-TNF-α activity in the compound library of indole derivatives. 11 out of 41 indole derivatives inhibited the TNF-α effect. Among them, Mitochonic Acid 35 (MA-35), 5-(3, 5-dimethoxybenzyloxy)-3-indoleacetic acid, showed the

γ-Aminobutyric Acid Alleviates Discoloration of Or Mutant Cauliflower via BoMYB24-Mediated Transcriptional Regulation of BoLCYB.

Orange (Or) mutant cauliflower is prone to discoloration during storage, which affects its commercial value. γ-Aminobutyric acid (GABA) treatment can effectively delay the discoloration of Or mutant cauliflower during storage; however, its mechanism of action remains unclear. Here, we aimed to identify key genes and candidate transcription factors associated with discoloration in Or cauliflower treated with GABA during storage. Transcriptome analysis revealed that GABA regulated BoLCYB, with BoMYB24 identified as the candidate transcription factor in this pathway. BoMYB24 binds the BoLCYB prom

Analysis of phytohormones in vermicompost using a novel combinative sample preparation strategy of ultrasound-assisted extraction and solid-phase extraction coupled with liquid chromatography-tandem mass spectrometry.

Vermicompost (VC), a widely used premium organic fertilizer, is the by-product of symbiotic interactions between earthworms and microorganisms living within them. It has been postulated that phytohormones are plausible "magic compounds" in VC that are responsible for making them such good fertilizers. Thus, a novel approach involving ultrasound-assisted extraction (UAE) and solid-phase extraction (SPE) was developed as a fast and efficient sample preparation method to screen for different classes of phytohormones in VC by liquid chromatography-tandem mass spectrometric (LC-MS/MS) analysis. Plant growth regulators and fertilizer increase phytoextraction efficiency of Tagetes minuta under lead-contaminated soil.

Soil contamination with heavy metals severely threatens food security and human health worldwide. Heavy metal contamination must be controlled through sustainable and economical technologies. To enhance the phytoextraction of lead (Pb), the plants with resistance against Pb stress and maximum production of biomass could serve as a potential candidate for the phytoremediation of Pb-contaminated soils. The current study was conducted to illuminate the effect of gibberellic acid (GA3), indole acetic acid (IAA), and fertilizers (NPK) on plant growth and Pb phytoextraction capabilities of Tagetes m

Design, synthesis, and structure-activity relationships of ω-aryloxy-α-hydroxyalkanoates as selective lateral root inhibitors.

We synthesized a series of ω-aryloxy-α-hydroxyalkanoates and evaluated their effects on Arabidopsis thaliana root architecture. We found that compounds with an odd number of methylene linkers (such as JAX-64) exhibited highly potent overall inhibitory activity for primary root growth and lateral root formation. Structural tuning-such as introducing an ortho-methoxy group or converting the ester to an amide-effectively attenuated the severe primary root inhibition while maintaining sufficient lateral root suppression, yielding highly selective inhibitors. It may be considered that these optimiz

The plant-like protein phosphatase PPKL regulates parasite replication and morphology in Toxoplasma gondii.

BACKGROUND: The protozoan parasite Toxoplasma gondii encodes dozens of phosphatases, among which a plant-like phosphatase absent from mammalian genomes named PPKL, which is involved in regulating brassinosteroid signaling in Arabidopsis, was identified in the genome. Among the Apicomplexa parasites, T. gondii is an important and representative pathogen in humans and animals. PPKL was previously identified to modulate the apical integrity and morphology of the ookinetes and parasite motility and transmission in another important parasite, Plasmodium falciparum. However, the exact function of PP

Colonization with non-mycorrhizal culturable endophytic fungi enhances orchid growth and indole acetic acid production.

BACKGROUND: Symbiotic associations of endophytic fungi have been proved by possessing an ability to produce hormones and metabolites for their host plant. Members of the Orchidaceae are obligate mycorrhizal species but a non-mycorrhizal association needs more investigation for their ability to promote plant growth and produce plant growth hormones. In the present study, endophytic fungi were isolated from the roots of Dendrobium longicornu Lindl., to investigate the root colonizing activity and role in plant growth and development. RESULTS: Among 23 fungal isolates were identified both by mor

Detoxification of phenanthrene in Arabidopsis thaliana involves a Dioxygenase For Auxin Oxidation 1 (AtDAO1).

Polycyclic aromatic hydrocarbon (PAH) contamination has a negative impact on ecosystems. PAHs are a large group of toxins with two or more benzene rings that are persistent in the environment. Some PAHs can be cytotoxic, teratogenic, and/or carcinogenic. In the bacterium Pseudomonas, PAHs can be modified by dioxygenases, which increase the reactivity of PAHs. We hypothesize that some plant dioxygenases are capable of PAH biodegradation. Herein, we investigate the involvement of Arabidopsis thaliana At1g14130 in the degradation of phenanthrene, our model PAH. The At1g14130 gene encodes Dioxygen

Auxins, the hidden player in chloroplast development.

Throughout decades of plant research, the plant hormones known as auxins have been found to be of vital importance in most plant development processes. Indole-3-acetic acid (IAA) represents the most common auxin in plants and can be synthesized from its tryptophan precursor, which is synthesized in the chloroplast. The chloroplast constitutes an organelle of great relevance to plants since the photosynthesis process by which plants get most of their energy is carried out there. The role of auxins in photosynthesis has been studied for at least 50 years, and in this time, it has been shown that

Indole-3-acetic acid synthesized through the indole-3-pyruvate pathway promotes Candida tropicalis biofilm formation.

We previously found that the elevated abundance of the fungus Candida tropicalis is positively correlated with the bacteria Escherichia coli and Serratia marcescens in Crohn's disease patients and the three pathogens, when co-cultured, form a robust mixed-species biofilm. The finding suggests that these three pathogens communicate and promote biofilm formation, possibly through secretion of small signaling molecules. To identify candidate signaling molecules, we carried out a metabolomic analysis of the single-species and triple-species cultures of the three pathogens. This analysis identified

Hormonal control of second flushing in Douglas-fir shoots.

Spring-flushing, over-wintered buds of Douglas-fir (Pseudotsuga menziesii (Mirb.) Franco) produce new buds that may follow various developmental pathways. These include second flushing in early summer or dormancy before flushing during the following spring. Second flushing usually entails an initial release of apical dominance as some of the current-season upper lateral buds grow out. Four hypotheses concerning control of current bud outgrowth in spring-flushing shoots were tested: (1) apically derived auxin in the terminal spring-flushing shoot suppresses lateral bud outgrowth (second flushin

Etodolac. A reappraisal of its pharmacology and therapeutic use in rheumatic diseases and pain states.

Etodolac is a nonsteroidal anti-inflammatory drug (NSAID) effective in the treatment of rheumatoid arthritis, osteoarthritis and ankylosing spondylitis, and in the alleviation of postoperative pain. Etodolac also provides relief of other types of pain, including that arising from gouty conditions and traumatic injury. In all indications, etodolac appears to be at least as effective as other NSAIDs. The incidence of clinical adverse effects other than abdominal pain and dyspepsia is similar to that observed with placebo, and etodolac has been associated with a low rate of gastrointestinal ulcer

Protein S-nitrosylation: roles for nitric oxide signaling in the regulation of stress-dependent phytohormones.

Nitric oxide (NO) is integral to modulating a wide array of physiological processes in plants, chiefly through its interactions with phytohormones. This review explores the complex dynamics between NO with four principal phytohormones: abscisic acid (ABA), salicylic acid (SA), auxin, and jasmonic acid (JA). The primary focus is on NO-mediated reversible redox-based modifications with a specific emphasis on S-nitrosylation. The impact of NO-induced S-nitrosylation is profound as it regulates crucial proteins involved in hormone signaling pathways, thereby influencing their synthesis, stability,

Revealing Differential Expression of Phytohormones in Sorghum in Response to Aphid Attack Using the Metabolomics Approach.

Sorghum (Sorghum bicolor) is an important multipurpose crop grown worldwide, but like many other crops, it is often threatened by insect pests. Sugarcane aphid (SCA, Melanaphis sacchari Zehntner), for example, is one of the most severe pests in sorghum, which causes plant damage and yield loss. The main objective of this study was to assess the effect of phytohormones on host plant resistance to aphid attack. Two sorghum genotypes, BTx623 (susceptible) and Tx2783 (resistant), were selected for a comparative analysis of differential expression of a group of phytohormones in response to aphid in
