Characterization of simple sequence repeats (SSRs) from Phlebotomus papatasi (Diptera: Psychodidae) expressed sequence tags (ESTs).

BACKGROUND: Phlebotomus papatasi is a natural vector of Leishmania major, which causes cutaneous leishmaniasis in many countries. Simple sequence repeats (SSRs), or microsatellites, are common in eukaryotic genomes and are short, repeated nucleotide sequence elements arrayed in tandem and flanked by non-repetitive regions. The enrichment methods used previously for finding new microsatellite loci in sand flies remain laborious and time consuming; in silico mining, which includes retrieval and screening of microsatellites from large amounts of sequence data from sequence data bases using micros…

PCR-based detection of Leishmania major kDNA within naturally infected Phlebotomus papatasi in southern Iran.

The annual incidence of cutaneous leishmaniasis (CL) in Iran rose by 43% over a five year period, from 2002 to 2006; most of these cases were caused by Leishmania major. Two complementary standard and nested polymerase chain reactions (PCRs) were used to detect parasites within their natural vector, Phlebotomus papatasi. Twelve different sand fly species were morphologically identified. The most abundant species (31.3%) was P. papatasi. Leptomonads were found in nine (2.4%) phlebotomines. Twenty (5.3%) sand fly species were found positive for Leishmania-genus DNA using standard PCR. The infect…

First Findings and Molecular Data of Phlebotomus mascittii (Diptera: Psychodidae) in the Cantabrian Cornice (Northern Spain).

Phlebotomus (Transphlebotomus) mascittii Grassi, 1908 (Diptera: Psychodidae) has been found in several European countries. In Spain, sporadic records were reported in the early '80s in Catalonia (Northeast Spain), and it was never detected again. Recent entomological surveys carried out between 2004 and 2020 revealed the presence of several specimens of P. mascittii in Spain. The species identification was confirmed by both morphological and molecular analyses. The analyzed specimens belonged to the haplotype (COI_2) defined by one polymorphic site compared to other European specimens. Phlebot…

A scalable and reproducible manufacturing process for Phlebotomus papatasi salivary protein PpSP15, a vaccine candidate for leishmaniasis.

Cutaneous leishmaniasis is a parasitic and neglected tropical disease transmitted by the bites of sandflies. The emergence of cutaneous leishmaniasis in areas of war, conflict, political instability, and climate change has prompted efforts to develop a preventive vaccine. One vaccine candidate antigen is PpSP15, a 15 kDa salivary antigen from the sandfly Phlebotomus papatasi that facilitates the infection of the Leishmania parasite and has been shown to induce parasite-specific cell-mediated immunity. Previously, we developed a fermentation process for producing recombinant PpSP15 in Pichia pa…

First molecular identification of Leishmania major in Phlebotomus papatasi in an outbreak cutaneous leishmaniasis area in Iraq.

Cutaneous leishmaniasis (CL) is transmitted by Phlebotomine sand fly vectors, among which Phlebotomus papatasi is prevalent in Western Asia, Northern Africa and Southern Europe, and it is known as a vector for Leishmania major parasite in the world. However, in Iraq, morphological studies showed that P. papatasi is a predominant sand fly species and hypothesised to transmit CL causing Leishmania species including L. major and L. tropica. Few studies have found Leishmania species in sand flies in mixed pools of samples in this country. Accurate identification of sand flies as vectors of Leishma…

Understanding Phlebotomus perniciosus abundance in south-east Spain: assessing the role of environmental and anthropic factors.

BACKGROUND: Leishmaniosis is associated with Phlebotomus sand fly vector density, but our knowledge of the environmental framework that regulates highly overdispersed vector abundance distributions is limited. We used a standardized sampling procedure in the bioclimatically diverse Murcia Region in Spain and multilevel regression models for count data to estimate P. perniciosus abundance in relation to environmental and anthropic factors. METHODS: Twenty-five dog and sheep premises were sampled for sand flies using adhesive and light-attraction traps, from late May to early October 2015. Temp…

Implication of vector characteristics of Phlebotomus argentipes in the kala-azar elimination programme in the Indian sub-continent.

BACKGROUND: Visceral leishmaniasis (VL), also known as kala-azar in the Indian sub-continent (ISC), is a major public health concern in Bangladesh, India, and Nepal, where it is caused by Leishmania donovani transmitted by the sand fly Phlebotomus argentipes. Various ecological parameters including air temperature, rainfall, wind speed, relative humidity, soil moisture, pH, and organic carbon are known to influence the oviposition of female sand flies, as well as the survival and development of larvae. However, more detailed knowledge on vector behavior, such as biting times, breeding places, …

[Effects of the development of irrigation systems in the arid areas on the establishment of Phlebotomus (Larroussius) perfiliewi Parrot, 1939].

In Tunisia, Phlebotomus (Larroussius) perfiliewi Parrot, 1939, is highly suspected to be involved in the transmission of sporadic cutaneous leishmaniasis. The geographical distribution of this sandfly species is limited to the northern part of Tunisia and more specifically to the humid, sub-humid and semi-arid bio-geographical areas. To date, the geographical distribution of P. perfiliewi and subsequently of the sporadic cutaneous leishmaniasis extends to the arid bio-geographical areas located in central Tunisia. This extension is related to the intense development of irrigation systems in th…

Sub-additive effect of conspecific eggs and frass on oviposition rate of Lutzomyia longipalpis and Phlebotomus papatasi.

Oviposition behavior is a fairly neglected aspect in our understanding of the biology of sand flies. In this study, we used a comparative approach using both new- and old-world species (Lutzomyia longipalpis and Phlebotomus papatasi) in choice and no-choice oviposition chambers to evaluate the effect of old sand fly colony remains (frass), conspecific eggs, and their combination on oviposition rates of these sand flies. We also tested the effect of egg washing with de-ionized water on oviposition rates. In both choice and no-choice experiments, sand fly species laid more eggs on a substrate co…

Characterization of the antibody response to the saliva of Phlebotomus papatasi in people living in endemic areas of cutaneous leishmaniasis.

Important data obtained in mice raise the possibility that immunization against the saliva of sand flies could protect from leishmaniasis. Sand fly saliva stimulates the production of specific antibodies in individuals living in endemic areas of parasite transmission. To characterize the humoral immune response against the saliva of Phlebotomus papatasi in humans, we carried out a prospective study on 200 children living in areas of Leishmania major transmission. We showed that 83% of donors carried anti-saliva IgG antibodies, primarily of IgG4 isotype. Positive sera reacted differentially wit…

Natural infection of Phlebotomus argentipes with Leishmania and other trypanosomatids in a visceral leishmaniasis endemic region of Nepal.

Monitoring Leishmania infection in sand flies is important for understanding the eco-epidemiology of kala-azar and assessing the impact of t … nia group. Our results highlight the diversity of flagellate infections likely to be encountered in Phlebotomus argentipes populations. Our methodology allows clear discrimination of Leishmania from other trypanosomatids and should be applied on larger insect samples or in longitudinal studies.

The efficacy of indoor CDC light traps for collecting the sandfly Phlebotomus argentipes, vector of Leishmania donovani.

The efficacy of light traps for collecting sandflies (Diptera: Psychodidae) varies both inter-specifically and intra-specifically (by gender and physiological status) as a result of significant differences in phototropic and other behavioural characteristics. The efficacy of miniature CDC light traps for collecting Phlebotomus argentipes Annandale & Brunetti, a vector of Leishmania donovani Laveran & Mesnil (Kinetoplastida: Trypanosomatidae), was assessed in the Indian state of Bihar. Sandflies were collected during the night from 16 houses in each of three villages over 3 months (four times a…

Laboratory demonstration of the acquisition and development of leishmania major in the sand fly Phlebotomus kazeruni (Diptera: Psychodidae).

Phlebotumus kazeruni, a blood-feeding, xerophilic sand fly species found broadly throughout North Africa and Western Asia, is a suspected vector of cutaneous leishmaniasis (CL). Following successful laboratory colonization of this species, we employed the murine (BALB/c) infection model to determine whether our Sinai strain of P. kazeruni was able to successfully acquire, develop, and transmit a Sinai strain of Leishmania major. Groups of female sand flies were fed 1) by membrane, hamster blood containing culture-produced L. major promastigotes, 2) by membrane, hamster blood containing a suspe

Delayed-type hypersensitivity to Phlebotomus papatasi sand fly bite: An adaptive response induced by the fly?

The saliva of bloodsucking arthropods contains a large array of pharmacologically active compounds that assist hematophagy. Arthropod saliva is also responsible for causing uncomfortable allergic responses in its vertebrate hosts. In this article, we investigate whether the sand fly Phlebotomus papatasi, known to produce a strong delayed-type hypersensitivity (DTH) in humans, could benefit from, and possibly adaptively induce, this response in their vertebrate hosts. In this study, we show that flies fed on humans to completion nearly twice as fast in DTH sites as compared with normal skin sit…

Protective efficacy of lambdacyhalothrin-impregnated bednets against Phlebotomus orientalis, the vector of visceral leishmaniasis in Sudan.

Field investigations on the sandfly Phlebotomus orientalis (Diptera: Psychodidae), the vector of Leishmania donovani causing visceral leishmaniasis (VL) in Sudan, were undertaken in two villages (Bellow and Elgamel) and Dinder National Park, to determine the protective value of bednets (polyester, 100 denier) impregnated with lambda-cyhalothrin 10 mg a.i./m2 pyrethroid insecticide. After exposure to treated netting for 30 s, P. orientalis females all died within 1 h. When field-tested in Acacia woodland, treated bednets provided complete protection from bites of the vector. Numbers of P. orien…

Phlebotomus-transmitted toscana virus infections of the central nervous system: a seven-year experience in Tuscany.

Toscana virus (TOSv) is a recently discovered Phlebotomus-transmitted human pathogen involved in acute infections of the central nervous system (CNS) occurring during the summer in natural foci in Italy. The purpose of this prospective study was to investigate the role of this virus in 170 patients with meningitis-meningoencephalitis of suspected viral origin, admitted to the Departments of Infectious Diseases at the Siena Hospital from 1990 to 1996. Infections caused by tick-borne encephalitis virus (TBEv) and TOSv or other neurotropic viruses were routinely diagnosed by means of conventional…
