Ruehssiosides A-R, 18 Previously Undescribed Pregnane Glycosides from Ruehssia caatingae: Structural Elucidation, Potential Anti-Inflammatory Activity, and Insights into Structure-Activity Relationships.

Nature has always inspired the discovery of novel compounds to treat human inflammatory processes, which are related to many diseases in acute and chronic conditions. Found in some Apocynaceae species, pregnane glycosides (PGs) have been attracting attention due to their promising anti-inflammatory potential. Ruehssia is a neotropical genus endemic to arid regions and, in Brazil, the Caatinga serves as a major hotspot for its occurrence. However, no in-depth chemical or pharmacological investigations have yet been conducted on Ruehssia species. Based on this, the present work sought to isolate

Effects of polymorphisms in pregnane X receptor and ABC transporters on afatinib in Japanese patients with non-small cell lung cancer: pharmacogenomic-pharmacokinetic and exposure-response analysis.

PURPOSE: Because of the large interindividual variability of afatinib pharmacokinetics and adverse events, we evaluated the effects of polymorphisms in pregnane X receptor (NR1I2) and ABC transporters (ABCB1, ABCG2, and ABCC2) on the pharmacokinetics of afatinib. METHODS: The steady-state area under the concentration-time curve (AUC)0-24 of afatinib was analyzed using blood sampling just prior to and at 1, 2, 4, 6, 8, 12, and 24 h on day 15 after administration. RESULTS: The median oral clearance (CL/F) of afatinib in patients with the NR1I2 7635A allele was significantly lower than those in

Induction of CYP3A4 and MDR1 gene expression by baicalin, baicalein, chlorogenic acid, and ginsenoside Rf through constitutive androstane receptor- and pregnane X receptor-mediated pathways.

The herbal products baicalin, baicalein, chlorogenic acid, and ginsenoside Rf have multiple pharmacological effects and are extensively used in alternative and/or complementary therapies. The present study investigated whether baicalin, baicalein, chlorogenic acid, and ginsenoside Rf induced the expression of the cytochrome P450 3A4 (CYP3A4) and multi-drug resistance 1 (MDR1) genes through the pregnane X receptor and constitutive androstane receptor pathways. Real time PCR, western blotting, and a luminescent assay were used to assess the induction of gene expression and activity of CYP3A4 and

Comparison of the effects of sodium phenobarbital in wild type and humanized constitutive androstane receptor (CAR)/pregnane X receptor (PXR) mice and in cultured mouse, rat and human hepatocytes.

Phenobarbital (PB), a constitutive androstane receptor (CAR) activator, produces liver tumours in rodents by a mitogenic mode of action involving CAR activation. In this study, the hepatic effects of sodium phenobarbital (NaPB) were compared in male C57BL/6J wild type (WT) mice and in humanized mice, where both the mouse CAR and pregnane X receptor (PXR) have been replaced by their human counterparts (hCAR/hPXR mice). Investigations were also performed in cultured male C57BL/6J and CD-1 mouse, male Sprague-Dawley rat and male and female human hepatocytes. The treatment of WT and hCAR/hPXR mice

Pregnane X receptor deletion induces a lean phenotype in chow-fed mice through a liver-ileum-muscle axis involving 6α-hydroxylated bile acids and Fgf15.

OBJECTIVES: The pregnane X receptor (Pxr; Nr1i2) plays a role in metabolic regulation. However, its contribution to systemic energy homeostasis under physiological conditions remains unclear. Here, we investigated the metabolic consequences of Nr1i2 deficiency in chow-fed mice. METHODS: Phenotyping was performed in adult whole-body Nr1i2 knockout (Nr1i2-/-) chow-fed mice and their wild-type littermates. Body composition, metabolic parameters, skeletal muscle mass and myofiber size were assessed. Ileal gene expression, circulating Fgf15 levels, and bile acid composition were analyzed. Intestin

Conicasterol E, a small heterodimer partner sparing farnesoid X receptor modulator endowed with a pregnane X receptor agonistic activity, from the marine sponge Theonella swinhoei.

We report the isolation and pharmacological characterization of conicasterol E isolated from the marine sponge Theonella swinhoei. Pharmacological characterization of this steroid in comparison to CDCA, a natural FXR ligand, and 6-ECDCA, a synthetic FXR agonist generated by an improved synthetic strategy, and rifaximin, a potent PXR agonist, demonstrated that conicasterol E is an FXR modulator endowed with PXR agonistic activity. Conicasterol E induces the expression of genes involved in bile acids detoxification without effect on the expression of small heterodimer partner (SHP), thus sparing

Schisandrin B inhibits hepatic inflammation and steatosis via activation of pregnane X receptor signaling in methionine and choline-deficient high fat diet-induced steatohepatitis mouse model.

Schisandrin B (Sch B), a key bioactive lignan extracted from Schisandra chinensis (Turcz.) Baill, exhibits lipid lowering and anti-inflammatory properties in vivo and in vitro. However, few studies investigate the anti-metabolic dysfunction associated steatohepatitis (MASH) effect of Sch B. In this study, we employed a methionine and choline-deficient high fat diet (MCDHF)-induced MASH mouse model and palmitate acid (PA)-induced MASH cell models to evaluate the anti-inflammatory and anti-steatosis effects of Sch B. We also explore the molecular targets of Sch B for its anti-MASH effect. The re

Plasma membrane receptors for the cancer-regulating progesterone metabolites, 5alpha-pregnane-3,20-dione and 3alpha-hydroxy-4-pregnen-20-one in MCF-7 breast cancer cells.

Recent observations indicate that the progesterone metabolite, 5alpha-pregnane-3,20-dione (5alphaP), which is produced at higher levels in tumorous breast tissue, promotes cell proliferation and detachment, whereas 3alpha-hydroxy-4-pregnen-20-one (3alphaHP), which is produced at higher levels in nontumorous breast tissue, suppresses proliferation and detachment of MCF-7 breast cancer cells. The objective of the current study was to determine the presence and characteristics of binding sites for these endogenous putative cancer-regulating steroid hormones. Radiolabeled 5alphaP and 3alphaHP were

Comparison of the hepatic and thyroid gland effects of sodium phenobarbital and pregnenolone-16α-carbonitrile in wild-type and constitutive androstane receptor (CAR)/pregnane X receptor (PXR) knockout rats.

The hepatic and thyroid gland effects of the constitutive androstane receptor (CAR) activator sodium phenobarbital (NaPB) and the pregnane X receptor (PXR) activator pregnenolone-16α-carbonitrile (PCN) were examined in male Sprague-Dawley wild-type (WT) and knockout (KO) rats lacking both hepatic CAR and PXR receptors (CAR KO/PXR KO rats). The treatment of WT rats for 7 d with 500 ppm NaPB in the diet and 100 mg/kg/d PCN by gavage resulted in increased relative liver weight, hepatocyte hypertrophy, increased hepatocyte replicative DNA synthesis (RDS) and induction of cytochrome P450 CYP2B and

Chronic Intermittent Ethanol Exposure Alters Stress Effects on (3α,5α)-3-hydroxy-pregnan-20-one (3α,5α-THP) Immunolabeling of Amygdala Neurons in C57BL/6J Mice.

The GABAergic neuroactive steroid (3α,5α)-3-hydroxy-pregnan-20-one (3α,5α-THP, allopregnanolone) is decreased in various brain regions of C57BL/6J mice following exposure to an acute stressor or chronic intermittent ethanol (CIE) exposure and withdrawal. It is well established that there are complex interactions between stress and ethanol drinking, with mixed literature regarding the effects of stress on ethanol intake. However, there is little research examining how chronic ethanol exposure alters stress responses. The present work examined the impact of CIE exposure and withdrawal on changes

Pregnane X receptor, constitutive androstane receptor, or peroxisome proliferator-activated receptor α activation does not affect yes-associated protein phase separation under physiological and hyperosmotic conditions.

Yes-associated protein (YAP) is a critical factor of the Hippo pathway, which plays a key role in regulating organ size. Pregnane X receptor (PXR), constitutive androstane receptor (CAR), and peroxisome proliferator-activated receptor α (PPARα) are key members of the nuclear receptor superfamily, known for mediating diverse physiological and biological processes. Previous studies have demonstrated that activation of PXR, CAR, or PPARα promotes hepatomegaly and liver regeneration via YAP signaling. YAP has been reported to undergo phase separation under hyperosmotic stress, leading to enhanced

Xenobiotic-induced hepatocyte proliferation associated with constitutive active/androstane receptor (CAR) or peroxisome proliferator-activated receptor α (PPARα) is enhanced by pregnane X receptor (PXR) activation in mice.

Xenobiotic-responsive nuclear receptors pregnane X receptor (PXR), constitutive active/androstane receptor (CAR) and peroxisome proliferator-activated receptor α (PPARα) play pivotal roles in the metabolic functions of the liver such as xenobiotics detoxification and energy metabolism. While CAR or PPARα activation induces hepatocyte proliferation and hepatocarcinogenesis in rodent models, it remains unclear whether PXR activation also shows such effects. In the present study, we have investigated the role of PXR in the xenobiotic-induced hepatocyte proliferation with or without CAR activation

Purification and characterization of a novel dimeric 20 alpha-hydroxysteroid dehydrogenase from Tetrahymena pyriformis.

Tetrahymena pyriformis was found to exhibit high NADPH-dependent 20-oxosteroid reductase activity that converted 17 alpha-hydroxyprogesterone into 17 alpha,20 alpha-dihydroxypregn-4-en-3-one. The enzyme was purified 400-fold from the cytosolic fraction. The purified enzyme with a specific activity of 6.4 mumol/min per mg of protein had an isoelectric point of 4.9 and M(r) of 68,000, and was composed of two subunits of equal size. The N-terminal sequence was determined to be LAKTVPLNDGTNFPIFGG. The enzyme reduced pregnanes and pregnanes possessing a 17 alpha-hydroxy group to a greater extent th

Sustained Drug-Drug Interaction Between Cyclosporine and Apalutamide in a Patient With Metastatic Hormone-Sensitive Prostate Cancer: A Case Report and Evaluation of CYP3A4 Induction via Pregnane X Receptor Activation by Apalutamide.

BACKGROUND: Apalutamide (Apa) is a key therapeutic agent for prostate cancer. Despite its efficacy, Apa is known to induce several drug-metabolizing enzymes including cytochrome P450 3A4 (CYP3A4), raising concerns about drug-drug interactions (DDIs). This study reports a rare case of a sustained DDI between Apa and cyclosporine (CsA)-a CYP3A4 substrate-in a patient with metastatic hormone-sensitive prostate cancer (mHSPC) and primary pure red cell aplasia (PRCA). We further investigated whether Apa could activate pregnane X receptor (PXR), a key nuclear receptor that regulates CYP3A4 expressio

Activation of pregnane X receptor-organic anion transporting polypeptide 1a/b /P-glycoprotein/multidrug resistance protein 2 axis mediates the accelerated blood and liver clearance of PEGylated liposomes.

Previous studies revealed that pregnane X receptor (PXR) was involved in the "accelerated blood clearance (ABC)" phenomenon induced by repeated injections of PEGylated liposomes, and observed the quickly reduced hepatic accumulation of the second dose in rats at 12 hours after the last injection, compared with the increased hepatic accumulation characteristic of the ABC phenomenon. Among the downstream target genes of PXR, organic anion transporting polypeptide 1a/b (Oatp1a/b), P-glycoprotein (P-gp), and multidrug resistance protein 2 (Mrp2) are responsible for regulating the absorption, distr

Commiphora myrrha resin extract-modulated cytochrome P-450 2C9 enzyme expression in cultured Hep G2 cells is associated with resin extract-derived metabolites binding to Pregnane X receptor.

BACKGROUND: The use of natural health products (NHP) is constantly increasing worldwide. Among the most NHP consumed in the Arabian Peninsula are the resins of Commiphora myrrha (C. myrrha) tree. Consumption of C. myrrha may result in herb-drug interactions mediated by drug-metabolizing cytochrome P-450 (CYP) enzymes in the liver, which could lead to serious health consequences for patients. This study aims to determine the herb-drug interactions based on the induction of predominant CYP 2C9 by a non-toxic boiled aqueous extract of C. myrrha and to unveil the potential involvement of xenobioti

Withdrawal from 3alpha-OH-5alpha-pregnan-20-One using a pseudopregnancy model alters the kinetics of hippocampal GABAA-gated current and increases the GABAA receptor alpha4 subunit in association with increased anxiety.

In the present study, we have characterized properties of steroid withdrawal using a pseudopregnant rat model. This paradigm results in increased production of endogenous progesterone from ovarian sources and as such is a useful physiological model. "Withdrawal" from progesterone induced by ovariectomy on day 12 of pseudopregnancy resulted in increased anxiety, as determined by a decrease in open arm entries on the elevated plus maze compared to control rats and pseudopregnant animals not undergoing withdrawal. Similar findings were obtained 24 hr after administration of a 5alpha-reductase blo

Steroid modulation of the GABAA receptor complex: electrophysiological studies.

The effect of some endogenous and synthetic steroids on the operation of inhibitory and excitatory amino acid neurotransmitter receptors was examined. Anaesthetic pregnane steroids (e.g. alphaxalone, 5 alpha-pregnan-3 alpha-ol-20-one, 5 alpha-pregnane-3 alpha,21-diol-20-one) potentiated GABAA receptor-mediated whole-cell currents recorded from bovine chromaffin cells. The threshold concentration for enhancement was 10-30 nM. Potentiation was stereoselective and was mediated by a steroid-induced prolongation of the burst duration of the GABA-activated channel. Additionally, the pregnane steroid
