Hypothyroidism and thyroxin substitution affect the n-3 fatty acid composition of rat liver mitochondria.

The effects of hypothyroidism and of daily treatment for up to 21 days with thyroxin (T4, 0.5 micrograms/100 g body weight) on the fatty acid composition of total lipid, phosphatidylethanolamine, and phosphatidylcholine of rat liver mitochondria were studied. The fatty acid compositions of hypothyroid and euthyroid (control) rats of similar age were compared. The n-6 and n-3 polyunsaturated fatty acids (PUFA) were affected differently by the hypothyroid state. The levels of linoleic (18:2n-6), gamma-linolenic (18:3n-6) and dihomo-gamma-linolenic acids (20:3n-6) were higher in hypothyroid rats …

Omega-3 fatty acids and cardiovascular disease.

PURPOSE OF REVIEW: Omega-3 fatty acids are gaining acceptance in the cardiovascular field. The present review describes the most recent studies and developments in the field. RECENT FINDINGS: Marine omega-3 fatty acids, that is eicosapentaenoic and docosahexaenoic acids, prevent fatal myocardial infarction and sudden cardiac death by their antiarrhythmic effects and presumably also by their effect on infarct size, the latter mediated by plaque stabilization, improvements in endothelial function and other mechanisms. In contrast, a cardioprotective effect of alpha-linolenic acid, a plant-deriv

Reducing cellular inflammation and oxidation by supercritical CO2 extracts from edible-medicinal mushrooms.

Extracts from Hericium erinaceus, Ganoderma lucidum and Tremella fuciformis obtained with supercritical CO2 showed high cellular antioxidant activity (CAA) when tested on Caco-2 cell cultures. Extract obtained in separator 2 (5 MPa) after 3 h extraction at 30 MPa and 40 °C from G. lucidum showed a remarkable higher CAA than noticed for the other species. In addition, extracts to THP-1 macrophages reduced secretion of TNF-α and particularly IL-6 (80-90 % applied at 15 μg/mL). The G. lucidum extract was also more effective than those from the other two species except for IL-1β. The use of ethano

Monitoring the optimal infusion of intravenous lipids. Detection of essential fatty acid deficiency.

Since increased amounts of the trienoic acid C20:3w9 are produced in patients with essential fatty acid (EFA) deficiency (EFAD) of the linol … leic acid deficiency was not corrected by the treatment. Plasma levels of the EFA, ie, linoleic and linolenic acids, cannot be predicted from the lipid treatment or from the patient's clinical course; therefore, periodic measurements of EFA are required to monitor the effects of therapy. To provide the most effective therapy, the amounts and proportions of linole …

Profiles of the fatty acids in the plasma membrane of human brain tumors.

Several studies demonstrated that certain fatty acids have specific effects on tumor cells. n-3 series fatty acids (alpha-linolenic acid, eicosapentaenoic acid and docosahexaenoic acid) may suppress the carcinogenesis, whereas n-6 series fatty acids (arachidonic acid, linoleic acid) may exert tumor promoting effects. In this study, 19 patients with various brain tumors and 12 control brain tissues were studied. n-3, n-6, n-9 unsaturated fatty acids and certain saturated fatty acids levels were measured in the plasma membrane of tumor or control brain tissues by capillary gas chromatography. Identification of radical adducts formed in the reactions of unsaturated fatty acids with soybean lipoxygenase using continuous flow fast atom bombardment with tandem mass spectrometry.

Structures of alpha-(4-pyridyl-1-oxide)-N-tert-butylnitrone (4-POBN) radical adducts formed in the reactions of soybean lipoxygenase with linoleic acid, linolenic acid, and arachidonic acid were determined using continuous flow fast atom bombardment (CF-FAB) combined with tandem mass spectrometry. The radical adducts of these lipoxygenase-dependent reactions were: n-octanoic acid radical, 12,13-dihydroxylinoleic acid radical, 12,13-epoxylinoleic acid radical, and n-pentyl radical from linoleic acid; n-octanoic acid radical, ethyl radical, and cis/trans and/or positional isomers (1- and 3-pente

Unsaturated fatty acids show clear elicitation responses in a modified local lymph node assay with an elicitation phase, and test positive in the direct peptide reactivity assay.

The Organisation for Economic Co-operation and Development (OECD) Test Guidelines (TG) adopted the murine local lymph node assay (LLNA) and guinea pig maximization test (GPMT) as stand-alone skin sensitization test methods. However, unsaturated carbon-carbon double-bond and/or lipid acids afforded false-positive results more frequently in the LLNA compared to those in the GPMT and/or in human subjects. In the current study, oleic, linoleic, linolenic, undecylenic, fumaric, maleic, and succinic acid and squalene were tested in a modified LLNA with an elicitation phase (LLNA:DAE), and in a direc

Impact of the addition of 4-vinyl-derivatives of ferulic and sinapic acids on retention of fatty acids and terpenoids in cold-pressed rapeseed and flaxseed oils during the induction period of oxidation.

In the western diet there is an oversupply of n-6 fatty acids. This adverse trend can be balanced by the consumption of rapeseed and flaxseed oils rich in α-linolenic acid (n-3). However, the high share of this fatty acid contributes to low oxidative stability of oil. Oxidation decreases n-3 fatty acid and other bioactive compounds contents, which adversely affects oil nutritional value. In this study, the impact of ferulic and sinapic acids vinyl derivatives on the fatty acids and oil terpenoids (sterols, tocols, carotenoids, squalene) retention at the end of induction period during accelerat

Fatty acid changes in rat brain ethanolamine phosphoglycerides during and following chronic exposure to trichloroethylene.

Rats were exposed by continuous inhalation to a moderate level of trichloroethylene (1720 mg/m3). The fatty acid pattern of brain ethanolamine phosphoglycerides was examined during exposure and after an additional exposure-free period. Alterations in the fatty acid pattern were noted after 30 days of exposure. An increased ratio of linoleic acid-derived (n-6) to linolenic acid-derived (n-3) fatty acids was observed in the cerebral cortex, the hippocampus, and the brain stem. Of the major fatty acids, arachidonic acid (20:4(n-6)) was increased in the cerebral cortex and the brain stem, while do

Localization of arachidonate 12-lipoxygenase in canine brain tissues.

The cytosol fraction from a thoroughly irrigated canine cerebrum was subjected to immunoaffinity chromatography using a monoclonal antibody against porcine leukocyte 12-lipoxygenase. Arachidonate 12-lipoxygenase eluted from the column with some retardation. The enzyme, with a specific activity of 9 nmol/min/mg of protein, converted arachidonic acid to 12(S)-hydroperoxy-5,8,10,14-eicosatetraenoic acid. The enzyme was active not only with arachidonic acid, but also with linoleic and alpha-linolenic acids. In contrast, 12-lipoxygenase of canine platelets was almost inactive with linoleic and alph…

Fluorescence formation from the interaction of DNA with lipid oxidation degradation products.

To clarify the mechanism of fluorescence formation between DNA and lipid degradation products in the presence of ferric chloride and ascorbic acid, a number of carbonyl compounds and decomposition products of pure methyl linolenate hydroperoxides were examined. Keto derivatives of methyl ricinoleate, linoleate, and oleate, alkanals and 2-alkenals produced little or no fluorescence with DNA in the presence of ferric chloride-ascorbic acid. 2,4-Alkadienals were more active and 2,4,7-decatrienal was the most active. Mixtures of volatile aldehydes prepared from linolenate hydroperoxide decomposed

Emergence of omega-3 fatty acids in biomedical research.

Shortly after the discovery that linoleic acid was an essential fatty acid in 1930, α-linolenic acid also was reported to prevent the fatty acid deficiency syndrome in animals. However, several prominent laboratories could not confirm the findings with α-linolenic acid, and as a result there was a loss of interest in omega-3 fatty acids in lipid research. Even the findings that a prostaglandin can be synthesized from eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) is necessary for optimum retinal function generated only limited interest in omega-3 fatty acids. The breakthrough came

Potato tubers exhibit both homolytic and heterolytic hydroperoxide fatty acid-cleaving activities.

The action of a crude potato-tuber extract on 9- and 13-hydroperoxides of linoleic and linolenic acids was investigated. HPLC analysis revealed that 50% of the 9-hydroperoxide isomers and almost all the 13-hydroperoxide isomers were rapidly enzymically metabolized. No degradation of fatty acid hydroperoxides was observed with a thermally denatured enzymic extract. GC-MS identification of the volatiles formed by the reaction revealed that no volatiles were detected from the 9-hydroperoxide isomers, whereas 13-hydroperoxide of linolenic acid was cleaved into (Z)-3-hexenal, pentenols or dimers of…

Fatty Acids Increase GDF15 and Reduce Food Intake Through a GFRAL Signaling Axis.

UNLABELLED: In contrast to the well-defined biological feedback loops controlling glucose, the mechanisms by which the body responds to changes in fatty acid availability are less clearly defined. Growth differentiating factor 15 (GDF15) suppresses the consumption of diets high in fat but is paradoxically increased in obese mice fed a high-fat diet. Given this interrelationship, we investigated whether diets high in fat could directly increase GDF15 independently of obesity. We found that fatty acids increase GDF15 levels dose dependently, with the greatest response observed with linolenic aci
