Quantifying the risks and benefits of natalizumab in relapsing multiple sclerosis.

Using published data, we quantified the risk and benefits of natalizumab in relapsing multiple sclerosis using quality-adjusted life years (QALYs) as a metric. Over the first 2 years of therapy, the negative health effects from progressive multifocal leukoencephalopathy were small (loss of 0.001 QALYs) relative to the positive effects on relapses and disability resulting in 0.033 QALYs (12 quality-adjusted days) gained. For context, we performed an analogous calculation for interferon beta-1a, which also had a net health benefit of 0.033 QALYs (12 quality-adjusted days).

Cloning and Optimization of Intracellular Expression of Human Interferon β-1a in Pichia pastoris GS115.

BACKGROUND: Interferon-beta (IFN-β) is a cytokine with a wide range of biological and pharmaceutical applications, including multiple sclerosis (MS), cancer, some autoimmune disorders, and viral infectious diseases. Thus, many studies have been performed to develop novel strategies for the high-yield production of functional IFN-β in a cost-effective approach. Here, we aimed to improve the intracellular expression of IFN-β-1a in Pichia pastoris. MATERIALS AND METHODS: The gene of IFN-β-1a was successfully sub-cloned into the pPICZA vector. The recombinant vector was transfected to P. pastoris

Successful treatment of a woman with relapsing neuromyelitis optica by interferon beta.

Neuromyelitis optica (NMO) is an antibody-mediated disease with the target antigen being the water channel aquaporin-4 (AQP4). More than 90% of NMO patients have relapse. Optimal treatments to prevent recurrence have not been established. Previous reports suggest that interferon beta (IFNB) has not been effective in NMO. However, we present a case of NMO with positive anti-myelin oligodendrocyte glycoprotein (MOG)(27-38) IgG and anti-AQP4 IgG treated with IFNB-1a successfully. The effect of IFNB on NMO and the role of anti-MOG antibodies in NMO deserve further study.

Effectiveness of first-line treatment for relapsing-remitting multiple sclerosis in Brazil: A 16-year non-concurrent cohort study.

BACKGROUND: Relapsing-remitting multiple sclerosis (RRMM) is a chronic, progressive, inflammatory and immune-mediated disease that affects the central nervous system and is characterized by episodes of neurological dysfunction followed by a period of remission. The pharmacological strategy aims to delay the progression of the disease and prevent relapse. Interferon beta and glatiramer are commonly used in the Brazilian public health system and are available to patients who meet the guideline criteria. The scenario of multiple treatments available and in development brings the need for discussi

Abrogating doxorubicin-induced chemobrain by immunomodulators IFN-beta 1a or infliximab: Insights to neuroimmune mechanistic hallmarks.

Chemobrain is a well-established clinical syndrome that impairs patient's daily function, in particular attentiveness, coordination and multi-tasking. Thus, it interferes with patient's quality of life. The putative pharmacological intervention against chemobrain relies on understanding the molecular mechanisms underlying it. This study aimed to examine the potential neuroprotective effects of two immunomodulators: Interferon-β-1a (IFN-β-1a), as well as Tumor necrosis function-alpha (TNF-α) inhibitor; Infliximab in doxorubicin (DOX)-induced chemobrain in rats. Besides, the current study target

Urinary tract infections, urosepsis, and multiple sclerosis disease modifying therapies: a disproportionality analysis of the FDA Adverse Event Reporting System.

Infection-related complications, including urinary tract infections (UTIs) and urosepsis, account for a significant proportion of morbidity and mortality in patients with advanced multiple sclerosis (MS). However, the role of specific disease-modifying therapies (DMTs) in UTI and urosepsis risks remains underexplored. We identified 12,997 UTI and 651 urosepsis reports associated with various MS in the FDA Adverse Event Reporting System. Ocrelizumab exhibited the greatest safety signal for UTI, while natalizumab, alemtuzumab, and interferon beta-1a were also associated with disproportionately h

Magnetization transfer ratio in new MS lesions before and during therapy with IFNbeta-1a.

OBJECTIVE: The authors examined the effect of 6.0 MIU interferon beta-1a (IFNbeta-1a) administered IM each week on the evolution of monthly magnetization transfer ratio (MTR) within new gadolinium-enhancing (Gd+) lesions in patients with very early relapsing-remitting (RR) MS. BACKGROUND: IFNbeta is an effective disease-modifying treatment for patients with RRMS. Among other effects, it has been shown to decrease the number of new Gd+ and T2-weighted lesions. MTR is a putative marker for irreversible tissue damage and evolution of MTR within a lesion may reflect recovery of tissue damage. It
