Targeting the triad: phenotypic plasticity, tumor microenvironment and bone microenvironment in prostate cancer bone-metastatic events.

The incidence and mortality rates of prostate cancer are increasing annually, indicating that it poses a serious threat to men's health. Previous studies have demonstrated that bone-metastatic tumor cells undergo four hallmark bone-metastatic events, including colonization, dormancy, reactivation and bone reconstruction. However, most advanced patients experience four stages of progression and undergo advanced bone reconstruction-the "vicious circle"-which is irreversible. These patients have a poor prognosis and can even die. Therefore, determining how various components in the tumor and bone

Nanozyme-Driven Tumor Microenvironment Remodeling for Potentiated Phototherapy-Immunotherapy.

Tumor hypoxia critically drives the development of an immunosuppressive tumor microenvironment (TME) and accelerates tumor progression. Besides, hypoxia seriously restricts O2-dependent reactive oxygen species (ROS) generation, thereby severely compromising the therapeutic efficacy of photodynamic therapy (PDT). To address these challenges, we developed a multifunctional manganese (Mn)-based nanozyme (MLDHI) loaded with indocyanine green (ICG) to reprogram the immunosuppressive TME for potentiated phototherapy combined with immunotherapy. Upon intratumoral injection, the Mn4+ doped within the …

Heterogeneous tumor microenvironment - A hallmark of ameloblastoma invasive phenotype.

Ameloblastoma is a benign odontogenic tumor with an aggressive growth phenotype orchestrated by a complex and heterogeneous tumor microenvironment. This review addresses how tumor cells, cancer-associated fibroblasts, mesenchymal stem cells, endothelial cells, and immune cells interact with non-cellular elements especially the extracellular matrix and hypoxic niches to drive invasive growth and recurrence. Several genetic changes associated with ameloblastoma activate mitogen-activated protein kinase (MAPK), Hedgehog (HH), Wnt/β-catenin, and less commonly PI3K/AKT signaling pathways. These pat…

Tumor microenvironment and signaling pathways in melanoma brain metastasis.

Melanoma has one of the highest propensities to metastasize to the brain, and despite major advances with immune checkpoint inhibitors, brain metastases remain a leading cause of morbidity and mortality. The biological mechanisms governing brain colonization, immune evasion, and resistance to therapy are incompletely understood. This review summarizes recent advances in the understanding of the tumor microenvironment and signaling pathways involved in melanoma brain metastasis, with emphasis on mechanisms that may provide novel therapeutic opportunities. We performed a review of preclinical an…

Tumor microenvironment of head and neck squamous cell carcinomas.

The character of the tumor microenvironment is a relevant prognostic and predictive biomarker across a wide range of malignancies. The composition, density, and functional capacity of tumor-infiltrating immune cells are especially crucial for selecting suitable immunotherapy. Head and neck squamous cell carcinomas are considered immunologically hot tumors, with high numbers of tumor-infiltrating effector and regulatory T cells. Higher T cell counts, along with a better prognosis, were observed in patients with head and neck squamous cell carcinomas associated with human papillomavirus infectio…

Acidic tumor microenvironment-modulated nanoparticle potentiates gastric cancer photoimmunotherapy.

INTRODUCTION: Immunotherapy, especially anti-PD-1 antibodies (α-PD-1), has revolutionized the landscape of cancer treatment. However, the response rate of α-PD-1 for Gastric Cancer (GC) remains relatively low. The acidic immunosuppressive Tumor Microenvironment (TME) greatly hinders the efficacy of α-PD-1. Thus, therapeutic strategies targeting the acidic TME in GC are highly desired. OBJECTIVES: This study aimed to investigate the effect of FX-11@PEG-Ce6 on the therapeutic efficacy of photoimmunotherapy for gastric cancer. METHODS: We developed FX-11 encapsulated with PEG-Ce6 nanoparticles …

Tumor microenvironment-based classification for predicting gastric cancer prognosis.

BACKGROUND: The tumor microenvironment (TME), consisting of tumor-associated stroma and tumor-infiltrating lymphocytes (TIL), is crucial for prognostic information in gastric cancer (GC). Despite its potential, routine clinical adoption remains limited. METHODS: In a study of 320 GC patients, virtual staining and image processing were applied to hematoxylin & eosin-stained slides. This method quantified the tumor-stroma ratio (TSR) and TIL, leading to a TME-based prediction model (TMEPATH) using a scoring system derived from univariate Cox regression. Subgroups were categorized to predict GC …

Targeted Degradation of sGRP78 Alleviates the Immunosuppressive Tumor Microenvironment.

There is an urgent need for appropriate methods to identify patients who are sensitive to neoadjuvant therapy (NAT) for precise stratified treatment. Based on clinical proteomics screening and further validation in patient cohorts, we reported that soluble glucose-regulatory protein 78 (sGRP78) could be a serological biomarker to infer the poor chemotherapy response in breast cancer. Consequently, an "sGRP78 index" was established to accurately predict patient responses to NAT. When exploring the linked mechanisms on how sGRP78 confer the resistance, it was found that in high sGRP78 index tumo

Mesothelial and immune cells interplay in the tumor microenvironment.

Mesothelial cells (MCs) constitute a dynamic mesothelium in which their numerous crucial functions synergistically interact with other cells to maintain serosal integrity and homeostasis. Previous studies have confirmed the crucial role of interactions between MCs and tumor cells in tumorigenesis and progression in the tumor microenvironment (TME). However, recent research has found that MCs can induce an immunosuppressive microenvironment by secreting various cytokines, chemokines, and exosomes which recruit immunosuppressive cells or interact with immune cells, thus contributing to tumor pro…

Oncogenic drivers shape the tumor microenvironment in human gliomas.

Gliomas are aggressive and heterogeneous brain tumors with limited treatment options. While oncogenic mutations in gliomas have been well-characterized, their impact on the tumor microenvironment remains poorly understood. To investigate how genomic alterations may influence the glioma microenvironment, we performed an integrative multiomic and spatial transcriptomic analysis of 93 glioma samples (46 IDH-mutant gliomas and 47 IDH-wildtype glioblastoma) from 69 patients representing both primary and recurrent stages. Using whole-genome sequencing, chromatin conformation capture (Hi-C), RNA-seq,…

Tumor Microenvironment-responsive Nanocatalyst for Targeted Chemodynamic Cancer Therapy.

To address the challenges of insufficient hydrogen peroxide (H2O2) levels, rapid Fe3+ precipitation, and a slow Fenton reaction cycle, tumor-activated, self-accelerating CDT nanocatalysts are synthesized, comprising poly (lactic-co-glycolic acid) (PLGA)-encapsulated Ca-Fe peroxide clusters and polyarginine (R). Nanocatalysts are camouflaged with cancer cell membranes (CCM) to enhance tumor targeting. Additionally, polyarginine tailored the PLGA responsiveness to low H2O2 levels (50-100 µm). H2O2 triggered the degradation of PLGA, releasing CaFe clusters to produce Fe3+/Fe2+ and additional H2O2

Tumor Microenvironment-Responsive Polymer Delivery Platforms for Cancer Therapy.

Most chemotherapeutic and bioimaging agents struggle with inadequate bioavailability, primarily due to their limited biocompatibility and lack of specificity in targeting, leading to low or decreased anticancer efficacy and inaccurate imaging. To surmount these obstacles, the development of stimuli-responsive polymer delivery platforms, predominantly leveraging the tumor microenvironment (TME), has emerged as a promising strategy. Therapeutic and diagnostic agents can be released controllably at the tumor site by virtue of the bond cleavage or hydrophobic to hydrophilic transformation of TME-s…

What's new about the tumor microenvironment of urothelial carcinoma?

Urothelial carcinoma is a significant global health concern that accounts for a substantial part of cancer diagnoses and deaths worldwide. The tumor microenvironment is a complex ecosystem composed of stromal cells, soluble factors, and altered extracellular matrix, that mutually interact in a highly immunomodulated environment, with a prominent role in tumor development, progression, and treatment resistance. This article reviews the current state of knowledge of the different cell populations that compose the tumor microenvironment of urothelial carcinoma, its main functions, and distinct in…

Castration promotes the browning of the prostate tumor microenvironment.

BACKGROUND: Adipose tissue has gained attention due to its potential paracrine role. Periprostatic adipose tissue surrounds the prostate and the prostatic urethra, and it is an essential player in prostate cancer progression. Since obesity is directly related to human tumor progression, and adipose tissue depots are one of the significant components of the tumor microenvironment, the molecular mediators of the communication between adipocytes and epithelial cells are in the spotlight. Although periprostatic white adipose tissue contributes to prostate cancer progression, brown adipose tissue (…

Mass Spectrometry-Based Tissue Imaging of the Tumor Microenvironment.

Multiplex ion beam imaging (MIBI) and imaging mass cytometry (IMC) enable highly multiplexed antibody (40+) staining of frozen or formalin fixed, paraffin-embedded (FFPE) human or murine tissues through detection of metal ions liberated from primary antibodies by time-of-flight mass spectrometry (TOF). These methods make detection of more than 50 targets theoretically possible while maintaining spatial orientation. As such, they are ideal tools to identify the multiple immune, epithelial, and stromal cell subsets in the tumor microenvironment and to characterize spatial relationships and tumor…

Reprogramming of sentinel lymph node microenvironment during tumor metastasis.

Metastasis is a major cause of death in patients with cancer. The two main routes for cancer cell dissemination are the blood and lymphatic systems. The underlying mechanism of hematogenous metastasis has been well characterized in the past few decades. However, our understanding of the molecular basis of lymphatic metastasis remains at a premature stage. Conceptually, cancer cells invade into lymphatic capillary, passively move to collecting lymphatic vessels, migrate into sentinel lymph node (SLN;, the first lymph node to which cancer cells spread from the primary tumor), and enter the blood
