Archaeal tyrosinase as a versatile biocatalyst for lignin-derived aromatic compounds valorization.

Biomass-derived aromatic compounds, including those obtained from lignin (which is the most abundant aromatic biopolymer on Earth), are valuable for sustainable chemical production. Various lignin-degrading approaches have been developed to cleave recalcitrant bonds. The incorporation of biocatalysts that operate under environmentally friendly and mild conditions with high substrate specificity is considered one of the emerging strategies for lignin valorization. In this study, an archaeal tyrosinase (Tyr-CNK), derived from the marine archaeon Candidatus nitrosopumilus koreensis, is characteri…

Acid-Catalyzed Solvolysis of Softwood in Caprylyl Glycol to Produce Lignin Derivatives.

Glycol lignin (GL) is produced via the acid-catalyzed solvolysis of softwood meal using glycols such as polyethylene glycol. The physicochemical and thermal properties of GL are expected to be controllable by varying the glycol type. In this study, caprylyl glycol (CG), which is a biobased glycol derived from the caprylic acid component of coconut and palm oils, was used to investigate the effects on the acid-catalyzed solvolysis of softwood. The reactions were performed at 140, 150, and 160 °C for 30-120 min to obtain lignin derivatives (CG-lignin: CGL). The chemical and physical properties o…

Effect of time series on the degradation of lignin by Trametes gibbosa: Products and pathways.

Lignin is the third most abundant organic resource in nature. The utilization of white-rot fungi for wood degradation effectively circumvents environmental pollution associated with chemical treatments, facilitating the benign decomposition of lignin. Trametes gibbosa is a typical white-rot fungus with rapid growth and strong wood decomposition ability. The lignin content decreased from 23.62 mg/mL to 17.05 mg/mL, which decreased by 27 % in 30 days. The activity of manganese peroxidase increased steadily by 9.44 times. The activities of laccase and lignin peroxidase had the same trend of chang…

On the fractionation of lignin oligomers by stepwise gradient reversed-phase liquid chromatography.

With the increased interest in lignin valorization, the analytical challenge to separate a complex mixture of a vast number of phenolics has made chromatography an indispensable step in lignin analysis. High-resolution separations, such as gas chromatography, reversed-phase liquid chromatography and supercritical fluid chromatography have typically been targeting low-molecular-weight compounds, while larger lignin oligomers have received less attention. These compounds have proven to be difficult to separate due to the inherent complexity of the high-molecular-weight fraction of lignins, in fa…

A hydrotrope pretreatment for stabilized lignin extraction and high titer ethanol production.

The biomass pretreatment strategies using organic acids facilitate lignin removal and enhance the enzymatic digestion of cellulose. However, lignin always suffers a severe and irreversible condensation. The newly generated C-C bonds dramatically affect its further upgrading. In this study, we used a recyclable hydrotrope (p-Toluenessulfonic acid, p-TsOH) to dissolve lignin under mild condition and stabilized lignin with a quenching agent (formaldehyde, FA) during extraction, achieving both value-added lignin extraction and efficient enzymatic saccharification of cellulose. Approximately 63.7% …

Production of Aromatic Compounds by Catalytic Depolymerization of Technical and Downstream Biorefinery Lignins.

Lignocellulosic materials are promising alternatives to non-renewable fossil sources when producing aromatic compounds. Lignins from Populus salicaceae. Pinus radiata and Pinus pinaster from industrial wastes and biorefinery effluents were isolated and characterized. Lignin was depolymerized using homogenous (NaOH) and heterogeneous (Ni-, Cu- or Ni-Cu-hydrotalcites) base catalysis and catalytic hydrogenolysis using Ru/C. When homogeneous base catalyzed depolymerization (BCD) and Ru/C hydrogenolysis were combined on poplar lignin, the aromatics amount was ca. 11 wt.%. Monomer distributions chan…

Recent Advances in the Application of Functionalized Lignin in Value-Added Polymeric Materials.

The quest for converting lignin into high-value products has been continuously pursued in the past few decades. In its native form, lignin is a group of heterogeneous polymers comprised of phenylpropanoids. The major commercial lignin streams, including Kraft lignin, lignosulfonates, soda lignin and organosolv lignin, are produced from industrial processes including the paper and pulping industry and emerging lignocellulosic biorefineries. Although lignin has been viewed as a low-cost and renewable feedstock to replace petroleum-based materials, its utilization in polymeric materials has been …

Fractionation of enzymatic hydrolysis lignin by sequential extraction for enhancing antioxidant performance.

The heterogeneity of lignin chemical structure and molecular weight results in the lignin inhomogeneous properties which also covers the antioxidant performance. In order to evaluate the effects of lignin heterogeneity on its antioxidant activity, four lignin fractions from enzymatic hydrolysis lignin were classified by sequential organic solvent extraction and further evaluated by DPPH (1,1-Diphenyl-2-Picrylhydrazyl) free radical scavenging capacity and reducing power analysis. The characterization including FTIR, 1H NMR and GPC showed that the fractionation process could effectively separate…

[Changes of phenols and lignin contents in alfalfa leaf damaged by Odontothrips loti].

In order to investigate the changes of secondary metabolites content of alfalfa induced by thrips Odontothrips loti damaging, two alfalfa strains, one resistant to thrips (R-1) and the other susceptible to thrips (I-1) , were chosen to measure the phenols and lignin contents of alfalfa leaves under infestation with thrips at different densities (0, 1, 3, 5, 7 thrips x branch(-1), and 0 thrip x branch(-1) as control). After infestation 7 days, the polyphenols, tannin and condensed tannin contents increased in both leaves of R-l and I-i with the increasing thrips density, the simple phenols cont…

Halide-mediated electrochemical functionalization of lignin for high-performance flexible humidity sensors.

Lignin is an abundant, renewable aromatic biopolymer that remains largely underutilized despite its rich chemical potential. In this work, lignin is valorized through a mild electrochemical functionalization process using chloride ions as a redox mediator on a boron-doped diamond electrode. This halide-mediated oxidation introduces oxygen-containing functional groups, while simultaneously inducing particle fragmentation and increasing surface accessibility. These structural modifications significantly enhance the hydrophilicity and interfacial interaction of lignin with gold electrodes. The el…

Lignin macromolecular structural stability drives sapwood-heartwood color homogeneity in Populus deltoides.

Radial xylem maturation in woody plants typically involves programmed cell death (PCD) and accumulation of colored secondary metabolites, leading to distinct color convergent between sapwood and heartwood. In fast-growing P. deltoides, however, the xylem exhibits remarkable color convergence along the radial gradient. Through the integration of anatomical observations, chemical analyses, and advanced spectroscopy (FTIR, Raman, 2D-HSQC NMR), color convergence was analyzed from two coordinated regulatory features: (1) a non-condensed lignin structure characterized with an elevated syringyl/guaia…

Preparation and performance of rapid water activation lignin-based wood adhesive films.

To advance lignin utilization in wood adhesives, this work presents the development of poly(vinyl alcohol)-lignin (PVAL) films by solution casting. Water spray activation rapidly switched the film from a dormant to an adhesive state, enabling immediate hot-pressing of plywood. Bonding performance was assessed by tensile strength testing. A systematic parametric study was conducted on the PVA concentration, lignin addition amount (relative to PVA), pH value, and film thickness to determine the optimal formulation. This was shown to be formed by a 12 wt% aqueous PVA solution, 50 wt% lignin relat…

Activity-Coefficient-Guided Heuristics for Deep Eutectic Solvent Fractionation of Kraft Lignin.

The valorization of technical lignin requires fractionation strategies that deliver chemically and thermally distinct lignin streams with predictable structure-property relationships while minimizing hazardous solvent use. Here, we propose a preliminary, exploratory thermodynamic design framework for lignin fractionation based on deep eutectic solvents (DESs), using activity coefficients as quantitative descriptors of solvent-solute interactions. Six chloride-based DESs were synthesized and characterized through freezing-point depression, FTIR spectroscopy, and Schröder-Le Chatelier analysis. …

Structural transformation strategy of lignin to humic acid during biological acclimated composting.

Humic acid (HA) generated during straw-based fertilization is a key component in restoring the quality of black soil. However, strategies for manipulating lignin structure to regulate HA formation during acclimated composting remain unclear. In this study, rice straw (RS), willow branches (TB), and pine needles (PN), each characterized by distinct lignin structures, were composted to investigate the microbial conversion pathways from lignin to HA. Four representative functional groups in lignin were identified: aryl-ether CO bonds, side-chain CH groups, dimeric OH groups, and benzene-ring skel…

Toward high-performance supercapacitors: Electrodes from solvent-fractionated lignin with enhanced activity.

The presence of lignin-rich oxygen-containing groups significantly increases the potential of supercapacitors. However, the heterogeneity of lignin limits their use in industry. In this study, eucalyptus wood was used as the raw material to obtain highly reactive fractionated lignin through solvent extraction and fractionation processes. After fractionation, the weight-average molecular weight decreased from 27,760 to 19,665, and the polydispersity decreased from 2.70 to 2.11. The fractionated lignin was subsequently used to partially replace phenol, synthesizing a series of lignin-based pheno…

Understanding Lignin Radical Dynamics: Quenching Radicals by Solvent and Thermal Induced Mobility.

Lignin contains persistent free radicals (PFRs) stabilized by restricted molecular mobility in a glassy state. Herein, we investigate quenching effects in softwood kraft lignin PFRs by solvation and thermal treatment. Electron paramagnetic resonance (EPR) spectroscopy and temperature-modulated differential scanning calorimetry (TM-DSC) were used to characterize changes. Room-temperature methanol or acetone swelling reduced the paramagnetic signal to 48% and 71% of the original intensity. Similarly, heating lignin above its glass transition temperature (Tg) mobilized trapped radicals, leading t…

Lightly colored and reactive maleic acid hydrotropic lignin: Characterization and polymeric applications.

Acid hydrotropic fractionation using maleic acid, a US FDA approved indirect food additive, can rapidly (20 min) dissolve wood lignin at low temperatures (70-110 °C). This study characterized the physical and chemical properties, i.e., color, molecular weight, solubility, and chemical structure of maleic acid hydrotropic fractionation dissolved wood lignin (MAHL) obtained under various fractionation conditions. The study also demonstrated valorization of MAHL for two polymeric applications through direct mixing with (1) commercial sunscreen for its light color and UV protection capability, and…

Improvement in lignin peroxidase oxidative stability via surface display by Bacillus subtilis.

Lignin peroxidase (LiP) is a key enzyme involved in lignin degradation. However, the production of natural LiP is limited by the low enzyme-producing capacity of native fungal hosts and the poor stability of the enzyme, hindering its industrialization. To overcome these limitations, we prepared an immobilized enzyme by fusing the LiP gene with the Bacillus subtilis (B. subtilis) spore coat protein CotB and displaying it on the bacterial surface. This approach aims to improve enzymatic stability and industrial applicability. Enzymatic characterization showed that the immobilized LiP exhibited o…
