Modulation of peroxisomal compartment by Epstein-Barr virus.

Viruses utilize clever strategies of interacting with various cellular factors, to remodel an organelle function, for the establishment of successful infection. In recent decades, numerous studies revealed the exploitation of the peroxisomal compartment by viruses. Epstein-Barr virus (EBV) is a ubiquitous virus linked with various cancers and neurological disorders. Till now, there is no report regarding the impacts of EBV infection on peroxisomal compartment. Therefore, we investigate the modulation of peroxisomal proteins in EBV transformed cell lines and during acute EBV infection. EBV posi

Dysfunctional peroxisomal lipid metabolisms and their ocular manifestations.

Retina is rich in lipids and dyslipidemia causes retinal dysfunction and eye diseases. In retina, lipids are not only important membrane component in cells and organelles but also fuel substrates for energy production. However, our current knowledge of lipid processing in the retina are very limited. Peroxisomes play a critical role in lipid homeostasis and genetic disorders with peroxisomal dysfunction have different types of ocular complications. In this review, we focus on the role of peroxisomes in lipid metabolism, including degradation and detoxification of very-long-chain fatty acids, b

Clinical and Biochemical Pitfalls in the Diagnosis of Peroxisomal Disorders.

Peroxisomal disorders are a heterogeneous group of genetic metabolic disorders, caused by a defect in peroxisome biogenesis or a deficiency of a single peroxisomal enzyme. The peroxisomal disorders include the Zellweger spectrum disorders, the rhizomelic chondrodysplasia punctata spectrum disorders, X-linked adrenoleukodystrophy, and multiple single enzyme deficiencies. There are several core phenotypes caused by peroxisomal dysfunction that clinicians can recognize. The diagnosis is suggested by biochemical testing in blood and urine and confirmed by functional assays in cultured skin fibrobl

[Peroxisome proliferator activated receptors and cardiovascular disorders].

Peroxisome proliferator activated receptors (PPARs) are transcription factors only recently discovered. Nevertheless, the interest surrounding their study has involved and involves a continuously growing number of researchers. This is due to the role that PPARs play in the understanding of the pathophysiology of clinical conditions such as obesity, diabetes, atherosclerosis, and angiogenesis. Lipid and glucidic metabolism, synthesis of cytokines, adhesion molecules, coagulation factors, fibrinolysis, are only few of the several processes controlled by PPARs. In this review the more recent acqu

Peroxisome proliferator-activated receptors in cutaneous biology.

Spontaneous cerebrospinal fluid (CSF) rhinorrhoea, often linked to benign intracranial hypertension (BIH), arises from skull base defects and carries a risk of serious complications such as meningitis. While acetazolamide can temporarily reduce intracranial pressure, recurrence remains problematic. This prospective study aims to assess the relationships between body mass index (BMI), defect size, disc edema severity, and recurrence duration, and to compare outcomes of conservative versus surgical management. A prospective observational study was conducted on 30 adults with spontaneous CSF rhin

Peroxisome proliferator-activated receptors in inflammation control.

Peroxisome proliferator-activated receptors (PPARs) are ligand-activated transcription factors belonging to the nuclear receptor superfamily. PPARalpha is highly expressed in liver, skeletal muscle, kidney, heart and the vascular wall. PPARgamma is predominantly detected in adipose tissue, intestine and macrophages. PPARs are activated by fatty-acid derivatives and pharmacological agents such as fibrates and glitazones which are specific for PPARalpha and PPARgamma respectively. PPARs regulate lipid and lipoprotein metabolism, glucose homeostasis, cell proliferation and differentiation, and ap

Pendular nystagmus in patients with peroxisomal assembly disorder.

BACKGROUND: Pendular nystagmus commonly occurs in congenital and acquired disorders of myelin. OBJECTIVE: To characterize the nystagmus in 3 siblings with an infantile form of an autosomal recessive peroxisomal assembly disorder causing leukodystrophy. DESIGN: We examined visual function and measured eye movements using infrared oculography. We noted changes in eye speed and frequency before and after the administration of gabapentin to 1 patient. RESULTS: All 3 siblings showed optic atrophy and pendular nystagmus that was predominantly horizontal, at a frequency of 3 to 6 Hz, with phase sh

Peroxisomal localization of alpha-oxidation in human liver.

It was found that alpha-oxidation in rat liver is a peroxisomal process, consisting of an activation, a 2-hydroxylation, and a reaction leading to the production of formate. alpha-Oxidation of 3-methyl-substituted fatty acids was quantified by measuring the production of formate and CO2, and the production of a 2-hydroxy-3-methylacyl-CoA-intermediate was demonstrated. We wanted to extend these findings to human liver, in view of the controversy over the subcellular localization of alpha-oxidation in man. In homogenates from human liver, rates of alpha-oxidation were highest when measured in th

Characterization of peroxisome-deficient mutants of Hansenula polymorpha.

In the methylotrophic yeast Hansenula polymorpha, approximately 25% of all methanol-utilization-defective (Mut-) mutants are affected in genes required for peroxisome biogenesis (PER genes). Previously, we reported that one group of per mutants, termed Pim-, are characterized by the presence of a few small peroxisomes with the bulk of peroxisomal enzymes located in the cytosol. Here, we describe a second major group of per mutants that were observed to be devoid of any peroxisome-like structure (Per-). In each Per- mutant, the peroxisomal methanol-pathway enzymes alcohol oxidase, catalase and

[Clinical and molecular aspects of peroxisome-deficient disorders].

Peroxisome-deficient disorders including Zellweger syndrome (ZS), neonatal adrenoleukodystrophy (NALD) and infantile Refsum disease (IRD) are characterized by the absence of peroxisomes associated with secondary multiple enzyme deficiencies and by a defect in the neuronal migration. Collaborative complementation studies revealed the presence of at least 9 genetic groups among these disorders. Clinical phenotypes did not correlate with the genetic grouping. Responsible gene for one of these groups (group F) was determined as peroxisome assembly factor-1 (PAF-1) by the functional cloning using p

Dehydroepiandrosterone-induced peroxisome proliferation in the rat liver.

We have investigated the effect of dehydroepiandrosterone (DHEA), a naturally occurring steroid hormone, on hepatic peroxisomes in the rat. Dietary administration of DHEA at a concentration of 0.45% for 2 weeks resulted in a 200% increase in the liver weight and a more than 5.0-fold increase in the volume density of peroxisomes. SDS-polyacrylamide gel electrophoresis of the postnuclear fraction of livers showed a marked increase in the 80,000-molecular weight polypeptide. Northern blot analysis of total RNA showed 2- and 12-fold increases, respectively, in the levels of catalase and peroxisoma

Peroxisomal dysfunction in chondrodysplasia punctata, rhizomelic type.

The rhizomelic type of chondrodysplasia punctata (RCDP) is recognizable at birth because of the typical phenotype and radiological features. Most patients die young, some survive until their teens but all are severely retarded. Recent studies showed RCDP to be a peroxisomal disorder. Peroxisomal investigations may be important in defining the prognosis for an individual patient, and are definitely of use in antenatal diagnosis.

Infantile Refsum's disease: a peroxisomal storage disorder?

An 18-month-old infant presented with a history of arrest of neurological development from the age of eight months, with progressive ataxia, deafness, retinitis pigmentosa and hepatomegaly. Biochemical investigations revealed an elevated plasma phytanic acid level and deficiency of phytanic acid oxidase in skin fibroblasts. Histopathological findings in a liver biopsy were similar to those reported in infantile phytanic acid storage disease. Unexpected findings were the presence of elevated levels of plasma pipecolic acid, and elevated plasma long-chain fatty acid ratios, biochemical findings

Peroxisomal Lipid Metabolism as a Therapeutic Target in Leukemia.

Lipid delivery and metabolism profoundly shape cancer cell fate, energy balance, redox control, and therapy resistance. Peroxisomes regulate fatty acid oxidation (FAO) and oxidative homeostasis with growing relevance in hematologic malignancies. Recent studies show peroxisomal FAO (pFAO) is selectively elevated in acute myeloid leukemia (AML) and chronic lymphocytic leukemia (CLL), sustaining cancer cell survival under metabolic and therapeutic stress. Elevated ACOX1, ABCD1/2, CROT, and PEX5 expression underscores leukemia-specific peroxisomal vulnerability; their pharmacologic or genetic inhi

The role of peroxisome proliferator-activated receptors in endometriosis.

Endometriosis constitutes the most common cause of chronic pelvic pain in female patients and is associated with infertility. Although there is no known cause for the disease, it is a heritable condition that is determined by numerous genetic, epigenetic, and environmental aspects. Peroxisome proliferator-activated receptors (PPARs) represent nuclear receptor proteins that control gene expression. By using the MEDLINE and LIVIVO databases we conducted a literature review in order to look into the role of PPARs in the endometriosis pathophysiology and succeeded in revealing 36 pertinent publica

The Role of Peroxisome Proliferator-Activated Receptors in Preeclampsia.

Preeclampsia is a common pregnancy-related hypertensive disorder. Often presenting as preexisting or new-onset hypertension complicated by proteinuria and/or end-organ dysfunction, preeclampsia significantly correlates with maternal and perinatal morbidity and mortality. Peroxisome proliferator-activated receptors (PPARs) are nuclear receptor proteins that regulate gene expression. In order to investigate the role of PPARs in the pathophysiology of preeclampsia, we conducted a literature review using the MEDLINE and LIVIVO databases. The search terms "peroxisome proliferator-activated receptor
