Estimation of tsetse challenge and its relationship with trypanosomosis incidence in cattle kept under pastoral production systems in Kenya.

In an on-farm trial conducted amongst the Maasai pastoralists in Nkuruman and Nkineji areas of Kenya between April 2004 and August 2005 desi … dy weight. Treatments administered by the owners over the sampling intervals were recorded as well. Tsetse flies were trapped at the time of sampling using baited stationary traps and apparent tsetse density estimated as flies per trap per day (FTD). A fixed proportion (10%) of the flies was dissected and their infection status determined through microscopy. B …

Tsetse fly ecology and risk of transmission of African trypanosomes related to a protected forest area at a military base in the city of Abidjan, Côte d'Ivoire.

African trypanosomoses, whose pathogens are transmitted by tsetse flies, are a threat to animal and human health. Tsetse flies observed at the military base of the French Forces in Côte d'Ivoire (FFCI base) were probably involved in the infection and death of military working dogs. Entomological and parasitological surveys were carried out during the rainy and dry seasons using "Vavoua" traps to identify tsetse fly species, their distribution, favorable biotopes and food sources, as well as the trypanosomes they harbor. A total of 1185 Glossina palpalis palpalis tsetse flies were caught, corre…

Interactions Between Tsetse Endosymbionts and Glossina pallidipes Salivary Gland Hypertrophy Virus in Glossina Hosts.

Tsetse flies are the sole cyclic vector for trypanosomosis, the causative agent for human African trypanosomosis or sleeping sickness and African animal trypanosomosis or nagana. Tsetse population control is the most efficient strategy for animal trypanosomosis control. Among all tsetse control methods, the Sterile Insect Technique (SIT) is one of the most powerful control tactics to suppress or eradicate tsetse flies. However, one of the challenges for the implementation of SIT is the mass production of target species. Tsetse flies have a highly regulated and defined microbial fauna composed …

Enhancing tsetse fly refractoriness to trypanosome infection--a new IAEA coordinated research project.

To date, IAEA-supported Sterile Insect Technique (SIT) projects for tsetse and trypanosomiasis control have been in areas without human sleeping sickness, but future projects could include areas of actual or potential human disease transmission. In this context it would be imperative that released sterile tsetse flies are incompetent to transmit the disease-causing trypanosome parasite. Therefore, development of tsetse fly strains refractory to trypanosome infection is highly desirable as a simple and effective method of ensuring vector incompetence of the released flies. This new IAEA Coordin…

A review of recent knowledge of the ecology of the main vectors of trypanosomiasis.

In this survey of recent ecological research on the main vectors of trypanosomiasis in those countries of East, Central and West Africa that are not predominantly French-speaking, the authors, after outlining the distribution of tsetse flies and the type of country in which they occur, discuss the direct and indirect effects of climate on these insects-particularly on their physiological water balance and on pupal fat reserves-and their recent advances into new areas. They review the considerable work that has been done on the resting habits and breeding-sites of different Glossina species, kn…

Population genetics of Trypanosoma brucei circulating in Glossina palpalis palpalis and domestic animals of the Fontem sleeping sickness focus of Cameroon.

BACKGROUND: Human African Trypanosomiasis is still a public health threat in Cameroon. To assess Trypanosoma brucei strains circulating in the Fontem sleeping sickness focus, we conducted a genetic structure study using microsatellites to assess genotypes circulating in both tsetse flies and domestic animals. METHOD: For this study, pyramidal traps were set up and 2695 tsetse flies were collected and 1535 (57%) living flies were dissected and their mid-guts collected. Furthermore, blood samples were collected from 397 domestic animals (pigs, goats, sheep and dogs). DNA was extracted from midg…

Risk of transmission of sleeping sickness in the four historical epicentre villages of the Bipindi focus, Cameroon.

BACKGROUND: Bipindi is a historic focus of human African trypanosomiasis (HAT). With the number of cases declining to near-elimination levels, understanding the current entomological risk is crucial to guiding elimination strategies. METHODS: Two entomological surveys in four historical sentinel villages used pyramidal traps to capture tsetse flies. Dissected midguts were analyzed by polymerase chain reaction (PCR) for trypanosome infection. Blood meal origins were identified using cytochrome B gene heteroduplex analysis. RESULTS: Glossina palpalis palpalis was accounted for 99.0% of 708 cap…

Tsetse immune responses and trypanosome transmission: implications for the development of tsetse-based strategies to reduce trypanosomiasis.

Tsetse flies are the medically and agriculturally important vectors of African trypanosomes. Information on the molecular and biochemical nature of the tsetse/trypanosome interaction is lacking. Here we describe three antimicrobial peptide genes, attacin, defensin, and diptericin, from tsetse fat body tissue obtained by subtractive cloning after immune stimulation with Escherichia coli and trypanosomes. Differential regulation of these genes shows the tsetse immune system can discriminate not only between molecular signals specific for bacteria and trypanosome infections but also between diffe…

Maintaining zebu Maure cattle in a tsetse infested area of Mali. II. Epidemiological considerations.

Scheduled monitoring of tsetse and other biting flies at Tienfala Forest, Republic of Mali from December 1979 until June 1981 yielded more than 14,500 specimens. Comparisons of the monthly totals of tsetse and other biting flies with the monthly incidence of bovine trypanosomiasis in sentinel cattle suggested that biological transmission by a single riverine species of tsetse, Glossina palpalis gambiensis, accounted for most if not all disease transmission at the study site. The data obtained also suggested that human decisions had contributed to the magnitude if not the degree of the disease

Acute Trypanosoma vivax infection of Ethiopian cattle in the apparent absence of tsetse.

Investigations of acute trypanosomiasis in a group of 1,200 cattle provided strong evidence for mechanical transmission by haematophagous flies other than tsetse. Although recently introduced cattle may have had previous contact with tsetse flies the incident occurred in an area remote from their known range. Mortality exceeded 6% before the outbreak was controlled by chemotherapy. Mechanically transmitted Trypanosoma vivax infection may be of widespread occurrence in eastern Ethiopia.

Surface sialic acids taken from the host allow trypanosome survival in tsetse fly vectors.

The African trypanosome Trypanosoma brucei, which causes sleeping sickness in humans and Nagana disease in livestock, is spread via blood-sucking Tsetse flies. In the fly's intestine, the trypanosomes survive digestive and trypanocidal environments, proliferate, and translocate into the salivary gland, where they become infectious to the next mammalian host. Here, we show that for successful survival in Tsetse flies, the trypanosomes use trans-sialidase to transfer sialic acids that they cannot synthesize from host's glycoconjugates to the glycosylphosphatidylinositols (GPIs), which are abunda…

Evaluation of the relative roles of the Tabanidae and Glossinidae in the transmission of trypanosomosis in drug resistance hotspots in Mozambique.

BACKGROUND: Tsetse flies (Diptera: Glossinidae) and tabanids (Diptera: Tabanidae) are haematophagous insects of medical and veterinary importance due to their respective role in the biological and mechanical transmission of trypanosomes. Few studies on the distribution and relative abundance of both families have been conducted in Mozambique since the country's independence. Despite Nicoadala, Mozambique, being a multiple trypanocidal drug resistance hotspot no information regarding the distribution, seasonality or infection rates of fly-vectors are available. This is, however, crucial to unde…

Glossina fuscipes fuscipes in the trypanosomiasis endemic areas of south eastern Uganda: apparent density, trypanosome infection rates and host feeding preferences.

A study was undertaken in three districts in south eastern Uganda endemic for human and animal trypanosomiasis, to investigate the status of the vector tsetse fly population. Apparent density (AD) of tsetse was between 2 and 21 flies/trap/day across the three districts, with Glossinia fuscipes fuscipes identified as the predominant species. Trypanosomes were observed in G.f. fuscipes with an infection rate, as determined by microscopy, of 1.55% across the three studied areas. However, trypanosome infections were only identified in female flies giving an infection rate of 2.39% for the female t

Friends or foes: Unraveling the tsetse fly-Spiroplasma symbiosis.

Tsetse flies (Glossina spp.) transmit African trypanosomes, the causative agents of human African and African animal trypanosomiases (HAT and AAT, respectively). These neglected tropical diseases impose significant public health and economic burdens across sub-Saharan Africa. Trypanosome transmission by tsetse flies is influenced by multiple factors, including host genetic background, ecological factors, and interactions with heritable microbial endosymbionts. Spiroplasma glossinidia has recently emerged as an important modulator of tsetse reproductive fitness and vector competence, making it …

Species distribution modeling to predict tsetse fly (Glossina spp.) habitat suitability in Kenya.

BACKGROUND: African animal trypanosomosis (AAT) and human African trypanosomosis (HAT) are transmitted and spread primarily by tsetse flies (Glossina spp.) in sub-Saharan Africa. The animal disease poses significant challenges to agropastoral systems, including in Kenya, where 38 out of 47 counties are infested with eight species of Glossina. Climate change and human activities can also aggravate these infestations, putting rural-scale farmers who rely on agropastoral systems at a greater risk. Geographical gaps in existing entomological datasets limit a comprehensive understanding of tsetse f…

[Updating the northern tsetse distribution limit in Chad in the context of global change].

BACKGROUND - RATIONALE: Tsetse flies (Diptera: Glossinidae) are obligate bloodfeeders that occur exclusively in Sub-Saharan Africa, where they are the vectors of trypanosomes causing HAT (human African trypanosomiasis) and AAT (African animal trypanosomiasis). In Chad, tsetse flies occur only in the most southern part of the country because of its favorable bioclimatic conditions. However, despite the importance of HAT and AAT in this country, very little is known about the current tsetse distribution, in particular its northern limit, which is of key importance for the surveillance of these d…
