Season does not influence embryo recovery rate and conceptus size until day 14 after ovulation in the horse.

Although the horse is a seasonal breeding species, a considerable number of mares continue to cycle throughout autumn and winter. Slower equine embryo growth during the non-breeding season has been hypothesized, and because smaller embryo size is beneficial for cryopreservation, embryo collection outside the breeding season could be an interesting approach for the production of frozen horse embryos. In the present retrospective study, we have therefore analysed embryo recovery rates and conceptus size in mares (n = 30) throughout the year. Conceptus diameter was either size determined after co

Phosphate metabolism and foetal growth in the rat. III. Net transfer of 31P and 32P inorganic phosphate from the maternal plasma to the placenta after foetectomy.

In primigravid rat females at the 12th day or later stages of gestation one foetus is surgically removed and the amniotic sac allowed to retract freely around the remaining placenta in situ. One hour to 10 days after foetectomy [32P]ortho-phosphate is injected intraperitoneally. The post-foetectomy placenta and matched normal placenta and foetus in the contralateral horn are taken after labelling periods of one or three hours and analysed for calcium, inorganic phosphate, organic-bound acid-soluble and organic-bound acid-insoluble phosphates in their [31P] and [32P] forms. The acutely foetecto

Material properties of the embryonic small intestine during buckling morphogenesis.

During embryonic development, tissues undergo dramatic deformations as functional morphologies are stereotypically sculpted from simple rudiments. Formation of healthy, functional organs therefore requires tight control over the material properties of embryonic tissues during development, yet the biological basis of embryonic tissue mechanics is poorly understood. The present study investigates the mechanics of the embryonic small intestine, a tissue that is compactly organized in the body cavity by a mechanical instability during development, wherein differential elongation rates between the

Is hull cleaning wastewater a potential source of developmental toxicity on coastal non-target organisms?

Chemical contaminants can be discharged by vessel hull cleaning processes, such as scraping, jet spraying, and painting, all of which produce readily transportable contaminants into the marine environment, where they are referred to as 'hotspots' of contamination in coastal areas. However, many countries have not yet established effective evaluation methods for disposal of waste mixtures or management guidelines for areas of hull cleaning. To define the toxic effects of wastewater from vessel hull cleaning in dry docks on resident non-target organisms, we investigated the chemical concentratio

Immunofluorescent studies on titin and myosin in developing hearts of normal and cardiac mutant axolotls.

Homozygous recessive cardiac mutant gene c in the axolotl, Ambystoma mexicanum, results in a failure of the embryonic heart to initiate beating. Previous studies show that mutant axolotl hearts fail to form sarcomeric myofibrils even though hearts from their normal siblings exhibit organized myofibrils beginning at stage 34-35. In the present study, the proteins titin and myosin are studied using normal (+/+) axolotl embryonic hearts at stages 26-35. Additionally, titin is examined in normal (+/c) and cardiac mutant (c/c) embryonic axolotl hearts using immunofluorescent microscopy at stages 35

Histological studies on developing chick embryos treated with aminoguanidine sulfate.

Fertile eggs were injected 10 or 20 mg/egg of aminoguanidine sulfate (AGS) into the albumen on the 5th day of incubation and examined macroscopically and histopathologically. The marked abnormalities were observed in liver, gallbladder and spleen while other organs were not different from the control. The earliest change was an increase of mitotic cells in the liver which reached to the peak at 2 days after the AGS injection. The increase of the ratio of cells in metaphase was characteristic. Decrease of weight, retardation of the development, fatty degeneration, necrosis and increase of conne

Emergence of the Endoderm in the Mammalian Embryo: From Cell Fate Specification to Gut Tube Formation.

The endoderm is the embryonic tissue that gives rise to vital organ systems in both the developing embryo and adult animal. Cells with a definitive (embryonic) endoderm identity emerge from the pluripotent epiblast at gastrulation, and go on to form the gut tube, which develops into the epithelial component of the digestive and respiratory tracts. In this review, we discuss the mechanisms driving endoderm emergence and morphogenesis, including key epithelial-mesenchymal and mesenchymal-epithelial transitions, the mechanical forces driving gut tube formation, and the patterning events that subd

Nkx2-5(+)islet1(+) mesenchymal precursors generate distinct spleen stromal cell subsets and participate in restoring stromal network integrity.

Secondary lymphoid organ stromal cells comprise different subsets whose origins remain unknown. Herein, we exploit a genetic lineage-tracing approach to show that splenic fibroblastic reticular cells (FRCs), follicular dendritic cells (FDCs), marginal reticular cells (MRCs), and mural cells, but not endothelial cells, originate from embryonic mesenchymal progenitors of the Nkx2-5(+)Islet1(+) lineage. This lineage include embryonic mesenchymal cells with lymphoid tissue organizer (LTo) activity capable also of supporting ectopic lymphoid-like structures and a subset of resident spleen stromal c

Spatiotemporal transcriptome atlas of human embryos after gastrulation.

The comprehensive spatiotemporal atlas of gene expression during early human embryonic development is critical for insights into embryogenesis1, organogenesis2 and disease origins3,4. Here, leveraging Stereo-seq technology, we generated spatial transcriptomic profiles across 77 sagittal sections of 13 whole-human embryos ranging from Carnegie stage 12 to 23, integrated with single-nucleus RNA sequencing to elucidate gene expression patterns within defined cellular contexts, revealing the cellular heterogeneity that drives organ-specific differentiation. Our study has established a regulatory p

Lessons Learned from Somatic Cell Nuclear Transfer.

Somatic cell nuclear transfer (SCNT) has been an area of interest in the field of stem cell research and regenerative medicine for the past 20 years. The main biological goal of SCNT is to reverse the differentiated state of a somatic cell, for the purpose of creating blastocysts from which embryonic stem cells (ESCs) can be derived for therapeutic cloning, or for the purpose of reproductive cloning. However, the consensus is that the low efficiency in creating normal viable offspring in animals by SCNT (1-5%) and the high number of abnormalities seen in these cloned animals is due to epigenet

polished rice mediates ecdysone-dependent control of Drosophila embryonic organogenesis.

In many animals, progression of developmental stages is temporally controlled by steroid hormones. In Drosophila, the level of ecdysone titer oscillates and developmental stage transitions, such as larval molting and metamorphosis, are induced at each of ecdysone peaks. Ecdysone titer also peaks at the stage of mid-embryogenesis and the embryonic ecdysone is necessary for morphogenesis of several organs, although the regulatory mechanisms of embryonic organogenesis dependent on ecdysone signaling are still open questions. In this study, we find that absence or interruption of embryonic ecdyson

Cerebellar Vermis and Midbrain Hypoplasia Upon Conditional Deletion of Chd7 from the Embryonic Mid-Hindbrain Region.

Reduced fibroblast growth factor (FGF) signaling from the mid-hindbrain or isthmus organizer (IsO) during early embryonic development results in hypoplasia of the midbrain and cerebellar vermis. We previously reported evidence for reduced Fgf8 expression and FGF signaling in the mid-hindbrain region of embryos heterozygous for Chd7, the gene mutated in CHARGE (Coloboma, Heart defects, choanal Atresia, Retarded growth and development, Genitourinary anomalies and Ear defects) syndrome. However, Chd7+/- animals only exhibit mild cerebellar vermis anomalies. As homozygous deletion of Chd7 is embry

Globins and nitric oxide homeostasis in fish embryonic development.

Since the discovery of new members of the globin superfamily such as Cytoglobin, Neuroglobin and Globin X, in addition to the most well-known members, Hemoglobin and Myoglobin, different hypotheses have been suggested about their function in vertebrates. Globins are ubiquitously found in living organisms and can carry out different functions based on their ability to bind ligands such as O2, and nitric oxide (NO) and to catalyze reactions scavenging NO or generating NO by reducing nitrite. NO is a highly diffusible molecule with a central role in signaling important for egg maturation, fertili

Etiologic theories of idiopathic scoliosis: the breaking of bilateral symmetry in relation to left-right asymmetry of internal organs, right thoracic adolescent idiopathic scoliosis (AIS) and vertebrate evolution.

In the search to understand the etiology and pathogenesis of adolescent idiopathic scoliosis (AIS) some workers have focused on mechanisms initiated in embryonic life including a disturbance of bilateral (left-right or mirror-image) symmetry highly conserved in vertebrates. The normal external bilateral symmetry of vertebrates results from a default process involving mesodermal somites. The normal internal asymmetry of the heart, major blood vessels, lungs and gut with its glands is also highly conserved among vertebrates. It results from the breaking of the initial bilateral symmetry by a bin

Localization and quantitation of 125I-epidermal growth factor binding in mouse embryonic tooth and other embryonic tissues at different developmental stages.

We have shown earlier that epidermal growth factor (EGF) inhibits morphogenesis and cell differentiation in mouse embryonic teeth in organ culture. This inhibition depends on the stage of tooth development so that only teeth at early developmental stages respond to EGF (A-M. Partanen, P. Ekblom, and I. Thesleff (1985) Dev. Biol. 111, 84-94). We have now studied the quantity and pattern of EGF binding in teeth at various stages of development by incubating the dissected tooth germs with 125I-labeled EGF. Although the quantity of 125I-EGF binding per microgram DNA stays at the same level, locali

Exploring hyperglycemia's impact on embryonic development: Insights from the chicken embryo model.

Hyperglycemia during pregnancy is a growing health concern due to its association with congenital anomalies. While rodent models have been used to study this condition, their variability and inability to control the embryonic microenvironment is a limitation. Having this in mind, this study explores the chicken embryo (Gallus gallus) as an alternative model for studying the effects of hyperglycemia on early embryonic development. For that purpose, fertilized chicken eggs were exposed to glucose (0.2 and 0.4 mmol) from embryonic day 1 (E1) onwards. On embryonic day 5 (E5), glucose levels, devel

Single-cell RNA sequencing: revealing human pre-implantation development, pluripotency and germline development.

Early human development is a dynamic, heterogeneous, complex and multidimensional process. During the first week, the single-cell zygote undergoes eight to nine rounds of cell division generating the multicellular blastocyst, which consists of hundreds of cells forming spatially organized embryonic and extra-embryonic tissues. At the level of transcription, degradation of maternal RNA commences at around the two-cell stage, coinciding with embryonic genome activation. Although numerous efforts have recently focused on delineating this process in humans, many questions still remain as thorough

Functional cargos of exosomes derived from Flk-1+ vascular progenitors enable neurulation and ameliorate embryonic anomalies in diabetic pregnancy.

Источник пометил публикацию как отозванную

Various types of progenitors initiate individual organ formation and their crosstalk orchestrates morphogenesis for the entire embryo. Here we show that progenitor exosomal communication across embryonic organs occurs in normal development and is altered in embryos of diabetic pregnancy. Endoderm fibroblast growth factor 2 (FGF2) stimulates mesoderm Flk-1+ vascular progenitors to produce exosomes containing the anti-stress protein Survivin. These exosomes act on neural stem cells of the neuroepithelium to facilitate neurulation by inhibiting cellular stress and apoptosis. Maternal diabetes cau

Impact of glyphosate-based herbicide on early embryonic development of the amphibian Xenopus laevis.

BACKGROUND: Worldwide, amphibian populations are declining drastically. One reason might be the use of pesticides including herbicides. The herbicide glyphosate is an inhibitor of the 5-enolpyruvylshikimate-3-phosphate (EPSP) synthase of the plant shikimate pathway, preventing the formation of aromatic amino acids and thus inducing plant death. Due to this specific action, GBH are considered nontoxic to non-target organisms. However, GBH impairs embryonic development of chickens, amphibians and fishes. So far, no detailed tissue- and organ-specific analysis of the effects of GBH during develop
