In denitrifying bacteria, nitric oxide (NO) is an electron acceptor and a free intermediate produced during anaerobic respiration. NO is also a signal for transcriptional regulation of the genes encoding nitrite (Nir), nitric oxide (Nor) and nitrous oxide reductases (N2OR). We hypothesise that the timing and strength of the NO signal necessary for full nir expression are key factors in the bet-hedging strategy of Paracoccus denitrificans, and that systems scavenging NO under hypoxia reduce the probability of nir induction. We show that the flavohemoglobin Hmp scavenges NO in aerobic cultures a

The greenhouse gas nitrous oxide (N2O) has strong potential to drive climate change. Soils are a major source of N2O, with microbial nitrification and denitrification being the primary processes involved in such emissions. The soybean endosymbiont Bradyrhizobium diazoefficiens is a model microorganism to study denitrification, a process that depends on a set of reductases, encoded by the napEDABC, nirK, norCBQD, and nosRZDYFLX genes, which sequentially reduce nitrate (NO3-) to nitrite (NO2-), nitric oxide (NO), N2O, and dinitrogen (N2). In this bacterium, the regulatory network and environment

Pelosinus spp. are fermentative firmicutes that were recently reported to be prominent members of microbial communities at contaminated subs … ction. Transcriptional studies demonstrated differential expression of hydrogenases and nitrate and nitrite reductases. Overall, the unexpected metabolic capabilities and gene content reported here broaden our perspective on what biogeochemical and ecological roles this species might play as a prominent member of microbial communities in subsurface environments.

The coordination chemistry of the one-electron reduced form of nitric oxide, termed a nitroxyl or nitrosyl hydride (NO- or HNO), is described with special focus on its interaction with hemes and heme model complexes. Nitroxyl intermediates have been proposed in the catalytic cycles of several heme-based nitrite and nitric oxide reductases; in fungal cytochrome P450nor, a short-lived nitroxyl-adduct has been observed during catalytic turnover. Ferrous-nitroxyl adducts were first identified in electrochemical reductions of nitrosyl porphyrins and heme proteins, but only recently have these speci

A genetic approach was used to study (dissimilatory) ferric iron (Fe3+) reduction in Shewanella putrefaciens 200. Chemical mutagenesis procedures and two rapid plate assays were developed to facilitate the screening of Fe3+ reduction-deficient mutants. Sixty-two putative Fe3+ reduction-deficient mutants were identified, and each was subsequently tested for its ability to grow anaerobically on various compounds as sole terminal electron acceptors, including Fe3+, nitrate (NO3-), nitrite (NO2-), manganese oxide (Mn4+), sulfite (SO3(2-)), thiosulfate (S2O3(2-)), trimethylamine N-oxide, and fumara

Phosphorus is generally considered as the prime limiting nutrient responsible for cyanobacterial blooms. However, recent research is drawing attention to the importance of bioavailable nitrogen (N) in freshwater eutrophication. This study investigated the bioavailability of NO3(-)-N, NO2(-)-N, NH4(+)-N and Urea-N under different concentrations of 1.2, 3.6 and 6.0mgL(-1) to Microcystis aeruginosa. Overall, Urea-N ranked the first in promoting M. aeruginosa growth, followed by NO3(-)-N and NO2(-)-N. However, the algal growth cultured in NH4(+)-N was depressed under test N levels. The bioavailabi

Biological treatment of cyanide-contaminated wastewater is mediated by microbial consortia in which different organisms perform distinct, functionally specialized roles. This study investigated microbial communities involved in gold mine wastewater treatment with integrated aerobic-anoxic reactors seeded with consortia from an alkaline soda lake, Lake Chitu. Whole-genome sequencing of isolates (WGS) and metagenomic sequencing of the bioreactor were performed to characterize the consortia, resulting in the identification of 23 non-redundant genomes, comprising 14 whole-genome sequencing isolate

Engineered hemoproteins can selectively incorporate nitrogen from nitrene precursors like hydroxylamine, O-substituted hydroxylamines, and organic azides into organic molecules. Although iron-nitrenoids are often invoked as the reactive intermediates in these reactions, their innate reactivity and transient nature have made their characterization challenging. Here we characterize an iron-nitrosyl intermediate generated from NH2OH within a protoglobin active site that can undergo nitrogen-group transfer catalysis, using UV-vis, electron paramagnetic resonance (EPR) spectroscopy, and high-resolu

Methionine (Met) can inhibit plant diseases caused by phytopathogens. However, the effect of Met on gray mold resulted from Botrytis cinerea in tomato is still unclear. This study showed 5 mM Met alleviated disease development of gray mold, enhanced chitinase (CHI) and β-1, 3-glucanase (GNS) activities and the expression of SlCHI, SlGNS, SlPR1 and SlNPR1 in tomatoes, rather than inhibited the growth of B. cinerea directly. Moreover, ethylene biosynthesis and signal transduction before pathogen inoculating were induced by 5 mM Met. Interestingly, Met reduced the nitrosylation levels of ACS4 and

The fungicide chlorothalonil (CHT) has been widely used in the tea orchard due to its high-efficiency and sterilization. It has been reported that repeated application of CHT inhibits soil nitrification process. However, the acute impact of CHT on soil denitrification and associated N2O emissions is unclear. This study evaluated nitrate (NO3-) removal, denitrifying gene abundance and denitrifying enzyme activity of tea orchard soil after a 72-h-exposure to CHT. It was found that increasing CHT from 5 to 25 mg kg-1 suppressed the NO3- removal efficiency from 74.6% to 54.1%, but increased N2O em

Plants are increasingly recognized as active contributors to nitrous oxide (N2O) fluxes. However, the physiological and biochemical processes underlying these plant-mediated N2O emissions remain poorly defined. This review synthesizes current understanding of direct N2O production pathways within plants, including chloroplast- and mitochondrial-associated processes involving nitrate and nitrite reductases, as well as cytochrome-mediated reactions under specific conditions. Plant-microbe interactions in the rhizosphere and phyllosphere further influence N2O dynamics. Symbiotic nitrogen-fixing b…

Pseudomonas aeruginosa infects the lungs of patients with cystic fibrosis. Sputum expectorated from the lungs of patients contains low levels of oxygen, indicating that P. aeruginosa may be oxygen-deprived during infection. During in vitro growth under oxygen-limiting conditions, a P. aeruginosa reference strain increases expression of a cytochrome oxidase with a high affinity for oxygen, and of nitrate and nitrite reductases that enable it to use nitrate instead of oxygen during respiration. Here, we quantified transcription of the genes encoding these three enzymes in sputum samples from 18 …

Sodium nitrite (NaNO2 ) is widely used in the food industry as a preservative and colorant in meat and fish products. Industrialization and improper agricultural practices have greatly increased human exposure to high nitrite levels, mainly through contaminated drinking water, causing various health disorders. We have investigated the protective effect of carnosine (CAR) and N-acetyl cysteine (NAC) on NaNO2 -induced toxicity in rat blood. CAR is a bioactive dipeptide found in mammalian muscle while NAC is a synthetic sulfhydryl amino acid and an important precursor of glutathione. Animals were

There is an emerging paradigm that certain foods promote nitric oxide (NO) production from the stepwise reduction of nitrate to nitrite to NO, providing an endothelium independent source of bioactive NO. We hypothesize that a unique formulation containing nitrate-rich beetroot along with Hawthorn berry shown to have a robust nitrite reductase activity would improve NO status in humans and modify cardiovascular risk factors. The trial was conducted at the Houston Institute for Clinical Research in Houston, Texas. Inclusion criteria for this double-blinded, placebo-controlled study were patients

Enriching anaerobic ammonium oxidation (ANAMMOX) bacteria from conventional municipal activated sludge remains difficult despite reported feasibility, particularly without highly enriched anammox cultures (HeAnAC). This study examined ANAMMOX start-up in an anaerobic-membrane-bioreactor fed ammonium (NH4+) and nitrite (NO2-) under organic carbon-free conditions. Initial nitrogen removal (NH4+: 78%; NO2-: 98%) declined sharply after day 70 (<11% and <28%, respectively), coinciding with stable hydrazine synthase gene (hzs) abundance, elevated DNRA nitrite reductase gene (nrfA) abundance, and hig

The sole known heme enzyme of the parasitic protist Giardia intestinalis is a flavohemoglobin (gFlHb) that acts as a nitric oxide dioxygenase (NOD) and protects the organism from the free radical nitric oxide. To learn more about the properties of this enzyme, we measured its nitric oxide dioxygenase, NADH oxidase, and cytochrome c reductase activities and compared these to the activities of the E. coli flavohemoglobin (Hmp). The turnover number for the NOD activity of gFlHb (23 s-1) is about two-thirds of that of Hmp (34 s-1) at pH 6.5 and 37 °C. The two enzymes differ in their sensitivity to

Nitrogenous wastewater is a major environmental problem. Denitrification refers to the sequential reduction of nitrate (NO3-) and nitrite (NO2-) to nitric oxide (NO) and nitrous oxide (N2O) to form N2. Oxygenic denitrification could transform NO into N2 and O2 via NO dismutase (Nod), an unusual O2-forming process to reduce N2O emission during nitrogen removal. After confirming the presence of the nod gene in three samples including anaerobically-activated sludge, farmland soil, and paddy soil, the targeted enrichment experiment was conducted. The abundance of key denitrification genes rose by

The purification and biochemical characterization of the respiratory membrane-bound nitrate reductase from Sinorhizobium meliloti 2011 (Sm NR) is reported together with the optimal conditions for cell growth and enzyme production. The best biomass yield was obtained under aerobic conditions in a fed-batch system using Luria-Bertani medium with glucose as carbon source. The highest level of Sm NR production was achieved using microaerobic conditions with the medium supplemented with both nitrate and nitrite. Sm NR is a mononuclear Mo-protein belonging to the DMSO reductase family isolated as a

Two parallel pilot-scale reactors were operated to investigate pollutant removal performance and metabolic pathways in elemental sulfur-driven autotrophic denitrification (SDAD) process under low temperature and after addition of external electron donors. The results showed that low temperature slightly inhibited SDAD (average total nitrogen removal of ∼4.7 mg L-1) while supplement of sodium thiosulfate (stage 2) and sodium acetate (stage 3) enhanced denitrification and secretion of extracellular polymeric substances (EPS), leading to the average removal rate of 0.75 and 1.01 kg N m-3 d-1, res

Biodegradable microplastics (MPs) are proposed as sustainable alternatives to conventional MPs, yet their distinct effects on soil microbial communities and ecological functions remain insufficiently understood. This study compares the impacts of biodegradable polylactic acid (PLA) and conventional polyvinyl chloride (PVC) MPs on soil microbial assemblages and nitrogen cycling. Fluorescein diacetate hydrolase (FDAse) activity was temporarily stimulated by 2 % (w/w) PLA and PVC MPs, while 7 % (w/w) PVC MPs initially inhibited FDAse activity before promoting it. PLA MPs (2 % and 7 %, w/w) dramat
