A novel angiotensin I-converting enzyme inhibitory peptide derived from the trypsin hydrolysates of salmon bone proteins.

When fish are processed, fish bone becomes a key component of the waste, but to date very few researchers have sought to use fish bone to prepare protein hydrolysates as a means of adding value to the final product. This study, therefore, examines the potential of salmon bone, through an analysis of the benefits of its constituent components, namely fat, moisture, protein, and ash. In particular, the study seeks to optimize the process of enzymatic hydrolysis of salmon bone with trypsin in order to produce angiotensin-I converting enzyme (ACE) inhibitory peptides making use of response surface…

In Vitro and in Vivo Studies on the Angiotensin-Converting Enzyme Inhibitory Activity Peptides Isolated from Broccoli Protein Hydrolysate.

In the present study we purified and identified peptides from broccoli protein hydrolysates and evaluated their angiotensin-converting enzyme (ACE) inhibitory activity in vitro and hypotensive effect in vivo. Three ACE inhibitory peptides were isolated and identified as IPPAYTK, LVLPGELAK, and TFQGPPHGIQVER, and their inhibitory IC50 values were 23.5, 184.0, and 3.4 μM, respectively. We then investigated the effect of gastrointestinal digestion on ACE inhibitory activity. We detected almost two times the ACE inhibitory activity of the peptide LVLPGELAK following simulated digestion than prior …

Plasma concentrations and ACE-inhibitory effects of tryptophan-containing peptides from whey protein hydrolysate in healthy volunteers.

PURPOSE: The tryptophan-containing dipeptides isoleucine-tryptophan (IW) and tryptophan-leucine (WL) are angiotensin-converting enzyme (ACE)-inhibitors in vitro. These peptides are released by enzymatic hydrolysis of bovine whey protein. To exhibit ACE inhibition in vivo, peptides need to be absorbed into the circulatory system. This study aimed to determine the in vivo ACE-inhibitory potency of a whey protein hydrolysate (MPH), containing IW and WL, and to quantify plasma concentrations of these peptides after oral administration of MPH in healthy volunteers. Additionally, changes in blood pr

Short communication: Casein hydrolysate and whey proteins as excipients for cyanocobalamin to increase intestinal absorption in the lactating dairy cow.

Bioavailability of vitamin B12 is low in humans and animals. Improving vitamin B12 absorption is important for optimal performance in dairy cows and for increasing vitamin B12 concentrations in milk for human consumption. However, when supplemented in the diet, 80% of synthetic vitamin B12, cyanocobalamin (CN-CBL), is degraded in the rumen of dairy cows and only 25% of the amount escaping destruction in the rumen disappears from the small intestine between the duodenal and ileal cannulas. In pigs, vitamin B12 from milk is more efficiently absorbed than synthetic CN-CBL. The objective of this s

Influence of dietary inclusion of black soldier fly larvae protein hydrolysates on nutritional indicators and cardiac-related biomarkers of adult Beagle dogs.

Protein hydrolysates derived from insects have been explored in recent years for their bioactive functionalities, namely antioxidant and antihypertensive activities, although the investigation of in vivo effects remains scarce. The present study aimed to assess the effects of dietary inclusion of black soldier fly (Hermetia illucens) larvae (BSFL) hydrolysates on nutritional status and cardiac biomarkers of adult Beagle dogs. Three complete diets were used: a control diet containing 3% shrimp hydrolysate (D_Shrimp), and two experimental diets in which the shrimp hydrolysate was replaced by BSF…

Photobacterium damselae subsp. piscicida challenge reveals dietary animal protein hydrolysates modulate early immune, oxidative and barrier responses in European seabass (Dicentrarchus labrax).

Reducing dependence on marine-derived ingredients like fishmeal, while maintaining the robustness of carnivorous species, like European seabass (Dicentrarchus labrax), remains a challenge for sustainable aquaculture. Photobacterium damselae subsp. piscicida (Phdp) outbreaks are still responsible for significant economic losses, but functional ingredients capable of modulating immune responses may enhance resilience to a Phdp challenge. In this study, fish were fed four balanced diets for 89 days: a plant-based control (CTRL) and three diets including three protein hydrolysates (SHARK, FISH and…

Structures, antioxidant, and angiotensin I-converting enzyme (ACE)-inhibitory activities of peptides derived from protein hydrolysates of three phenolics-rich legume genera.

Lentil, black soybean, and black turtle bean are rich in phenolic antioxidants but belong to different scientific genera with different protein structures. This study's objective was to compare the characteristics of peptides derived from the protein hydrolysates of these legumes. Proteins were isolated, cooked, and subjected to in vitro digestion with pepsin, trypsin, and chymotrypsin. Hydrolyzed peptides were fractionated by ultrafiltration (UF), anion-exchange chromatography, and gel-permeation chromatography (GPC). GPC-eluted peptides of <3 kDa with high angiotensin I-converting enzyme (AC…

Protective effects of oyster protein hydrolysates on alcohol-induced liver disease (ALD) in mice: based on the mechanism of anti-oxidative metabolism.

Many bioactivities of hydrolysates from oyster (Crassostrea gigas) muscle have been reported, while there is no knowledge about their protective effects on alcohol-induced liver disease (ALD). In the present study, the anti-oxidative activities in vitro and molecular weight distribution of oyster protein hydrolysates (OPH) were detected and the OPH released by alcalase (AOPH) was used to treat C57BL/6 mice. C57BL/6 mice were treated with a Lindros control diet to establish an ethanol-exposed model. The content of small-weight components (<2.0 kDa) of OPH reached 90.85%. AOPH showed more potent…

Olive leaf protein hydrolysates yield gastro-resistant peptides with antioxidant and anti-inflammatory potential: peptidomics, in vitro validation and molecular docking analyses.

Olive (Olea europaea L.) leaves are an abundant olive-oil by-product and a promising feedstock for sustainable valorisation. An olive leaf protein isolate (OLPI) from olive-leaf powder (OLP) was enzymatically hydrolysed to yield seven hydrolysates (OLPHs). All showed notable antioxidant activity as whole hydrolysate matrices (EC₅₀ = 0.11-0.28 mg mL-1); likely reflecting the combined contribution of released peptides and co-extracted phenolic compounds; the 15-min Alcalase product (OLPH15A) showed high activity with the shortest processing time. Its INFOGEST digest (dOLPH15A) attenuated LPS-ind

The cytoprotective role of whey protein hydrolysates against CdCl2-induced toxicity in fibroblasts by the Electric Cell-substrate Impedance Sensing (ECIS) method.

INTRODUCTION AND OBJECTIVE: Cadmium (Cd) is a toxic mineral element with a long half-life that causes excessive accumulation in tissues, especially the kidney. It influences cell development, proliferation, and death by activating intracellular signalling pathways. Currently, treatment options include chelation agents and antioxidant therapy. Researchers are investigating natural compounds that might protect against Cd-induced toxicity while providing less negative effects. The aim of the study is to look into the protective properties of bovine milk whey protein hydrolysates against Cd-induce…

Effect of Protease Type and Peptide Size on the In Vitro Antioxidant, Antihypertensive and Anti-Diabetic Activities of Eggplant Leaf Protein Hydrolysates.

Solanum macrocarpon (eggplant) leaf protein isolate (ELI) was hydrolyzed using four different enzymes to produce hydrolysates from alcalase (AH), chymotrypsin (CH) pepsin (PH) and trypsin (TH). CH had an overall stronger antioxidant property and was separated using ultrafiltration membranes into <1, 1-3 and 3-5 kDa peptide fractions. Gel-permeation chromatography confirmed conversion of the ELI (average of 22 kDa) into protein hydrolysates that contained smaller peptides (<6 kDa). A total of 23 peptides consisting of tri and tetrapeptides were identified from the CH, which is a wider spectrum …

Transcriptome analysis reveals the promoting mechanism of corn steep liquor protein hydrolysate on cordycepin biosynthesis of Cordyceps militaris.

Cordycepin is an important nucleoside analogue and the principal active compound found in Cordyceps militaris. It exhibits anticancer, antiviral, immunomodulatory, and various other biological activities. However, the low cordycepin yields and high manufacturing costs limit its large-scale application. To develop a low-cost nutrient regulator to improve cordycepin production, we investigated the effect of corn steep liquor protein hydrolysate (CSLPH) on cordycepin biosynthesis during submerged fermentation of C. militaris, and the response mechanism of C. militaris to CSLPH utilization was rev

Effect of 2'-fucosyllactose and hydrolysis degree on oxidation, Maillard reaction, and physical stability of whey protein hydrolysate emulsions.

Processing of liquid infant formulas inevitably triggers undesirable side reactions affecting product quality and nutritional value. This study investigated the effects of whey protein hydrolysis (WPH) at different degrees of hydrolysis (DH; 0%, 6%, 8%, and 10%) and 2'-fucosyllactose (2'-FL) on the physical stability, protein oxidation, lipid oxidation, and Maillard reaction in infant formula model emulsions. Increasing DH progressively enhanced DPPH and ABTS radical scavenging activities, resulting in lower peroxide values and thiobarbituric acid reactive substances, thereby improving lipid o

Threadfin bream surimi gel containing squid fin protein hydrolysate: Textural properties, acceptability, and volatile profile.

Gel properties of threadfin bream surimi gels with squid fin protein hydrolysate (SFPH) at various concentrations (0, 1, 2, 3, and 4%; w/w) were determined. The gel without SFPH (CON) had the highest breaking force (BrF) and deformation (DeF) as compared to those with SFPH (p < 0.05). Among the gel with SFPH, gel containing 1 and 2% SFPH (SFPH-1 and SFPH-2, respectively) had the highest BrF, whereas the lowest value was obtained when 4% SFPH was used (SFPH-4) (p < 0.05). The whiteness of all samples was slightly decreased with an upsurging amount of SFPH. However, water holding capacity was in
