Anorectal pruritus after intravenous hydrocortisone sodium succinate and sodium phosphate.

A double-blind study demonstrated that single intravenous doses of 100, 200, or 400 mg of hydrocortisone sodium succinate and hydrocortisone sodium phosphate were similar in eosinophil suppression, elevation of glucose, white blood count differential shifts (polymorphonuclear cells, lymphocytes, and monocytes), and urinary excretion of sodium and potassium but not in incidence of side effects. More subjects receiving hydrocortisone sodium phosphate experienced systemic or localized adverse effects than those receiving hydrocortisone sodium succinate. The most common side effect was burning or

A tissue factor influencing succinic dehydrogenase activity in tuberculous guinea pigs.

The alterations in succinic dehydrogenase activity during the course of infection with Mycobacterium tuberculosis strains H37Rv and BCG, as well as with dead bacilli, has been described. A tissue factor has been described which is prepared from normal bovine kidney and will restore enzyme activity in vitro. This factor is prepared by chromatographic analysis and contains a nucleotide with an ultraviolet absorption maxima at 250 mmicro (pH 3) and a free sulfhydryl group. The role of this factor in the response to tuberculosis is discussed.

Energy Metabolic Reprogramming Characterized by Succinate Accumulation Aggravates Necrotizing Enterocolitis.

Necrotizing enterocolitis (NEC) is a severe inflammatory intestinal disorder of preterm infants associated with substantial morbidity and mortality. This study investigated energy metabolic alterations in NEC and the contribution of succinate to disease progression. Fecal energy metabolites were quantified by high-performance liquid chromatography-mass spectrometry in preterm infants and neonatal mice. Neonatal C57BL/6 mice were assigned to the Control, NEC, NEC+dimethyl malonate (DMM), NEC+succinate (Suc), and NEC+DMM+Suc groups. Ileal tissues were analyzed by metabolomics, transcriptomics, h

Bioconversion of the C1 substrate methanol to succinic acid with engineered Komagataella phaffii.

Succinic acid is a versatile platform chemical produced either by petrochemistry or via microbial fermentation of sugars. In light of increasing anthropogenic CO2 emissions, sustainable alternatives using non-food, carbon-neutral substrates are urgently needed. Here, we report the efficient biosynthesis of succinic acid from methanol-a promising C1 feedstock derivable from CO2-using the native methylotrophic yeast Komagataella phaffii. By integrating dicarboxylic acid transporters from Aspergillus niger and Schizosaccharomyces pombe, alongside a heterologous cytosolic reductive tricarboxylic a

ACOD1-dependent succinate signalling acts as a metabolic checkpoint for osteoclast differentiation.

Immune cells undergo metabolic reprogramming in response to inflammatory stimuli. The immuneresponsive gene 1 (Irg1) encodes aconitate decarboxylase (ACOD1), which generates itaconate from cis-aconitate in the TCA cycle. Itaconate inhibits succinate dehydrogenase, resulting in succinate accumulation. Stable ACOD1 overexpression in RAW264.7 cells shifted cellular metabolism towards glycolysis, as indicated by enhanced mTOR activation, increased 4E-BP1 phosphorylation, and reduced ATP levels. ACOD1 cells displayed impaired osteoclastogenesis with reduced expression of osteoclast-associated genes

Succinate-mediated metabolic alterations and signaling in RA: mechanisms and therapeutic implications.

Rheumatoid Arthritis (RA) is a chronic autoimmune disease characterized by persistent synovial inflammation. Recent studies have revealed that immune metabolites, particularly succinate, play a critical role in its pathogenesis. In this review, we systematically analyze the molecular mechanisms underlying succinate accumulation and elucidate how succinate drives inflammatory and immune dysregulation in RA. Hypoxia and inflammation microenvironment drive metabolic alterations, leading to succinate accumulation via three primary mechanisms: 1) succinate retention caused by impaired mitochondrial

Potential for pathogenic protein acylation in succinate dehydrogenase-deficient pheochromocytoma and paraganglioma.

Historically, the study of cellular metabolism and the cellular proteome have largely focused on specific proteins and metabolites in certain cellular pathways. With the advent of new "omics" technologies, researchers have been able to probe disruptions and alterations in cellular homeostasis on a near proteome-wide and metabolome-wide basis. Yet the truly interconnected and interdependent nature of the cellular metabolome and proteome has only become more evident with the discovery of proteome-wide protein post-translational modifications (PTMs) such as lysine acylation by acetyl-CoA or succi

Suppressing MASH fibrotic progression by blocking succinate-GPR91 signaling in HSCs.

BACKGROUND AND AIMS: The succinate receptor GPR91 is highly expressed in HSCs, with its expression further elevated during metabolic dysfunction-associated steatohepatitis (MASH)-induced fibrotic progression. However, convincing in vivo data on whether blocking GPR91 signaling leads to fibrotic regression in MASH are lacking. APPROACH AND RESULTS: MASH models were induced by choline-deficient amino acid-defined diet feeding along with lipopolysaccharide injection (CDAA-LPS) plus i.p. injection of succinate or by high-fat and high-calorie diet plus high fructose and glucose in drinking water (

Succinate preserves CD8+ T cell fitness to augment antitumor immunity.

Succinate, a tricarboxylic acid cycle intermediate, accumulates in tumors with succinate dehydrogenase (SDH) mutations. Although succinate is recognized for modulating CD8+ T cell cytotoxicity, its impact on T cell differentiation remains poorly understood. Here, we reveal that succinate accumulation in tumors lacking the SDH subunit B (SDHB) enhanced tumor-reactive CD8+ T cell-mediated immune responses. Sustained succinate exposure promoted CD8+ T cell survival and facilitated the generation and maintenance of stem-like subpopulations. Mechanistically, succinate enhanced mitochondrial fitness

Imaging Pathway of a Pediatric Patient with Succinate Dehydrogenase B-Deficient Paraganglioma.

Pheochromocytoma and paraganglioma are rare in children, at only 1 in every 50,000 cases. Even though some cases are sporadic, they have been connected to syndromes such as von Hippel-Lindau, multiple endocrine neoplasia types IIa and IIb, neurofibromatosis type 1, and hereditary pheochromocytoma-paraganglioma syndromes. A genetic mutation causes around 60% of pheochromocytomas and paragangliomas in children under 18. Methods: A 15-y-old child with a 6-y history of back discomfort is presented. The justification for using 2 functional imaging modalities, 68Ga-DOTATATE PET/CT and 123I-meta-iodo

In vitro biocompatibility of polylactide and polybutylene succinate blends for urethral tissue engineering.

Surgical treatment of urothelial defects with autologous genital or extragenital tissue grafts is susceptible to complications. Tissue engineering utilizing novel biomaterials and cells such as human urothelial cells (hUC) for epithelial regeneration and adipose stromal cells (hASC) for smooth muscle restoration might offer new treatment options for urothelial defects. Previously, polylactide (PLA) has been studied for urethral tissue engineering, however, as such, it is too stiff and rigid for the application. Blending it with ductile polybutylene succinate (PBSu) could provide suitable mecha

Intraperitoneal administration of nanoparticles containing tocopheryl succinate prevents peritoneal dissemination.

Intraperitoneal administration of anticancer nanoparticles is a rational strategy for preventing peritoneal dissemination of colon cancer due to the prolonged retention of nanoparticles in the abdominal cavity. However, instability of nanoparticles in body fluids causes inefficient retention, reducing its anticancer effects. We have previously developed anticancer nanoparticles containing tocopheryl succinate, which showed high in vivo stability and multifunctional anticancer effects. In the present study, we have demonstrated that peritoneal dissemination derived from colon cancer was prevent
