Simultaneous detection of multiple mycotoxins in Radix Dipsaci and estimation of exposure risk for consumers.

Like many traditional Chinese herbal medicines, preparations from Radix Dipsaci are at risk of contamination by harmful mycotoxins; however, there have been no reports of actual contamination. In this study, we developed an analytical method to simultaneously detect eight mycotoxins in Radix Dipsaci and estimate the exposure risk for consumers. We have developed an analytical method utilizing ultra-high performance liquid chromatography and tandem mass spectrometry to accurately determine the levels of AFB1, AFB2, AFG1, AFG2, OTA, ZEN, T-2 and ST mycotoxins in 45 batches of Radix Dipsaci sourc…

Rapid microwave-assisted dispersive micro-solid phase extraction of mycotoxins in food using zirconia nanoparticles.

Mycotoxins are a group of secondary fungi metabolites present in foods that cause adverse effects in humans and animals. The objective of this study was to develop and validate a reliable and sensitive method to determine the presence of fumonisin B1, aflatoxin B1, ochratoxin B, T-2 toxin, ochratoxin A and zearalenone. A rapid, effective process, which involves microwave-assisted dispersive micro-solid phase extraction (MA-d-μ-SPE), has been proposed for the extraction and detection of 6 mycotoxins in peach seed, milk powder, corn flour and beer sample matrixes, for subsequent analysis by ultr…

Modified Fusarium Mycotoxins in Cereals and Their Products-Metabolism, Occurrence, and Toxicity: An Updated Review.

Mycotoxins are secondary fungal metabolites, toxic to humans, animals and plants. Under the influence of various factors, mycotoxins may undergo modifications of their chemical structure. One of the methods of mycotoxin modification is a transformation occurring in plant cells or under the influence of fungal enzymes. This paper reviews the current knowledge on the natural occurrence of the most important trichothecenes and zearalenone in cereals/cereal products, their metabolism, and the potential toxicity of the metabolites. Only very limited data are available for the majority of the identi…

Dietary exposure to mycotoxins of the Hong Kong adult population from a Total Diet Study.

Dietary exposure of Hong Kong adults to mycotoxins and their metabolites including aflatoxins (AFs), ochratoxin A (OTA), fumonisins (FNs), deoxynivalenol (DON), acetyldeoxynivalenols (AcDONs) and zearalenone (ZEA) was estimated using the Total Diet Study (TDS) approach to assess the associated health risk to the local people. Sixty commonly consumed food items, collected in four seasons, were sampled and prepared as consumed. These mycotoxins were primarily found at low levels. The highest mean levels (upper bound) were: AFs, 1.50 µg kg(-)(1) in legumes, nuts and seed; OTA, 0.22 µg kg(-)(1) in…

Mycotoxins in root extracts of American and Asian ginseng bind estrogen receptors alpha and beta.

The estrogenic activity of ginseng has been the subject of conflicting reports. Cell proliferation, induction of estrogen-responsive genes, and isolated cases of adverse reactions such as postmenopausal vaginal bleeding and gynecomastia have been reported after ginseng treatment. Other studies report antiproliferative effects with no induction of estrogen-responsive genes. We developed estrogen receptor (ER) alpha and ER alpha competitive binding assays using recombinant receptors and [(3)H]-17 alpha-estradiol to detect phytoestrogens in extracts of Asian ginseng root (Panax ginseng C. Individual and combined effects of the Fusarium mycotoxins fumonisin B1 and moniliformin in broiler chicks.

The individual and combined effects of feeding fumonisin B1 (FB1; 0, 100, 200 mg FB1/kg) and moniliformin (M; 0, 100, 200 mg M/kg) were evaluated using a 3 x 3 factorial arrangement of treatments. Significant mortality (P < 0.05) occurred in chicks fed all diets containing 200 mg M/kg (50%-65%). Compared with controls and chicks fed FB1, both feed intake and body weight gain were decreased (P < 0.05) in chicks fed diets containing 100 mg M/kg. Chicks fed M had heavier heart weights (P < 0.05) than control chicks or chicks fed FB1. Compared with controls, chicks fed diets containing 200 mg M/kg

Mycotoxins produced by toxic Fusarium isolates obtained from agricultural and nonagricultural areas (Arctic) of Norway.

Twenty-five isolates of F. acuminatum, 38 of F. avenaceum, 1 of F. culmorum, 31 of F. oxysporum and 56 of F. sambucinum were obtained in 198 … vely and caused uterine enlargement in rats. Most of these isolates produced no known trichothecene mycotoxins that could account for the toxicity that was demonstrated in the rat feeding tests. All but F. avenaceum N26B produced fusarin C (1.5 ppm) but caused no toxic effects in rat feeding test. None of the isolates produced fusarochromanone (TDP-1). Thirt …

Combined exposure to emerging Fusarium mycotoxins produces concentration-dependent synergistic toxicity in Jurkat T cells.

INTRODUCTION: Beauvericin (BEA), enniatin B (ENN B), and moniliformin (MON), mainly produced by Fusarium species, are often described as emerging mycotoxins due to their frequent detection in cereals and cereal-based products and their current lack of regulatory limits. Despite increasing evidence regarding their individual toxicities-including immunotoxic, genotoxic, reproductive, and cardiotoxic effects-information on their combined toxicity and the underlying mechanisms remains limited. To address this gap, this study aimed to evaluate the cytotoxic effects, reactive oxygen species (ROS) ge…

Occurrence and Co-Occurrence of Regulated and Emerging Mycotoxins in Foods Marketed to U.S. Toddlers.

Foodborne mycotoxins are toxic secondary metabolites produced by filamentous fungi. At sufficient concentrations, ingested mycotoxins have been found to disrupt the microbiome and exert toxic effects on gastrointestinal, hepatic, renal, and other tissues. Toddlers are particularly vulnerable to ill effects due to increased intake relative to lower body weight and immature detoxification, metabolic, and immune function. Despite this susceptibility, their adverse health effects remain under-recognized in public health sectors, and data on mycotoxin contamination in foods marketed to young childr…

Dynamics of Bacterial and Fungal Communities Related to Mycotoxin Production in Stored Wheat Grain.

Wheat grain deterioration during storage poses a critical threat to food security, primarily driven by fungal succession and mycotoxin contamination. In this study, we simulated high-humidity (97%) and warm (28°C) storage conditions to characterize the dynamics of bacterial and fungal communities and their correlation with mycotoxin production. We identified three distinct phases of mycotoxin accumulation: a non-toxigenic early stage (D0-D6), an AFB1-only middle stage (D15-D25), and a multi-mycotoxin late stage (D35-D50) characterized by the co-occurrence of AFB1, ZEN, and DON. High-throughput

Detriment of mycotoxin contamination in feed and nutrition-based prevention and control strategies: A review.

The health of humans and livestock is significantly compromised by the pervasive and serious issue of mycotoxin contamination in crops. Exploring ways to eliminate mycotoxin contamination is a global concern. In vivo biological prevention and treatment of mycotoxin damage demand a comprehensive understanding of its harm mechanisms, which is essential for developing comprehensive strategies. This study aimed to systematically review the mechanisms by which mycotoxin contamination damages the organism, causing oxidative stress, cellular apoptosis, autophagy, ferroptosis, and via the gut-liver ax

Lycopene Attenuates T2 Mycotoxin-Induced Hepatotoxicity and Dysbiosis by Activating PPAR Signaling.

Exposure to T2 toxin is known to induce hepatotoxicity and gut dysbiosis, yet effective dietary interventions remain underexplored. This study investigates the hepatoprotective and microbiota-modulating effects of lycopene against T2 toxin-induced toxicity in mice. Mice were exposed to T2 toxin with or without lycopene supplementation at low and high doses. The hepatic function, oxidative stress markers, inflammatory gene expression, detoxification pathway activity, and gut microbiota composition were assessed using histological, biochemical, and molecular analyses. T2 toxin exposure resulted

Ferroptosis in Mycotoxin-Induced Toxicity: Molecular Mechanisms, Intervention Implications, and Future Directions.

Chronic exposure to mycotoxin-contaminated food and feed induces systemic toxicity in humans and animals, with emerging evidence identifying ferroptosis as a key mechanism driving this pathology. Key mycotoxins, including deoxynivalenol, aflatoxin B1, T-2 toxin, fumonisin B1, zearalenone, ochratoxin A, and patulin, trigger ferroptosis through interconnected pathways: iron metabolism disruption, lipid peroxides accumulation, Glutathione Peroxidase 4 (GPX4) antioxidant system imbalance, Ferroptosis Suppressor Protein 1 (FSP1) pathway modulation, and mitochondrial homeostasis disruption. This rev…

Corneal transcriptomic signatures of mycotoxin exposure reveal distinct immune and metabolic responses.

Fungal keratitis (FK) is a vision-threatening infection in which Fusarium species not only invade the cornea but may also release mycotoxins that exacerbate tissue injury. Here, we established a corneal exposure model to delineate the direct effects of Fusarium-derived mycotoxins independent of live fungal infection, using deoxynivalenol (DON), fumonisin B1 (FB1), and T-2 toxin as representative compounds. Hstological analyses revealed that all three mycotoxins markedly impaired corneal epithelial repair, induced stromal disorganization, and promoted inflammatory cell infiltration. At the mole…
