Methods for the fractionation and estimation of serum kallikrein in the blood of healthy and rhesus monkeys infected with Plasmodium knowlesi are discussed. A total mean increase of 4·5 ng. bradykinin equivalents/μg. protein content of kallikrein was shown to be present in the systemic circulation in the P. knowlesi-infected animal. A remarkable fall in the bradykininogen (BKG) content in infected animal is also observed. The kallikrein from an infected animal is observed to be more active than that from control in enhancing capillary permeability, and in the development of small vessels damag…

Monkey erythrocytes infected with schizonts of Plasmodium knowlesi are agglutinated when mixed with serum taken from chronically infected monkeys (Eaton, 1938). This reaction has proved highly specific and capable of distinguishing antigenic variants of one strain. Chronic infections of P. knowlesi in rhesus monkeys are maintained by a succession of antigenically distinct populations, each population stimulating a specific agglutinin response. Additional agglutinins which are not variant specific are found at low titre in sera taken after an infection has persisted for a month or more. The pos…

Plasmodium infection induces extensive remodeling of host erythrocytes through parasite-exported proteins that interact with host cell components. Among these, tryptophan-rich antigens (TRAgs) are conserved across Plasmodium species and have been proposed as vaccine candidates, but their molecular functions remain largely unexplored. In this study, we characterized Plasmodium knowlesi tryptophan-rich antigen 40.1 (PkTRAg40.1) to investigate its localization, biochemical properties, host interactions, and immunogenicity. Recombinant PkTRAg40.1 showed no detectable binding to intact human erythr…

BACKGROUND: Thailand is committed to eliminating malaria by 2024. From 2013 to 2020, the total number of malaria cases have decreased, from 37,741 to 4474 (an 88.1% reduction). However, infections with Plasmodium knowlesi, a monkey malarial pathogen that can also infect humans, have been increasingly observed. This study focused on the molecular analysis of P. knowlesi parasites causing malaria in Thailand.
METHODS: Under Thailand's integrated Drug Efficacy Surveillance (iDES), which includes drug-resistance monitoring as part of routine case-based surveillance and responses, specimens were c…

Population genetic analysis revealed that Plasmodium knowlesi infections in Malaysian Borneo are caused by 2 divergent parasites associated with long-tailed (cluster 1) and pig-tailed (cluster 2) macaques. Because the transmission ecology is likely to differ for each macaque species, we developed a simple genotyping PCR to efficiently distinguish between and survey the 2 parasite subpopulations. This assay confirmed differences in the relative proportions in areas of Kapit division of Sarawak state, consistent with multilocus microsatellite analyses. Analyses of 1,204 human infections at Kapit…

Most malaria in Malaysia is caused by Plasmodium knowlesi parasites through zoonotic infection from macaque reservoir hosts. We obtained genome sequences from 28 clinical infections in Peninsular Malaysia to clarify the emerging parasite population structure and test for evidence of recent adaptation. The parasites all belonged to a major genetic population of P. knowlesi (cluster 3) with high genomewide divergence from populations occurring in Borneo (clusters 1 and 2). We also observed unexpected local genetic subdivision; most parasites belonged to 2 subpopulations sharing a high level of d…

In vitro studies of sexual blood stages of the most fatal malaria species, Plasmodium falciparum, have revealed key processes by which gametocytes develop and transmit infection from humans to anopheline mosquitoes. However, most malaria cases outside sub-Saharan Africa are caused by other Plasmodium spp., frequently Plasmodium vivax and Plasmodium knowlesi, a zoonotic parasite of macaque monkeys. Gametocytes of P. vivax and P. knowlesi exhibit distinct morphology, faster development, and a shorter life span compared with gametocytes of P. falciparum, reflecting the evolutionary separation and…

Plasmodium knowlesi is increasingly recognized as a major cause of malaria in Southeast Asia. Anopheles leucosphyrous group mosquitoes transmit the parasite and natural hosts include long-tailed and pig-tailed macaques. Despite early laboratory experiments demonstrating successful passage of infection between humans, the true role that humans play in P. knowlesi epidemiology remains unclear. The threat posed by its introduction into immunologically naïve populations is unknown despite being a public health priority for this region. A two-host species mathematical model was constructed to analy…

Infections of humans with the zoonotic simian malaria parasite Plasmodium knowlesi occur throughout Southeast Asia, although most cases have occurred in Malaysia, where P. knowlesi is now the dominant malaria species. This apparently skewed distribution prompted an investigation of the phylogeography of this parasite in 2 geographically separated regions of Malaysia, Peninsular Malaysia and Malaysian Borneo. We investigated samples collected from humans and macaques in these regions. Haplotype network analyses of sequences from 2 P. knowlesi genes, type A small subunit ribosomal 18S RNA and cy…

Plasmodium knowlesi is a potentially life-threatening zoonotic malaria parasite due to its relatively short erythrocytic cycle. Microscopic identification of P. knowlesi is difficult, with "compacted parasite cytoplasm" being one of the important identifying keys. This report is about a case of hyperparasitaemic human P. knowlesi infection (27% parasitaemia) with atypical amoeboid morphology. A peninsular Malaysian was admitted to the hospital with malaria. He suffered anaemia and acute kidney function impairment. Microscopic examination, assisted by nested PCR and sequencing confirmed as P. k…

We describe a PCR-confirmed case of Plasmodium knowlesi infection with a high parasitemia level and clinical signs of severe malaria in a migrant worker from Malaysian Borneo in the Netherlands. Investigations showed that commercially available rapid antigen tests for detection of human Plasmodium infections can detect P. knowlesi infections in humans.

Two parasite-derived antigens (designated band 1 and band 2) were identified upon double-diffusion analysis of culture medium from reinvading culture of P. knowlesi. Band 1 antigen showed characteristics with the R-antigens of P. falciparum while the other was similar to the P. falciparum L-antigens. Both antigens appeared to be largely particulate in character and did not display any obvious variant-specificity. Analysis of plasma from rhesus monkeys infected with the same antigenic variant showed the presence of band 1 antigen only.

We describe 2 cases of Plasmodium knowlesi malaria in persons from Israel who traveled to Thailand in 2023. One patient, likely infected in northwestern Thailand, might signal geographic expansion into areas not previously associated with human infection. The infection's rarity in travelers, diagnostic challenges, and potential severity underscore the importance of clinical awareness.

Plasmodium knowlesi malaria infection in humans has been reported throughout southeast Asia. The communities at risk are those living in areas where Macaque monkeys and Anopheles mosquito are present. Zoonotic malaria control is challenging due to the presence of the reservoir host and the possibility of human-vector-human transmission. Current control measures, including insecticide-treated nets (ITNs) and indoor residual spraying (IRS), are insufficient to address this threat due to gaps in protection associated with outdoor and early evening vector biting and social and economic activities,…

Despite recent success in reducing the regional incidence of Plasmodium falciparum malaria, cases of zoonotic malaria are on the rise in Southeast Asia. The Cambodian National Malaria Surveillance Program has previously relied on rapid diagnostic tests and blood smear microscopy with confirmatory polymerase chain reaction (PCR) testing in a subset of cases to further distinguish P. falciparum, P. malariae, P. ovale, and P. vivax species. Here, metagenomic next-generation sequencing identified P. knowlesi mono-infection in six Cambodian patients initially diagnosed with P. malariae by blood sme
