Uterine Leiomyoma and Reproductive Tract Infections Detected by Polymerase Chain Reaction.

BACKGROUND AND OBJECTIVE: For nearly a century, it has been suspected that reproductive tract infections play an etio- logic role in uterine leiomyoma. However, no epidemiologic study of leiomyoma has used the polymerase chain reaction (PCR) to compare uterine tissues from cases and non-cases, and to investigate associations between uterine leiomyoma and infections detected by PCR. METHODS: In this case-control study, 92 leiomyoma tissues from cases, and 94 myometrial tissue from controls were screened by PCR for cytomegalovirus, Chlamydia trachomatis, herpes simplex virus-1, 2, and human pap…

Rapid detection of Borrelia burgdorferi strains by nested polymerase chain reaction.

This study was carried out to evaluate the nested PCR for specific detection of different strains of B. burgdorferi. Five strains of B. burgdorferi including ACA-1, B-31, 2B45, 3B45, 7B49 obtained from different countries were used in this study. The strains of B. hermsii, Escherichia. coli and T. pallidum were also included as control strains. Two pairs of nested PCR primers were used to amplify the gene encoding the Osp-A protein of B. burgdorfer under standard PCR condition. In a two stage procedure, nested PCR yielded a positive reaction for five tested strains of B. burgdorferi. None the

Detection of schistosomes polymerase chain reaction amplified DNA by oligochromatographic dipstick.

The applications of highly specific and sensitive molecular techniques based on polymerase chain reaction (PCR) have constituted a valuable tool for the diagnosis of schistosomiasis and also for the detection of schistosome infections in the snail intermediate hosts. The common method of detecting PCR amplicons is gel electrophoresis in the presence of ethidium bromide, a carcinogen, which is followed by UV transillumination. Other methods, which are available for detecting PCR products, are real-time PCR, PCR-enzyme-linked immunosorbent assay (PCR-ELIZA) and mass spectrometry but they are cum…

Application of polymerase chain reaction fingerprinting to differentiate Ornithobacterium rhinotracheale isolates.

Ornithobacterium rhinotracheale (ORT) is an infectious respiratory pathogen of chickens, turkeys, and wild birds. There are 18 serotypes of ORT reported worldwide. In this study, enterobacterial repetitive intergenic consensus (ERIC) polymerase chain reaction and random amplified polymorphic DNA assay with Universal M13 primer-based fingerprinting techniques were investigated for their ability to differentiate ORT isolates. The authors examined 50 field isolates and 8 reference strains of ORT for their genetic differences. The fingerprint patterns were compared with serotyping results of ORT b…

Diagnosis of intra-oral MALT lymphoma using seminested polymerase chain reaction.

Diagnosis of mucosa-associated lymphoid tissue (MALT) lymphoma based on histological examination alone is difficult. We report three patients with histologically suspected MALT lymphoma who developed lymphoproliferative lesions of the sublingual gland. Seminested polymerase chain reaction (PCR) analysis applied to formalin-fixed and paraffin-embedded specimens showed clonal rearrangement of immunoglobulin heavy chain genes in two patients and a polyclonal characteristic in one. The clinical findings and Southern blot analysis confirmed the accuracy of the diagnosis. The molecular method descri…

Detection of pathogenic leptospiral DNA in urine by polymerase chain reaction.

Diagnosis of leptospirosis is currently based on serological tests detecting antibodies against the spirochete. The standard method is the microscopic agglutination test (MAT), which is serovar-specific, requires a period of antibody development, and increases the risk of exposure to viable organisms. Therefore, the present study aimed to develop a rapid, sensitive, specific, and safe method based on the PCR technique to detect pathogenic Leptospira in urine samples. Nested PCR using two sets of primers, external and internal primers, were shown to specifically amplify 16S rRNA target of patho

Detection of Babesia equi (Laveran, 1901) by nested polymerase chain reaction.

We describe a nested polymerase chain reaction (PCR) for the detection of Babesia equi in equine infected erythrocytes using oligonucleotides designed on the published sequence of a B. equi merozoite antigen gene (ema-1). A 102bp DNA fragment is specifically amplified from B. equi but not from Babesia caballi, Babesia bovis or Babesia bigemina DNA. In a mock infection we were able to detect down to six infected cells in 10(8) equine erythrocytes or to detect the parasite in blood with an equivalent parasitemia of 0.000006%. Furthermore, gene polymorphism was found by performing a PCR-RFLP (PCR…

Mycoplasma pneumoniae pericarditis demonstrated by polymerase chain reaction and electron microscopy.

We report a case of pericarditis occurring in a 17-year-old girl who received a bone marrow transplant. The presence of Mycoplasma pneumoniae in the pericardial fluid was demonstrated by polymerase chain reaction, and Mycoplasma-like organisms were detected by electron microscopy. Because no other infectious agent could be found in the pericardial fluid, M. pneumoniae was presumed to be the etiologic agent of the pericarditis.

Nested polymerase chain reaction in the diagnosis of congenital cytomegalovirus infection.

OBJECTIVE: Human cytomegalovirus infection is highly prevalent in Indian population. It is the commonest congenitally acquired infection causing various anomalies. The diagnosis of infection in neonates is difficult as IgM may not be detected in all cases. The polymerase chain reaction is reported as alternative and better option in these patients. However, there is lack of data to substantiate this preference in a resource poor country like India. METHODS: Blood samples from 930 neonates/fetuses were first tested for specific anti-CMV IgM antibodies using mu-capture enzyme linked immunosorbe…

Quantification of bacteria in oral samples by competitive polymerase chain reaction.

Information about the total amount of bacteria in oral samples contributes to assessment of an individual's risk of contracting dental caries or developing periodontitis and the prediction of that individual's clinical course. Since existing techniques are often time-consuming and expensive, it seemed attractive to look for alternative methods for the quantification of eubacteria. With their high specificity and sensitivity, polymerase chain-reaction (PCR) techniques have the potential of supplying fast and reliable results. We developed a method of competitive PCR for the quantification of eu

Combined polymerase chain reaction approach for clonality detection in lymphoid neoplasms.

The present study analyzes the efficiency of a combination of four immunoglobulin heavy chain (IgH) gene polymerase chain reaction (PCR) primer systems and a multiplex T-cell receptor gamma chain (TRG) gene PCR for detection of clonality in 409 samples (234 paraffin sections, 175 bone marrow aspirates) of different lymphomas. Using the four IgH PCR systems together, clonality was detected in all samples of B-cell chronic lymphocytic leukemias, hairy cell leukemias, common acute lymphoblastic leukemias, and Burkitt-like B-cell lymphomas. Clonality was detected in all bone marrow aspirates with …

Improved detection of Dirofilaria repens DNA by direct polymerase chain reaction.

Diagnosis of human infection by Dirofilaria repens, depends mainly on microscopic evaluation of tissue cross-sections and the macroscopic characteristics of the worm. Tissue degeneration and/or poor specimen preparation practices however, often render many cases of subcutaneous dirofilariasis elusive to such morphological diagnostic approaches. The early PCR protocols, developed to satisfy these complex diagnostic needs, failed to amplify dirofilariae DNA from formalin preserved material. To overcome these difficulties, we developed an improved PCR protocol using a set of primers designed to a

Detection of Japanese encephalitis virus by reverse transcription/polymerase chain reaction.

A simple and rapid reverse transcription/polymerase chain reaction (RT-PCR) assay for detection of Japanese encephalitis virus (JEV) envelope (E) gene sequences in various biological samples is described. The assay successfully amplified JEV E gene sequences from infected cell cultures, Aedes aegypti larvae, mosquitoes and mouse blood. The sensitivity of the assay was currently 1 ng of JEV RNA and could be increased up to 1 pg on the background of 1 microgram of cellular RNA by biotinylation of the PCR product, Southern blot analysis and streptavidin/alkaline phosphatase detection.

Detection and identification of enteroviruses RNA by using polymerase chain reaction.

For rapid diagnosis of enteroviral infection in clinic practice, we developed a reverse transcription and polymerase chain reaction (RT-PCR) assay. Primers homologous to the conserved 5' non-coding region were designed by analyzing enteroviral genomes, and then they were used to enzymatically amplify RNA from 31 prototype enteroviral strains and enteroviruses (EV) in cerebrospinal fluid (CSF) of 34 cases of aseptic meningitis and 11 cases of aseptic encephalitis. The RT-PCR products generated with these enteroviral primers were analyzed by agar gel electrophoresis and dot blot hybridization an…
