A novel amide-bonded silica stationary phase was prepared starting from N-Boc-phenylalanine, cyclohexylamine and spherical silica gel (4 µm, 60 Å). The amide ligand was synthesised with high yield. The resulting amide bonded stationary phase was characterised by SEM, IR and elemental analysis. The resulting selector bearing a polar amide group is used for the reversed-phase chromatography separation of different classes of thirteen phytohormones (plant hormones). The chromatographic behaviours of these analytes on the amide-silica stationary phase were compared with those of RP-C18 column unde

Liquid-liquid phase separation (LLPS), a fundamental biophysical process driven by multivalent weak interactions, regulates diverse cellular physiological functions. Ion channels are critical transmembrane protein complexes governing electrophysiological signaling, yet their phase separation characteristics remain largely uncharacterized. Here, we systematically assessed the phase-separation propensity of 140 human ion-channel proteins from the PhaSePred database, and 69 human ion channel proteins were predicted to have a high propensity for LLPS (prediction score ≥ 0.5). Based on the self-ass…

Liquid-liquid phase separation (LLPS) of α-Synuclein (α-Syn) is recognized as an early biophysical event driving pathological aggregation in Parkinson's disease (PD). Although 10%-15% of PD cases are due to familial mutations, the remaining cases are sporadic, often linked to various factors, like pesticides and metals. For example, rotenone and dopamine are known to be involved in PD pathology and are suggested to cause changes in α-Syn protein homeostasis, although the mechanism by which they influence α-Syn and cellular toxicity is largely unknown. In this work, we demonstrate that both dop…

The mitotic kinase Aurora-A is frequently overexpressed in cancers and contributes to tumor progression and therapy resistance, yet the mechanisms underlying its role in drug resistance remain unclear. Here, we show that sorafenib treatment triggers Aurora-A phase separation, leading to its recruitment into stress granules (SGs), membraneless organelles that promote cancer cell survival. Aurora-A facilitates robust SGs assembly, thereby conferring sorafenib resistance. Mechanistically, upon sorafenib-induced SGs formation, Aurora-A binds RNA via positively charged lysine/arginine residues with…

We numerically study motility-induced phase separation of active particles in two-dimensional convection rolls. We analyze local packing-fraction distributions, density fluctuations, the corresponding phase diagrams, and diffusivity curves to characterize the interplay between self-propulsion, global packing fraction, and advection strength. In the weak-flow regime, the system exhibits phase separation characterized by bimodal density distributions, slowly decaying density fluctuations, and a sharp reduction in diffusivity. Increasing advection suppresses clustering by enhancing particle trans…

Personal thermal management (PTM) textiles can reduce building energy consumption and improve personal comfort, yet their practical application is constrained by the inherent trade-off between flexibility, thermal insulation, and mechanical strength. Herein, porous single-component polyimide (PI) fibers were fabricated via coagulation bath-modulated wet spinning of 3,3',4,4'-benzophenone tetracarboxylic dianhydride (BTDA)-4,4'-oxydianiline (ODA) poly(amic acid) (PAA). By tuning the EtOH/H2O ratio (20/80-60/40) and winding speed (2.6-13.1 mm/s), the fiber cross-sectional morphology evolves from

The characteristic length scale of phase-separated organic thin film blends is a critical structural parameter governing the performance and functionality of organic electronic devices. The arrested morphologies of vapor-deposited organic thin films result from the interplay between thermodynamic driving forces and kinetic constraints during deposition. Here, we aim to isolate the role of kinetic effects in phase separation by varying the deposition rate at a constant substrate temperature for a co-deposited molecular glass blend of N,N'-bis(3-methylphenyl)-N,N'-diphenylbenzidine (TPD) and Dis…

BACKGROUND: Makorin-2 (MKRN2) is an E3 ubiquitin ligase involved in multiple biological processes, yet its role in neurological disorders remains poorly understood. This study aims to elucidate how MKRN2 regulates the RNA-binding protein CSDE1-a molecule linked to autism-related genes-and to explore the functional implications of this interaction in neurodevelopment.
METHODS: Using mass-spectrometry screening, we identified CSDE1 as a direct substrate of MKRN2. Ubiquitination sites were validated through mutagenesis of conserved lysine residues. Liquid-liquid phase separation (LLPS) assays we…

A butterfly-shaped B ← N open molecular box, (TPX)(BMCT)2(PhCl)4, is constructed via a [4 + 2] coordination mode, and the activated form (TPX)(BMCT)2 is obtained after removal of chlorobenzene guests. The two methyl groups on the central benzene ring of the tetradentate pyridine ligand create steric hindrance, forcing the dative B ← N bonds to become nearly parallel and achieving perfect geometric complementarity with the bidentate boronate ester. After activation, the characteristic infrared bands of the open molecular box remain unchanged, confirming its structural integrity. Static gravimet

Alzheimer's disease (AD) is one of the most common neurodegenerative diseases, and effective clinical treatment currently remains lacking. Ferulic acid (FA) has shown clinical effects in the treatment of AD, but the underlying mechanism of action remains unclear. Here, we showed that FA significantly enhances glycolytic metabolism by inhibiting Tau-441 liquid-liquid phase separation (LLPS). Through molecular docking and solution experiments, we demonstrated that FA binds to Tau-441 via hydrogen bonds, significantly reducing its β-sheet content, thereby effectively preventing the formation of T…

We investigate the dynamical pathways of a morphological transition in a two-dimensional active lattice gas undergoing motility-induced phase separation. The transition is between two locally stable morphologies of the liquid cluster: a system-spanning "slab" and a compact "droplet." We generate trajectories of this transition in both directions using forward flux sampling. We find that the droplet-to-slab transition always follows a similar mechanism to its equilibrium counterpart, but the reverse (slab-to-droplet) transition depends on rare nonequilibrium fluctuations. At low Péclet numbers …

Plasmodium falciparum malaria relies on antigenic variation mediated by mutually exclusive expression of var genes. Repressed var genes cluster together and are bound by PfHP1 (Plasmodium falciparum heterochromatin protein 1), but the regulatory mechanism remains unclear. Here, we show that PfHP1 undergoes phase separation in vitro and compartmentalizes DNA. This process is tunable by RNA and PfH2A.Z. Our single-molecule assays show that PfHP1 preferentially compacts AT-rich DNA. We identify point mutations in the intrinsically disordered region of PfHP1 that disrupt phase separation and DNA c…

Protein phase separation is a fundamental process in organizing membraneless organelles and is implicated in pathological conditions. Importantly, this process is dynamic and depends on conditions such as concentration, temperature, and solvent composition. However, current machine learning models infer phase separation propensity solely from amino acid sequences, failing to capture these context-dependent behaviors. Here we show that LLPSense, a machine learning framework that integrates protein language model embeddings with environmental parameters, achieves accurate, condition-aware predic…

Tau protein undergoes liquid-liquid phase separation (LLPS), forming dynamic condensates that act as intermediates in the transition to neurofibrillary tangles in Alzheimer's disease (AD). This review highlights tau's dynamic and reversible LLPS behavior, an underexplored aspect of tau's role, particularly as an early pathogenic driver in AD. We summarize the molecular mechanisms and regulatory factors governing tau LLPS, with a focus on its physiological functions in microtubule stability, synaptic activity, and cellular stress responses. Additionally, we discuss how metal ions, RNA, and neur…

The development of hydrophobic poly(ethylene glycol) (PEG) hydrogels, which are typically hydrophilic, could significantly enhance their application as artificial extracellular matrices. In this study, we designed PEG hydrogels with enhanced hydrophobicity through gel-gel phase separation (GGPS), a phenomenon that uniquely enhances hydrophobicity under ambient conditions, and we elucidated the pivotal role of elasticity in this process. We hypothesized that increased elasticity would amplify GGPS, thereby improving the hydrophobicity and cell adhesion on PEG hydrogel surfaces, despite their in…

Coacervation, a common method for microspheres preparation, has not been given sufficient attention and study. This study aims to clarify the key factors in the phase separation step. Firstly, based on the thermodynamics of polymer solutions, the phase equilibrium time, binodal line, and the relationship between the concentration of lactic-glycolic acid copolymer (PLGA) in the coacervate phase (CPLGA) and the concentration of polydimethylsiloxane (PDMS) in the PLGA-lean phase (CPDMS) were determined. It was found that the lactic:glycolic ratio (L:G) significantly influences phase separation, a…
