Multispectral and molecular dynamics investigation of GABA-induced gelation behavior in walnut protein isolate.

Walnut protein isolate (WPI) is a high-quality plant protein source. However, its application in gel-based foods is limited by weak gelling properties. The present study elucidated the molecular mechanisms by which γ-aminobutyric acid (GABA) enhanced the gelation of WPI. The addition of GABA (0.80%) significantly increased the water holding capacity of WPI gels (from 61.9% to 85.2%) and improved the textural properties, including hardness and springiness. Mechanistically, GABA induced protein unfolding (transitioning α-helix to ordered β-sheets), exposing buried hydrophobic groups. This facili

Multi-omics analysis of microbial succession and metabolite dynamics in cold-stored walnut kernels.

Fresh walnut kernels are highly susceptible to microbial contamination and quality deterioration during cold storage due to their high moisture content and active physiological metabolism, posing challenges to storage safety. This study systematically investigated the microbial community succession, metabolite dynamics, and quality changes of dehusked fresh walnut kernels during 90 days of cold storage at 4 °C. The results showed that the bacterial community was dominated by Proteobacteria (83.35 %-87.13 %) throughout storage, with the initially dominant Paucibacter showing a slight decrease i

Chemical Characterization and Beneficial Effects of Walnut Oil on a Drosophila melanogaster Model of Parkinson's Disease.

A nutritional approach could be a promising strategy to prevent or decrease the progression of neurodegenerative disorders such as Parkinson's disease (PD). The neuroprotective role of walnut oil (WO) was investigated in Drosophila melanogaster treated with rotenone (Rot), as a PD model, WO, or their combination, and compared to controls. WO reduced mortality and improved locomotor activity impairment after 3 and 7 days, induced by Rot. LC-MS analyses of fatty acid levels in Drosophila heads showed a significant increase in linolenic (ALA) and linoleic acid (LA) both in flies fed with the WO-e

Walnut peptide alleviates obesity, inflammation and dyslipidemia in mice fed a high-fat diet by modulating the intestinal flora and metabolites.

INTRODUCTION: Obesity is a chronic disease in which the body stores excess energy in the form of fat, and intestinal bacterial metabolism and inflammatory host phenotypes influence the development of obesity. Walnut peptide (WP) is a small molecule biopeptide, and the mechanism of action of WP against metabolic disorders has not been fully elucidated. In this study, we explored the potential intervention mechanism of WP on high-fat diet (HFD)-induced obesity through bioinformatics combined with animal experiments. METHODS: PPI networks of Amino acids and their metabolites in WP (AMWP) and "ob

Toxicological Effects of Aqueous Extract From African Walnut ( Tetracarpidium conophorum) Leaves in Rats.

Tetracarpidium conophorum leaves are used in traditional medicine for the treatment of male infertility, without considering its toxicity and side effects. In this study, we investigated the effects of T conophorum leaves on some biochemical parameters such as alanine aminotransferase, aspartate aminotransferase, bilirubin, albumin, creatinine, and uric acid. Histology of the liver and kidney were also assessed. The result revealed that the alanine aminotransferase and aspartate aminotransferase levels of the control group were not significantly different from the experimental groups. There wa

Walnut-Derived Extracellular Vesicles Orchestrate a Pre-Regenerative Niche via c-Myc Mediated Metabolic Reprogramming.

Peripheral nerve injury (PNI) remains a major regenerative challenge, in part because the post-injury microenvironment can disrupt Schwann cell (SCs) homeostasis. Walnuts (Juglans regia) have long been used in ethnomedicine for perceived neurotrophic or neuroprotective benefits, a view historically linked to their resemblance to the brain. To examine whether this traditional concept can be leveraged as a nanotherapeutic approach, we isolated walnut-derived extracellular vesicles (WEVs) and evaluated their effects on peripheral nerve repair. We found that WEVs are readily internalized by SCs an…

Characterization of Walnut Oil and Evaluation of Its Neuroprotective Effects in an In Vitro Model of Parkinson's Disease.

Parkinson's disease (PD) is a common neurodegenerative disorder marked by the degeneration of dopaminergic neurons and the buildup of α-synuclein aggregates. The current treatments focus on symptom relief, with no drugs available to halt disease progression. This has prompted interest in plant-based extracts as alternative therapies. This study examines the neuroprotective and antioxidant effects of walnut oil (WO), extracted from Juglans regia L., in an in vitro PD model using the neurotoxin rotenone (ROT). WO, rich in polyunsaturated fatty acids (PUFAs), including linoleic acid (LA) and α-li…

Insight into succinylated modified walnut protein isolate: structural and functional properties, and curcumin release behavior.

BACKGROUND: Due to its high glutenin content, walnut protein isolate (WPI) exhibits low solubility, limiting its applications in the food industry. RESULTS: WPI was modified with succinic anhydride (SA), and the potential of the resulting curcumin-loaded emulsions was evaluated. The degree of succinylation of WPI was measured as 89.52 ± 0.66% when 40% SA was used. Succinylation enhanced the surface charge of WPI, reduced its surface hydrophobicity and disulfide bond content, and involved both lysine residues and free sulfhydryl in the acylation reaction. Structural analyses, including Fourier

Workflow for characterizing volatile oxidation compounds in walnut oil by headspace solid-phase microextraction comprehensive two-dimensional gas chromatography time of flight mass spectrometry coupled with tile-based Fisher ratio analysis and other chemometric tools.

Due to their high fat content, it is well known that walnuts are highly susceptible to oxidation and thermal degradation. These processes produce various compounds, including aromatics, aldehydes, acids, ketones, and alcohols, resulting in poor quality walnuts and shorter shelf life. The goal of this work was to demonstrate a high-level analytical workflow to follow and capture the kinetic volatile fingerprint of oxidized walnut oil through development of a method using headspace solid-phase microextraction sampling followed by comprehensive two-dimensional gas chromatography time-of-flight ma

Efficient and effective immobilization of tetracycline and copper from wastewater by zero-valent iron fabricated hydrochar derived from walnut peel.

Antibiotics and heavy metals often coexist as non-point-source contaminants in wastewater and their quite contrary physiochemical properties make their co-removal processes challenging. In this work, a bifunctional zero-valent iron-modified hydrochar derived from walnut peel (MWPHC) was synthesized, which was then applied for the simultaneous removal of tetracycline (TC) and Cu(II) from wastewater. Based on the characterizations, Fe0 species were successfully distributed on the surface of the walnut peel substrates. The TC and Cu(II) could be synergistically immobilized, and bridging effects w
