ProTx-I and ProTx-II: gating modifiers of voltage-gated sodium channels.

The tarantula venom peptides ProTx-I and ProTx-II inhibit voltage-gated sodium channels by shifting their voltage dependence of activation to a more positive potential, thus acting by a mechanism similar to that of potassium channel gating modifiers such as hanatoxin and VSTX1. ProTx-I and ProTx-II inhibit all sodium channel (Nav1) subtypes tested with similar potency and represent the first potent peptidyl inhibitors of TTX-resistant sodium channels. Like gating modifiers of potassium channels, ProTx-I and ProTx-II conform to the inhibitory cystine knot motif, and ProTx-II was demonstrated to…

The epileptic seizures induced by BmK I, a modulator of sodium channels.

In the present study, the susceptibility to rat epileptic seizures induced by the intrahippocampal administration of BmK I, a modulator of sodium channels purified from the venom of Chinese scorpion, has been investigated. The results showed that the strong epileptic behaviors and discharges in the hippocampus were evoked by BmK I dose-dependently. The hippocampal c-Fos expression displayed two peak waves in a specific spatio-temporal pattern elicited by BmK I. The whole cell patch clamp recordings showed that the inactivation of sodium currents in rat cultured hippocampal neurons was prolonge…

Voltage-gated sodium channels as primary targets of diverse lipid-soluble neurotoxins.

Voltage-gated Na(+) channels are heteromeric membrane glycoproteins responsible for the generation of action potentials. A number of diverse lipid-soluble neurotoxins, such as batrachotoxin, veratridine, aconitine, grayanotoxins, pyrethroid insecticides, brevetoxins and ciguatoxin, target voltage-gated Na(+) channels for their primary actions. These toxins promote Na(+) channel opening, induce depolarization of the resting membrane potential, and thus drastically affect the excitability of nerve, muscle and cardiac tissues. Poisoning by these lipid-soluble neurotoxins causes hyperexcitability

Tracking sodium channels in live cells: confocal imaging using fluorescently labeled toxins.

One particularly important class of ion channels in excitable cells are the voltage-dependent sodium channels (VDSC). Knowledge of the distribution of VDSC in living cells is important for studies of neuronal excitability, development, and plasticity. Here, we demonstrate a new method for visualizing the spatial distribution of VDSC in living cells. To illustrate the capabilities of the technique, the distribution of VDSC in GH3 cells was revealed with fluorescent derivatives of the alpha-type and beta-type scorpion toxins in conjunction with laser scanning confocal microscopy. Cells exhibited…

Investigation of the modulation of glutamate release by sodium channels using neurotoxins.

The modulation of neurotransmitter release by calcium channels is well established, yet, sodium channels were regarded mainly as charge carriers. Many lines of evidence suggest a more fine-tuning role played by sodium channels. Using rat cerebrocortical isolated nerve endings (synaptosomes) and two toxins that have separate sites of action on sodium channels and provoke distinct changes in channel kinetics, we were able to show that depending on the rate of increase in channel conductance, the outcome in terms of neurotransmitter release and calcium channel types coupled to that event are diff…

Voltage-dependent blockade of normal and mutant muscle sodium channels by benzylalcohol.

1. We studied the effects of benzylalcohol on heterologously expressed wild type (WT), paramyotonia congenita (R1448H) and hyperkalaemic periodic paralysis (M1360V) mutant alpha-subunits of human skeletal muscle sodium channels. 2. Benzylalcohol blocked rested channels at -150 mV membrane potential, with an ECR(50) of 5.3 mM in wild type, 5.1 mM in R1448H, and 6.2 mM in M1360V. When blockade was assessed at -100 mV, the ECR(50) was reduced in R1448H (2 mM) compared with both wild type (4.3 mM; P<0.01) and M1360V (4.3 mM). 3. Membrane depolarization before the test depolarization significantly …

Lidocaine induces a slow inactivated state in rat skeletal muscle sodium channels.

1. Local anaesthetics such as lidocaine (lignocaine) interact with sodium channels in a manner that is exquisitely sensitive to the voltage-dependent conformational state of the ion channel. When depolarized in the presence of lidocaine, sodium channels assume a long-lived quiescent state. Although studies over the last decade have localized the lidocaine receptor to the inner aspect of the aqueous pore, the mechanistic basis of depolarization-induced 'use-dependent' lidocaine block remains uncertain. 2. Recent studies have shown that lowering the extracellular Na+ concentration ([Na+]o) and m…

Altered properties of neuronal sodium channels associated with genetic resistance to pyrethroids.

Genetic resistance to pyrethroid insecticides involves nervous system insensitivity linked to regulatory and structural genes of voltage-sensitive sodium channels. We examined the properties and relative density of sodium channels in central neurons of susceptible and pyrethroid-resistant (Pyr-R) insects that were homozygous for the amino acid substitution V421M in the I-S6 transmembrane segment. Pyr-R sodium channels show approximately 21-fold lower sensitivity to the synthetic pyrethroid permethrin and a approximately 2-fold increased sensitivity to the alpha-scorpion toxin LqhalphaIT. Pyr-R…

Interactions between anemone toxin II and veratridine on single neuronal sodium channels.

The nature of the known positive cooperativity between alkaloid and alpha-polypeptide toxins on macroscopic sodium currents was studied at the single-channel level. We have previously characterized the single-channel function of veratridine (VTD)-modified and anemone toxin II (ATX)-modified channels from lobster leg nerve. VTD and ATX are known to potentiate each other's effects in stimulating 22Na flux into vesicles containing sodium channels from lobster leg nerve. These channels, therefore, provided an excellent model for further investigation of the interactions between the toxins. A varie…

Gate-dependent blockade of sodium channels by phenothiazine derivatives: structure-activity relationships.

Voltage-clamp studies of myelinated nerve fibers that are designed to determine structural criteria regarding selective drug blocking of open and inactive states of the sodium channel are described. A series of phenothiazines were studied. It was shown that two of these drugs (ethmozine and ethacizine, at 5 microM) require open channels for blocking action and the other two (chlorpromazine and chloracizine, at 5 microM) can effectively block inactive channels. A size criterion, which looks at the spanning width at the aromatic end of these molecules, can explain this qualitative difference in

Ionizing radiation alters the properties of sodium channels in rat brain synaptosomes.

The effect of ionizing radiation on neuronal membrane function was assessed by measurement of neurotoxin-stimulated 22Na+ uptake by rat brai … robe. These results demonstrate an inhibitory effect of ionizing radiation on the voltage-sensitive sodium channels in rat brain synaptosomes. This effect of radiation is not dependent on changes in the order of membrane lipids.

Neurotoxin-sensitive sodium channels in neurons developing in vivo and in vitro.

Fetal mouse brain cells were investigated by 22Na+ flux assays with the aim to determine the ontogenetic time course of appearance of functional voltage-sensitive sodium channels. Their pharmacological properties were assessed by measurement of the response to known neurotoxins, acting at site 1, 2, or 3 of the Na+ channel. Brain cell suspensions, prepared at 11-19 d of prenatal development in vivo, and fetal brain neurons in culture were explored. In vivo neurotoxin-sensitive Na+ influx becomes detectable at 12 d of gestation, in concordance with the time of appearance of saturable binding si…

Efficacy of sodium channel blockers in treating myotonia is independent of action potential sodium current block.

The prevailing view holds that the therapeutic benefit of sodium channel blockers arises from use-dependent inhibition of the transient sodium current (NaT) that is responsible for action potentials. This assumption underlies decades of drug development but has never been directly tested. Our goal was to test this assumption to guide the development of more effective sodium channel blockers. We used a mouse model of myotonia congenita, an ion channelopathy of skeletal muscle, as a system to study mechanism and efficacy of both use-dependent and closed sodium channel blockers. Sodium channels are required during in vivo sodium chloride hyperosmolarity to stimulate increase in intestinal endothelial nitric oxide production.

NaCl hyperosmolarity increases intestinal blood flow during food absorption due in large part to increased NO production. We hypothesized that in vivo, sodium ions enter endothelial cells during NaCl hyperosmolarity as the first step to stimulate an increase in intestinal endothelial NO production. Perivascular NO concentration ([NO]) and blood flow were determined in the in vivo rat intestinal microvasculature at rest and under hyperosmotic conditions, 330 and 380 mosM, respectively, before and after application of bumetanide (Na(+)-K(+)-2Cl(-) cotransporter inhibitor) or amiloride (Na(+)/H(+

Scorpion toxins affecting sodium current inactivation bind to distinct homologous receptor sites on rat brain and insect sodium channels.

Sodium channels posses receptor sites for many neurotoxins, of which several groups were shown to inhibit sodium current inactivation. Receptor sites that bind alpha- and alpha-like scorpion toxins are of particular interest since neurotoxin binding at these extracellular regions can affect the inactivation process at intramembranal segments of the channel. We examined, for the first time, the interaction of different scorpion neurotoxins, all affecting sodium current inactivation and toxic to mammals, with alpha-scorpion toxin receptor sites on both mammalian and insect sodium channels. As sp…

Relative role of alkalosis and sodium ions in reversal of class I antiarrhythmic drug-induced sodium channel blockade by sodium bicarbonate.

BACKGROUND: Hypertonic sodium salts are used to treat sodium channel-blocking drug cardiotoxicity. The relative roles of alkalinization and increased sodium concentration ([Na+]o) for various drugs are incompletely known. METHODS AND RESULTS: The effects of four class I drugs on action potential characteristics of canine Purkinje fibers at equieffective concentrations (disopyramide 30 mumol/L, mexiletine 80 mumol/L, flecainide 7 mumol/L, imipramine 5 mumol/L) were studied in the presence of normal Tyrode solution and one altered solution (increased [Na+]o, increased bicarbonate concentration,

Decoding Voltage-Gated Sodium Channels Structure: Molecular Mechanisms Linking Architecture, Function, and Disease.

Voltage-gated sodium (NaV) channels are critical transmembrane proteins responsible for the initiation and propagation of electrical signals in excitable tissues. They consist of a large α subunit and auxiliary β subunits. Together, the voltage sensing, ion selectivity, and rapid gating transitions regulate neuronal, muscular, and cardiac excitability. Recent cryo-electron microscopy breakthroughs have transformed the understanding of NaV structure-function relationships, revealing the asymmetric organization of voltage-sensing domains (VSD), the architecture of the DEKA selectivity filter, th

Electrophysiological Modeling and Molecular Dynamics Simulations of Cardiac Sodium Channels in Brugada Syndrome.

Brugada syndrome (BrS) is a rare but potentially fatal genetic cardiac disorder, primarily caused by mutations in sodium and potassium ion channels, leading to ventricular arrhythmias and sudden cardiac death. Despite advances in computational modeling, the precise effects of ionic environment and channel kinetics on BrS-related action potentials remain incompletely understood. In this study, we developed a flexible electrophysiological model of cardiac sodium channels using a modifiable Richards activation function to simulate major BrS phenotypes and optimized the shape parameter (h) using p…

Effect of pain-related sodium channels SCN9A and SCN10A polymorphisms in migraine chronification.

OBJECTIVE: Based on the frequency of attacks, migraine is classified into two subtypes: episodic (EM) and chronic (CM). Migraine progression from EM to CM is supposed to be affected by various factors, which is known as "migraine chronification. However, the pathophysiology of migraine chronification is multifactorial and still not fully understood. Ion channels are hypothesized to play a role in migraine pathophysiology and are essential for generating and suppressing action potentials, which lie under the pain and related symptoms. Two sodium channel genes, SCN9A and SCN10A, have been report
