Experience-dependent regulation of tissue-type plasminogen activator in the mouse barrel cortex.

It has been suggested that tissue-type plasminogen activator (tPA), a serine protease, plays a key role in regulating the extracellular matrix core proteins, thereby impacting the structural plasticity in the cerebral cortex. Much is known about its role in regulating plasticity in the visual cortex. However, its permissive role has not been demonstrated to generalize to other cerebral cortical areas. By utilizing a combination of immunofluorescent histochemistry and confocal microscopy, we demonstrate that endogenous tPA is indeed present in the somatosensory cortex, and its expression is exp

Rapid changes in thalamic firing synchrony during repetitive whisker stimulation.

Thalamic firing synchrony is thought to ensure selective transmission of relevant sensory information to the recipient cortical neurons by rendering them more responsive to temporally correlated input spikes. However, direct evidence for a synchrony code in the thalamus is limited. Here, we directly measure thalamic firing synchrony and its stimulus-induced modulation over time, using simultaneous single unit recordings from individual thalamic barreloids in the rat somatosensory whisker/barrel system. Employing whisker deflections varying in velocity or frequency and a cross-correlation appro

Divergent movement of adjacent whiskers.

The current view of whisker movement is that approximately 25 whiskers on each side of the face move in synchrony. To determine whether whiskers are constrained to move together, we trained rats to use two whiskers on the same side of the face in simple behavioral tasks and videotaped the whiskers during the task. Here we report that the movement of adjacent whiskers is usually synchronous but can diverge: 1) the distance between whiskers can vary dramatically during movement; 2) one whisker can move while the second one remains stationary; 3) two whiskers can simultaneously move in opposite d

The pial vasculature of the mouse develops according to a sensory-independent program.

The cerebral vasculature is organized to supply the brain's metabolic needs. Sensory deprivation during the early postnatal period causes altered neural activity and lower metabolic demand. Neural activity is instructional for some aspects of vascular development, and deprivation causes changes in capillary density in the deprived brain region. However, it is not known if the pial arteriole network, which contains many leptomeningeal anastomoses (LMAs) that endow the network with redundancy against occlusions, is also affected by sensory deprivation. We quantified the effects of early-life sen

Developmental and tissue-specific sulfonylurea receptor gene expression.

We have studied the developmental regulation of mouse sulfonylurea receptor (SUR) gene expression throughout several embryonic stages as well as in the adult mouse. To this end we used a 229-bp mouse complementary DNA corresponding to the 3'-end of the SUR gene for in situ hybridization and solution hybridization/ribonuclease protection assays. We found that the SUR gene was expressed as early as embryonic day 12 in the developing pancreas, heart, and central nervous system. These tissues maintained significant levels of SUR messenger RNA (mRNA) throughout development. In addition, SUR mRNA wa

Accelerated recovery following polyamines and aminoguanidine treatment after facial nerve injury in rats.

Accelerated axon regeneration is of paramount importance for improved functional recovery after motor nerve injuries. Following injury of their axon neurones undergo a series of changes, termed the axon reaction, aimed at survival and regeneration of a new axon. We and others have found that early treatment with exogenous polyamines can enhance neuronal survival and accelerate the rate of axon regeneration and functional recovery after sympathetic and motor (sciatic) nerve injuries. Results of the present study corroborate the previous findings and demonstrate that after facial nerve injury in

Morphofunctional regeneration by mesenchymal stem cell and IGF-1 inoculation in a model of facial nerve crush injury in rats.

OBJECTIVE: To analyze the morphofunctional regeneration process of facial nerve injury in the presence of insulin-like growth factor-1 and mesenchymal stem cells. METHODS: Fourteen Wistar rats suffered unilateral facial nerve crushing and were randomly divided into two groups. All received insulin-like growth factor-1 inoculation, but only half of the animals received an additional inoculation of mesenchymal stem cells. The animals were followed for 90 days and facial nerve regeneration was analyzed via spontaneous facial motor function tests and immunohistochemistry in the nerve motor nucleu

The neurobiology and behavior of the American water shrew (Sorex palustris).

American water shrews (Sorex palustris) are aggressive predators that dive into streams and ponds to find prey at night. They do not use eyesight for capturing fish or for discriminating shapes. Instead they make use of vibrissae to detect and attack water movements generated by active prey and to detect the form of stationary prey. Tactile investigations are supplemented with underwater sniffing. This remarkable behavior consists of exhalation of air bubbles that spread onto objects and are then re-inhaled. Recordings for ultrasound both above and below water provide no evidence for echolocat

Nest box-deployed bait for delivering oral vaccines to white-footed mice.

Although a wide range of interventions are available for use in reducing the public health burden of Lyme disease, additional tools are needed. Vaccinating mouse reservoirs may reduce the prevalence of spirochetal infection due to the powerful vector and reservoir competence-modulating effects of anti-outer surface protein A (OspA) antibody. A delivery system for an oral immunogen would be required for field trials of any candidate vaccine. Accordingly, we tested candidate bait preparations that were designed to be environmentally stable, attractive to mice, and non-nutritive. In addition, we

The human umbilical cord-mesenchymal stem cell secretome regulates hair growth and cycle transition by promoting methylthioadenosine synthesis via the PI3K/AKT/mTOR pathway.

BACKGROUND: The human umbilical cord (hUC)-mesenchymal stem cell (MSC) secretome (SCT) is a cell-free therapy that may emerge as a novel therapeutic strategy for hair loss prevention. Here, we aimed to elucidate the underlying mechanisms through which SCT regulates hair growth and cycle transition. METHODS: Using C57BL/6 mice, ex vivo follicles, and cell experiments, we studied the effects and mechanisms of SCT on hair growth and cycling using untargeted metabolomics and phosphoproteomics. A three-month double-blind clinical study was conducted to validate the effects of SCT on human hair. R

An Experimental Rat Model for Simultaneous Induction of Peripheral Neuropathy and Myelotoxicity by Docetaxel Administration: Evaluating the Protective Role of Dimethyl Fumarate.

Docetaxel is extensively used for treating different types of cancer; however, its clinical efficacy is primarily limited by myelotoxicity and peripheral neuropathy, adverse effects that often lead to treatment discontinuation. This study aimed to establish a preclinical model in Wistar rats for the simultaneous induction of myelotoxicity and peripheral neuropathy associated with docetaxel administration, enabling the evaluation of potential chemopreventive agents. Four distinct docetaxel administration schemes were assessed by performing behavioral nociceptive tests and complete blood cell co

Functional ultrasound and brain connectivity reveal central nervous system compromise in Trembler-J mice model of Charcot-Marie-Tooth disease.

The Charcot-Marie-Tooth-1E (CMT1E) disease is typically described as a peripheral neuropathy in humans, causing decreased nerve conduction, spastic paralysis, and tremor. The Trembler-J (TrJ) mice serve as a high fidelity model of this disease. Here, we use functional ultrasound (fUS) and functional connectivity (FC) to analyze TrJ mice's brain activity during sensory stimulation and resting state experiments against wild type (WT) mice - the healthy counterpart. fUS is an imaging technique that measures cerebral blood volume (CBV) temporal changes. We study these changes in the primary somato

Bone Marrow-Derived ABCC6 Is an Essential Regulator of Ectopic Calcification In Pseudoxanthoma Elasticum.

Physiological calcification of soft tissues is a common occurrence in aging and various acquired and inherited disorders. ABCC6 sequence variations cause the calcification phenotype of pseudoxanthoma elasticum (PXE) as well as some cases of generalized arterial calcification of infancy, which is otherwise caused by defective ENPP1. ABCC6 is primarily expressed in the liver, which has given the impression that the liver is central to the pathophysiology of PXE/generalized arterial calcification of infancy. The emergence of inflammation as a contributor to the calcification in PXE suggested that

Blocking the glial function suppresses subcutaneous formalin-induced nociceptive behavior in the rat.

This study examined whether glial cells in the trigeminal nucleus caudalis (Sp5C) were necessary for orofacial nociception and nociceptive processing induced by subcutaneously (s.c.) injection of 5% formalin into left mystacial vibrissae. The immunohistochemical, immunoelectron microscopical methods and behavior assessment were used in this study. Two hours after administration of carbenoxolone (CBX, a gap junction blocker) or fluorocistrate (FCA, a glail metabolic inhibitor) into the cerebellomedullary cistern, the nociceptive behavior and scratching-cumulative time reduced significantly (P<0

A novel mouse type I intermediate filament gene, keratin 17n (K17n), exhibits preferred expression in nail tissue.

Inactivating the type I keratin 17 gene (mK17) causes severe but reversible hair loss in a strain-dependent fashion in mouse (McGowan et al, Genes Dev. 16:1412, 2002). Missense mutations in human K17 give rise to two dominantly inherited disorders apparented to ectodermal dysplasias, pachyonychia congenita (PC), and steatocystoma multiplex (SM). In contrast to the null phenotype in mouse, marked lesions are seen in the nail and nail bed and sebaceous glands of PC and SM patients, respectively. In an effort to understand the lack of nail involvement in mK17 null mice, we discovered that the gen

Temperature responses and other effects of 5-hydroxytryptophan and 5-hydroxytryptamine when acting from the liquor space in unanaesthetized rabbits.

1. In unanaesthetized rabbits 5-hydroxytryptophan (5-HTP) and 5-hydroxytryptamine (5-HT) were injected into the cisterna magna or into the cannulated left lateral cerebral ventricle while rectal temperature was recorded.2. 5-HTP injected intracisternally in a dose of 1.5-3 mg produced a fall in temperature often followed by a rise beyond the pre-injection level. With 6 mg the main effect was a rise in temperature. The intraventricular injection of 1-2 mg 5-HTP usually produced a fall followed by a rise.3. 5-HT injected intracisternally in a dose of 0.2 mg produced a fall in temperature similar

Whisker-based spatial cognition in mice.

Object localization has been well characterized in the visual and auditory systems, yet how tactile inputs are transformed into a percept of object location in external space remains unclear. Here, we developed a whisker-based categorization task in head-restrained mice that requires judgments of object position in the horizontal plane. Mice categorized object positions with minimal bias relative to the task category boundary and achieved submillimeter spatial acuity by sampling the object with multiple whiskers, a process that depended on the whisker-related primary somatosensory cortex. Bilateral transport of neuronal tracer after unilateral administration: Investigation of potential peripheral mechanism.

Previous studies have shown pain attenuation after the administration of botulinum neurotoxin type A into the whisker pad contralateral to the nerve injury. It further showed localization in the neurons of the bilateral trigeminal ganglia, indicating a possible connection between the trigeminal nerves. Thus, this study aimed to investigate the potential link between the right and left trigeminal ganglia, considering their anatomical proximity and the possible neural interactions between these structures. The fluorogold (FG) neuronal tracer was localized in the neurons of both the right and lef

Trigeminal Sensory Supply Is Essential for Motor Recovery after Facial Nerve Injury.

Recovery of mimic function after facial nerve transection is poor. The successful regrowth of regenerating motor nerve fibers to reinnervate their targets is compromised by (i) poor axonal navigation and excessive collateral branching, (ii) abnormal exchange of nerve impulses between adjacent regrowing axons, namely axonal crosstalk, and (iii) insufficient synaptic input to the axotomized facial motoneurons. As a result, axotomized motoneurons become hyperexcitable but unable to discharge. We review our findings, which have addressed the poor return of mimic function after facial nerve injurie

Development and sensory experience dependent regulation of microglia in barrel cortex.

The barrel cortex is within the primary somatosensory cortex of the rodent, and processes signals from the vibrissae. Much focus has been devoted to the function of neurons, more recently, the role of glial cells in the processing of sensory input has gained increasing interest. Microglia are the principal immune cells of the nervous system that survey and regulate the cellular constituents of the dynamic nervous system. We investigated the normal and disrupted development of microglia in barrel cortex by chronically depriving sensory signals via whisker trimming for the animals' first postnat
