The cochlear macrophage in presbycusis: inflammaging, neuroinflammation, and targets for therapeutic modulation.

Age-related neurodegenerative diseases and age-related hearing loss are increasing in prevalence globally. Immune cells are novel mediators linking these diseases through shared mechanisms of chronic inflammation and maladaptive repair. In the cochlea, resident macrophages and infiltrating immune cells respond to injury, noise exposure, and aging by adopting activated phenotypes that disrupt synaptic integrity, impair myelination, and promote hair cell and neuronal loss. Similarly, in the central nervous system, microglia, the resident immune cells of the brain undergo age- and disease-associa

Beyond the Audiogram: Psychosocial Barriers to Hearing Aid Use in Presbycusis.

This cross-sectional study investigated audiological and psychosocial factors associated with hearing aid use preferences among 57 individuals with presbycusis who accepted (n = 37) or refused (n = 20) hearing aids. The refusal group was older and had greater hearing loss. Social loneliness was significantly higher among individuals refusing hearing aids, whereas depression and emotional loneliness did not differ between groups. Exploratory logistic regression showed that older age, male gender, and higher social loneliness were associated with hearing aid refusal. These findings suggest that …

Degree centrality and functional connections in presbycusis with and without cognitive impairments.

Presbycusis is a major public issue that affecting elderly adults. However, the neural substrates between normal cognition and cognitive deficits in these patients need to be illustrated. 47 patients with presbycusis and 33 well-matched healthy controls were recruited in present study. Each subject underwent pure-tone audiometry (PTA), MRI scanning and cognition evaluations, then we found 22 patients with cognitive deficits and 25 patients with common cognition. We analyzed the Degree centrality (DC) characteristics among three groups, and try to recognize key nodes which contribute significan…

Vulnerability to chronic stress and the phenotypic heterogeneity of presbycusis with subjective tinnitus.

Age-related functional reserve decline and vulnerability of multiple physiological systems and organs, as well as at the cellular and molecular levels, result in different frailty phenotypes, such as physical, cognitive, and psychosocial frailty, and multiple comorbidities, including age-related hearing loss (ARHL) and/or tinnitus due to the decline in auditory reserve. However, the contributions of chronic non-audiogenic cumulative exposure, and chronic audiogenic stress to phenotypic heterogeneity of presbycusis and/or tinnitus remain elusive. Because of the cumulative environmental stressor…

Association of genetic variations in the mitochondrial DNA control region with presbycusis.

BACKGROUND: The prominent role of mitochondria in the generation of reactive oxygen species, cell death, and energy production contributes to the importance of this organelle in the intracellular mechanism underlying the progression of the common sensory disorder of the elderly, presbycusis. Reduced mitochondrial DNA (mtDNA) gene expression and coding region variation have frequently been reported as being associated with the development of presbycusis. The mtDNA control region regulates gene expression and replication of the genome of this organelle. To comprehensively understand of the role …

Investigation of auditory brainstem function in elderly diabetic patients with presbycusis.

We performed brainstem auditory evoked potential (BAEP) examinations in 100 patients older than 60 years and having type I diabetes mellitus and presbycusis. The aim of our investigation was to compare the BAEP results of this group with those of healthy controls with presbycusis and to look for possible correlations between alteration of the auditory brainstem function and the aging of elderly diabetic patients. Absolute and interpeak latencies of all waves were prolonged significantly in the study group of diabetic patients. The amplitudes of all waves I through V were diminished in the stud…

Outer- and middle-ear contributions to presbycusis in the Brown Norway rat.

This paper examines the contribution of the outer and middle ears to the hearing loss associated with presbycusis in Brown Norway rats. Animals were formed into two groups; young adults (2-3 months old) and aged animals (approximately 34 months old). Auditory brainstem response (ABR) thresholds were obtained with the outer ear intact or surgically removed. Tympanic membrane (TM) velocity transfer functions were measured from the umbo with the outer ear removed. The length of the auditory meatus, TM surface area, and TM thickness were quantified. The ABR thresholds were 17-26 dB less sensitive …

Speech recognition in noise and presbycusis: relations to possible neural mechanisms.

This study is part of ongoing efforts to characterize and determine the neural bases of presbycusis. These efforts utilize humans and animals in sets of overlapping hypotheses and experiments. Here, 50 young adult and elderly subjects, with normal audiometric thresholds or high-frequency hearing loss, were presented three types of linguistic materials at suprathreshold levels to determine speech recognition performance in noise. The study sought to determine how peripheral and central auditory system dysfunctions might be implicated in the speech recognition problems of elderly humans. There w…

Results of visual information processing tests in elderly people with presbycusis.

In a group of 21 elderly subjects with mild to moderate hearing loss of presbycusis type, a series of tests of visual information processing was performed. Cortical-evoked potentials were recorded in response to flash and to checker-board pattern stimulation. Further, five speech reading tests were administered. The results obtained in this group of elderly subjects were compared to those of a group of young normal-hearing subjects and those of a group of young subjects with congenital hearing impairment. The visual-evoked potentials showed significantly smaller N1P2-amplitude in elderly subje…

[Time resolution ability of the ear in presbycusis and sensorineural hearing disorders].

In psychoacoustical and electrophysiological experiments the time resolution ability in monaural and binaural hearing was investigated for defined signal parameters. The binaural stimulation is characterized by signals of different structure (dichotic stimulation). With regard to different functional changes in the peripheral and central areas of the hearing organ caused by presbycusis, these investigations confirm that there are similar changes in time resolution processes which may influence speech discrimination in presbycusis to a high degree. In patients with sensorineural hearing loss ti…

[Implication of mitochondrial apoptosis in neural degeneration of cochlea in a murine model for presbycusis].

HYPOTHESIS: Pathologies of senescence, in particular those of neurosensory organs represent an important health problem. The improvement of the life expectation entails the fast increase of the frequency of the presbyacusis in the population. The biological and molecular causes of this degenerative pathology of the inner ear are linked to the disappearance of the sensory cells (inner and outer hair cells) and are associated to nervous damages of the spiral ganglion in the cochlea. We were interested in mechanisms causing the cochlear degeneration in a model of mouse CD 1 presenting prematurely

Distortion product otoacoustic emissions in the CBA/J mouse model of presbycusis.

CBA mice do not exhibit age-related loss of auditory sensitivity or cochlear pathology until relatively late in life. Therefore, this strain is believed to be an excellent animal model for the examination of the effects of age on the cochlea. To evaluate the effects of age on outer hair cell function, 2f1-f2 distortion product otoacoustic emissions (DPOAEs) were measured for f2 between 8 and 16 kHz in CBA/J mice between 1 and 25 months of age. CBA mice exhibited mild age-related changes in DPOAE level and detection threshold at 17 months of age, and changes of 20-40 dB by 25 months of age. The

Effects of presbycusis and other types of hearing loss on auditory brainstem responses.

Auditory brainstem responses were studied in 373 healthy individuals of various ages and with sensorineural hearing loss of differing degrees. 209 subjects were elderly and had a hearing loss which included a component of presbycusis. The remainder were young or middle-aged with cochlear hearing loss. Altogether 606 ears were tested. The old individuals had generally longer ABR wave latencies than the young subjects. The I-V interpeak latency (IPL) was also prolonged in the older age groups compared with the group of younger individuals, except for subjects with pronounced hearing loss. The re…
