Ferroptosis, necroptosis, and pyroptosis in calcium oxalate crystal-induced kidney injury.

Kidney stones represent a highly prevalent urological disorder worldwide, with high incidence and recurrence rates. Calcium oxalate (CaOx) crystal-induced kidney injury serves as the foundational mechanism for the formation and progression of CaOx stones. Regulated cell death (RCD) such as ferroptosis, necroptosis, and pyroptosis are essential in the pathophysiological process of kidney injury. Ferroptosis, a newly discovered RCD, is characterized by its reliance on iron-mediated lipid peroxidation. Necroptosis, a widely studied programmed necrosis, initiates with a necrotic phenotype that res

Urine metabolomic changes in cats with renal disease or calcium oxalate uroliths.

Two common problems in the urinary system of cats are renal disease (RD) and calcium oxalate (CaOx) stones. The objective of this study was to assess urine metabolomic parameters of cats with these diseases to determine metabolic abnormalities and differences between the groups. The urine metabolic profile for each cat was determined, along with their lifetime history of stone incidence and renal disease. In order to reduce the data for analysis, factor analysis/factor loading was used allowing statistical hypothesis testing and the selection of significant metabolites from 488 identified meta

Exogenous uromodulin and tubular responses in a model of oxalate-induced kidney injury.

Crystalline nephropathies are associated with kidney injury. Uromodulin (Umod), a glycoprotein produced in the kidneys, regulates salt transport, protecting against urinary tract infections, kidney stones, and kidney injury, contributing to innate immunity. After cleavage by the protease hepsin, Umod is secreted into the tubular lumen. We hypothesize that exogenous Umod may reduce injury associated with crystalline nephropathy. Both in vivo and in vitro models were used. Eight-week-old C57BL/6J male mice were treated with sodium oxalate (NaOx, 9 mg/100 g body wt) and/or Umod (5 µg/animal) and

Associations of Oxalate Consumption and Some Individual Habits with the Risk of Kidney Stones.

Kidney stone is a highly recurrent disease in the urinary tract system. Most kidney stones are calcium stones, usually consisting of either calcium oxalate or calcium phosphate. Supersaturation of soluble calcium, oxalate, phosphate, and citrate in the urine is the basis for calcium stone formation. Genetics, diet, low physical activity, and individual habits contribute to the formation of kidney stones. In this review, the associations of the risk of kidney stones with oxalate consumption and some individual habits, such as smoking, alcohol drinking, and opium consumption, are summarized.

The association of calcium oxalate deposition in kidney allografts with graft and patient survival.

BACKGROUND: Whether calcium oxalate (CaOx) deposition in kidney allografts following transplantation (Tx) adversely affects patient outcomes is uncertain, as are its associated risk factors. METHODS: We performed a retrospective cohort study of patients who had kidney allograft biopsies performed within 3 months of Tx at Brigham and Women's Hospital and examined the association of CaOx deposition with the composite outcome of death or graft failure within 5 years. RESULTS: Biopsies from 67 of 346 patients (19.4%) had CaOx deposition. In a multivariable logistic regression model, higher serum

P2X7 Receptor Stimulation Is Not Required for Oxalate Crystal-Induced Kidney Injury.

Oxalate crystal-induced renal inflammation is associated with progressive kidney failure due to activation of the NLRP3/CASP-1 inflammasome. It has been suggested previously that purinergic P2X7 receptor signaling is critical for crystal-induced inflammasome activation and renal injury. Therefore, we investigated the role of the P2X7 receptor in response to crystal-induced cytokine release, inflammation, and kidney failure using in vitro and in vivo models. Dendritic cells and macrophages derived from murine bone marrow and human peripheral blood mononucleated cells stimulated with calcium-oxa

Quantum Spin Liquid from a Three-Dimensional Copper-Oxalate Framework.

The quantum spin liquid (QSL) state is of great interest in relation to quantum computation and superconductivity and the search for new QSL materials is a current challenge in chemistry. Existing inorganic and molecular QSL compounds have two-dimensional structures, with spins arranged on triangular and kagome lattices, whereas three-dimensional structures with QSL characteristics are rare. In the copper-oxalate framework compound [(C2H5)3NH]2Cu2(C2O4)3, Cu(II) is coordinated with three bisbidentate oxalate bridges to form a three-dimensional (10,3) lattice and this produces a strong antiferr

A retrospective study of the prevalence of calcium oxalate crystals in veterinary Aspergillus cases.

Fungi in the genus Aspergillus are some of the most common fungal pathogens in veterinary species, primarily affecting the respiratory tract. In both human and veterinary cases, calcium oxalate crystals have been documented in sites of Aspergillus infection. Cases in multiple species (16 birds, 15 horses, 5 dogs, 1 ox, and 1 dolphin) were identified that had either positive cultures for Aspergillus sp., or had conidiophores present that could be identified as belonging to the genus Aspergillus. Histologic slides were examined to confirm the presence of oxalate crystals and how often they were

Feline Calcium Oxalate Urolithiasis: Risk factors and rational treatment approaches.

PRACTICAL RELEVANCE: Uroliths occur commonly in the bladder and/or urethra of cats and can be lifethreatening if urethral obstruction occurs. Calcium oxalate accounts for 40-50% of urocystoliths and these stones are not amenable to medical dissolution; therefore, removal by surgery or minimally invasive techniques is required if uroliths must be treated. Medical protocols for prevention involve decreasing urine saturation for minerals that form uroliths. ETIOPATHOGENESIS: Formation of uroliths is not a disease, but rather a complication of several disorders. Some disorders can be identified a

Acute oxalate nephropathy following kidney transplantation: Report of three cases.

Calcium oxalate (CaOx) crystal deposition is a common finding immediately after kidney transplantation. However, small depositions of CaOx could be benign while extensive depositions lead to poor graft outcome. Here we report three cases with end-stage renal disease (ESRD), bilateral nephrolithiasis, and unknown diagnosis of primary hyperoxaluria (PH) who underwent a renal transplant and experienced an early-onset graft failure. Although an acute rejection was suspected, renal allograft biopsies and subsequent allograft nephrectomies showed extensive CaOx deposition, which raised a suspicion o

Plasma oxalate following kidney transplantation in patients without primary hyperoxaluria.

BACKGROUND: Patients with primary hyperoxaluria may need repeated kidney transplants due to damage from oxalic acid (oxalate) deposits. However, oxalate may also be potentially harmful in all transplant recipients. Determinants of oxalate following transplantation have not been well studied. METHODS: Two hundred and twelve recipients admitted for transplantation were included in the study. Blood samples for measurement of oxalate and other relevant laboratory parameters were collected at baseline and subsequently 10 weeks after transplantation. For oxalate determination, samples were obtained

Calcium oxalate deposition in renal allografts: morphologic spectrum and clinical implications.

Many aspects of calcium oxalate (CaOx) deposition in renal transplant biopsies are not known. Review of all renal transplant biopsies performed in a 7-year period showed that CaOx deposition could be classified into three groups. Group I: Seven biopsies within a month post-transplant displayed rare CaOx foci against a background of acute tubular necrosis or acute cell-mediated rejection. At follow-up, five grafts functioned well and two failed due to chronic allograft nephropathy. CaOx in this context was an incidental finding secondary to a sudden excretion of an end-stage renal disease-induc

Calcium oxalate crystal deposition in necrotizing otomycosis caused by Aspergillus niger.

Numerous calcium oxalate crystals were present within fruiting heads of Aspergillus niger and among necrotic debris in a case of bilateral invasive otomycosis occurring in a diabetic female with end stage renal disease. This is the first report of in vivo calcium oxalate crystal deposition associated with Aspergillus niger at this anatomic site. The presence of localized oxalate crystals within necrotic tissue from the external auditory canal is presumptive of otomycosis caused by Aspergillus niger, and may serve as a diagnostic clue to the etiologic agent before histologic demonstration of hy

Effect of pyridoxine supplementation on plasma oxalate concentrations in patients receiving dialysis.

Plasma oxalate and erythrocyte glutamic oxaloacetate transaminase activity (EGOT) (an indicator of nutritional status with respect to pyridoxine) were measured in 21 patients maintained on regular continuous ambulatory peritoneal dialysis or haemodialysis before and after a 4-month period of supplementation with pyridoxine, 100 mg day-1. Prior to supplementation 10/21 patients showed subnormal EGOT activity, although the increment in activity on addition of pyridoxal-5-phosphate in vitro was within the normal range in all cases. Mean plasma oxalate was 31.5 mumol l-1 (SEM 2.9) prior to supplem

Calcium oxalate crystals in the thyroid. Their identification, prevalence, origin, and possible significance.

Calcium oxalate crystals are not encountered in normal animal tissues, except for the human thyroid, where they were found in 79 of 100 routine consecutive autopsies. They appear during childhood, and numbers of crystals increase with age. In diffuse hyperplasia, prevalence was higher, but crystals were fewer than expected. In adenomas and carcinomas, crystals were decreased except for three cases with a striking focal increase. None was found in 22 adult primate thyroids. After Clorox digestion of human thyroids, calcium oxalate dihydrate was identified by x-ray diffraction and infrared spect

Calcium oxalate crystal growth in normal urine: role of contraceptive hormones.

Oral contraceptives inhibited the growth of calcium oxalate crystals in female urine, and the growth rate inhibition depended on the dose of ethinyl-oestradiol in the oral contraceptive agent. The results suggest that the crystallization kinetics of calcium oxalate in urine could be under the control of oestrogenic hormones by an unidentified mechanism.

Calcium oxalate microcrystalline-associated arthritis in end-stage renal disease.

Chronic renal failure is known to be associated with secondary hyperoxalemia and the deposition of calcium oxalate in visceral organs, bones, and cartilage. We report the identification of calcium oxalate crystals in the synovial fluid of three patients with chronic renal failure. In one patient, calcium oxalate crystals were also identified within synovium and cartilage. Crystals were pleomorphic and bipyramidal. Some crystals were rod-like and had positive birefringence, thus tending to be confused with calcium pyrophosphate dihydrate when observed with only compensated polarized light micro
