Calciproteins regulate cyclic AMP content and melatonin secretion in trout pineal photoreceptors.

Photoreceptor cells of the fish pineal transduce photoperiodic information into the rhythmic secretion of melatonin. The nocturnal rise in melatonin secretion has been associated with an increase in cyclic AMP (cAMP) production and with an entry of Ca2+ ions through L-type voltage-dependent channels. It is shown here that two inhibitors of calciproteins, W7 and calmidazolium, inhibit melatonin secretion and, to a lesser extent, cAMP levels in cultured trout pineal photoreceptors. Kinetic studies indicated that melatonin secretion was affected earlier than cAMP in cells cultured in the presence…

Anti-inflammatory Activities of Extracellular Vesicles-Derived from Rainbow Trout (Oncorhynchus mykiss) Plasma.

Extracellular vesicles (EVs) are nanoscale, lipid bilayer membrane-bound particles carry bioactive cellular cargos such as protein, lipids, nucleic acids. They are considered as key mediators of intercellular communication, potential biomarkers, and play role in numerous pathophysiological processes including modulating innate and adaptive immune responses. In this study, plasma-derived EVs were isolated from rainbow trout (Oncorhynchus mykiss) and determined the physicochemical-molecular properties to investigate their anti-inflammatory activities. Isolated EVs had inherent cup shaped morphol…

Enantioselective toxicokinetics drive the differential toxicity of chiral 6PPD-quinone in rainbow trout.

The tire-derived contaminant N-(1,3-dimethylbutyl)-N'-phenyl-p-phenylenediamine quinone (6PPD-Q) is widespread in aquatic ecosystems and poses significant ecological risk. As a chiral compound, its enantiomers may exhibit differential toxicity, yet the underlying mechanisms remain limited. Here, we found that (+)-6PPD-Q was 2.4-fold more toxic to rainbow trout than (-)-6PPD-Q (p < 0.05). In addition, in vitro metabolism assays using liver S9 fraction showed that (+)-6PPD-Q was metabolized 2.5-fold faster than (-)-6PPD-Q. Semi-quantitative nontarget analysis revealed that hydroxylated metabolit…

Inversion Polymorphisms Carried by Introduced Rainbow Trout Disrupt Hybrid Zones Between Native Species.

Genomic mechanisms explaining why some hybrid zones remain stable while others collapse into hybrid swarms are poorly understood. In Montana's Kootenai River Basin, native interior redband trout (Oncorhynchus mykiss gairdneri; IRT) and westslope cutthroat trout (O. lewisi; WCT) are naturally sympatric and historically exhibited limited hybridization. However, the introduction of allopatric coastal rainbow trout (O. m. ssp.; CRT) has coincided with hybrid swarm formation. Using novel IRT linkage maps and 3489 individuals genotyped at 189,984 SNPs, we characterized the role of genome structure i…

Saprolegnia species isolated from rainbow trout: molecular identification, specific PCR and drug sensitivity.

Saprolegniosis is a common infectious disease caused by members of the genus Saprolegnia. This genus contains several species, some of which can cause severe infection in vulnerable fish such as rainbow trout Oncorhynchus mykiss. This study reports the isolation and identification of 4 Saprolegnia species, namely S. parasitica, S. diclina, S. aenigmatica and S. asterophora, from a rainbow trout farm. The isolates were identified by amplification and sequencing of the internal transcribed spacer region. Among the isolates, S. parasitica and S. diclina were more common. S. diclina was found only…

Genomic and transcriptomic analysis of the Whirling disease-resistant Gunnison River Rainbow Trout.

Whirling disease is a debilitating disease of Rainbow Trout caused by Myxobolus cerebralis. The parasite invasion leads to skeletal deformities, neurological impairment, and high mortality. Since its introduction to North America, M. cerebralis has severely impacted wild trout populations in several regions. In this study, we focus on a promising Whirling disease-resistant Rainbow Trout strain developed in the Gunnison River, Colorado. We analyzed the genomes and transcriptomes of this resistant strain at different time points after challenge with M. cerebralis. Signature selection analysis re…

Transcriptional and epigenetic responses to acute hypoxia exposure in rainbow trout (Oncorhynchus mykiss).

Rainbow trout, which are comparatively intolerant to low oxygen, face increasing hypoxic events. This study investigated their metabolic and molecular responses to acute hypoxia and investigated whether O2-dependent epigenetic marks mediate these responses. Adult rainbow trout were exposed to normoxia (10.5 mg/L dissolved oxygen (DO)), 50 % hypoxia (5.25 mg/L DO) and 25 % oxygen saturation (2.61 mg/L DO) for 4 h. Plasma metabolites and cortisol were measured, as were transcripts of hypoxia-responsive genes involved in oxygen sensing, glucose/lactate metabolism, and lipid metabolism in liver, w…

An inactivated vaccine against infectious pancreatic necrosis virus in rainbow trout (Oncorhynchus mykiss).

Infectious pancreatic necrosis virus (IPNV), belonging to the genus Aquabirnavirus within the family Birnaviridae, causes huge economic loss to the global salmonid industry every year. Recently, outbreaks of disease caused by genogroup I IPNV were found in many rainbow trout (Oncorhynchus mykiss) farms worldwide. An inactivated vaccine was prepared using a genogroup I IPNV isolate with an optimized procedure as incubation with β-propanolactone (BPL) at the final concentration of 0.5% at room temperature for 48 h. The inactivated vaccine was used to immunize rainbow trout, and the protection ef…

Reproductive and thyroid endocrine axis cross-talk in rainbow trout (Oncorhynchus mykiss) alevins.

The goal of this study was to characterize morphological and molecular effects in rainbow trout alevins after waterborne exposures to 17β-estradiol (E2; 0.0008 to 0.5 μg/L), triiodothyronine (T3; 0.52 to 65 μg/L), and various co-treatments for 21 to 23 days. Interestingly, there was no consistent evidence that E2 alone influenced growth, development or deformity rates, however, 65 μg/L T3 alone expedited development, and both 13 μg/L and 65 μg/L alone caused a unique opercular deformity not previously reported. In addition, some potentiation between E2 and T3 at lower concentrations suggests s…

Molecular and physiological responses predict acclimation limits in juvenile brook trout (Salvelinus fontinalis).

Understanding the resilience of ectotherms to high temperatures is essential because of the influence of climate change on aquatic ecosystems. The ability of species to acclimate to high temperatures may determine whether populations can persist in their native ranges. We examined physiological and molecular responses of juvenile brook trout (Salvelinus fontinalis) to six acclimation temperatures (5, 10, 15, 20, 23 and 25°C) that span the thermal distribution of the species to predict acclimation limits. Brook trout exhibited an upregulation of stress-related mRNA transcripts (heat shock prote…

Massive infection of Cystidicoloides ephemeridarum in brown trout Salmo trutta with skeletal deformities.

We investigated the cause of skeletal deformities found in brown trout from the Aspromonte mountain area in Reggio Calabria, Italy. Toxicological, histopathological and parasitological analyses were carried out on 14 fish with evident macro-morphological alterations from 2 different locations in the same river, and 4 control fish without morphological alterations from a different river (far from the first river but still within the area under study). Histopathological and radiological observations confirmed severe skeletal deformities in the specimens investigated. Parasitological examinations…

Aeromonas salmonicida activates rainbow trout IgM+ B cells signalling through Toll like receptors.

As B cells are singularly equipped with a B cell receptor (BCR) and a range of innate receptors, they are able to integrate both antigen-specific and innate signals, with the latter being essential to reach an adequate level of activation. Whether teleost B cells sense pathogens through innate mechanisms has not yet been explored, despite the fact that fish B cells display a wider array of innate receptors than many mammalian B cell subsets. Hence, in the current study, we have investigated the effects of inactivated Aeromonas salmonicida, a Gram negative rainbow trout pathogen, on trout splen…

Nasal Vaccination Drives Modifications of Nasal and Systemic Antibody Repertoires in Rainbow Trout.

Bony fish represent the most basal vertebrate branch with a dedicated mucosal immune system, which comprises immunologically heterogeneous microenvironments armed with innate and adaptive components. In rainbow trout (Oncorhynchus mykiss), a nasopharynx-associated lymphoid tissue (NALT) was recently described as a diffuse network of myeloid and lymphoid cells located in the olfactory organ of fish. Several studies have demonstrated high levels of protection conferred by nasal vaccines against viral and bacterial pathogens; however, the mechanisms underlying the observed protection are not well…

Genome-wide methylation study of diploid and triploid brown trout (Salmo trutta L.).

The induction of triploidization in fish is a very common practice in aquaculture. Although triploidization has been applied successfully in many salmonid species, little is known about the epigenetic mechanisms implicated in the maintenance of the normal functions of the new polyploid genome. By means of methylation-sensitive amplified polymorphism (MSAP) techniques, genome-wide methylation changes associated with triploidization were assessed in DNA samples obtained from diploid and triploid siblings of brown trout (Salmo trutta). Simple comparative body measurements showed that the triploid…

Natural mineral particles are cytotoxic to rainbow trout gill epithelial cells in vitro.

Worldwide increases in fluvial fine sediment are a threat to aquatic animal health. Fluvial fine sediment is always a mixture of particles whose mineralogical composition differs depending on the sediment source and catchment area geology. Nonetheless, whether particle impact in aquatic organisms differs between mineral species remains to be investigated. This study applied an in vitro approach to evaluate cytotoxicity and uptake of four common fluvial mineral particles (quartz, feldspar, mica, and kaolin; concentrations: 10, 50, 250 mg L(-1)) in the rainbow trout epithelial gill cell line RTg…

Modulation of hypothalamic-pituitary-interrenal axis function by social status in rainbow trout.

Juvenile rainbow trout (Oncorhynchus mykiss) form stable dominance hierarchies when confined in pairs. These hierarchies are driven by aggressive competition over limited resources and result in one fish becoming dominant over the other. An important indicator of low social status is sustained elevation of circulating cortisol levels as a result of chronic activation of the hypothalamic-pituitary-interrenal (HPI) axis. In the present study it was hypothesized that social status modulates the expression of key proteins involved in the functioning of the HPI axis. Cortisol treatment and fasting …

Cloning, expression analysis and bioactivity studies of rainbow trout (Oncorhynchus mykiss) interleukin-22.

This report describes the cloning and characterisation of rainbow trout (Oncorhynchus mykiss) interleukin (IL)-22, and presents studies of the functional activity of its recombinant protein for the first time in a non-mammalian species. The predicted IL-22 coding region consists of 522 nucleotides which translates into a 173 amino acid protein, that contains an IL-10 family signature which is reasonably well conserved with other vertebrate IL-22 molecules. Expression analysis in tissues from healthy fish revealed a higher constitutive expression of IL-22 in mucosal tissues, suggesting a potent…
