Metabolomic investigation of pathogenesis of myxosporean emaciation disease of tiger puffer fish Takifugu rubripes.

Serum biochemical analysis was undertaken to study the pathophysiological details of emaciation disease of the tiger puffer fish Takifugu rubripes (Temminck and Schlegel). Serum parameters were measured by biochemical analysis using automated dry chemistry and gas chromatography/mass spectrometry (GC/MS). Serum concentrations of albumin, amylase, calcium, creatinine, glucose and total protein were significantly lower in the emaciated fish when compared with those of normal fish. Regression analyses found close correlation between concentrations of total protein, albumin, amylase, glucose and p

Pathologic Changes Associated With Respiratory Compromise And Morbidity Due To Massive Interlamellar Henneguya Exilis Infection In Channel × Blue Hybrid Catfish.

There are multiple Henneguya spp. (Myxozoa: Myxobolidae) endemic to North American catfish aquaculture that affect the gills of channel catfish and their hybrids. These parasites are morphologically similar, and confusion exists regarding the predilection sites and pathologic changes associated with different species. In the spring of 2018, channel (Ictalurus punctatus) female × blue (Ictalurus furcatus) male hybrid catfish from 2 separate commercial operations in northwest Mississippi were submitted for diagnostic assessment in response to observed morbidity and reduced feeding activity. Fish…

Hoferellus matosi sp. nov. (Bivalvulida: Myxobilatidae) parasitizing Ageneiosus ucayalensis Castelnau (Siluriformes: Auchenipteridae) from the Coaracy Nunes Hydroelectric Reservoir in Amap, Eastern Amazon, Brazil.

The microparasite species Hoferellus matosi sp. nov. was found parasitizing the urinary bladder of the fish species Ageneiosus ucayalensis collected from the Coaracy Nunes hydroelectric reservoir in Ferreira Gomes, Amap, Brazil. Plasmodia and myxospores were identified in fragments of the urinary bladder and characterized through microscopic and molecular analyses. Myxospores were subspherical in sutural view, with fine striations or projections on the posterior portion of the valves, measuring an average length and width of 8.5 (8.28.8) m and 7.9 (7.68.2) m, respectively. Each spore contained

Enteromyxum leei (Myxosporea: Bivalvulida) as the cause of myxosporean emaciation disease of farmed olive flounders (Paralichthys olivaceus) and a turbot (Scophthalmus maximus) on Jeju Island, Korea.

A life-threatening emaciation disease of unknown cause(s) is affecting the farming of olive flounders (Paralichthys olivaceus) and turbots (Scophthalmus maximus) on Jeju Island, Korea. As this is one of the major industries in the region, it is of great concern to local farmers trying to develop successful and sustainable aquaculture. We examined 16 olive flounders and one turbot cultured at three farms located in the southern part of Jeju Island, which manifested moderate to severe emaciation such as thinning of the body with notable appearance of bony ridges of the skull on heads. Fresh muco

CD10+ Cells and IgM in Pathogen Response in Lumpfish (Cyclopterus lumpus) Eye Tissues.

Lumpfish (Cyclopterus lumpus), a North Atlantic "cleaner" fish, is utilized to biocontrol salmon louse (Lepeophtheirus salmonis) in Atlantic salmon (Salmo salar) farms. Lumpfish require excellent vision to scan for and eat louse on salmon skin. The lumpfish eye immune response to infectious diseases has not been explored. We examined the ocular response to a natural parasite infection in wild lumpfish and to an experimental bacterial infection in cultured lumpfish. Cysts associated with natural myxozoan infection in the ocular scleral cartilage of wild adult lumpfish harbored cells expressing …

Acquired protective immune response in a fish-myxozoan model encompasses specific antibodies and inflammation resolution.

The myxozoan parasite Enteromyxum leei causes chronic enteritis in gilthead sea bream (GSB, Sparus aurata) leading to intestinal dysfunction. Two trials were performed in which GSB that had survived a previous infection with E. leei (SUR), and naïve GSB (NAI), were exposed to water effluent containing parasite stages. Humoral factors (total IgM and IgT, specific anti-E. leei IgM, total serum peroxidases), histopathology and gene expression were analysed. Results showed that SUR maintained high levels of specific anti-E. leei IgM (up to 16 months), expressed high levels of immunoglobulins at th…

Under control: how a dietary additive can restore the gut microbiome and proteomic profile, and improve disease resilience in a marine teleostean fish fed vegetable diets.

BACKGROUND: The constant increase of aquaculture production and wealthy seafood consumption has forced the industry to explore alternative and more sustainable raw aquafeed materials, and plant ingredients have been used to replace marine feedstuffs in many farmed fish. The objective of the present study was to assess whether plant-based diets can induce changes in the intestinal mucus proteome, gut autochthonous microbiota and disease susceptibility of fish, and whether these changes could be reversed by the addition of sodium butyrate to the diets. Three different trials were performed using

Effects of Enteromyxum scophthalmi experimental infection on the neuroendocrine system of turbot, Scophthalmus maximus (L.).

Enteromyxum scophthalmi is an intestinal myxosporean parasite responsible for serious outbreaks in turbot Scophthalmus maximus (L.) culture, in North-western Spain. The disease affects the digestive tract, provokes severe catarrhal enteritis, emaciation and high rates of mortality. The digestive parasitization triggers a response with the coordinate participation of immune and neuroendocrine systems through the action of peptides released by enteroendocrine cells and present in nervous elements, acting as neuro-immune modulators. The present study was designed to assess the response of the tur

Monomyxum ligophori n. sp. in a ParasiteBlitz: monopisthocotylans as myxozoan hosts in South Carolina and monophyly of a cosmopolitan hyperparasitic clade.

A ParasiteBlitz event offers a brief, intense opportunity to discover diverse parasite species and to reveal life cycles of heteroxenous parasite taxa. In this study, we describe Monomyxum ligophori n. sp., a hyperparasitic myxozoan (Monomyxidae) proliferating in two dactylogyrid monopisthocotylan flatworms (Ligophorus saladensis, Ligophorus mugilinus) infecting mugilid fishes (Mugil cephalus, Mugil curema) on the Atlantic coast of North America. Furthermore, we used DNA barcoding to infer the parasite's complex life cycle, matching its hyperparasitic myxospore stages with actinospore stages i…

Redescription of Chloromyxum ellipticum Li & Nie, 1973 (Myxosporea: Chloromyxidae) infecting the gall bladder of grass carp Ctenopharyngodon idellus Valenciennes, 1844, supplemented by morphological and molecular characteristics.

The traditional taxonomy of the genus Chloromyxum Mingazzini, 1890 has been intensively challenged to be paraphyletic by recent ribosomal DNA (rDNA)-based phylogenetic analysis. Undersampling to get rich sequence data to infer more scientific phylogenetic relationships makes scientists conservatively assign all non-marine elasmobranch-infecting species as Chloromyxum sensu lato. Although complex ridge pattern on the spore surface observed by scanning electron microscopy was thought to be critical for the identification of Chloromyxum species, insufficient data also prevent this ultrastructural

Production of a novel monoclonal antibody applicable for an immunochromatographic assay for Kudoa septempunctata spores contaminating the raw olive flounder (Paralichthys olivaceus).

Kudoa septempunctata, a myxosporean parasite of the olive flounder (Paralichthys olivaceus), causes foodborne gastroenteritis after ingestion of contaminated raw flounder. Available methods to detect K. septempunctata require expensive equipment, well-trained personnel, and lengthy procedures. Here we generated a novel monoclonal antibody (MAb 15G11) against K. septempunctata and used it to produce a prototype immunochromatographic assay (prototype Kudoa-ICA). Within 15min, the prototype Kudoa-ICA detected ≥1.0×105spores/mL in a spore suspension and ≥2.0×104spores/g of P. olivaceus muscle. The

Study of the distribution of active caspase-3-positive cells in turbot, Scophthalmus maximus (L.), enteromyxosis.

Enteromyxosis caused by Enteromyxum scophthalmi is one of the parasitizations with a higher economic impact on turbot, Scophthalmus maximus (L.), aquaculture. This myxosporean produces severe catarrhal enteritis with abundant inflammatory infiltrates in the lamina propria-submucosa (LP), epithelial detachment and leucocyte depletion of the lymphohaematopoietic organs. Some advances made on the pathogenesis pointed to a role of apoptosis in the enteromyxosis. Therefore, the main aim of this work was to employ the TUNEL assay and the anti-(active caspase-3) immunohistochemical assay to detect ap

Parasites and hepatic lesions among pink salmon, Oncorhynchus gorbuscha (Walbaum), during early seawater residence.

Juvenile pink salmon, Oncorhynchus gorbuscha (Walbaum), in the Broughton Archipelago region of western Canada were surveyed over 2 years for sea lice (Lepeophtheirus salmonis and Caligus clemensi), gross and microscopic lesions and evidence of infections with viruses and bacteria. The 1071 fish examined had an approximate ocean residence time no longer than 3 months. A high prevalence of degenerative liver lesions, renal myxosporean parasites and a low prevalence of skin lesions and sea lice were observed. No indications of viral or bacterial diseases were detected in either year. The monthly
