An aeroponic system to characterize maize root exudates in relation to N and P nutrition and arbuscular mycorrhizal symbiosis.

Root exudates play major roles in the recruitment of plant microbiota. The metabolic composition of root exudates varies according to plant developmental stage, nutrient availability, (a)biotic stresses, and interaction with the root-associated microbiota, including arbuscular mycorrhizal fungi (AMF), which play a key role in plant mineral nutrition and stress tolerance. While it is well established that AMF can perceive plant root exudate compounds, little is known about plant root exudate modifications in response to AMF inoculation. Here, we developed an aeroponic-based culture system suita

Direct nitrogen, phosphorus and carbon exchanges between Mucoromycotina 'fine root endophyte' fungi and a flowering plant in novel monoxenic cultures.

Most plants form mycorrhizal associations with mutualistic soil fungi. Through these partnerships, resources are exchanged including photosynthetically fixed carbon for fungal-acquired nutrients. Recently, it was shown that the diversity of associated fungi is greater than previously assumed, extending to Mucoromycotina fungi. These Mucoromycotina 'fine root endophytes' (MFRE) are widespread and generally co-colonise plant roots together with Glomeromycotina 'coarse' arbuscular mycorrhizal fungi (AMF). Until now, this co-occurrence has hindered the determination of the direct function of MFRE

Transcriptome Analysis Provides Novel Insights into the Capacity of the Ectomycorrhizal Fungus Amanita pantherina To Weather K-Containing Feldspar and Apatite.

Ectomycorrhizal (ECM) fungi, symbiotically associated with woody plants, markedly improve the uptake of mineral nutrients such as potassium (K) and phosphorus (P) by their host trees. Although it is well known that ECM fungi can obtain K and P from soil minerals through biological weathering, the mechanisms regulating this process are still poorly understood at the molecular level. Here, we investigated the transcriptional regulation of the ECM fungus Amanita pantherina in weathering K-containing feldspar and apatite using transcriptome sequencing (RNA-seq) and validated these results for diff

Potato aphid Macrosiphum euphorbiae performance is determined by aphid genotype and not mycorrhizal fungi or water availability.

Intra- and interspecific variation in plant and insect traits can alter the strength and direction of insect-plant interactions, with outcomes modified by soil biotic and abiotic conditions. We used the potato aphid (Macrosiphum euphorbiae Thomas) feeding on cultivated Solanum tuberosum and wild Solanum berthaulti to study the impact of water availability and plant mutualistic arbuscular mycorrhizal (AM) fungi on aphid performance and susceptibility to a parasitoid wasp (Aphidius ervi Haliday). Plants were grown under glass with live or sterile AM fungal spores and supplied with sufficient or

Effects, tolerance mechanisms and management of salt stress in grain legumes.

Salt stress is an ever-present threat to crop yields, especially in countries with irrigated agriculture. Efforts to improve salt tolerance in crop plants are vital for sustainable crop production on marginal lands to ensure future food supplies. Grain legumes are a fascinating group of plants due to their high grain protein contents and ability to fix biological nitrogen. However, the accumulation of excessive salts in soil and the use of saline groundwater are threatening legume production worldwide. Salt stress disturbs photosynthesis and hormonal regulation and causes nutritional imbalance

Geographical Variation in Community Divergence: Insights from Tropical Forest Monodominance by Ectomycorrhizal Trees.

Convergence occurs in both species traits and community structure, but how convergence at the two scales influences each other remains unclear. To address this question, we focus on tropical forest monodominance, in which a single, often ectomycorrhizal (EM) tree species occasionally dominates forest stands within a landscape otherwise characterized by diverse communities of arbuscular mycorrhizal (AM) trees. Such monodominance is a striking potential example of community divergence resulting in alternative stable states. However, it is observed only in some tropical regions. A diverse suite o

Zinc phosphate transformations by the Paxillus involutus/pine ectomycorrhizal association.

In this research, we investigate zinc phosphate transformations by Paxillus involutus/pine ectomycorrhizas using zinc-resistant and zinc-sensitive strains of the ectomycorrhizal fungus under high- and low-phosphorus conditions to further understand fungal roles in the transformation of toxic metal minerals in the mycorrhizosphere. Mesocosm experiments with ectomycorrhizas were performed under sterile conditions with zinc phosphate localized in cellophane bags: zinc and phosphorus mobilization and uptake by the ectomycorrhizal biomass were analyzed. In the presence of a phosphorus source, an ec

Nutritional influences on the solubilization of metal phosphate by ericoid mycorrhizal fungi.

Four ericoid mycobionts (two isolates of Hymenoscyphus ericae, and two dark, sterile ericoid mycobionts isolated from metal-contaminated mine sites) were grown on solid agar plates supplemented with zinc phosphate (0.25 %) containing different forms of nitrogen (nitrate, ammonium or alanine) and different concentrations of carbon (glucose) and phosphorus (K2HPO4). The influence of nutrient variation on solubilizing ability of the fungi was assessed by measuring the zones of solubilization appearing beneath the growing colonies. All four mycobionts were capable of zinc phosphate solubilization

Underground Baristas: ecology of mycorrhizal fungi in Cerrado coffee cultivation.

The symbiosis between arbuscular mycorrhizal fungi (AMF) and coffee plants can improve nutrient use and crop resilience in low-fertility tropical soils. In this study, the activity and diversity of AMF in the rhizosphere of 35 Coffea arabica genotypes grown under Brazilian Cerrado conditions were characterized. Spore density, root colonization, and community composition were assessed by morphological characterization of field-collected spores and by multivariate analyses (CCA, PCA, and hierarchical clustering). No significant differences in spore density or root colonization were detected amon

Mechanism of ion homeostasis mediated by arbuscular mycorrhizal fungi in plant responses to saline-alkaline stress.

Against the backdrop of global climate change, soil salinization and alkalization have emerged as major environmental constraints on sustainable agricultural development. Salt-alkali stress primarily disrupts cellular ion homeostasis, resulting in excessive accumulation of ions such as Na+ and Cl- and deficiencies of essential nutrient ions including K+ and Ca2+, with negative consequence on plant growth and development. Arbuscular mycorrhizal fungi (AMF), a widespread group of beneficial soil microorganisms, could establish symbiotic associations with most terrestrial plants and enhance the t

Plant-associated microbial communities vary across compartments, mycorrhizal types, and species identities in alpine meadows of the Qinghai-Tibetan Plateau.

Plant-microbe symbioses can enhance plant adaptability. However, whether root-associated microbes form specific associations with plants of different mycorrhizal types remains controversial. Furthermore, it is currently unknown whether plants with different mycorrhizal types show heterogeneity in their phyllosphere microbial communities. In this study, we independently conducted in two alpine meadows, simultaneously collecting root and phyllosphere samples from plants with different mycorrhizal types, and used amplicon sequencing to characterize patterns of microbial community variations. Development of a high-throughput spore germination test to assess the toxicity of pesticides on arbuscular mycorrhizal fungi.

Pesticides are essential agricultural inputs that help securing crop yields. However, they can affect non-target soil microorganisms, including arbuscular mycorrhizal (AM) fungi, that are potential indicators of the toxicity of pesticides on the soil microbiota. Here, we developed a fast-track high-throughput spore germination test, for AM fungi produced in vitro. This test allows the determination of EC50 values and the nature of the effects of pesticides on AM fungal spores (fungicidal or fungistatic). First, 19 active ingredients were tested on Rhizophagus intraradices MUCL 49410. Secondly,

Enhancements in morphology, biochemicals, nutrients, and L-Dopa in Faba bean through plant growth promoting rhizobacteria and arbuscular mycorrhizal Fungi.

Faba beans are among the main food sources grown since ancient times; seeds contain higher protein (23-41%) than other legumes and are rich in healthy secondary bioactive components. This study was carried out to determine the effects of plant growth-promoting rhizobacteria (PGPR) and arbuscular mycorrhizal fungi (AMF) on the biofortification potential of faba beans. The study utilized Bacillus subtilis, Bacillus megaterium, and an AMF mix to examine changes in growth parameters, chlorophyll content, bioactive compounds, and mineral nutrients. AMF and PGPR treatments significantly enhanced var

Resistance to Plant Parasites in Tomato Is Induced by Soil Enrichment with Specific Bacterial and Fungal Rhizosphere Microbiome.

Commercial formulations of beneficial microbes have been used to enrich the rhizosphere microbiome of tomato plants grown in pots located in a glasshouse. These plants have been subjected to attacks by soil-borne parasites, such as root-knot nematodes (RKNs), and herbivores, such as the miner insect Tuta absoluta. The development of both parasites and the symptoms of their parasitism were restricted in these plants with respect to plants left untreated. A mixture, named in the text as Myco, containing plant growth-promoting rhizobacteria (PGPR), opportunistic biocontrol fungi (BCF), and arbusc

Removal of sulfamethoxazole and Cu, Cd compound pollution by arbuscular mycorrhizal fungi enhanced vertical flow constructed wetlands.

The combined pollution of antibiotics and heavy metals (HMs) has a serious impact on the water ecological environment. Previous researches mainly focused on the removal of antibiotics or HMs as single pollutants, with limited investigation into the treatment efficiencies and underlying mechanisms associated with their co-occurring pollution. In this study, 16 micro vertical flow constructed wetlands (MVFCWs) were constructed to treat composite wastewater consisting of sulfamethoxazole (SMX), copper (Cu) and cadmium (Cd), involving two different inoculation treatments (arbuscular mycorrhizal fu

Arbuscular mycorrhizal fungi reduce potassium, cadmium and ammonium losses but increases nitrate loss under high intensity leaching events.

BACKGROUND: Nutrients and heavy metals can be lost from soils via leaching, and arbuscular mycorrhizal fungi (AMF) can influence these events. Soil column experiments were carried out to examine whether leaching intensity and AMF can alter nutrient and Cd uptake in white clover plants and the extent of their losses through leaching. RESULTS: The presence of AMF significantly increased shoot and total biomass, as well as increased N, P, Cu and Zn uptake independent of water amount applied; while root P and Cu uptakes were promoted by AMF at any water amount treatments. Higher water amounts led

The possible involvement of salicylic acid and hydrogen peroxide in the systemic promotion of phenolic biosynthesis in clover roots colonized by arbuscular mycorrhizal fungus.

Arbuscular mycorrhizal fungal (AMF) colonization can induce both the local and the systemic increase in phenolic accumulation in hosts. However, the signaling molecules responsible for the systemic induction is still unclear. In this study, a split-root rhizobox system was designed to explore these molecules, with one half of clover (Trifolium repense) roots colonized by AMF, Funneliformis mosseae (formerly known as Glomus mosseae), and the other not (NM/M). Plants with two halves both (M/M) or neither (NM/NM) inoculated were also established for comparison. The contents of phenols and the acc
