Impact of DNA-stored mutations on virological response in virologically suppressed PWH in the switch to modern regimens.

OBJECTIVE: The clinical relevance of genotypic resistance testing (GRT) performed on viral DNA is still debated. Here, we investigate the role of archived resistance on the virological rebound (VR) after switching treatment. METHODS: This retrospective study enrolled virologically suppressed people with HIV (PWH) included in the ARCA database with one proviral GRT performed in the 6 months prior to the switch to a modern regimen. Baseline genotypic susceptibility scores (GSS) from HIV-1 DNA GRT (DNA-GSS) and, for a subgroup, from historical plasma GRT (hRNA-GSS) were calculated based on the s

Cultures Through Time: Forging a Xeno-Free Future for Cell Culture-Based Virology.

As a cornerstone of modern science, cell lines are the foundational platforms for key medical advances. They enable vaccinology (through vir … he far-reaching influence of cell culture systems as the expression powerhouse that sustains modern virology, whilst focusing on recent cell-engineering methods and optimization strategies in culture media that have facilitated this shift.

Beyond efficiency: automation as an embedded decision architecture in clinical microbiology and virology.

Automation has become a cornerstone of modern clinical microbiology and virology laboratories, extending beyond operational efficiency. Diagnostic workflow design influences the precision, reproducibility, and reliability of laboratory results. This review examines laboratory automation as an integrated workflow framework that standardizes diagnostic processes and supports clinical decision-making through improved generation and application of diagnostic information. Drawing on classical microbiology, high-throughput molecular diagnostics, and pandemic-scale testing, it highlights how automati…

History, Virology, Pathogenesis, Epidemiology, and Laboratory Diagnosis of Dengue Virus: A Focused Review.

Dengue virus (DENV) is a mosquito-borne flavivirus with four antigenically distinct serotypes and represents a leading cause of acute febrile illness in tropical and subtropical regions worldwide. This focused review summarizes the historical evolution of dengue disease descriptions, virus discovery, vectors and transmission dynamics, viral structure, mechanisms of pathogenesis-including antibody-dependent enhancement (ADE) and non-structural protein-1 (NS1)-mediated endothelial dysfunction-clinical classification, current epidemiology with emphasis on India, and laboratory diagnostic approach

Lassa fever: A comprehensive review of virology, clinical management, and global health implications.

Lassa fever (LF) is a serious acute viral hemorrhagic illness that is endemic to West Africa where it affects an estimated two million people and results in up to 10000 deaths each year. The disease is caused by the Lassa virus (LASV), part of the Arenaviridae family, and is primarily transmitted through contact with urine or feces of infected Mastomys natalensis rodents. Human-to-human transmission, particularly in healthcare and community settings, further amplifies the risk of spread. Since its discovery in 1969, LF continues to be a neglected tropical disease with significant health impact

Spaceflight Virology: What Do We Know about Viral Threats in the Spaceflight Environment?

Viruses constitute a significant part of the human microbiome, so wherever humans go, viruses are brought with them, even on space missions. In this mini review, we focus on the International Space Station (ISS) as the only current human habitat in space that has a diverse range of viral genera that infect microorganisms from bacteria to eukaryotes. Thus, we have reviewed the literature on the physical conditions of space habitats that have an impact on both virus transmissibility and interaction with their host, which include UV radiation, ionizing radiation, humidity, and microgravity. Also,

Enterovirus D-68 Molecular Virology, Epidemiology, and Treatment: an Update and Way Forward.

Enterovirus D68 (EV-D68) is a single-stranded positive-sense RNA virus, and it is one of the family members of Picornaviridae. Except for EV-D68, the entire family Picornaviridae has been illustrated in literature. EV-D68 was first discovered and isolated in California, USA, in 1962. EV-D68 has resulted in respiratory disorders' outbreaks among children worldwide, and it has been detected in cases of various neurological diseases such as acute flaccid myelitis (AFM). A recent study documented a higher number of EV-D68 cases associated with AFM in Europe in 2016 compared to the 2014 outbreak. E

Occult hepatitis B infection and hepatocellular carcinoma: Epidemiology, virology, hepatocarcinogenesis and clinical significance.

Occult hepatitis B infection (OBI) refers to a condition where replication-competent HBV DNA is present in the liver, with or without HBV DNA in the blood, in individuals with serum HBsAg negativity assessed by currently available assays. The episomal covalently closed circular DNA (cccDNA) in OBI is in a low replicative state. Viral gene expression is mediated by epigenetic control of HBV transcription, including the HBV CpG island methylation pathway and post-translational modification of cccDNA-bound histone, with a different pattern from patients with chronic HBV infection. The prevalence

Biology and virology of the human malignant lymphomas: 1st Milford D. Schulz Lecture.

Permanent cell lines have been established from twelve diffuse histiocytic lymphomas (SU-DHL-1 to -12), three American Burkitt's lymphomas (SU-AmB-1 to -3), two acute lymphoblastic leukemias (SU-ALL-1 and -2), and three diffuse undifferentiated lymphomas (SU-DUL-1, -2, and -3). The cultured cells displayed neoplastic characteristics, as manifested by heterotransplantability in congenitally athymic nude mice and by the presence of cytogenetic abnormalities in early passage generations. Functional and marker studies revealed that the three American Burkitt's lymphomas, as well as several of the

Predicting Virological Response to HIV Treatment Over Time: A Tool for Settings With Different Definitions of Virological Response.

OBJECTIVE: Definitions of virological response vary from <50 up to 1000 copies of HIV-RNA/mL. Our previous models estimate the probability of HIV drug combinations reducing the viral load to <50 copies/mL, with no indication of whether higher thresholds of response may be achieved. Here, we describe the development of models that predict absolute viral load over time. METHODS: Two sets of random forest models were developed using 50,270 treatment change episodes from more than 20 countries. The models estimated viral load at different time points following the introduction of a new regimen fr

Hantavirus Emergence in a Changing World: Virology, Pathogenesis, Surveillance, and One Health Preparedness.

Hantaviruses are emerging rodent-borne pathogens that pose increasing global public health concerns due to their association with hemorrhagic fever with renal syndrome (HFRS) and hantavirus cardiopulmonary syndrome (HCPS), both of which can result in substantial morbidity and mortality. Environmental change, climate variability, urbanization, and land-use transformation are increasingly recognized as critical drivers of hantavirus emergence and transmission. This review summarizes current evidence regarding hantavirus virology, epidemiology, pathogenesis, clinical manifestations, diagnostics, …

GSK3640254 Is a Novel HIV-1 Maturation Inhibitor with an Optimized Virology Profile.

HIV-1 maturation inhibitors (MIs) offer a novel mechanism of action and potential for use in HIV-1 treatment. Prior MIs displayed clinical efficacy but were associated with the emergence of resistance and some gastrointestinal tolerability events. Treatment with the potentially safer next-generation MI GSK3640254 (GSK'254) resulted in up to a 2-log10 viral load reduction in a phase IIa proof-of-concept study. In vitro experiments have defined the antiviral and resistance profiles for GSK'254. The compound displayed strong antiviral activity against a library of subtype B and C chimeric viruses
